Effect of Planting Time on Seed Production of Vegetable Soybean at Different Locations (풋콩 재배지에 따른 파종시기가 종자생육에 미치는 영향)

  • This investigation was conducted to determine the effect of the optimum planting date and place for good seed production on growth variation, grain yields, different of variety, infection of seed by fungi, and seed germination and vigor after room storage. Early maturity Korean soybean variety, Keunolkong, and introduced vegetable soybean variety, Fukura-edamame, were planted at field of YAES. and high cool land of Sajapyong in Milyang(altitude, 850m) on May 15 (early), June 15 (mid.), and July 15 (late) in 1991. The emergence and vegetative period gradually increased in the early planting date. The grain yield, seed weight, pod number, healthy grain yield also increased when Keunolkong was planted on early date. The healthy seed rate, Keunolkong increased in the early planting date, whereas that of Fukura also increased in the late planting date. Infection ratio of grain to phomopsis seed decay (Phomopsis spp. ' Diaporthe phaseolorum) in Fukura steadily increased in the early planting date. Infection ratio of grain to purple seed stain (Cercospora kikuchii) generally increased in the mid and late planting date. The seed germination and seedling vigor after room storage from five to six months gradually increased in the late planting date. And seed germination of Fukura rapidly decreased in the early planting date. Therefore, the optimum planting date for good seed production in early maturity vegetable soybean was June 15 in terms of harvesting time avoid a high temprature and humidity.

Impact of Environmental Factors on in vitro Interactions and Niche Overlap between Aspergillus ochraceus and other Storage Fungi (Aspergillus ochraceus와 다른 저장균간의 in vitro 상호작용 및 Niche Overlap에 미치는 환경요인의 영향)

  • The effect of water activity ($a_w,\;0.9{\sim}0.995$) and temperature ($18{\sim}30^{\circ}$C) on in vitro growth and interactions between ochratoxin-producing Aspergillus ochraceus and six other fungi (Alternaria alternata, Aspergillus candidus, A. flavus, A. niger, Eurotium amstelodami, E. rubrum) isolated from maize grain were investigated. A. ochraceus and each six other species were paired and their interactions given a numerical score to obtain an index of dominance ($I_D$) for each species. Generally A. ochraceus was very competitive and dominant against other fungi. It was, however, dominanted by Alternaria alternata and A. niger at high $a_w\;(0.995\;a_w)$, and mutually antagonistic when paired with E. amstelodami and E. rubrum at low $a_w\;(0.9\;a_w)$. The growth rates of each species were also calculated under the same range of environmental conditions. They were markedly influenced by aw and temperature. At high temperature ($30^{\circ}C$), A. ochraceus grew most rapidly under slightly drier conditions ($0.95\;a_w$), while A. alternata, A. flavus and A. niger did at high water availability level ($0.995\;a_w$). At $18^{\circ}C\;and\;25^{\circ}C$, and high $a_w$ level ($0.995\;a_w$), A. alternata grew fastest, while A. candidus, E. amstelodami and E. rubrum grew very slowly. Using Biolog plates the effect of $a_w$ and temperature on utilization patterns of carbon sources in maize was evaluated. The niche overlap index (NOI) relative to A. ochraceus was determined and compared with that of each interacting species. Under high water available condition ($0.995\;a_w$). the NOI of A. ochraceus was often >0.9, indicative of the coexistence with other interacting species. However, against E. amstelodami and E. rubrum at $18^{\circ}C$, the species had NOI <0.8, indicative of occupation of different niches. At low $a_w\;(0.95\;a_w)$, NOI for A. ochraceus was <0.8 when paired with A. alternata and A. niger also suggested the occupation of different niches.

  • Twenty seven specimens of deteriolated storage rice were collected all over Korea. Types of deteriolation were classified according to color outlooks, and for 48 grains of each specimen the causative storage microorganisms were isolated and identified. The following results were observed; 1. 27 specimens of deteriolated rice were classified according to color outlooks into 7 types: reddish yellow 1, light reddish yellow 3, light grayish yellow 4, light red 6, dark gray 7, light gray 3, and rice weevil type 3. 2. The most common storage microorganisms group which infected deteriolated Korean rice were Aspergillus glaucus group, especially species of A. amstelodami, A. chevalieri, A. montevidensis, and A. ruber, which were frequently associated with Penicillium, Brevibactereum, and Bacillus. 3. As a specific case sometimes a specimen of deteriolated rice was infected chiefly by one deminant species of microorganism. Five cases were observed: that is, by P. islandicum, P. lanosum, B. lentus, Pseudomonas cohaerans, Brev. lipolyticum. 4. No definite relationship was observed between color outlook types and the deteriolation causing microorganisms. Only the heavily infected rice by Penicillium islandicum expressed discernible reddish yellow color indicative of the infection by this mold. 5. Mycotoxin problem could be noted in one specimen of deteriolated imported rice heavily infected by P. islandicum. Other mycotoxin producing fungi, A. flavus, A. ochraceus, A. fumigatus were also detected, but their growth frequencies were so low that it might not be serious problem.

Seed Germination, Seedling Growth, and Herbicidal Responses of Bidens tripartita L. (가막사리 (Bidens tripartita L.)의 종자발아, 유묘의 생장 및 제초제반응)

  • This study was conducted to investigate seed germination, seedling growth, and responses to herbicides of Bidens tripartita L. When the field-collected seeds were stored under a dry-room temperature, dry-low temperature, wet-low temperature, or dry-high temperature condition, no seeds were germinated in a growth chamber with 14 hr photoperiod up to 35 days after the storage. Exceptionally, however, some seeds stored under a wet-room temperature condition were germinated after 25 days of the storage. This might be due to the fact that the seed coats were damaged by fungi which developed during the storage. Seeds stored under a wet-low temperature condition (stratification) began to be germinated after 3 months of the storage and the germination rate increased with a prolonged stratification. Almost all seeds were germinated after 9 months of the stratification. These results suggest that the dormancy of B. tripartita L. seeds relate to the seed coat and thus several attempts were made to induce seed germination through damaging or weakening the seed coat. Freezing($-20^{\circ}C$), drying($100^{\circ}C$), or swelling($40^{\circ}C$) of the seeds was not effective to induce the germination. Treatments of concentrated sulfuric acid, $KNO_3$, or gibberellin to the seeds had no effect on inducing the germination. However, ethrel had a stimulatory effect on the germination of the seeds with an optimum concentration of 250ppm. A seed cutting was also effective to induce the germination, but seedlings from the seeds had cutted cotyledons. Germination of the stratified seeds varied with the temperature condition to which they were subjected, but not with light. The germination rate was the highest at 35 - $40^{\circ}C$. Although the seeds were not able to germinate under a submerged condition, seedlings after 2-leaf-stage exhibited better growth under a submerged or a subirrigated condition than under an upland condition. Among the herbicides tested, pyrazosulfuton-ethyl, linuron, and bentazone were found to be effective for controlling B. tripartita L., having more herbicidal effect with an earlier application.

Monitoring of Aflatoxins on Commercial Herbal Medicines (유통생약의 아플라톡신 모니터링)

  • This study was performed to investigate contamination levels of aflatoxins, the secondary metabolites produced by fungi Aspergillus flavus and A. parasiticus, in herbal medicine. Herbs is susceptible to these fungi infections through its growth harvest, transport and storage. This study determine the aflatoxin $B_1$, $B_2$, $G_1$ and $G_2$ levels by HPLC-florescence detector coupled with photochemical enhancement in 558 samples herbal medicine distributed in Korea and China. Also, We checked a transfer ratio of aflatoxins from raw herbal medicines to herbal medicine extract. Hot water extraction of herbal medicines was prepared by air pressure and high pressure condition. The analytical method for aflatoxins was validated in this method. In results recoveries of the analytical method were ranged from 67.4% to 96.2% and, limits of detection and quantitation for aflatoxins were $0.015{\sim}0.138\;{\mu}g/kg$ and $0.046{\sim}0.418\;{\mu}g/kg$, respectively. According to the results of monitoring on aflatoxins in herbal medicine, aflatoxins 1.7 ug/kg $B_1$ and 0.9 ug/kg $G_1$ were detected in only one sample of Strychni Ignatii Semen, and 0.8 ug/kg $G_1$ in Strychni Semen. About 13.6~51.3% of aflatoxins were transferred to hot water extract. Although the detected levels are under the permitted levels for aflatoxins in herbal medicine, these amounts should be considered in regard to overall daily exposure to mycotoxins.

Inhibitory Effect of Grapefruit Seed Extract Mixture on the Physiological Function of Botrytis cinerea (식물성 항균소재 처리가 Botrytis cinerea의 생리기능에 미치는 저해효과)

  • Botanical antimicrobial agent-grapefruit seed extract mixture (BAAG) have an unknown compounds which exhibit the antibiotic activities aganist microorganisms including bacteria and fungi. We have examined the effects of BAAG on the physiological function of Botrytis cinerea which was isolated from necrotic lesions of decayed fruits and vegetables such as cucumbers, grapes, tomatoes, and red peppers during storage. In the results of enzymatic activities related to the energetic metabolism there was no inhibitory effect of BAAG on the activities of several enzymes in vitro including glucose 6-phosphate dehydrogenase and malate dehydrogenase, while there was inhibitory effect of BAAG on the activities of hexokinase and succinate dehydrogenase. O-nitrophenyl-$\beta$-D-galactopyranoside(ONPG), the artificial substrate of $\beta$-galactosidase was hydrolyzed in the presence of BAAG, indicating that the membrane was pertubated by the BAAG. From the results we suggested that the antibiotic activity of BAAG is due to the change of membrane permeability of the cell. BAAG was fractionated and purified by silica gel and sephadex column chromatography. Among active fractions two peaks were identified as naringin and limonin when they were analyzed by by NMR and Fast atomic bombardment.

  • 62 specimens of deteriorated rice which were collected all over Korea, were classified according to their color outlooks, and the causitive microorganisms were isolated and identified. The following results were observed; 1. 62 specimens of deteriorated rice were classified according to color outlooks into 7 types; reddish yellow, light reddish yellow, light gray yellow, light reddish yellow, light gray yellow, light red, ligt gray, dark gray and rice weevil type. 2. The isolated microorganisms from 62 specimens of deteriorated rice were 44 species, 5 genera of molds, 1 species of yeast, and 14 species, 4 genera of bacteria. 3. The frequently observed microorganisms which caused the deterioration were Asp. glaucus group, Asp. oryzae, Asp. candidus, and Asp. versicolor. Among bacteria, Bacillus was dominant. 4. The relationship between color outlook types and the deterioration causing microrrganisms was not definite, but Pen. islandicum from reddish yellow, Asp. candidus and Asp. clavatus from light reddish yellow, Asp. glaucus group, Asp. candidus, Asp. versicolor and Asp. fumigatus from light red, Asp. oryzae from light gray yellow, and Asp. glaucus group and Pencillium species from light gray and dark gray were chiefly isolated. 5. As mycotoxin producing fungi, Asp. fumigatus, Pen. citrinum, Pen. islandicum, Asp. flavus, and Asp. ochraceus were detected, but their growth frequencies were not very high to be problems except Pen, islandicum which infected an imported rice sample heavily.

  • In this study, there has been investigated the simulation of irradiation dose using Monte Carlo methodology and experimental substantiation for the biological control of wooden cultural property. In the evaluation of fungal contamination on wooden cultural property, Dongyae, from exhibition storage, Aureobasidium pullulans was mainly identified. But these microorganisms were completely inactivated by 20 kGy gamma irradiation. For dosimetry simulation of Dongyae, Monte Carlo methodology with MCNP was used. The real dosimetry was measured using alanin dosimeters (at 7 different points on the front plan and 7 points on the back plan). Simulated and experimental results are compared and good agreement is observed. These result shows that irradiation can offer biologic control of wooden cultural property by optimal irradiation dose through high penetration power and Monte Carlo simulation.

Prevention from microbial post-harvest injury of fruits and vegetables by using grapefruit seed extract, a natural antimicrobial agent (천연항균제처리에 의한 과채류의 선도유지 및 병해방지에 관한 연구 -저장중 병리적 장해 방지를 중심으로-)

  • In order to retain the freshness of fruits and vegetables and to reduce the rate of disease damage, grafruit seed extract (GPSE), natural microorganism control agent, was applied during the preservation process of fresh fruits and vegetables. GFSE showed an effective inhibitory action against plant putrefactive bacteria and fungi which were involved in the decay of fruits and vegetables. Minimal inhibitory concentrations for GFSE against the microbes were in the range of 50 to 2,000 ppm. Direct observation of microbial cells and spores using electron microscopy showed their function was destroyed by the treatment of the dilute solutions of GFSE. Fresh Welsh onions, onions and red peppers treated with GFSE and stored in polyethylene film (0.1 mm) retained better quality in color and texture than the non-treated control. GFSE was efficient in controlling the germination of potatoes. It was observed that GFSE would reduce disease damages and have bactericidal and fungicidal properties during the storage of such fruits and vegetables as zucchinis, cucumbers, tomatoes and mandarin oranges.

Occurrence and control of postharvest diseases of apple (사과저장병의 발생 및 방제)

  • The occurrence of postharvest disease of apple was surveyed from 1995 to 1997 in the major apple-producing area in Korea. Ten genera including Alternaria spp., Botryosphaeria dothidea, Botrytis cinerea, Fusarium spp., and Penicillium spp. were isolated from the decayed apple fruits. Of these, B. cinerea, Penicillium spp. and Fusarium spp. were frequently isolated and were highly pathogenic to apple fruits. Optimum temperature of mycelial growth for Penicillium spp. ranged from 10 to $30^{\circ}C$ and that of mycelial growth for B. cinerea and Alternaria spp. ranged from 5 to $30^{\circ}C$. Optimum temperature of sporulation of Penicillium spp. and Alternaria spp. ranged $15{\sim}25^{\circ}C$ and $10{\sim}20^{\circ}C$, respectively. Occurrence of postharvest disease of apple increased in neglecting selection of wounded fruits before storing apples. Most of these fungi causing postharvest diseases such as Penicillium spp, Botrytis cinerea and Alternaria spp. were isolated from healthy fruits sampled at apple orchard. These results suggested that posthavest diseases of apple were originated from apple fruits contaminated from apple orchard and occurred during storage. In addition, five fungicides including prochloraz EC, fenari EC, captan WP, benomyl WP and folpet WP suppressed posthavest diseases when they sprayed on apple fruits at 30 days before harvesting.
