Effect of Sulfate and Heavy Metals on Methanogenic Activation of in the Anaerobic Digestion of Tannery Wastes (피혁폐수의 혐기성 소화시 황산염과 중금속이 메탄균 활성에 미치는 영향)

  • For treating tannery wastewater containing high sulfate and heavy metals, test was performed to assess their performance, competition between SRB (sulfate reducing bacteria) and MPB (methane producing bacteria), and the activity of MPB according to change of chromium concentrations. COD removal efficiency was above 70% at VLR (volumetric loading rate) of 2.0 gCOD/I.day and HRT (hydraulic retention time) of 18hrs at $35^{\circ}C$. In the competition between SRB and MPB, about 15% of the removed COD was utilized by SRB in the begining, but it became 43% at the end. It indicated that MPB was strongly suppressed by the occurrence of significant sulfate reduction since a large electron flow was uptaken by SRB. For the entire experiment, removal efficiencies of chromium concentration were more than 90%. Despite high removal efficiencies of chromium concentration, performance of reactor did not change significantly during the experimental periods. Expecially, chromium (III) is tannery wastewater is less toxic than chromium (VI).

  • The effect of varying sulfate concentration on continuous fermentative hydrogen production was studied using enriched mixed microflora in continuously fed reactor. Glucose was used as a model substrate for carbohydrates, and hydraulic retention time (HRT) was maintained at 1, 0.5, 0.25 day, respectively. Sulfate concentration was 0${\sim}$20,000 mg/L and the operating pH was maintained at 5.5. The experimental results indicate that hydrogen production is not affected by high sulfate concentration and shorter HRT of 0.25 day enhance hydrogen production. At HRT 1, 0.5, 0.25 day, the hydrogen production rate and hydrogen yield were 2.6, 4.6, 9.4 L/day, and 2.0, 1.8, 1.6 mol $H_2$/mol glucose, respectively. Residual sulfate content was 96${\sim}$98, 95${\sim}$97, and 94${\sim}$97% at HRT 1, 0.5, 0.25 day which show that no sulfate reduction occurred in the reactor during the experiments. Results of Fluorescence In Situ Hybridization (FISH) may indicate the presence of HPB (hydrogen producing bacteria) under all experimental conditions. However, SRB (sulfate reducing bacteria) were not found.

  • CBCT is useful for improving the accuracy of the treatment site, but Repeated use increases the exposure dose. In this study, we aimed to provide basic data for dose reduction in CBCT implementation by dataization the simulating and dose reduction effect using shielding substance. Material in this study, Analyzation the photon beam by simulate the CBCT Through MCNPX and then calculate the absorption dose of body organ at shooting moment of thoracic abdominal position as target UF-Revise simulated body. At this time. Dose reduction effects at this time were evaluated according to the texture of materials and presence of shielding materials( lead, antimony, barium, sulfate, tungsten, bismuth). When CBCT was taken without shielding, the dose was calculated to be high in the breast and spine, and the dose in the esophagus and lung was calculated to be low. The doses according to the shield material were calculated as barium sulfate, antimony, bismuth, lead, and tungsten. The shielding rate was the highest in the thymus (73.6%) and the breast (59.9%) compared with the dose reduction according to presence or absence of the shield. However, it showed the lowest shielding rate in lung (2.1%) and spine (12.6%).

Microbial Community Composition Associated with Anaerobic Oxidation of Methane in Gas Hydrate-Bearing Sediments in the Ulleung Basin, East Sea (동해 울릉분지 가스 하이드레이트 매장 지역의 메탄산화 미생물 군집 조성 및 분포)

  • To elucidate the microbial consortia responsible for the anaerobic methane oxidation in the methane hydrate bearing sediments, we compared the geochemical constituents of the sediment, the rate of sulfate reduction, and microbial biomass and diversity using an analysis of functional genes associated with the anaerobic methane oxidation and sulfate reduction between chimney site (UBGH2-3) on the continental slope and non-chimney site (UBGH2-10) on the basin of the Ulleung Basin. From the vertical profiles of geochemical constituents, sulfate and methane transition zone (SMTZ) was clearly defined between 0.5 and 1.5 mbsf (meters below seafloor) in the UBGH2-3, and between 6 and 7 mbsf at the UBGH2-10. At the UBGH2-3, the sulfate reduction rate (SRR) in the SMTZ exhibited was appeared to be $1.82nmol\;cm^{-3}d^{-1}$ at the depth of 1.15 mbsf. The SRR in the UBHG2-10 showed a highest value ($4.29nmol\;cm^{-3}d^{-1}$) at the SMTZ. The 16S rRNA gene copy numbers of total Prokaryotes, mcrA, (methyl coenzyme M reductase subunit A), and dsrA (dissimilatory sulfite reductase subunit A) showed the peaks in the SMTZ at both sites, but the maximum mcrA gene copy number of the UBGH2-10 appeared below the SMTZ (9.8 mbsf). ANME-1 was a predominant ANME (Anaerobic MEthanotroph) group in both SMTZs of the UBGH2-3 and -10. However, The sequences of ANME-2 were detected only at 2.2 mbsf of the UBGH2-3 where high methane flux was observed because of massive amount of gas hydrate at shallow depth. And Desulfosarcina-Desulfococcus (DSS) that is associated with ANME-2 was detected in 2.2 mbsf of the UBHG2-3. Overall results demonstrate that ANME-1 and ANME-2 are considered as significant archaeal groups related to methane cycle in the subsurface sediment of the East Sea, and ANME-2/DSS consortia might be more responsible for methane oxidation in the methane seeping region than in non-seeping region.

A Study on the Chemical and Dyeing Properties of Rhus Verniciflua Extract (옻나무 추출액의 염색성)

  • This study was carried out to investigate K/S values, surface color, the fastness to washing, bacteria reduction rate of the silk and cotton fabrics dyed with Rhus verniciflua extract under the various dyeing conditions. The optimum dyeing temperature, dyeing time, dyeing pH and repetition of the silk fabrics dyed with Rhus verniciflua extract were $l00^{\circ}C, 30min., pH 5, five times repetition respectively, but in the cotton fabrics, it were $60^{\circ}C . 30min., pH 7, one times repetition. It were colored orangish Yellow in the silk fabrics and colored bright yellow in the cotton fabrics dyed with Rhus verniciflua extract. Surface color(munsell value) was not changed by the mordanting agents but those of the silk showed high tone when mordanting with stannous chloride, and it was decolored and darked when mordanting with ferric sulfate. The fastness to washing in the silk fabrics dyed with mordanting agents improved in 4~5 grade, but the cotton fabrics were 3~4 grade, so washing fastness of the silk and cotton fabrics were significantly improved when washing with the neutral detergent. The bacteria reduction rate of the silk fabrics increased drastically by dyeing of Rhus verniciflua extract.

Thiobenzamide S-oxidation in Perfused Rat Liver: Ex Vivo Determination of Hepatic Flavin-Containing Monooxygenase Activity

  • An ex vivo assay determining the flavin-containing monooxygenase (FMO) activity in perfused rat liver has been developed by assessing the rate of thiobenzamide S-oxide (TBSO) formation from the infused thiobenzamide (TB). The hepatotoxicity by TB or TBSO was not a critical factor for maintaining the FMO activity for up to 50 min. The FMO activity expressed in nmoles TBSO produced/g liver/min was the same for the recycling and non-recycling perfusion. This implies that reduction of the oxidized TBSO back to the parent compound (TB) is negligible. Hydrolysis of the collected perfusates with either ${\beta}-glucuronidase$ or arylsulfatase did not increase the TBSO level and thus, TBSO does not appear to undergo conjugation either to glucuronide or sulfate esters. Thus, measuring the rate of TB S-oxidation in the isolated perfused liver with 1 mM TB for 50 min provides a useful tool for evaluation of the hepatic FMO activity in the absence of hepatic necrosis and without the interferences caused by further conjugation or back reduction of the TBSO to the parent TB.

A Study on Formation of Ni-Tl-P deposits by Electroless Plating (무전해도금에 의한 Ni-Tl-P 피막형성에 관한 연구)

  • This study investigated the bath compositions and plating conditions and crystal structure used for achieving nickel-thallium-phosphorus deposits by means electroless plating. The electroless nickel-thallium-phosphorus deposits were achieved with a bath using sodium hypophosphite as the reducing agent and sodium citrate as the complexing agent. The depositing rate was 10.5mg.$cm^{2-1}$ .$hr^{-1 }$ from the optimistic bath composition, 0.1M nickel sulfate, 0.005M thallium sulfate, 0.2M sodium hypophosphite, and 0.05M sodium citrat and the recommended plating conditions, pH 5.5 and $90^{\circ}C$. The composition of alloy deposits determined by X-ray analysis (EDS) that the Thallium was increased with major increasing concentration of complexing agent and thallium ion in bath solution, it decreased according to the increasing concentrations of reduction agent in the bath solution, Bit Phosphorus showed a contrary to the thallium. It was observed from X-ray diffraction analysis, Scanning Electron Microscopy and Transmission Electron Microscopy. The crystalline structure of deposits was amorphous at the first deposited state but it was changed $Ni-T1-Ni_{5}$ $P_2$ polycrystalline when subjected to 1 hour heat treatment of more than $350^{\circ}C$. TEM observation demonstrated that the microstructure was identical to the result of the XRD at as deposited but it became $Ni-Tl-Ni_{5}$ $P_2$ polycrystalline when heated. And grain size was 10-50nm.

Utilization of Bamboo Leaves as a New Resource of Natural Green Colorants

  • The objective of this study was to find an appropriate method and process for protecting the green color in bamboo leaves and subsequent extraction of the green colorants, chlorophyll. Various inorganic salts including cupric sulfate, ferric sulfate, and zinc chloride were employed as green color protectors. Accordingly, the effects of metal salts and treatment conditions on color protection were investigated to find appropriate protector and conditions. And also, the efficacy of bamboo colorants as a natural green dye was evaluated through dyeing and colorfastness tests. Antimicrobial activity of dyed fabrics was measured by shake flask method in terms of bacterial reduction rate using Staphylococcus aureus(ATCC 6538). On the basis of experimental results for stabilizing bamboo leaves colorants, it was confirmed that Cu was the most appropriate metal type considering dye uptake, photostability and light fastness, and its optimum concentration was 0.025%. After the stabilization, the colorants were extracted efficiently at NaOH aqueous solution of 1.00%. It was concluded that bamboo leave has a high potentiality as new resources to produce a natural green dye with antimicrobial functionality.

  • A series of pore water data were obtained during the different time over one year period between October 1987 and October 1988, from a site on a muddy intertidal flat, located in the Kyeong-gi Bay, west coast of Korea, The results have revealed that the tidal flat is an environment of active nutrient the subface supplied by the overlying seawater is almost completely removed from the pore water at depth of about 10 cm below the sediment surface. The nutrients such as ammonium and phosphate are produced through this process and subsequently accumulated in the pore water forming steep gradients near the sediment surface. Below the main sulfate redirection zone, a secondary peak of dissolved sulfate was often observed. Greal seasonal variation of the pore water nutrient profiles was observed, which was particularly clear in their maximum concentration as well as in their concentration gradient. The rate constants of sulfate reduction and nutrient regeneration, estimated by using a diagenetic model (Berner, 1980), differ by an order of magnitude between the summer and winter seasons. The difference in sediment temperature may account for most of the calculated variation. The C:N:P ratio, calculated from the pore water nutrient gradients also exhibits a slight seasonal difference. The organic matter being decomposed by sulfate reduction appears to be depleted in depleted in nitrogen, compared to the average marine organic matter.

  • Ni-graphite composite powders were prepared by reduction of $Ni^{++}$ from ammoniacal nickel sulfate solution on graphite core by hydrogen gas. Effect of reaction factors on the reduction rate and the properties of nickel layer were investigated by SEM, Optical Microscopy, size and chemical analysis. Induction period, a time lag between the injection of hydrogen gas and the start of the reduction, was 20~110 mins and affected by the reaction temperature and stirring speed. The reduction rate of $Ni^{++}$ was $4.5g/{\ell}/min$ at optimum condition and increased with increasing reaction temperature and stirring speed.
