Virulence and Plasmid Profiles of Salmonella gallinarum Isolated from Chickens (닭에서 분리한 Salmonella gallinarum의 병원성 및 Plasmid Profile)

  • The present study was conducted to investigate the antibiotic resistance of 24 Salmonella gallinarum isolated from 48 chicken samples of diagnosed fowl typhoid cases during the period from November 1998 to November 1999. And the isolates of S gallinarum were also tested for their invasion abilities to experimental infection as one of virulence tests, and the presence of virulence-related plasmid in S gallinarum isolates. The results obtained through this study were summarized as follows; 1. All of isolates from 48 cases of 24 farms were identified S gallinarum by biochemical and serological tests.2. Antimicrobial drug resistance test against 24 isolates showed that the isolates were resistant to Colistin(95.8%), and Penicillin(79.2%), Polymyxin B(75.0%), Streptomycin (65.2%), Gentamycin(54.2%), and Tetracycline(33.3%). 3. Mortality in chicken following peroral inoculation of four isolates of S gallinarum during 14-days inoculation pecked at 5 days(40%) after inoculation and all of experimental chickens died within 13 days after inoculation.4. Based on the pattern and number of isolated plasmid from each isolate, plasmid profiles were divided into five groups; group I with 3 plasmids, group II to group IV with 4 plasmids and group V with 5 plasmids.

Resistance on disinfectants of Clostridium chauvoei isolated from Kyongbuk province (경북지역에서 분리된 기종저균의 소독제에 대한 내성)

  • This study was investigated resistance on disinfectants and antibiotics of Clostridium chauvoei isolated from dairy farm in Kyongbuk province. The results obtained were summarized as follows ; C chauvoei isolated from dairy farm were susceptible to norfloxacin, penicillin, tetracycline, erythromycin, enrofloxacin, bacitracin, tyrosine, cephalothin and cefazolin but resistant to gentamicin, kanamycin, sulfamethoxazole+trimethoprim, amikacin, neomycin streptomycin, colistin. In effect on disinfectants, C chauvoei was inhibited completely to growth in mercuric bichloride ($HgCl_2$), harasol(sodium hypochloride 4-6%), long-life(high boiling tar acids et al), and phenol($C_6$$H_5$OH), but growth in all-stop(didecyl dimethyl ammonium chloride 10%), powercide(potassium monopersulphate 50% et al), antec vercon-s(triple salt 50% et al), and taego-51(6-alkyl-2.6-diaza-hexane-carbonic acid-1ㆍHCl et al). The effect of disinfectant was excellent in mercuric bichloride and harasol.

  • One hundred and sixty one Escherichia coli strains isolated from 24 swine (11 swine fed with feedstuffs containing 7.5mg/kg of tetracycline and 13 swine not received antibiotic) were studied for the drug resistance and distribution of R factors. About 42 per cent of E. coli strains isolated from pigs of a herd fed with tetracycline (TC)-containing feeds were resistant to TC, streptomycin(SM), sulfisomidine(SA), ampicillin (AP) and kanamycin (KM), alone or in combination thereof, but none of the swine not receiving antibiotic containing feedstuffs excreted E. coli resistant to these drugs, Among resistant strains, 18.2% were found to be singly resistant to TC, whereas 81.8% were resistant to two or more antibiotics. The most common pattern was the triple resistant to TC, SM and SA(30.3%), and follolwed by double ones to TC and SM(24.2%). About one half of resistant strains carried R factors which were tranferable to the recipients by conjugation. In spite of feeding with feedstuffs containing only TC, high incidences of multiple resistance and R factors were observed in the E. coli isolated from these swine.

Pattern of antimicrobial resistance and biochemical characteristics of Salmonella Typhimurium isolated from diseased pigs in Gyeongbuk province (경북지방 환돈에서 분리한 Salmonella Typhimurium의 생화학적 성상 및 약제내성 패턴)

  • Salmonella Typhimurium is a virulent pathogen for human and animal. We studied serotypes, biochemical characteristics, and antimicrobial resistance of S. Typhimurium isolated from diseased pigs in Gyeongbuk province over 1998 to 2008. One hundred sixteen isolates were identified as S. Typhimurium by biochemical characteristics and serotypes from 90 farms. The biochemical characteristics of S. Typhimurium isolates was production of $H_2S$, indole-negative, fermentation of mannitol, dulcitol, sorbitol, inositol, rhamnose, and maltose, and ornithine decarboxylase. At antimicrobial susceptibility test, the majority of isolates were highly susceptible to amoxicillin/clavulanic acid, cefepime, ciprofloxacin, while were highly resistant streptomycin, cephalothin, enrofloxacin, nalidixic acid, apramycin, chloramphenicol, tetracycline. The isolates were divided into 65 resistant patterns and 47 of the isolates were shown as a DT104 ASSSuT resistant phenotype.

Isolation and Characterization of Antibiotic and Heavy Metal-Resistant Pseudomonas aeruginosa from Different Polluted Waters in Sohag District, Egypt

  • Different polluted water samples were collected from a wastewater treatment plant, agricultural drainage canals, the River Nile, and irrigation canals. The samples were examined for the enumeration of Pseudomonas aeruginosa in the Sohag area, Egypt over a period of one year. A total of 240 isolates were collected and tested for their resistance to 12 common antibiotics and 6 heavy metals. The isolates were found to be less resistant to norfloxacin(1.7%), ofloxacin(4.6%), amikacin(9.6%), tobramycin (10.4), carbenicillin (15.4), and gentamycin (41.3%), yet more sensitive to rifampicin (75%), kanamycin (89.6%), ampicillin (90.8%), chloramphenicol (91.7%), streptomycin (92.9%), and tetracyclin(96.3%). In contrast, 7.1%, 12.9%, 25.4%, and 53.7% of the isolates were resistant to lead, cadmium, mercury, and zinc, respectively. None of the isolates had developed a resistance to silver or molybdenum. The high frequency of metal-antibiotic double resistance existed between lead and amikacin (56.5%), cadmium and ofloxacin (72.7%), zinc and norfloxacin (100%), and mercury and carbenicillin (94.6%). The high occurrence of antibiotic-resistant bacteria in natural water could be related to the widespread use of antibiotics, with possible public health hazard.

Antimicrobial resistance and distribution of tetracycline resistance determinants in Escherichia coli isolated from aquatic birds (수생조류에서 분리한 대장균의 항균제 내성 및 Tetracycline 내성인자의 분포)

  • One hundred and sixty nine Escherichia (E.) coli strains isolated from fecal samples of aquatic birds in Geumho river basin and Dalseong park were tested by agar dilution method to determine their susceptibility patterns to 14 antimicrobial agents. The distribution of tetracycline resistance determinants (tetA, tetB, tetC, tetD and tetE) were also examined by PCR in 76 tetracycline-resistant ($TC^r$) E. coli isolates. The high resistance was observed in tetracycline, cephalothin and ampicillin (45.0~36.7%). Resistance of E. coli isolates derived from Dalseong park to tetracycline, cephalothin, ampicillin and streptomycin (65.7~44.8%) were significantly higher than those isolated from Geumho river basin (31.4~14.7%). About seventy percent (70.4%) of the strains isolated were resistant to one or more drugs tested. Thirty (39.5%) of 76 $TC^r$ E. coli isolates which were resistant to one or more drugs transferred all or a part of their resistance patterns to the recipient strain of E.coli J53 by conjugation. All of $TC^r$ E. coli isolates contained at least one or more of 5 tet genes examined. The most common genes found in these isolates were tetA (60.6%) and followed by tetB (7.9%) and tetC (1.3%). However, tetD and tetE were not found in any of the isolates tested. Twenty one (27.6%) of $TC^r$ E. coli isolates had two determinants, tetA/tetB (20 strains), tetA/tetC (1 strain). And two strains (2.6%) contained three determinants (tetA/tetB/tetC).

  • We investigated patterns of antimicrobial resistance in Escherichia coli isolated from the water discharged from inland pollution sources in the Hansan-Geojeman and Jaranman-Saryangdo areas of Korea. A total of 217 strains of E. coli were isolated from 23 point-sources. These strains were tested for their susceptibility to 16 antimicrobial agents used in Korea for medical or veterinary therapy. The highest level of antibiotic resistance among the isolated strains was to tetracycline 10.6%, followed by ampicillin (3.2%), nalidixic acid (2.8%), rifampin (1.8%), trimethoprim (1.8%), trimethoprim/sulfamethoxazole (1.8%), chloramphenicol (1.4%), streptomycin (1.4%), cephalothin (0.5%) and gentamicin (0.5%). Resistance to at least one antimicrobial agent was present in 17.1% of the E. coli isolates. Only four of the isolated strains of E. coli showed multiple antibiotic resistance, which is defined as resistance to more than four antibiotics.

  • On hundred and forty stains of shigella cultures isolated from the twelve hygiene laboratories of cities and provincial general hospital laboratories in 1983 were tested for their resistance to thirteen antimicrobial drugs and their R-plasmid transfer. Antimicrobial drugs were used amikacin, ampicillin, cephalothin, chloramphenicol, gentamicin, kanamycin, nalidixic acid, rifampicin, streptamycin, tetracycline, tobramycin, cefoperazone and piperacillin. All strains were resistant to one or more of thirteen antimicrobial drugs but 94.3% were susceptible to amikacin, gentamicin and tobramycin of total isolated. The most strains commonly found resistance was to chloramphenicol (94%) followed by streptamycin (93%), tetracyline (92%) piperacillin (90%) ampicillin (83%), cefoperazone (42%), nalidixic acid (14%), cephalothin (17%), rifampicin (22%) and kanamycin (6%), sixty percent of strains among 140 were resistance to ampicillin, chloramphenicol, streptomycin, tetracycline at the same time. The transfer of drug resistance by conjugation was tested and ninety four strains (94.3%) were resistant to one or more drugs were found to transfer their drug resistance of E. coli. percentage of transfer frequency by conjugation was one strains (54%), the transfer frequency of drug resistance varied by donor strains and recipients, but not by selecting drugs. Resistance to nalidixic acid was not transferred by conjugation to recipients. Percentage of plasmid curing after the treatment of acriflavine, acridine orange was about 8%. Among strains cured two strains were tested compare original strains with them in biochemical properties in arginine dihydrolase and arabinose fermentation reaction. It was found to growth curves of No.2 shigella flexneri, serotype 1b, and its derivatives cured with acriflavine in $M{\ddot{u}}ller$ Hinton broth medium (pH 7.4, $38^{\circ}C$) by temperature Gradient Biophoto Recorder TN-1120 (Tokyo, Japan).

Analysis of Microorganisms and Antibiotic Resistance in Organic Dairy Farm (유기낙농가 사육환경 중 미생물 및 항생제 내성 분석)

  • Recently, antimicrobial resistance of microorganisms has been a major concern of its relation to food safety and national health, therefore, customer's needs to organic animal food is still increasing. In this study, we reviewed the usage of antimicrobials in animal farms and antimicrobial resistance of microorganisms isolated from organic dairy farm environments. The isolates from dairy farms were Acinetobacter sp., A. lwoffi, A. johnsonii, A. towneri, Aerococcus viridans, Aeromonas media, A. veronii, Bacillus pseudofirmus, B. pumilus, B. licheniformis, Corynebacterium glutamicum, Escherichia coli, Enterococcus faecium, Lysinibacillus fusiformis, Paenibacillus illinoisensis, Staphylococcus epidermidis, S. hominis, Streptococcus equinus, S. lutetiensis, and Saccharomyces cerevisiae. Indicator microorganisms isolated from organic dairy farms were tested for susceptibility to 20 types of antimicrobials. E. coli (ATCC 25922) isolated from dairy farm fence showed resistance to 8 types of antimicrobials, such as oxacillin, penicillin, vancomycin, etc., and E. faecium isolated from feces showed resistance to 9 types of antimicrobials, such as cephalothin, oxacillin, streptomycin, etc., respectively. However, these results showed less antimicrobial resistance compare with customary dairy farm.

Characteristics and Comparison of Antimicrobial Resistance Patterns of Escherichia coli Isolated from Fish Farms in the South Coast of Korea in 2018-2019 (2018-2019년 남해안 어류양식장에서 분리한 대장균(Escherichia coli)의 항생제 내성 특성 및 비교)

  • Antimicrobial resistance patterns of Escherichia coli strains isolated from 30 seawater samples and 30 Korean rockfish Sebastes schlegelii collected from fish farms off the South Coast of Korea in 2018-2019, were investigated. The isolated E. coli (154 strains) showed the highest antimicrobial resistance to tetracycline (44.8%) followed by chloramphenicol (38.3%), streptomycin (37.6%), and trimethoprim/sulfamethoxazole (35.7%). The antimicrobial resistance rate was higher in E. coli strains isolated from Korean rockfish than those from seawater. Multiple antibiotic resistance (MAR) against at least three antimicrobials (MAR index: ≥0.2) was observed in 35.6% and 79.2% of the seawater and Korean rockfish isolates, respectively. In conclusion, continuous monitoring is required to identify changes in antimicrobial resistance and their correlations in the aquatic environment and products.
