Studies on Heat Sensitivity of Egg Albumen III. Effects of Egg Albumen Concentration and Addition of Sugars on Heat Sensitivity of Egg Albumen (난백의 열감수성에 관한 연구 III. 난백의 농도와 당류의 첨가가 난백의 열감수성에 미치는 영향)

  • This study was undertaken to find out the effect of eeg albumen concentration and addition of sugars on the functional properties of egg albumen before and after heat treatment at $60^{\circ}C$ for 5 minutes. The turbidity was decreased until 8.3% protein concentration but increased as diluted and decreased again below 3.32% protein concentration before and after the heat treatment. The foaming power was peak at 8.3% protein concentration but decreased as diluted before and after the heat treatment. The foam stability was decreased as diluted before and after the heat treatment. The turbidity was not changed by addition of sucrose before the heat treatment and decreased after the heat treatment. The foaming power was decreased by addition of over 5% sucrose before the heat treatment and decreased by addition of over 2.5% sucrose after the heat treatment . The foam stability was increased by addition of over 5% sucrose before the heat treatment and increased by addition of sucrose after the heat treatment. The turbidity was increased by addition of glucose before the heat treatment and not changed after the heat treatment. The foaming power was decreased by the addition of glucose before the heat treatment and decreased by the addition of over 5% glucose after the heat treatment. The foam stability was decreased by the addition of glucose before and after the heat treatment.

The Nature of Viscous Polysaccharide Formed Kimchi Added Sucrose (Sucrose를 첨가한 김치의 발효시 생성되는 점성물질의 본성에 대하여)

  • Jangkimchi is a kind of Kimchi which is made with soy-sauce instead of salt. Occasionally, when sugar is added to Jangkimchi for condiment, the kimchi juice becomes viscous. In this study. the nature of the viscous material and the condition for producing viscous property in Kimchi juice were investigated. HPLC analysis showed that the viscous material in Jangkimchi is polysaccharide composed of glucose. Sucrose was more effective in forming viscous juice than glucose and the viscosity increased with the addition of sucrose up to 10%. Soy-sauce also played a role in increasing the viscosity of Kimchi juice compared with salt. Aerobic fermentation condition was found to be another factor to make the juice viscous. The population of Leuconostoc mesenteroides, well-known producer of viscous dextran was not different in the Kimchi juice prepared with the addition of sucrose from that without sucrose, which implys that the Jangkimchi preparation methods such as addition of sucrose and soy-sauce would do some effects on the production of viscous material in Jangkimchi.

  • The ability of neonates to perceive and react to pain, has been acknowledged recently. Recent researches have been shown that even short term pain can have lasting negative effects. We know that most of the anatomical pathway and neurotrans-mitter function necessary for pain perception are fully or nearly fully developed in the neonatal period. Many people are still reluctant to believe that pain felt by neonates may be as severe as that felt by older children or adults yet. The objective of the study is to assess and compare the analgesic effects of orally administered sucrose and pacifiers. And to determine the synergistic analgesic effects of sucrose and pacifiers. The tools for this study is Lawrence's NIPS (neonatal infant pain scale) with behavioural pain responses and index for pain physiological reponses as heart rate, respiratory rate and degree of SaO₂. The participants are 96 healthy infants of neonatal age 1-7days and randomised to receive heel prick before 2minutes to blood sampling and physiological test in Nursery of K Medical University Hospital on May-July, 2000. The experimental group assigned to one of three treatment groups : no treatment; a pacifier; placebo(2ml 25% sucrose). Collected data were analyzed with the SAS program using X²-test, ANOVA and Duncan's multiple range test as post hoc. The results were as follows. 1) Pain behaviour responses: The pain score of placebo group(2ml 25% sucrose) is lower than no treatment group and pacifier group significantly(P=.000). Placebo group is different from no treatment and pacifier group with Duncan's multiple range test. 2) Pain physiologic responses (1) heart rate: The heart rate of placebo group(2ml 25% sucrose) is different from that of no treatment group and pacifier group significantly(P=.000). The heart rate change of placebo group is less than the other groups. (2) respiratory rate: The respiratory rate of placebo group(2ml 25% sucrose) is different from that of no treatment group and pacifier group insignificantly (P=.2340). But, the respiratory rate of placebo group is lower than the other groups. (3) SaO₂: The SaO₂ score of placebo group(2ml 25% sucrose) is different from that of no treatment group and pacifier group insignificantly (P=.3265). But, the change of SaO₂ score of placebo group is less than the other groups. In conclusion, the sucrose placebo showed pain relief effect in behavior responses and less physiological responses. Accordingly, the sucrose placebo should be applied nursing intervention for simple pain management as heel prick.

Antimicrobial Effect of Free Available Chlorine on Postharvest Life of Cut Rose 'Brut' (절화 장미 'Brut'의 절화수명에 미치는 유리염소의 항균효과)

  • This study was conducted to investigate the antimicrobial effect of freely available chlorine (FAC) on the vase life of cut rose 'Brut' (Rosa hybrida L.). Postharvest treatments to extend the vase life of cut roses were divided into holding solution treatment and pulsing solution treatment. In holding solution treatment, the cut roses were treated with the preservative solutions containing FAC (0, 10, 20, and $40mg{\cdot}L^{-1}$) and sucrose (0 and 2%, w/v). In pulsing solution treatment, cut roses were dipped into the FAC solutions of 100, 200, and $400mg{\cdot}L^{-1}$ for 10 seconds. The longest vase life of cut roses was observed in the holding solution with FAC $20mg{\cdot}L^{-1}$ as 12 days, followed by pulsing with $400mg{\cdot}L^{-1}$ as 11 days, which were four or five days longer than the control. In addition, relative fresh weight and water uptake were the highest in the holding solutions with FAC 20 and $40mg{\cdot}L^{-1}$. The antimicrobial effect of FAC in vase solution was lasted for eight days after treatment, which was offset by sucrose addition. FAC contents in the FAC holding solution mixed with sucrose were exhausted by 88% two days after treatment, whereas only 15% of FAC was reduced in the holding solution without sucrose. This study indicated that FAC can be applied to extension of the postharvest life of cut roses by antimicrobial activity.

Effects of Free Alkali and Moisture on Sucrose Polyesters Synthesis (유리 알카리 및 수분이 sucrose polyesters 합성에 미치는 영향)

  • Effects of free alkali and moisture on sucrose polyesters (SPE)-possible non calorie fat substitute-synthesis were investigated using a model system composed of sodium oleate, sucrose, potassium carbonate and methyl oleate. Trace amounts of free alkali in sodium oleate were found to interefere with SPE synthesis. When free alkali content in sodium oleate was varied gradually from 0% to 5%(w/w), the yield of SPE production was reduced from 92% to 45.5%. The moisture absorbed in sodium oleate, sucrose and potassium carbonate during storage also interefered with SPE synthesis. The yield (92%) of SPE production with dried ($105^{\circ}C$.6 hrs) reactants and catalysts was higher than that (89%) of SPE production with non-dried. Soybean oil fatty acid sodium soaps (FASS) not containing free alkali could be manufactured with slightly less than molar ratio of sodium hydroxide to soybean oil fatty acid methyl esters (FAME). Practically, 91.7% yield of soybean oil SPE production was outcomed by minimizing free alkali and moisture which were remaining in sucrose, potassium carbonate, soybean oil FASS and soybean oil FAME.

Appropriate in Vitro Culture Conditions of Growing Medium for New Ever-bearing Strawberry 'Goha' (사계성 신품종 딸기 '고하'의 기내배양을 위한 배지의 적정 조건)

  • This study was carried out to determine suitable in vitro culture conditions of new ever-bearing strawberry ($Fragaria$ $ananassa$ Duch.), 'Goha'. Four-week old plantlets which were derived from the meristem culture were used in this study. Three different culture media including MS, Gamborg B5 and White medium were used for the plant culture and the medium concentrations were at the 5 levels of $1/3{\times}$, $1/2{\times}$, $1{\times}$, $2{\times}$ and $3{\times}$. Sucrose content ranged at four levels of 1, 3, 5 and 8% (w/v). Crown diameter on the MS medium was thicker (2.1 mm) than in other media. Fresh weight on the MS medium was 482 mg, which was heavier than 88 mg or 260 mg of Gamborg B5 or White medium, respectively. Particularly, 1/2MS medium was found to have higher growth rate than these in other treatments (fresh weight, dry weight and D/F rate). Shoot length in the treatment of 1% sucrose concentration was 3.6 cm which was the longest. Shoot length was in inverse proportion to the increasing concentration of sucrose. Fresh weight was increased up to 3% sucrose concentration, but decreased above 5% sucrose concentration. From the results, we found that the best condition for in vitro culture of new ever-bearing strawberry 'Goha' was 1/2MS medium supplemented with 1% sucrose concentration.

Effects of Sucrose and Glycerol during the Freezing Step of Cryopreservation on the Viability of Goat Spermatozoa

  • Four experiments were conducted to study the following: i) the influence of different concentrations of sucrose (0.15, 0.3 and 0.5 M with osmolality of 308, 500 and 760 mOsm/kg, respectively) in diluents and control diluent (370 mOsm/kg) on intensity of motility and progressive motility of goat sperm without rehydration and freezing step in four incubation periods (0, 0.5, 2 and 4 h after dilution); ii) the influence of gradual dilution (in 3 steps) on improvements in ascertained results of the first experiment; iii) cryoprotective effects of different concentrations of sucrose (0.15, 0.22, 0.29 and 0.37 M with osmolality of 450, 560, 740 and 920 mOsm/kg, respectively) plus 7% glycerol and 20% egg yolk in basic diluent (Tris-Citric acid-Fructose) and iv) the effect of two concentrations of sucrose (0.15 and 0.22 M) with and without glycerol (7%). In experiment 1, we obtained better results for control diluent, 0.15 and 0.3 M sucrose supplemented diluents with 0 and 0.5 h incubation periods. In experiment 2, apart from a slight improvement, similar tendencies to experiment 1 were observed. In experiment 3, we obtained the best result for diluent with 0.22 M sucrose with regard to intensity of motility, progressive motility, live sperm and normal acrosomes ($40{\pm}4%$, $3.1{\pm}0.2$, $37{\pm}4%$ and $37{\pm}4%$, repectively). In experiment 4, we obtained the best result for diluent with 0.22 M sucrose plus 7% glycerol in regard to intensity of motility, progressive motility and live sperm ($39{\pm}3%$, $3.6{\pm}0.4$ and $41{\pm}4%$, respectively). The characteristic normal acrosomes in diluents without glycerol, i.e. diluents with 0.15 and 0.22 M sucrose showed better results ($39{\pm}8$ and $42{\pm}6%$ respectively). With regard to the release of hyaluronidase enzyme there were no significant differences between diluents (p>0.05). The results of the diluents with 0.15 and 0.22 M sucrose without glycerol were slightly lower than those with glycerol ($69{\pm}11$ and $70{\pm}11$ vs. $72{\pm}11$ and $70{\pm}11{\times}120{\times}10^6$ units $ml^{-1}$, respectively). In conclusion, the use of concentrated sucrose solutions showed that goat sperm can tolerate osmolality up to 560 mOsm (0.22 M) in the freezing period. In addition, glycerol proved to be a necessary cryoprotective agent in the cryopreservation of goat sperm, particularly for intensity of motility, progressive motility and live sperm.

Optimization of Cell Culture Condition for Erythritol Production by Penicillium sp. KJ8l (Penicillium sp. KJ 81에 의한 Erythritol 생산 최적 배양 조건)

  • Erythritol is of interest as a low calorie sweetner. Penicillium sp. KJ8l was screened for erythritol producer in nature. The effect of culture conditions on erythritol production by Penicillium sp. KJ81 was examined. This strain produced about 12 g/l erythritol and a small amout of glycerol. Erythritol was not produced from mannitol, arabinose, sorbitol, and xylose but from glucose, sucrose, fructose, mannose, lactose, maltose, and galactose. This strain was able to produce erythritol in a medium containing 60% sucrose but demonstrated the highest productivity of erythritol in a 30% sucrose medium. The highest yield in Penicillium sp. KJ8l was obtained when 0.5% ammonium sulfate was added to the medium containing 30% sucrose and 0.5% yeast extract. Penicillium sp. KJ81 produced 28.2 g/l erythritol when this strain was cultured in the medium containing 30% sucrose, 0.5% yeast extract, 0.5%$(NH_{4})_{2}SO_{4}$ 0.1% $KH_{2}PO_{4}$ and 0.01% $MgCl_{2}$ under the condition of 1 vvm aeration and 200 rpm agitation at $37^{\circ}C$ for 10 days in 5ι jar fermentor.

Effects of Sucrose Treatment on the Morphology and Integration of foreign DNA into Bovine Oocytes (소 난자에서 형태와 외래 DNA Integration에 관한 Sucrose 처리의 효과)

  • The microinjection of retroviral vectors into the perivitelline spaces of MII-stage oocytes increased production of transgenic bovine embryos. However, oocytes have various sizes of perivitelline space, and there is the tendency that the oocyte membranes are damageable by micropipettes during the injection of retroviral vectors into perivitelline spaces or oocytes. Thus, it was not always possible to stably inject retroviral vector into perivitelline spaces of oocytes. Here we used sucrose to minimize the damage of the oocyte membrane. When the oocytes were suspended in 0.5% sucrose, poor quality oocytes showed rough cytoplasmic membranes, while good quality oocytes maintained smooth membranes. However, when the tatters were subjected to in vitro fertilization, no significant differences were observed in cleavage rates (82% of control Vs. 84% of sucrose treated oocytes). The Same trends were obtained from the oocytes fertilized after microinjection of LN$\beta$-EGFP and LNC-hGH genes into the perivitelline spares. The rates of cleavage and blastocyst from microinjection of LN$\beta$-EGFP genes were 81 and 25%, and from microinjection of LNC-hGM genes were 53 and 30%, respectively. The result indicated that microinjected oocytes could develop to the blastocyst stages after in vitro fertilization with no significant difference from control group. Moreover, the integration of hGH-gene (by PCR analysis) was detected in 52% of infected cleaved embryos and the expression of EGFP-gene (under a fluoresrence microscope) was also observed in 34% of infected blastocyst. These results indicated that 0.5% sucrose treatment could be an efficient method not only to select good quality embryos but also to inject retroviral vectors into perivitelline spares without any harm and hence improving developmental rates.

The Formation of Anthocyanin and Growth Response of Ginseng Cell Lin in vitro (기내배양 인삼세포주의 Anthocyanin 형함과 생장반응)

  • In order to clarify the characteristics of anthocyanln-producing cell line selected by in vitro grown cultures of ginseng (Panax ginseng C.A. Meyer), the calli of anthocyanin-Producing strain were cultured on media supplemented with different concentrations of 2,4-D and sucrose under light or dark condition. The light was found to be essential for anthocyanin synthesis. Anthocyanin synthesis of ginseng cell line was inhibited by the increase of 2,4-D in the medium. On the other hand, sucrose promoted the anthocyanin-production and the optimum concentration of sucrose for the highest production of anthocyanin was 5%. The growth of anthocyanin-producing tell line was best on the medium supplemented with 0.5 mg/l 2,4-D and 5% sucrose.
