  • The key parameter that affects the consolidation process of soil is the coefficient of permeability. The common assumption in the consolidation analysis is that the coefficient of permeability is porosity-dependent. However, various authors suggest that the strain-dependency of the coefficient of permeability should also be taken into account. In this paper, we present results of experimental and numerical analyses, with an aim to determine the strain-dependency of the coefficient of permeability. We present in detail both the experimental procedure and the finite element formulation of the two-dimensional axisymmetric numerical model of the oedometer test (standard and modified). We perform a set of experimental standard and modified oedometer tests. We use these experimental results to validate our numerical model and to define the model input parameter. Finally, by combining the experimental and numerical results, we propose the expression for the strain-dependent coefficient of permeability.

  • The strain data acquired from structural health monitoring (SHM) systems play an important role in the state monitoring and damage identification of bridges. Due to the environmental complexity of civil structures, a better understanding of the actual strain data will help filling the gap between theoretical/laboratorial results and practical application. In the study, the multi-scale features of strain response are first revealed after abundant investigations on the actual data from two typical long-span bridges. Results show that, strain types at the three typical temporal scales of $10^5$, $10^2$ and $10^0$ sec are caused by temperature change, trains and heavy trucks, and have their respective cut-off frequency in the order of $10^{-2}$, $10^{-1}$ and $10^0$ Hz. Multi-resolution analysis and wavelet shrinkage are applied for separating and extracting these strain types. During the above process, two methods for determining thresholds are introduced. The excellent ability of wavelet transform on simultaneously time-frequency analysis leads to an effective information extraction. After extraction, the strain data will be compressed at an attractive ratio. This research may contribute to a further understanding of actual strain data of long-span bridges; also, the proposed extracting methodology is applicable on actual SHM systems.

  • Heat dissipation technology using flow resonant phenomenon is a kind of new concept in heat transfer area. A vibration exciter is needed to generate air turbulence which has the natural shedding frequency of heat system. A mechanical vibrating device for the air flow oscillation is introduced, which is driven by a moving coil actuator. An analytical dynamic model for this mechanical vibration exciter is presented and its' validity is verified by the comparison with experimental data. Values of some unknown system parameters in the analytic model are estimated through the system identification approach. Based on this mathematical model, the vibration exciter using strain displacement estimator is developed. And in the experiment, the feedback control is used. During the experimental verification phase, it turns out the high modal resonant characteristics of vibrating plate are the major barrier against obtaining a high bandwidth vibration exciter.

  • The aim of the present research program was to isolate a strain of actinomycetes producing antifungal substance. Soil samples were collected from various sites in Korea and a number of actinomycetes were isolated from the soil samples by applying selective agar for actinomycetes. Among isolates, a strain (NA -52) producing antifungal substance against Pyricularia oryzae was selected. Chemotaxonomic and numerical identification were carried out for the isolate. Fifty taxonomic unit characters were tested and the data were analyzed numerically using TAXON program. The isolate was identified as a synonym of streptomyces diastaticus belong to cluster No. 19 (Streptomyces diastaticus). But it showed a low similarity to S. diastaticus in simple matching coefficients, hence it was considered as one new species in Streptomyces.

  • Damage detection based on measured vibration data has received intensive studies recently. Frequently, the damage to a structure may be reflected by a change of some system parameters, such as a degradation of the stiffness. In this paper, we apply a method to nondestructively locate and estimate the severity of damage in corrosion pipeline for which a few natural frequencies or mode shapes are available. The method is based on the strain modal sensitivity ratio (SMSR) and the orthogonality conditions sensitivities (OCS) applied to vibration features identified during the monitoring of the pipeline. The advantage of these methods is that it only requires measuring few modal parameters. The SMSR-based and OCS-based damage detection methods are illustrated using computer-simulated and laboratory testing data. The results show that the current method provides a precise indication of both the location and the extent of corrosion pipeline.

  • This paper presents a theoretical and experimental study on the damage identification of structures. In civil and aerospace, significant work has been done in the area of detecting damage in structures by using changes in the dynamic response of the structure. In this paper a method based on the changes in the strain energy of the structure will be discussed. To evaluate the effectiveness of the method it will be applied to both beam and LNG(liquefied natural gas) carrier.

  • Six strains of an obligate predatory bdellovibrio isolate that preys on Burkholderia glumae in rice paddy field water and rhizosphere soil, were identified and characterized. The numbers of Bdellovibrio cells varied from $3.2{\times}10^3$ to $9.2{\times}10^3$ plaque-forming unit/g after enrichment in cells of B. glumae. Prey range tests with six Bdellovibrio strains and 17 prey strains of rice-pathogenic, antibiosis-related, or nitrogen-fixing bacteria resulted in unique predation patterns in related prey cells. Strain BG282 had the widest prey range on 7 plant pathogenic bacteria among the 17 prey strains tested. However, no predation occurred with strains of Azospirillum brasilense, Paenibacillus polymyxa, Pseudomonas fluorescens, P. putida, and Serratia marcescens that are associated with antibiosis or nitrogen fixation in the rice ecosystem. Identification was confirmed by the presence of typical bdelloplast in the prey cells of B. glumae and by a PCR assay using B. bacteriovorus-specific primers. Furthermore, 16S rDNA sequencing of the six bdellovibrio strains showed a homology range of 97.2% to 99.2% to the type strain of B. bacteriovorus.

  • The purpose of this study is to develop species-specific DNA probes and polymerase chain reaction (PCR) primers for detection and identification of Prevotella nigrescens (P. nigrescens) 9336. This study procedure includes (1) whole-genomic DNA extraction of P. nigrescens 9336 (2) construction of the genomic DNA library, (3) screening of strain-specific DNA probe by reverse Dot Hybridization method, (4) confirmation of strain-specific DNA probe by Southern blot analysis, (5) determination of nucleotide sequences of strain-specific DNA probe. Thirty-five restriction fragments of P. nigrescens 9336 genomic DNA digested with the Hind III were obtained. Reverse dot hybridization and Southern blot analysis data showed that three of them, Pn10, Pn23, and Pn35, could be P. nigrescens 9336-specific DNA probes. These data indicated that these DNA probes could be useful in detection and identification of the P. nigrescens 9336.
