An Investigation of Elementary School Teachers학 Conceptions on Buoyancy (부력 개념에 관한 초등학교 교사들의 이해도 조사)

  • Elementary school teachers' understandings about buoyancy were investigated through the questionnaire method. The questionnaire was composed of 4 questions on hydraulic pressure and 8 questions on buoyancy. The questions on buoyancy asked about the correlation of buoyancy with following basic concepts, density of liquid, volume of submerged object and so forth. 295 teachers on the 22 elementary schools in Busan, Yangsan and Gimhae were selected through random sampling method. The results of this study were summarized as follows: On the correlation of the magnitude and direction of hydraulic pressure with the depth of water, a large portion of the respondents had a scientific conception. But on the correlation of hydraulic pressure with density, the relatively small portion of them appeared to have a scientific conception. The respondents, on the whole, had a scientific conception about the correlation of buoyancy with density of liquid. But they seemed to have naive conceptions about the correlation of buoyancy with the volume of a submerged object and with the depth of water, the amount of water in container and the reduced amount of water by the object from container. We found that the respondents were context dependent and tended to search for solutions for the questions of buoyancy using the concept of pressure in the water. From above results, we suggested that in the would-be revised elementary science text book, the contents of pressure in the water should be introduced after the concept of weight in the water was gained.

Immunomodulating Activity of the Exopolymer from Submerged Mycelial Culture of Phellinus pini

  • The immunomodulating activities and chemical characteristics of a water-soluble exopolymer from submerged mycelial culture of Phellinus pini were studied. Anticomplementary activity of this polymer was found to be $73.2\%$, and its activation system occurred through both classical and alternative pathways, where the classical pathway was detected to be the major one by crossed immunoelectrophoresis. Nitric oxide (NO) release ability and acid phosphatase activity of macrophage were increased by 1.6-fold ($100{\mu}g/ml$) and 3.4-fold ($500{\mu}g/ml$), respectively, and splenocyte proliferation in mixed lymphocyte reaction (MLR) was also increased by 2.6-fold ($200{\mu}g/ml$), compared to the control. The molecular weight of this polymer, determined by HPLC, was under 5 kDa. Total sugar and protein contents were 89.7 and 10.3%, respectively. Both sugar and amino acid compositions of the exopolymer were also analyzed.

Modeling and Simulation for the Initial Dynamics of a High Speed Underwater Vehicle Ejected from a Submerged Mother Ship (수중모함에서 사출되는 고속 수중운동체의 초기 거동 모델링 및 시뮬레이션)

  • Heavy-weight high speed underwater vehicle(HSUV) is launched from the submerged mother ship. For the safety point of view, it is important to confirm whether the HSUV would touch the launching mother ship. In this paper, the hydrodynamic force and moment were modeled by the polynomials of motion variables and the simple lift and drag acting on a plate and cylinder which consist of the HSUV's several parts. The mother ship was assumed as the Rankine half body to consider the flow field near the moving ship. Such hydrodynamic force and moment were included in the 6 DOF equations of motion of the HSUV and the dynamic simulations for the various conditions of the HSUV until the propeller activation were performed. Developed simulation program is expected to reduce the number of expensive sea trial test to develop safety logic of the HSUV at the initial firing stage.

Effects of reverse waves on the hydrodynamic pressure acting on a dual porous horizontal plate

  • The seaward reverse wave, occurring on the submerged dual porous horizontal plate, can contribute to the reduction of the transmitted wave as it reflects the propagating wave. However, the collision between the propagating and seaward reverse waves increases the water level and acts as a weight on the horizontal plate. This study investigated the characteristics of the wave pressure created by the seaward reverse wave through the analysis of experimental data. The analysis confirmed the following results: 1) the time series of the wave pressure showed reverse phase phenomena due to the collision, and the wave pressures acted simultaneously on both upper and lower surfaces of the horizontal plate; 2) the horizontal plate became repeatedly compressed and tensile before and after the occurrence of the seaward reverse wave; and 3) the seaward reverse wave created the total wave pressure to the maximum towards the direction of gravity, primarily on the upper plate. It was also confirmed that the wave distributions showed a similar trend to the wave steepness. Such outcome of the analysis will provide basic information to the structural analysis of the horizontal plate as a wave dissipater of the steel-type breakwater (STB).

Effect of sawdust on cordycepin production from the medicinal fungus Pesilomyces tenuipes in submerged culture

  • Cordycepin (3'-deoxyadenosine) is a nucleoside analog known for its diverse range of biological activities. This study investigated the effect of different types of sawdust on the production of the bioactive compound cordycepin. The results of the study showed that different types of wood sawdust affected the biosynthesis of cordycepin and a significant increase was observed when the conventional SDB medium was replaced with 1% NaOH treated pine sawdust. To optimize cordycepin production from Paecilomyces tenuipes in a medium containing 1% NaOH-pretreated pine sawdust, we employed Response Surface Methodology (RSM) in its Box-Behnken design (BBD) canonical form. The optimal conditions were determined as follows: a particle size of 109.5111-mesh (140 ㎛) for 1% NaOH-pretreated pine sawdust, an input weight of 21.1679 g/L, and an incubation time of 73.8423 hours. According to our model, this combination is expected to yield a maximum cordycepin content of 896.1428 ㎍/mL. Experimental validation of this prediction was performed using the suggested optimal conditions, resulting in an average cordycepin content of 922.6771 ㎍/mL across three replicates, thus confirming the model's accuracy.

Cultural Characteristics and Scale-up for Submerged Cultivation of Hericium erinaceum Through Air-lift and Jar Fermenter System (Air-lift 및 Jar Fermenter에 의한 Hericium erinaceum 심부배양의 배양특성 및 Scale-up)

  • For the study of Hericium erinaceum as a useful functional foods and materials, liquid cultivation under two different bioreactors(air-lift fermenter and jar fermenter) which was not studied systematically until now, was conducted as a method of mass cultivation for H. erinaceum. A batch cultivation in an air-lift fermenter and a jar fermenter was examined for enhancing the productivity because of small amounts of mycelial weight and slow growth in case of a liquid culture for H. erinaceum. We found that air lift fermenter system was more effective than jar fermenter for mycelial production of H. erinaceum, and mycelial morphology was a critical factor of the growth. By scale-up and cultivation based on morphological analysis, the conditions for mass production with 30 L and 500 L jar fermenter was 200 and 150 rpm of agitation speed at 1 vvm of aeration rate, respectively, and mycelial dry weight under these conditions was enhanced to about $13{\sim}14g/L$.

Development of the Automatic Turnout (자동 분수공의 개발)

  • Floating-type automatic turnout was developed for the purpose of reducing labor cost and labor-working hours related to turnout management. The point of automation is to use a flexible-float within the turnout. The weight of float is changed by emptying and filling with water at the beginning and ending of irrigation. The turnout is controlled to open and close small bole on the float bottom using electromagnets. With the weight control of float. the gate of turnout is opened by the empty float to begin irrigatiom and is closed by the filled float to stop irrigation. The turnout was designed to be operated by the main computer and to minimize electric power consumption by sending an electric current at the beginning and ending of irrigation. The functional experiment was succesfully carried out and the rating curves for both free overflow condition and submerged flow condition were derived.

  • Absorption of lightweight aggregate affects the properties of fresh and hardened concrete, so care must be taken. In this study, according to KS F 2533 absorption is measured to aggregate size, submerged time, holding time and practically ways to maintain a constant absorption was to seek. The findings for quality control of the lightweight aggregate concrete mixture is saturation of the surface-dry aggregate humidity 100% after 24 hours immersion in the environment has been stored for more than two days to absorb the state was able to define. Dry density at the surface of the lightweight aggregate and lightweight concrete mix design and placement is possible for it to apply.

  • This study relates to low and medium molecular weight isoflavone-${\beta}$-D-glucan produced by submerged liquid culture of Agaricus blazei, a method of producing the isoflavone-B-D-glucan using autolysis enzyme of Agaricus blazei mycelia, and use of the isoflavone-B-D-glucan for anti-cancer and immunoenhancing effect. In acordance with one aspect of the present study, it deals with a method of producing isoflavone-${\beta}$-D-glucan, which comprises the followings; 1) culturing and separating mushroom mycelia, 2) producing low-medium molecular weight isoflavone-${\beta}$-D-glucan from high molecular weight one. The cytotoxicity on human cnacer cell line (Caco-2, MCF-7), the expression of Cyclin D, Bcl-2, Bax protein, p21 protein, p53 protein in MCF-7 cells assessed by SDS-PAGE and immunoblotting, and other immuno related factors such as TNF-${\alpha}$ and IL-1B activities were examined. Structural identification of isoflavone-${\beta}$-D-glucan which showed cytotoxicity against cancer cell and immunoenhancing effects was carried by separation with DEAE-cellulose column chromatography, TLC, HPLC, IR, NMR. Clinical test for the cancer patients (n=119) for 6 month was carried out, and immunoenhancing factors (NK. cell number, ratio of T4/T8) were checked. We concluded the identified isoflavone-${\beta}$-D-glucan has immuno enhancing effects and could be useful for cancer chemoprevention.
