Effect of Different Irrigation Period on Photosynthesis and Growth Performances of Containerized Seedling of Eucalyptus pellita and Acacia mangium (관수 주기가 Eucalyptus pellita와 Acacia mangium 용기묘의 광합성 및 생장에 미치는 영향)

  • The objective of this study was to find optimal water condition of containerized seedling production of two tropical species for high seedling quality. This study was conducted to investigate photosynthesis, water use efficiency(WUE), Stomatal conductance(gs) and growth performances of containerized seedlings of Eucalyptus pellita and Acacia mangium growing under three different irrigation periods (1 time/1 day, 1 time/2 days and 1 time/3 days). E. pellita showed outstanding photosynthetic capacities at 1time/1day irrigation and A. mangium showed good photosynthetic capacity at all treatments. As irrigation period were shortened, gs of two species increased, while WUE of two species decreased. Root collar diameter and height of two species were the highest at 1 time/1 day irrigation, while the lowest at 1 time/3 days irrigation. As irrigation period were shortened, H/D ratio, biomass and seedling quality index (DQI) of two species increased but T/R ratio of two species showed the opposite tendency. These results showed that 1 time/1 day irrigation is optimal water condition of containerized seedling production of two tropical species and irrigation controlling is very important for growth and seedling quality of containerized seedling.

Effect of Light Transmittance Control on the Growth Status of Aerial Parts during the Growing Season of Panax ginseng (생육시기별 광량조절이 인삼의 지상부 생육에 미치는 영향)

  • This study was conducted to compare the growth status of aerial parts, photosynthesis and microclimate between fixing light transmittance (Control) and changing light transmittance (C.L.T.) during ginseng growing seasons. Control showed 8% light transmittance rate during growing seasons. But C.L.T. showed 18% light transmittance rate during early (April-June) and late growth stage (September-October) and 6% light transmittance rate middle growth stage(July-August). Air temperature, leaking water rate and soil water content of C.L.T. was higher than those of control during early and late growth stage. But Air temperature, leaking water rate and soil water content of C.L.T was lower than those of control during middle growth stage C.L.T. exhibited superiority in survival ratio, stem diameter, stem length, L.A.I. and stem angle compared to control. Chlorophyll content of C.L.T. was lower than that of control but S.L.W., stomatal opening and photosynthetic rates of C.L.T. was higher than those of control. Also Alternaria blight disease and defoliation of C.L.T. was lower than those of control.

A taxonomic study on six section subgenus Vigena Nees of Carex L. (Cyperaceae) in Korea (한국산 괭이사초아속(subgen. Vignea Nees) 6절의 분류 형질에 관한 연구)

  • Morphological characters of C. kobomugi, C. gibba, C. echinata. C. remotiuscula. C. maakii. C. curta, and C. loliacea of genus Carex were rexamined. The epidermal patterns of perigynium, achene and leaf were investigated using by a scanning electron microscope(SEM) and a light microscope(LM). Morphological characters. such as length and width of stem and leaf, sheath, bract, spike, scale, perigynium, beak of perigynium, length of spike peduncle, size, and frequency of stomatal complex of leaf, number of bract, shape of stem transection, scale and apex of scale, beak and base of perigynium, achene, epidermal cell and cell wall of perigynium, achene, leaf epidermal patterns (fundamental epidermal cell and cell wall, silica body, subsidiary cell), hair, papillae, prickle present/absent of perigynium and leaf were useful for the identification of observed seven taxa. Examined six taxa of section Macrocephalae, Gibbae, Stellulatae, Elongatae, Ovales, and Heleonates were distint from each other with respect of length and width stem, leaf, bract, perigynium, perigynium beak, lenght of spike peduncle, perigynium, and leaf. A key based on data was presented here.

Effects of the Light Source of LEDs on the Physiological and Flowering Response of Endangered Plant Silene capitata Kom. (LED광질에 따른 분홍장구채(Silene capitata Kom.)의 생리 및 개화 반응)

  • We examed physiological and flowering response of S. capitata, the endangered plant in Korea, under LED light conditions in plant factory to cultivate artificially for conservation. We cultivated S. capitata and measured its physiological responses and the number of flowers under red, blue, white, red+far-red mixed, red+blue mixed, and red+blue+white mixed light. The results showed that its photosynthetic rate and chlorophyll content were recorded relatively high in red+blue+white and red+blue mixed light respectively. Transpiration rate and stomatal conductance appeared relatively high in the white single light while water use efficiency was no difference. Photochemical efficiency of photochemical photosystem II by minimum and maximum chlorophyll fluorescence was the highest in the red+blue+white mixed light condition than other ones. The number of flowers of S. capitata was at its peak under the red light or red+far-red mixed light. Therefore, we conclude that the most efficient way to grow for flowering of S. capitata is to provide red light or red+far-red mixed light in the plant factory.

Physiological Response and Cadmium Accumulation of MuS1 Transgenic Tobacco Exposed to High Concentration of Cd in Soil: Implication to Phytoremediation of Metal Contaminated Soil (토양 중 고농도 카드뮴에 노출된 MuS1 형질전환 담배 (Nicotiana tabacum cv. Xanthi)의 생리적 반응 및 카드뮴 축적: 식물학적 오염토양정화를 위한 형질전환 식물 탐색)

  • The objective of this study was to understand the physiological response and cadmium accumulation of MuS1 transgenic tobacco exposed to high concentration of Cd in soil. For this, a pot experiment was carried out in a greenhouse for a month, with two lines of MuS1 transgenic tobaccos (S4 and S6) and non-transgenic tobacco cultivated in the soils spiked at three different Cd concentrations (0, 60 and 180 mg $kg^{-1}$). Both transgenic and non-transgenic tobacco showed visible toxic symptoms such as chlorosis and leaf roll as treated concentration increased. The net photosynthetic rates of MuS1 plants (S4 and S6) exposed at 180 mg $kg^{-1}$ Cd were 6.3 and $7.7{\mu}mol\;m^{-2}s^{-1}$, being higher than those of the non-transgenic plant ($4.8{\mu}mol\;m^{-2}s^{-1}$). Values of stomatal conductance of MuS1 transgenic plants (0.05 and 0.008 mmol $H_2O\;m^{-2}s^{-1}$) were also higher than those of non-transgenic plant (0.03 mmol $H_2O\;m^{-2}s^{-1}$). In addition, fresh and dry weights of MuS1 transgenic plants were heavier than those of non-transgenic plant. Likewise, MuS1 transgenic plants appeared to be better physiological performance than non-transgenic tobacco when exposed at high concentration of Cd in soil. With regard to metal accumulation, MuS1 transgenic tobaccos accumulated more Cd in their roots than non-transgenic tobacco implying that MuS1 transgenic tobacco is suggested to be used for phytostabilization of heavy metals.

Effect of Uniconazole and Silver Thiosulfate Treatment on Reduction of Ozone Injury in Snap Bean Plants (Uniconazole과 Silver Thiosulfate 처리(處理)가 강남콩의 오존피해(被害) 경감(輕減)에 미치는 효과(效果))

  • Studies were conducted to examine the effects of single or combined treatment of uniconazole [(E)-1-(4-chlorophenyl)-4, 4-dimethyl 2(1, 2,-4-triazol-1-yl)-1-penten-3-ol)] and silver thiosulfate (STS) on reducing ozone injury to snap beans (Phaseolus vulgaris L. 'Strike'). Two weeks after seeding, plants were given a soil drench of uniconazole(XE-1019) solution at concentrations of 0.001, 0.005 and 0.025 mg/pot, and then two days prior to ozone fumigation, 0.3 and 0.6 mM STS containing 0.01% Tween-20 were also sprayed. Uniconazole was effective in providing protection against ozone injury through increase activities of free radical scavengers such as superoxide dismutase (SOD) and peroxidase (POD) as well as the increase of chlorophyll content and stomatal resistance resulted from plant growth retardation. The phytoprotective effects of STS seemed to be related to its properly of blocking the ethylene action and increasing activities of SOD and POD. Even at low concentrations, a combined treatment with uniconazole drench, STS spray significantly reduced ozone injury compared to single application.

Confirmation of Drought Tolerance of Ectopically Expressed AtABF3 Gene in Soybean

  • Soybean transgenic plants with ectopically expressed AtABF3 were produced by Agrobacterium-mediated transformation and investigated the effects of AtABF3 expression on drought and salt tolerance. Stable Agrobacterium-mediated soybean transformation was carried based on the half-seed method (Paz et al. 2006). The integration of the transgene was confirmed from the genomic DNA of transformed soybean plants using PCR and the copy number of transgene was determined by Southern blotting using leaf samples from $T_2$ seedlings. In addition to genomic integration, the expression of the transgenes was analyzed by RT-PCR and most of the transgenic lines expressed the transgenes introduced. The chosen two transgenic lines (line #2 and #9) for further experiment showed the substantial drought stress tolerance by surviving even at the end of the 20-day of drought treatment. And the positive relationship between the levels of AtABF3 gene expression and drought-tolerance was confirmed by qRT-PCR and drought tolerance test. The stronger drought tolerance of transgenic lines seemed to be resulted from physiological changes. Transgenic lines #2 and #9 showed ion leakage at a significantly lower level (P < 0.01) than ${\underline{n}}on-{\underline{t}}ransgenic$ (NT) control. In addition, the chlorophyll contents of the leaves of transgenic lines were significantly higher (P < 0.01). The results indicated that their enhanced drought tolerance was due to the prevention of cell membrane damage and maintenance of chlorophyll content. Water loss by transpiration also slowly proceeded in transgenic plants. In microscopic observation, higher stomata closure was confirmed in transgenic lines. Especially, line #9 had 56% of completely closed stomata whereas only 16% were completely open. In subsequent salt tolerance test, the apparently enhanced salt tolerance of transgenic lines was measured in ion leakage rate and chlorophyll contents. Finally, the agronomic characteristics of ectopically expressed AtABF3 transgenic plants ($T_2$) compared to NT plants under regular watering (every 4 days) or low rate of watering condition (every 10 days) was investigated. When watered regularly, the plant height of drought-tolerant line (#9) was shorter than NT plants. However, under the drought condition, total seed weight of line #9 was significantly higher than in NT plants (P < 0.01). Moreover, the pods of NT plants showed severe withering, and most of the pods failed to set normal seeds. All the evidences in the study clearly suggested that overexpression of the AtABF3 gene conferred drought and salt tolerance in major crop soybean, especially under the growth condition of low watering.

Growth Performance and Photosynthesis of Two Deciduous Hardwood Species under Different Irrigation Period Treatments in a Container Nursery System (시설양묘과정에서 관수 주기 처리에 따른 두 활엽수종의 생장 및 광합성 기구 변화)

  • This study was conducted to investigate growth performance, photosynthesis, water use efficiency (WUE), and stomatal conductance ($g_s$) of container seedlings of Liriodendron tulipifera and Zelkova serrata growing under three different irrigation periods (1 time/1 day, 1 time/2 days and 1 time/3 days) for high seedling quality. The root collar diameter and height of L. tulipifera and Z. serrata seedlings were highest with 1 time/1 day irrigation, whereas they were lowest with 1 time/3 days irrigation. The two species showed low drought tolerance. As irrigation period was shortened, biomass and seedling quality index (SQI) of the two species increased. The ratio of height to root collar diameter (H/D) and the ratio of below to aboveground biomass (T/R) of the two species were lower with 1 time/3 days than at other irrigation periods. L. tulipifera and Z. serrata seedlings showed significantly higher photosynthetic capacity with 1 time/1 day irrigation. As irrigation period was shortened, $g_s$ of two species increased, while their WUE decreased significantly (P<0.05) These results show that 1 time/1 day irrigation provides the most optimal water condition for container seedling production of two species and irrigation controlling is very important for growth and quality of container seedlings.

Growth and Physiological Response in Container of Daphniphyllum macropodum by Shading Level (피음 수준에 따른 굴거리나무의 용기 내 생장 및 생리적 반응)

  • This study was carried out in order to closely examine the growth and the physiological response by shading levels of a container seedling of Daphniphyllum macropodum which is known as the species of having shade tolerance in a warm temperature evergreen broad-leaved tree. The shading treatment was regulated with the shading level of full sunlight, and 35%, 55%, 75% of full sunlight. As a result of surveying the height and the root collar diameter growth by the shading level, the height was the highest with 13.2 cm under 35% of shading. The root collar diameter was the highest with 4.99 mm under full sunlight. The organ-based (leaves, shoot and root) dry mass production and the whole dry mass production were higher with 1.38 g, 0.47 g, 0.87 g, 2.72 g, respectively, in order of 35% shading. As for chlorophyll a, b, and the total chlorophyll content, the highest chlorophyll content was shown under 75% of shading with the relatively highest shading level. The ratio of chlorophyll a to chlorophyll b (a/b) was the highest under 35% of shading. The photosynthetic rate, stomatal conductance and water use efficiency were the highest with $12.3{\mu}molCO_2{\cdot}m^{-2}{\cdot}s^{-1}$, $0.27molH_2O{\cdot}m^{-2}{\cdot}s^{-1}$, $3.53{\mu}molCO_2{\cdot}mmol^{-1}H_2O$, respectively, under 35% of shading. As a result of surveying the whole experiment, D. macropodum grows nicely by maintaining 35% shading.

Physiological Differences of Ilex rotunda and Illicium anisatum under Low Light Intensities (다른 광도에서 생육한 먼나무, 붓순나무의 생리적 차이)

  • We examined seedlings of two species (Ilex rotunda and Illicium anisatum) which have a different level of shade tolerance and raised them under different light regimes (full sunlight and 50% shading). After 12 months, we investigated chlorophyll content (Chl. a, Chl. b and Chl. a+b), photosynthetic systems (photosynthetic rate, light compensation point, dark respiration rate and quantum yield), intercellular $CO_2$ concentration and water use efficiency to show acclimation reaction to different light conditions. Seedlings grown under full sunlight showed lower chlorophyll content than those in the shading regime. There was a significant difference between the full sunlight and shade treatments in I. anisatum (shade tolerance species). I. rotunda (intermediate species) showed high photosynthetic rate and water use efficiency over PPFD $1000\;{\mu}mol\;m^{-2}s^{-1}$ to full sunlight. Also, I. anisatum grown under full sunlight showed lower photosynthetic rate and water use efficiency over a range of all PPFD. This result showed that I. rotunda has a more flexible reaction system than that of I. anisatum.
