Analysis of Zearalenone Contamination in Cereal-Based Products Using High Performance Liquid Chromatography-Fluorescence Detector and Ultra Performance Liquid Chromatography-Tandem Mass Spectrometry (곡류가공품 중 제랄레논 오염도 조사)

  • Zearalenone (ZEA) is an estrogenic mycotoxin mainly produced by Fusarium graminerum, a species which colonizes a wide variety of cereals, including wheat, barley and processed products. A survey of ZEA contamination was conducted on 141 dried confectioneries, 59 breads and rice cakes, 135 noodles and 101 other products, for a total of 432 commercial samples. Samples were analyzed by high performance liquid chromatography with fluorescence detection (HPLC-FLD) after immunoaffinity clean-up and was confirmed by liquid chromatography tandem mass spectrometry (LCMS/MS). The limits of detection and quantification were 2.0 and $6.0{\mu}g/kg$, respectively. The recovery ranged from 80.2% to 98.4% in the cereal based product. ZEA was detected in 38 samples (8.8% incidence), including 3 snack, 2 biscuit and 33 other cereal products. The ZEA contamination levels were in the range of $5.38-53.76{\mu}g/kg$. Finally, LC-MS/MS analysis of the contaminated samples was conducted to confirm the detected ZEA, and all 38 samples showing ZEA by HPLC-FLD were confirmed by LC-MS/MS.

Quality Characteristics of Jelly Prepared with Flowering Cherry(Prunus serrulata L. var. spontanea Max. wils.) Fruit Powder (버찌(Fruit of Prunus serrulata L. var. spontanea Max. wils.) 분말 첨가 젤리의 품질특성)

  • The purpose of this study was to investigate the effect of adding various concentrations of cherry powder(obtained from freezed dried fruit of Prunus serrulata L. var. spontanea Max. wils.) on the physicochemical, antioxidant, and sensory characteristics of jelly. The pH of the jelly decreased significantly with the addition of cherry powder. The sugar content of the jelly increased significantly (p<0.05) with cherry powder added more than 7%. The Hunter color L-values decreased significantly (p<0.05) by the addition of cherry powder, and a- and b-values increased by the addition of cherry powder compared to the control, but there were no constant changes with increasing amounts of cherry powder. For the textural characteristics, the addition of cherry powder significantly (p<0.05) increased the hardness, chewiness and gumminess but there were no significant difference (p<0.05) in the springness and cohesiveness. The antioxidative activity measured by DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity of jelly increased as the concentrations of cherry powder increased. The jellies containing 1 and 5% cherry powder had acceptable sensory properties, such as color, flavor, taste, texture, and overall quality. In consideration of the antioxidant properties and sensory characteristics, the highest quality improvement was obtained by adding 5% (w/w) of cherry powder into the jellies.

Investigation on Favorable Substrate Formulation for Bag Cultivation of Grifola frondosa (잎새버섯 봉지재배에 적합한 배지조성 연구)

  • This study was carried out to develop artificial culture method of Grifola frondosa in polypropylene bag. To find out the favorable substrate formulation of G. frondosa, physicochemical conditions were investigated. The T2 formulation (55 : 25 : 12 : 8) mixing ratio of oak sawdust, oak chip, dried bean-curd refuse and wheat bran showed the shortest time to complete the crop cycle and the highest yield (weight of fresh mushrooms harvested at maturity). Those physicochemical conditions were pH 4.3, 2.4% crude fat contents, 54.1% total carbon, 1.42% total nitrogen, 38.1 C/N ratio, 75.5% porosity and $0.21\;g/cm^3$ bulk density. Among the physicochemical factors, pH, crude fat and total nitrogen may affect mushroom yield. Therefore, development of favorable substrate would be benefit to increase production efficiency of G. frondosa, mushrooms and be commercial potential.

Anti-inflammatory activities of Scolopendra subspinipes mutilans in RAW 264.7 cells (RAW 264.7 세포에서 왕지네 추출물의 항염 활성)

  • Purpose: The dried body of Scolopendra subspinipes mutilans has long been used as a traditional Korean medicinal food, but little is known about its mechanisms of action. In this study, we investigated the anti-inflammatory activities of Scolopendra subspinipes mutilans and possible mechanisms in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Methods: Cytotoxicity of Scolopendra subspinipes mutilans extract (SSME) was measured by MTT assay, anti-inflammatory activities were analyzed by nitric oxide (NO) production, the expression of inducible NO synthase (iNOS) and the mRNA level of pro-inflammatory cytokines such as $interleukin-1{\beta}$ ($IL-1{\beta}$) and interleukin-6 (IL-6). Nuclear translocation of nuclear factor-kappa B ($NF-{\kappa}B$) p65 subunit and degradation of inhibitory kappa B ($I{\kappa}B$) were examined by western blot. Results: SSME inhibited LPS-induced NO production and iNOS expression without cytotoxicity. Up-regulation of LPS-induced pro-inflammatory cytokines, $IL-1{\beta}$ and IL-6 was dose dependently attenuated by SSME. Exposure of pyrrolidine dithiocarbamate, an $NF-{\kappa}B$ specific inhibitor, accelerated the inhibitory effects of SSME on NO production and iNOS expression in LPS-stimulated cells. Moreover, translocation of $NF-{\kappa}B$ from the cytosol to the nucleus and degradation of $I{\kappa}B$ were decreased by treatment with SSME in LPS-induced cells. Conclusion: These results suggest that the SSME might have the inhibitory effects on inflammation, partly through inhibition of the $NF-{\kappa}B$ signaling pathway.

Plant Growth Responses and Characteristics of Composting of Poultry Manure with Peatmoss and Cocopeat as Bulking Agent (수분조절제로서 피트모스 및 코코피트를 이용한 계분 발효 특성 및 시비효과)

  • This study was conducted to evaluate plant growth responses and characteristics of composting poultry manure with peatmoss and cocopeat as bulking agent. Treatments were designed as follows; only poultry manure (OP), sawdust+poultry manure (SP), peatmoss+poultry manure (PP), and cocopeat+poultry manure (CP). Period maintained $50^{\circ}C$ over in compost pile of PP and CP was 35 days that longer about 10 days than that of OP or SP, and water content of PP and CP after composting 45 days 50% that was higher about 10%. While poultry manure composted, EC values of PP and CP were higher than that of SP, and pH and OM/N ratio not significant. After finished composting poultry, physicochemical properties of dried all composts were matched on Korean fertilizer guideline. As compared with OP or SP, applications of PP and CP were improved growth and productivity of lettuce, kale and mustard leaf.

A Study on the Method of Health Promotion in Korean Medicine: Correlation between Night-shift Work and Yin-deficiency (건강증진을 위한 한의학적 방법론 연구 -야간근로와 음허증의 상관관계를 중심으로-)

  • This study regarded irregular life styles such as nigh-shifts as contrary to the norm advised in the rules of Yang-saeng to analyze the problems brought about in relation to the concept of 'Yin-deficiency syndrome'. Yin-deficiency survey was given to sales workers on a big shopping mall in Seoul to compare the measurements of daytime workers to those of nigh-shift workers. The measurement of complaining symptoms related to Yin-deficiency of daytime workers and night-shift workers were compared, In comparison of the daytime workers and the night-shift workers, night-shift workers showed higher measurements than the daytime workers in the item of irritable fever on the five Hearts, flushing of the zygomatic region in the afternoon, tidal fever, dizziness, insomnia, yellow and scanty urination, and constipation. Especially tidal fever, insomnia, and constipation showed statistically significant difference. The total of ten items consisting of Yin-deficiency-related symptoms showed statistically significant high score in night-shift workers than the daytime workers. 'Factors for deficiency-type Heat' consisting of irritable fever on the five Hearts, flushing of the zygomatic region in the afternoon, tidal fever, and dried mouth and throat showed statistically significant high score in night-shift workers than the daytime workers. 'Accompanying factors' consisting of night sweats, emaciation, dizziness, insomnia, yellowish and scanty urination, and constipation also showed statistically and significantly high score in night-shift workers than the daytime workers. From the above results that night-shift workers show high degree of Yin-deficiency than daytime workers, and those overworking irregularly also show high degree of Yin-deficiency than those who work for adequate amount of time regularly implies that sitting up at night for work and sleeping at daytime, excessive work, and irregular life styles all function as high-risk factor for Yin-deficiency.

  • Waste activated sludge(WAS) collected from domestic wastewater treatment plant is biomass that contains large quantities of organic matter. However, relevant literature show that the bio-hydrogen yield using WAS was too low. In this study, the effect of pretreatment of WAS on hydrogen yield was investigated. Pretreatment includes acid and alkali treatments, grinding, heating, ozone and ultrasound methods. After pretreatment organic matters of WAS were solubilized and soluble chemical oxygen demand(SCOD) was increased by 14.6 times. Batch experiments were conducted to investigate the effects of pre-treatment methods and buffer solution, hydrogen partial pressure, and sodium ion on hydrogen production from WAS by using heated anaerobic mixed cultures. Experimental results showed that addition of buffer solution, efficient pre-treatment method with alkali solution, and gas sparging condition markedly increased the hydrogen yield to 0.52 mmol $H_2/g$-DS.

Development of hiting sesamer thresher (타격식 참깨탈곡기 개발)

  • Generally, sesame are dried on the field after being harvested in Korea. Since harvesting season is rainy autumn and sesame drying is mainly affected sun and natural wind, it is difficult to dry sesame in time. Moreover, sesame threshing by hand is very laborious work. Therefore, tractor mounted sesame thresher which threshes sesame bundles fed in upside down and conveyed along threshing bar was developed to mechanize sesame threshing and to reduce labor cost in this research. the thresher was equipped with a threshing bar which beats sesame bundles and a three layer sieve which screens sesame. The results are summarized as follows ; 1 The sesame thresher was consisted of a hitting-stick, a feeding chain conveyor, a threshing bar, conveyor belt, and the three-layer shaking sieve. 2. In threshing test, prototype thresher showed maximum threshing ratio 98.5%, 98.7% at 14, 17 cpm beating rate respectively. 3. In screening test, prototype sieve showed maximum threshing ratio 97.2% at 12$^{\circ}$ of inclined angle and 220 cpm of sieve vibrating rate. 4. Prototype showed 98.7% of threshing ratio, 1.3% of threshing loss, 97.0% of screening ratio, 0.7% of screening loss on the rest condition of 15 sesame bundles/min of feeding rate, 14 cpm of beating rate, 220 cpm of sieve vibrating rate. 5. The working performance of prototype was 0.5hr/10a. It was 9.6 times more efficient than manual work. And, operation cost of prototype was saved by 45.9% compared to manual work.

Co-occurrence of Deoxynivalenol and Zearalenone in Cereals and their Products (곡류와 그 가공품에서 Deoxynivalenol과 Zearalenone의 분석)

  • Deoxynivalenol (DON) and zearalenone (ZEN) are naturally occurring toxins produced by Fusarium species, which may grow on cereals. The aims of this study were to determine the incidence and contamination levels of DON and ZEN in cereal products. Seventy samples of cereal products were randomly selected from retail outlets during 2005 and 2006. DON and ZEN were analyzed by using high performance liquid chromatography with fluorescence and UV-detector, respectively. Detection limits were $4.4{\mu}g\;kg^{-1}$ for DON and $3.4{\mu}g\;kg^{-1}$ for ZEN. DON and ZEN were detected in 37 and 17, respectively, of the 70 samples, but the levels found were very low. In particular, out of 70 samples, 12 samples of corn and barley were co-contaminated with DON and ZEN, with levels ranging from 5.6 to $1842.3{\mu}g\;kg^{-1}$ for DON and 12.1 to $174.9{\mu}g\;kg^{-1}$ for ZEN, respectively. However, DON and ZEN were not detected in breakfast cereals and wheat flour. The highest level was found in dried corn kernel samples that confirmed by LC-MS. This study show that DON and ZEN co-contaminate with low levels in cereal products.

Insulin Sensitizing and Insulin-like Effects of Water Extracts from Kalopanax pictus NAKAI in 3T3-L1 Adipocyte (엄나무 추출물이 3T3-L1 지방세포에서 인슐린 민감성과 인슐린 유사성 작용에 미치는 영향)

  • Effects of the water extract from Kalopanax pictus NAKAI on insulin-like action and glucose uptake in 3T3-L1 cells were investigated. The bark of K. pictus NAKAI was treated with hot water and the extract was freeze-dried. Total extract of K. pictus NAKAI was fractionated into 6 fractions with increasing gradients from 0 to 100% MeOH on Amberlite XDA-4. Treatment of 3T3-L1 fibroblasts with 1 ${\mu}g/ml$ and 10 ${\mu}g/ml$ of K. pictus NAKAI total extracts significantly increased the differentiation of the cells. When co-treated with inducers such a dexamethasone, 1-methyl-3-isobutylxanthine and insulin, the differentiation was increased at 1 ${\mu}g/ml$ of total extract, but not at 10 ${\mu}g/ml$. In 3T3-L1 adipocytes, glucose uptake was increased by 3.3 times with addition of 0.3 and 3 ${\mu}g/ml$ of Fr. 1 (0-10% MeOH) and Fr. 3 (30% MeOH) at 3 ng/ml insulin. In conclusion, K. pictus NAKAI contains such compounds that play a role of insulin-like action and insulin sensitizer.
