Effects of Green Tea Catechin on the Superoxide Dismutase, Glutathione Peroxidase and Xanthine Oxidase Activites of Kidney in Diabetic Rats (녹차 Catechin이 당뇨쥐 신장조직의 Superoxide Dismutase, Glutathione Peroxidase 및 Xanthine Oxidase)

  • The purpose of this study was to investigate the effect of green tea catechins on the antioxidative defense enzyme activity of kidney in diabetic rats. Sprague-Dawley male rats weighting 100$\pm$10g were randomly assigned to one normal and three STZ-induced diabetic groups; catechin free diet(DM-0C group), 0.25% catechin diet(DM-0.25C group) and 0.5% catechin diet(DM-0.5C group). Diabetes was induced by intravenous of 55mg/Kg body weight of STZ in sodium citrate buffer(pH 4.3) after 4 weeks feeding of experimental diets. Rats were sacrified at the 6th day of diabetic states. Superoxide dismutase(SOD) activity in kidney was decreased by 25% and 20% in DM-0C and DM-0.25C groups compared with normal group, DM-0.5C group was not significantly different when compared with normal group. Glutathione peroxidase(GSHpx) activity in kidney was were no significant differences the diabetic groups compared to normal group. Xanthin oxidase(XOD) activity was increased by 110% and 63% in DM-0C and DM-0.25C groups compared with normal group, DM-0.5C group was not significantly different when compared with normal group. The contents of superoxide radical(O$_2$)in kindney were 116% and 33%, respectively, higher in DM-0C and DM-0.25C groups than normal group. DM-0.5C group and normal groups were similar levels in their superoxide radical contents of kidneys. Levels of TBARS(thiobarbituric acid reactive substance) in kidney were increased by 62% in DM-0C group, when compared with normal group, but those of DM-0.5C group were similar to that of normal groups. These results indicate that free radical generation system was weakened and free radical scavenger system was enhance in kidney of STZ-induced diabetics rats by dietary catechin. Thereby it may reduce renal disorders such as oxidative damage and aging of tissue.

  • Superoxide dismutase which is metalloenzyme that decomposes superoxide radicals into hydrogen peroxide and molecular oxygen. Vitreoscilla has FeSOD. Expression of FeSOD to paraquat was largely constitutive. This suggests that the basal level of FeSOD is sufficient to provide protection against superoxide generated during normal aerobic metabolism. Induction of SOD by iron supports that insertion of the active site metal into the corresponding apoprotein. The effect of paraquat on induction by iron seemed that iron brought the synergism effect in SOD activity with paraquat. It suggests that the relief of growth inhibition is due to protection against the lethality of O$_2$afforded by the elevated SOD. There may be control of FeSOD activity posttranslationally. Posttranslation control of enzyme function is particularly feasible for a metalloenzyme, for which conversion of apo- to holoenzyme may be the rate-limiting or regulatory step.

Role of obioactin on toxoplasmacidal activity within mouse peritoneal macrophages (마우스 복강 macrophages내(內)의 살(殺)톡소플라즈마 활성에 있어서 obioactin의 역할)

  • The present study was undertaken to examine the effects of obioactin, lonomycin A, and MDP on toxoplasmacidal activities in glycogen-induced mouse peritoneal macrophages. The killing effect of obioactin on Toxoplasma multiplication was increased significantly in proportion to its concentrations. $O_2{^-}$ generation in obioactin-treated macrophages was also increased from twofold to threefold when compared with that of untreated control. Similarly, $H_2O_2$ continued to rise in parallel with increase of the concentration of obioactin. Lonomycin A-treated macrophages also exhibited a good effect of dose-response on toxoplasmacidal activities. However, $O_2{^-}$ and $H_2O_2$ were not generated significantly in lonomycin A-treated macrophages. Macrophages treated with muramyl dipeptide (MDP) were not found to inhibit the prolifi:ration of Toxoplasma but showed the enhancement of $O_2{^-}$ and $H_2O_2$, generation. The released lysozyme levels from macrophages into cultured media were decreased tn dose-dependent fashion by in vitro treatment of obioactin, lonomycin A, and MDP. The intracellular lysozyme levels appeared to be a constant value regardless of increasing the concentrations of obioactin, lonomycin A, and MDP. Therefore, these results suggest that Toxoplasma multiplication within macrophages treated with obioactin was inhibited by the generation of $O_2{^-}$ and $H_2O_2$ and that lysozyme per se within or released from macrophages had no effect on toxoplasmacidal activity.

  • Preterm infants are vulnerable to the oxidative stress due to the production of large amounts of free radicals, antioxidant system insufficiency, and immature oligodendroglial cells. Reactive oxygen species (ROS) play a pivotal role in the development of periventricular leukomalacia. The three most common ROS are superoxide ($O2^{\cdot-}$), hydroxyl radical ($OH^{\cdot}$), and hydrogen peroxide ($H_2O_2$). Under normal physiological conditions, a balance is maintained between the production of ROS and the capacity of the antioxidant enzyme system. However, if this balance breaks down, ROS can exert toxic effects. Superoxide dismutase, glutathione peroxidase, and catalase are considered the classical antioxidant enzymes. A recently discovered antioxidant enzyme family, peroxiredoxin (Prdx), is also an important scavenger of free radicals. Prdx1 expression is induced at birth, whereas Prdx2 is constitutively expressed, and Prdx6 expression is consistent with the classical antioxidant enzymes. Several antioxidant substances have been studied as potential therapeutic agents; however, further preclinical and clinical studies are required before allowing clinical application.

Effects of Mulberry (Morus alba L.) Leaf Extract on Oxygen Radicals and Their Scavenger Enzymes in Brain of SD Rats (뇌조직의 활성산소 및 그 제거효소에 미치는 뽕잎 추출물의 영향)

  • This study was designed to investigate the effects of mulberry (Morus alba L.) leaf extract (MLE) on oxygen radicals and their scavenger enzymes in brain membranes of rats. Sprague-Dawley (SD) male rats (160$\pm$10 g) were fed basic diet (control group), and experimental diets (MLE-100 and MLE-300 groups) added 100 and 300 mg/kg BW/day for 6 weeks. Hydroxyl radical (.OH) lecels resulted in significant decreases (13.4% and 21.1%, 12.0% and 13.4%, respectively) in brain mitochondria and microsome of MLE-100 and MLE-300 groups compared with control group. Superoxide radical ($O_2$) levels were significantly decreased about 12% in brain cytosol of MLE-300 group compared with control group. Lipid peroxide (LPO) levels were effectively inhibited (18.1% and 12.3%, respectively) in brain mitochondria and microsomes of MLE-300 groups compared with control group. Oxidized protein (OP) levels were significantly decreased (14.2%, and 10.9%, respectively) in brain mitochondria and microsomes of MLE-300 groups compared with control group. Mn-SOD activities in brain mitochondria were significantly increased (13.5% and 18.6%, respectively) in MLE-100 and MLE-300 groups, and Cu/Zn-SOD activities in brain cytosol were also effectively increased (about 17.7%) in MLE-300 groups compared with control group. GSHPx activities in brain cytosol were remarkably increased (17.2% and 23.9%, respectively) in MLE-100 and MLE-300 groups compared with control group. These results suggest that anti-aging effect of mulberry leaf extract (MLE) may play a pivotal role in attenuating a various age-related changes in brain.

Effects of Silk Fibroin Powder on Oxygen Radicals and Their Scavenger Enzymes in Serum of Rats (실크 피브로인 분말이 혈청중의 활성산소 및 제거효소에 미치는 영향)

  • This study was designed to investigate the effects of silk fibroin powder on oxygen radicals and their scavenger enzymes in serum of rats. Sprague-Dawley(SD) male rats (160${\pm}$10 g) were fed experimental diets (SFP-2.5 and SFP-5.0 groups)added 2.5 and 5.0 g/kg BW/day for 6 weeks. Triglyceride levels were remarkably inhibited (15∼25%) in SFP-2.5 and SFP-5.0 groups compared with control group. Hydroxyl radical ($.$OH) formations resulted in a marked decrease (15%∼25%) compared with control group, while superoxide radical and hydrogen peroxide formations resulted in a significant decrease (6∼12% and 10%). Lipid peroxide and oxidized protein (>C=O group) productions resulted in a significant decrease (6∼12% and 6%) compared with control group. Superoxide dismutase (SOD) and catalase (CAT) activities were remarkably increased (35∼55% and 40∼50%), but glutathione peroxidase (GSHPX) activities were significantly increased (13%) compared with control group. These results suggest that anti-aging effect of silk fibroin powder (SFP) may play a pivotal role in attenuating a various age-related changes.

  • This study was undertaken to determine whether Juglandis Semen extraction(JS) has a protective effect against the lung cell injury caused by oxidant, t-butythydroperoxide(t-BHP) and $H_2O_2$ in rabbit lung slices. JS significantly prevented an increase in water content induced by t-BHP. Similarly, JS significantly prevented the lipid peroxidation induced by t-BHP. Cellular concentration of glutathione, and the activities of catalase and superoxide dismutase were significantly not altered by 5% JS. However, JS at 5% concentration significantly increased the glutathione peroxidase activity in oxidant-treated and control tissues. JS decreased directly the production of superoxide or hydroxyl radical. These results indicate that JS prevents the cell injury and lipid peroxidation induced by oxidants in the lung. Such an antioxidant effect is attributed to enhancement of major endogenous antioxidant defence systems such as glutathione peroxidase and direct inhibition of oxygen free radical production.

How Extracellular Reactive Oxygen Species Reach Their Intracellular Targets in Plants

  • Reactive oxygen species (ROS) serve as secondary messengers that regulate various developmental and signal transduction processes, with ROS primarily generated by NADPH OXIDASEs (referred to as RESPIRATORY BURST OXIDASE HOMOLOGs [RBOHs] in plants). However, the types and locations of ROS produced by RBOHs are different from those expected to mediate intracellular signaling. RBOHs produce O2•- rather than H2O2 which is relatively long-lived and able to diffuse through membranes, and this production occurs outside the cell instead of in the cytoplasm, where signaling cascades occur. A widely accepted model explaining this discrepancy proposes that RBOH-produced extracellular O2•- is converted to H2O2 by superoxide dismutase and then imported by aquaporins to reach its cytoplasmic targets. However, this model does not explain how the specificity of ROS targeting is ensured while minimizing unnecessary damage during the bulk translocation of extracellular ROS (eROS). An increasing number of studies have provided clues about eROS action mechanisms, revealing various mechanisms for eROS perception in the apoplast, crosstalk between eROS and reactive nitrogen species, and the contribution of intracellular organelles to cytoplasmic ROS bursts. In this review, we summarize these recent advances, highlight the mechanisms underlying eROS action, and provide an overview of the routes by which eROS-induced changes reach the intracellular space.

Expression of a Cu-Zn Superoxide Dismutase Gene in Response to Stresses and Phytohormones in Rehmannia Glutinosa

  • Superoxide dismutases (SOD) are metalloenzymes that convert $O_2^-\;to\;H_2O_2$. Rehmannia glutinosa is highly tolerant to paraquat-induced oxidative stress. The primary objective of this study was to characterize regulation of SOD gene expression in R. glutinosa in response to oxidative stresses and hormones. A full-length putative SOD clone (RgCu-ZnSOD1) was isolated from the leaf cDNA library of R. glutinosa using an expressed sequence tag clone as a probe. RgCu-ZnSOD1 cDNA is 777 bp in length and contains an open reading frame for a polypeptide consisted of 152 amino acid residues. The deduced amino acid sequence of the clone shows highest sequence similarity to the cytosolic Cu-ZnSODs. The two to three major bands with several minor ones on the Southern blots indicate that RgCu-ZnSOD1 is a member of a small multi-gene family. RgCuZnSOD1 mRNA was constitutively expressed in the leaf, flower and root. The expression of RgCu-ZnSOD1 mRNA was increased about 20% by wounding and paraquat, but decreased over 50% by ethylene and $GA_3$. This result indicates that the RgCu-ZnSOD1 expression is regulated differentially by different stresses and phytohormones at the transcription level. The RgCu-ZnSOD1 sequence and information on its regulation will be useful in investigating the role of SOD in the paraquat tolerance of R. glutinosa.

Evaluation of Cigarette Quality by Measurement of Oxygen Free Radicals in Smoke (담배 연기 중 산소 자유 라디칼 측정에 의한 품질 평가)

  • To evaluate tobacco quality, several mathods including sensory test, or measurement of some toxic compounds such as tar, nicotine and carbon monoxide in cigarette smoke have been used. However, many detrimental effects of smoking on the physiological functions including respiratory system reported were turned out to be the action of reactive oxygen species. Therefore, the amounts of oxygen free radicals such as superoxide, hydroxyl radical, even hydrogen peroxide in the cigarette smoke are thought the very important factors. In the present study, we have determined the generation of superoxide and the content of hydrogen peroxide using superoxide dismutase and catalase in the gas and particulate phases obtained from cigarette smoke, respectively. In the aqueous extracts of total particulate materials, suproxide and hydrogen peroxide were detected, and there was an excellent correlation between oxygen tint of oxygen free radicals in cigarette smoke may be a useful index for evaluation of cigarette quality in the aspect of smoking and health.
