Responses of Pseudomonas sp. DJ-12 to Pollutant Stresses of Benzoate and 4-Chlorobenzoate

  • Aromatic hydrocarbons can be utilized as carbon and energy sources by some microorganisms at lower concentrations. However, they can also act as stresses to these organisms at higher concentrations. Pseudomonas sp. DJ-12 is capable of degrading 0.5 mM concentration of benzoate and 4-chlorobenzoate (4CBA). In this study, the exposure of Pseudomonas sp. DJ-12 to the pollutant stresses of benzoate and 4CBA at various concentrations was comparatively studied for its cellular responses, including survival tolerance, degradability of the aromatics, and morphological changes. Pseudomonas sp. DJ-12 utilized 0.5 to 1.0mM benzoate and 4CBA as carbon and energy sources for growth. However, the organism could not degrade the aromatics at concentrations of 3 mM or higher, resulting in reduced cell viability due to the destruction of the cell envelopes. Pseudomonas sp. DJ-12 cells produced stress-shock proteins such as DnaK and GroEL when treated with benzoate and 4CBA at concentrations of 0.5mM, or higher as sublethal dosage; Yet, there were differing responses between the cells treated with either benzoate or 4CBA. 4CBA affected the degradability of the cells more critically than benzoate. The DnaK and GroEL stress-shock proteins were produced either by 1mM benzoate with 5 min treatment or by higher concentrations after 10min. The proteins were also induced by 0.5mM 4CBA, however, it needed at least 20min treatment or longer. These results indicate that the chlorination of benzoate increased the recalcitrance of the pollutant aromatics and changed the conditions to lower concentrations and longer treatment times for the production of stress-shock proteins. of stress-shock proteins produced by the aromatics at sublethal concentrations functioned interactively between the aromatics for survival tolerance to lethal concentrations.

Stress response as a contributing factor in horses with laminitis

  • Background: Laminitis is a complex and debilitating disease of horses. Numerous predisposing factors contribute to laminitis development, however the exact pathogenesis remains undetermined. Serum T4, cortisol, and histamine are components of the innate stress response and could play a causative or contributory role. Stress hormone concentrations in laminitis are largely unknown. Objective: To evaluate parameters associated with stress response in horses with laminitis, and compare these to healthy horses and horses with gastrointestinal (GI) disease. Methods: Thirty-eight adult horses presenting for non-medical conditions, GI abnormalities, or clinical laminitis were prospectively enrolled. Horses were assigned to the appropriate disease group (healthy, GI disease, and laminitis) and had blood drawn on presentation to the hospital. Samples were analyzed for plasma endogenous adrenocorticotrophic hormone (eACTH), serum cortisol, serum thyroid hormone, and plasma histamine. Results: Stress hormone concentrations were significantly different between horses in the laminitis and GI disease groups. Plasma histamine levels were highest in horses with laminitis, compared with GI disease and controls. Both horses with laminitis and GI disease had increased plasma eACTH when compared to healthy horses. Horses with GI disease had higher serum cortisol concentrations than horses with laminitis or controls. Serum T4 was lower in horses with GI disease than in horses with laminitis and controls. Conclusions: Horses with laminitis had relative increases in both plasma histamine and eACTH concentrations. Serum T4 and cortisol concentrations of horses with laminitis did not differ significantly when compared to healthy horses. The role of stress hormones in equine disease warrants further investigation.

Relationship Between Depression Anxiety Stress Scale (DASS) and Urinary Hydroxyproline and Proline Concentrations in Hospital Workers

  • Objectives: Although increased reactive oxygen species (ROS) is caused by stress accelerates collagen degradation, there was no data on the relationship between stress and urinary hydroxyproline (Hyp) and proline (Pro), a good marker of collagen degradation. The purpose of this study was to evaluate the relationship between depression, anxiety, and stress (DAS) and concentrations of urinary Hyp and Pro. Methods: 97 hospital employees aged 20 to 58 were asked to fill out comprehensive self-administrated questionnaires containing information about their medical history, lifestyle, length of the work year, shit-work and DAS. Depression Anxiety Stress Scale (DASS) was applied to evaluate chronic mental disorders. Urine samples were analyzed using High Performance Liquid Chromatography (HPLC) with double derivatization for the assay of hydroxyproline and proline. Results: The mean value of Hyp and Pro concenturation in all subjects was $194.1{\pm}113.4\;{\mu}mol/g$ and $568.2{\pm}310.7\;{\mu}mol/g$. DASS values and urinary Pro concentrations were differentiated by sex (female > male, p < 0.05) and type of job (nurse > others, p < 0.05). In the stepwise multiple linear regressions, urinary Hyp and Pro concentrations were influenced by stress (Adjusted $r^2$ = 0.051) and anxiety and job (Adjusted $r^2$ = 0.199), respectively. Conclusions: We found that stress and anxiety were correlated with urinary Hyp and Pro concentrations. To identifying a definite correlation, further study in large populations will be needed.

Protective effects of blueberry drink on cognitive impairment induced by chronic mild stress in adult rats

  • BACKGROUND/OBJECTIVES: Stress-induced cognitive impairment is related to the suppression of hippocampal neurogenesis that results from an increase of oxidative stress. Therefore, the aim of this study was to investigate the effects of administration of a blueberry drink, having a high antioxidant power, on the cognitive performance of adult rats exposed to chronic mild stress. MATERIALS/METHODS: Twelve-week-old male Sprague-Dawley rats (n = 48) were randomly divided into four groups: control (CO), stress (ST), control + 5% blueberry drink (CO + B), and stress + 5% blueberry drink (ST + B). After eight weeks, the cognitive performance was assessed using a multiple T-maze water test. Levels of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), and ascorbic acid were measured in the brain, and catecholamine concentrations were measured in plasma. RESULTS: The brain weights of the rats from the ST and ST + B groups were significantly lower than those of the rats from the CO and CO + B groups. The cognitive performance of the ST group was impaired when compared to that of the CO group. This impairment was significantly improved by the blueberry drink supplementation (P < 0.05). The brain SOD and CAT concentrations were not influenced by the stress or by the blueberry drink. However, the brain levels of GPx and ascorbic acid were significantly lower in the ST group than those in the CO group and were increased by the blueberry drink supplementation. The plasma catecholamine concentrations were affected by chronic mild stress and by the blueberry drink. The plasma norepinephrine and dopamine concentrations were decreased by the chronic stress and improved by the blueberry drink supplementation. The plasma epinephrine level was only influenced by the stress. CONCLUSION: These findings suggest that the blueberry drink may protect against the cognitive impairment induced by chronic mild stress.

  • Internal stress of electrodeposited metals affect physical and mechanical characteristics of deposits. Internal stress of nickel deposits was reviewed intensively. Important outcomes are as follows. Substrate have an important effect on internal stress of electrodeposit. Origin of its internal stress could be explained mismatch of crystal lattice and coalescence of crystallites. When surface cleaning is not satisfying, instantaneous stress is low but the electrodeposited layer being thickened increasingly stress become to high and peeling phenomenon occurs. Effect of current density and temperature on internal stress is variable. Internal stress increases rapidly at pH 5 and above because of codepositing colloidal materials caused hydrolysis. Concentrations of nickel ion and $H_3BO_3$ ion affect little on internal stress and solution which contains impurities tend to increase stress. Especially impurities of $H_2O_2$ and iron ion have a great effect on internal stress. Additives are divided in two kind. One is increasing tensile stress another is increasing compressive stress. Concentrations of additives have a great effect on internal stress.

Paclobutrazol 처리가 Perennial Ryegrass의 생육 및 고온과 건조 Stress에 미치는 영향 (Effect of Paclobutrazol on Growth, and High Temperature and Drought Stress in Perennial Ryegrass)

  • This study was conducted to investigate the effect of paclobutrazol [(2 RS , 3 RS )1-(4- chlor-ophenyl )-4, 4- dimethyl -2- (1, 2, 4- triazol -1- yl )- pentan -3-01] on the tolerance of hi-gh temperature and drought stress as related to growth retardation , iranspiration rate , soil water content , nitrogen level and photosynthetic rate in perennial ryegrass ( Loliurn perenne L . ' Omega H , ). Plants were given a 30 ml soil drench of paclohutrazol at the concentrations of 0, 0.01, 0.1, 1.0, 10.. 0, mg / 6 .5cm- diameter pot . The rcsults were as follows : 1. Increasing concentrations of paclohutrazul reduced plant height , leaf area , fresh weight and dry weight , hut increased chlorophyll content per unit area . The number of tillers and leaf width were not affected hy the paclobutrazol concentrations . 2. The proper concentration of paclohatrazol on growth retardation in perennial ryegrass was about I mq /pot , hut leaf deformity and severe growth retardation were shown at high concentration of 10 mq / pot . 3. Perennial ryegrasses grown at 30˚C were shown significantly short plant height and low leaf nitrogen level compared with those grown at 20˚C. Increasing concentrations of paclohutrazol at 20˚C increased nitrogen level hut it could not increase nitrogen level at 30˚C . 4. During the drought stress , increasing temperatures significantly promoted transpiration rate and wilting time . It took about 5 days at 20˚C and 3 days at 30˚C to reach wilting time of leaves from water stress treatment . Soil water contents at wilting time of non-treated controls were averaged 6. 871% at 20˚C and 6. 17% at 30˚C 5. Paclohutrazol reduced transpiration rate at high temperature and drought stress . Wilting appeared at the lower water content of soil according to increasing concentrations of paclobutrazol at 30˚C hut there were no differences among concentrations of at 20˚C. 6.Paclohutrazol treatment at 1 rag /pot reduced injury rate of leaves from 67.1 % and 100 % in control plants to 15.7% and 80% at 20˚C and 3010, respectively. 7. Photosynthetic rate per unit area was significantly reduced at high temperature . Paclohutrazol stimulated photosynthetic rate with increase of concentrations at 20˚C but there was no increasing effect at 30˚C.

NaCl Stress에 따른 보리 유묘의 생육특성 및 세포학적 반응 (Growth and Histological Characteristics of Barley (Hordium vulgare L.) Seedling to NaCl Stress)

  • This study was conducted to determine the morphological responses of barley seedlings to NaCl stress and to investigate histological changes of cells with transmission electron microscope(TEM) after NaCl stress. Plant height and root length of 10-day old barley seedlings with NaCl stress were reduced and inhibition level was found to be more severe in the plant height than in the root length. The leaf length, leaf width and leaf area were shorter as well with NaCl stress than without NaCl stress. However, there was no difference in the number of roots between NaCl treatments. The weight of dry matter decreased at higher NaCl concentrations, especially at 100mM NaCl. The water content of shoots tend to decrease at higher NaCl concentrations, but there was no difference in the water content of roots, The reduced sugar content was greatly increase than starch. Cellulose content was higher in NaCl stressed-plant than control, and tended to decreased at higher NaCl concentrations. Lignin content also decreased NaCl stressed-plant but there was no tendency at NaCl stress concentrations. Electric conductivity of cell sap with seedlings was high with NaCl stressed-plant. Amount of cell sap gradually increased with time in the roots than in the shoots, The grana of chloroplasts was changed by 150mM NaCl concentration. The christe of mitochondria in root meristematic sells ruined in structure and cell wall of leaf and root was also ruined by NaCl stress.

Interfacial Stress Concentrations of Vertical Through-plate to H-beam Connections in CFT Column

  • This paper aims to evaluate the interfacial stress concentrations on connection between vertical through-plate and H-beam in CFT column. Full-scale experiments were performed on three specimens with varying thickness of the vertical through-plate to investigate the interfacial stress concentration factor in the connections. The specimens underwent brittle failure at the location where the steel beam is connected to the vertical through-plate before the steel beam reached its plastic moment. The strain data of the part were analyzed, and the sectional analyses were conducted to determine appropriate residual stress models. In addition, the stress concentration factor was quantified by comparing the analytical local behavior in which the stress concentration is not reflected and the experimental data reflecting the stress concentration. The results showed that the maximum reduction of the stress concentration factor due to an increase in the thickness of the vertical through-plate is 50.3%.

Job Stress and Neuropeptide Response Contributing to Food Intake Regulation

  • The purpose of the present study is to investigate the correlations between food intake behavior and job stress level and neuropeptide hormone concentrations. Job strain and food intake behavior were first identified using a self-reported questionnaire, concentrations of neuropeptide hormones (adiponectin, brain derived neurotrophic factor [BDNF], leptin, and ghrelin) were determined, and the correlations were analyzed. In the results, job strain showed significant correlations with adiponectin (odds ratio [OR], 1.220; 95% confidence interval [CI], 1.001~1.498; p < 0.05) and BDNF (OR, 0.793; 95% CI, 0.646~0.974; p < 0.05), and ghrelin exhibited a significant correlation with food intake score (OR, 0.911; 95% CI, 0.842~0.985, p < 0.05). These results suggest that job stress affects food intake regulation by altering the physiological concentrations of neuropeptide hormones as well as emotional status.

The Effects of Road Transportation on Some Physiological Stress Measures in Goats

  • A study to assess the physiological stress responses in goats that were subjected to road transportation was carried out using 10 Kacang crossbred does. Five does were transported in the morning with another five transported in the afternoon covering a distance of 46 km in an open-truck at an average speed of 55 km/h. Immediately following the road transportation, there were dramatic increases in neutrophil:lymphocyte ratios and plasma glucose concentrations but plasma cholesterol concentrations and body temperature were not affected. The neutrophil:lymphocyte ratios and plasma glucose concentrations appear to be reliable indicators of stress in goats.
