Behavior of Indoor Organic Pollutants Dissolved into the Ringer's Solution (수액제에 유입되는 실내 유기오염물질의 거동)

  • Recently, hospital acquired infections have an increase interest as a public problems, which are caused of indoor pollutants in hospital. Microorganisms, ethylene oxide, formaldehyde, and anesthetic gases are main hazardous pollutants in hospital. The possible pathways of the infection are a respiratory channel as well as a blood channel. The blood channel is concerned since these pollutants might be dissolved into the Ringer's solution. The objective of this research was to evaluate the removal efficiencies of adsorption trap for formaldehyde and microorganisms as indoor pollutants which permeated into the Ringer's solution. Dissolved formaldehyde in the solution was increased with the injection dose time. The amount of dissolved formaldehyde was 67.5 $\pm$ 9.5% in Ringer's solution when injection dose time was controlled about 7hrs. An adsorption trap was designed for preventing formaldehyde and microorganisms to be permeated into Ringer's solution. The adsorption trap was packed with 0.4g of active carbon (60/80 mesh) in a sterilized plastic tube (7.79 cm length, 0.46 cm i.d.) and both ends were packed with glass wool. Devised infusion set equipped with the adsorption trap showed 99.9% of removal efficiency for formaldehyde. Microorganism numbers detected on sterilized water for injection and 5% dextrose infusion used in the hospital were 2,695 $\times 10^3$ cells/l and 4,190 $\times 10^3$ cells/l, respectively. Removal efficiency by the adsorption trap was 92.3 $\pm$ 8.5% as for microorgnisms.

Effect of Casing Layer on Growth Promotion of the Edible Mushroom Pleurotus ostreatus

  • Various bacteria were isolated from the casing layer soil of the culture bed of P. ostreatus and their role in fruiting body induction of the edible mushroom, P. ostreatus, was investigated. Analysis of the bacterial community isolated from the casing layer soil revealed that the composition of genera and number of cultivable bacteria were different for each sterilizing treatment. Bordetella was predominant in the bulk soil whereas Flavobacterium was predominant after sterilization of the casing layer soil. Fluorescent Pseudomonas was predominant in the non-sterilized casing layer soil. Total number of the bacterial genera in the casing layer soil was higher than that in the bulk soil. In particular, an increase in the fluorescent Pseudomonas population was observed in the non-sterilized casing layer accompanied by induction of fruiting body and enhanced mushroom production yield. The results suggested that specific bacterial populations in the casing layer play an important role in the formation of primodia and the development of basidiome in P. ostreatus.

Effect of Homogenization Pressure on Plasmin Activity and Mechanical Stress-Induced Fat Aggregation of Commercially Sterilized Ultra High Temperature Milk during Storage

  • Commercially sterilized ultra high temperature (UHT) milk was manufactured at different homogenization pressures (20, 25, and 30 MPa), and changes in fat particle size, mechanical stress-induced fat aggregation, plasmin activity, and lipid oxidation were monitored during ambient storage of the UHT milk for up to 16 wk. The particle sizes of milk fat globules were significantly decreased as homogenization pressure increased from 20 to 30 MPa (p<0.05). The presence of mechanical stress-induced fat aggregates in milk produced at 20 MPa was significantly higher than for UHT milk produced at either 25 or 30 MPa. This difference was maintained all throughout the storage. There were no significant differences in plasmin activity, trichloroacetic acid (12%, w/v) soluble peptides, and the extent of lipid oxidation. Based on these results, an increase of homogenization pressure from 20 (the typical homogenization pressure employed in the Korea dairy industry) to 25-30 MPa significantly decreased mechanical stress-induced fat aggregation without affecting susceptibility to lipid oxidation during storage.

Effect of Non-sterilized Makgeolli Consumption on Blood Glucose Level and Survival Rate in Streptozotocin-induced Type I Diabetic Mice (Streptozotocin으로 유도된 제1형 당뇨 생쥐에서 생막걸리 투여가 혈당 및 생존률에 미치는 영향)

  • Makgeolli is a health beneficial food for diabetes compared to other alcoholic beverages. We examined the effect of Makgeolli on blood glucose level and survival rate in a streptozotocin (STZ)-induced diabetic mouse model. We force fed 30 male STZ-induced diabetic ICR mice Makgeolli consisting of 6% alcohol (DM-MAK), 6% ethanol (DM-EtOH), or distilled water (DM-DW) for 4 weeks. In the DM-MAK group, food intake and water intake were higher than those of other groups after 4 weeks. Body weight, however, was not different among the experimental groups. We also found no significant difference in blood glucose level among the experimental groups. In normal ICR mice fed Makgeolli for 1 week, the area of the blood glucose curve was higher than those of other groups fed 6% ethanol, 2% glucose, or distilled water. Survival rates of STZ-induced diabetic mice fed Makgeolli, 6% ethanol, or DW for 4 weeks were 100%, 25%, and 62.5%, respectively. In conclusion, Makgeolli had no beneficial effect on blood glucose in a STZ-induced diabetic mouse model, although their survival rate was high. These results show that Makgeolli has an effect on type 1 diabetes through other mechanisms than blood glucose control.

Validity Periods of Sterilization Products: Focus on Packing Materials and Storage Environments (멸균물품의 유효기간에 관한 연구: 포장재와 보관환경을 중심으로)

  • Purpose: The aim of this study was to provide a scientific basis for shelf life and re-establish the shelf life in a hospital environment by investigating the validity periods of various packaging materials. Methods: We selected six departments to store sterilized items, prepared 482 sterilized items, and stored them for 12 months. Each sample was tested using a microbiological culture in the laboratory every two weeks. When the result was positive, the sample was considered contaminated. The temperature and relative humidity were measured using an automatic control system to investigate the storage environment during the study period. Results: Except for two samples, which were positive in the 22nd and 28th weeks, 480 samples were negative. The temperature and relative humidity of the sample storage area were measured every week. The annual average temperature was 23.6±1.6℃, and the mean relative humidity was 35.1±15.2%. The cabinets used in this study were the two-open and four-closed types. Conclusion: This study confirmed the validity of the expiration date in a hospital environment. Based on the results, the nonwoven fabric remained sterile for more than nine months. No case of contamination of the paper-plastic pouch packaging was observed, owing to the microbial culture for two months.

Preliminary study on the diversity and quantity analysis of oral bacteria according to the sampling methods (구강 세균 채취법에 따른 세균의 다양성과 양 분석을 위한 예비 연구)

  • Objectives: Oral bacterial samples included subgingival, supragingival, and saliva plaques. As the diversity and number of microorganisms deffer depending on the area of the oral cavity and the method used, an appropriate and reliable collection method is important. The present study investigated oral bacterial sampling methods. Methods: Supragingival dental plaque was collected from the buccal and lingual tooth surfaces of study participants using sterilized cotton swabs. Plaques were collected from the subgingival area using a sterilized curette. Bacterial genomic DNA was extracted using MagNA Pure 96 DNA and Viral NA low-volume kits. Real-time polymerase chain reaction (PCR) was performed using the PowerCheckTM Periodontitis Pathogens Multiplex Real-time PCR kit. Results: Aggregatibacter actinomycetemcomitans, Prevotella intermedia, and Fusobacterium nucleatum of the orange complex were not observed in the subgingival biofilms of all study participants. For Porphyromonas. gingivalis, a significant correlation was observed between supragingival, subgingival, and total tooth surface biofilms. Compared to the supragingival and subgingival biofilmss, total tooth surface biofilm exhibited the highest bacterial count when the inswabbing method was used. Conclusions: Based on these findings, the supragingival swab method is recommended for oral bacterial research.

Cultivation of Oyster Mushroom[Pleurotus ostreatus (Jacq. ex Fr.) Kummer] at Medium with Ca(OH)2 Treatment (수산화칼슘[Ca(OH)2] 처리 배지를 이용한 느타리버섯의 재배)

  • This research is aimed to investigate the cultivation method of oyster mushroom[Pleurotus ostreatus (Jacq. ex Fr.) Kummer] at the non-sterilized medium with $Ca(OH)_2$ treatment. Therefore, experiments were carried out to develop non-sterilization method of medium by addition of $Ca(OH)_2$ for omission of heat sterilization progress of medium. General components, minerals and amino acid in Jiri wild type No. 1 (Pleurotus ostreatus) and production cost were analyzed. For the purpose of omission of heat sterilization progress, treatment ratio of $Ca(OH)_2$ (purity 95%) was 5%(w/w) of dry medium. Initial pH of this medium was 11 and then the pH was changed by 9 after the uniform mixing of the medium for half an hour. The various germs occurred 50% and 100% at pH 8 and pH 7 of the non-sterilized medium, respectively. Production of oyster mushroom increased by $2,030\;ton\;ha^{-1}$ when the main raw material used corn pith instead of waste cotton. The time required of mycelium culture was 30 days when hypha was cultured at the non-sterilized medium, and pinhead occurred when 2 or 3 days was passed after the time required of mycelium culture. Occurrence of pinhead was most rapid at the condition of $22{\sim}26^{\circ}C$, 65% humidity and pH $6.5{\sim}7.0$ and required of $22{\sim}28$ days at $70{\sim}80\;mm$ thickness of non-sterilized medium. Ca content in 1st harvest oyster mushroom was higher than that in 2nd harvest one, and its difference was $30.3\;mg\;kg^{-1}$. Amino acid content by stipe thickness of oyster mushroom was ranged from 411.2 to $343.9\;mg\;kg^{-1}$ both in a pileus and a stipe of 1st harvest mushroom, and from 402.4 to $498.2\;mg\;kg^{-1}$ and from 442.6 to $470.4\;mg\;kg^{-1}$ in those of 2nd harvest one, respectively. The results of the present study suggest that the non-sterilization medium by addition of $Ca(OH)_2$ is usable with the cultivation of oyster mushroom.

Comparative Study of Total Acidity Content of Makgeolli with Pre-Treatment and Analysis Methods (전처리과정과 분석방법에 따른 막걸리의 총산 함량 비교)

  • The total acidity (TA) of Makgeolli was affected by suspended solids and $CO_2$ produced during the fermentation process. Nine Makgeollis (four sterilized and five unsterilized Makgeollis) were collected in the market, and their TAs were compared before and after filtration and $CO_2$ removal. TAs of sterilized Makgeollis were 0.379~0.477%, which significantly decreased to 0.167~0.225% after filtration and 0.132~0.170% after $CO_2$ removal (P<0.05). TAs of unsterilized Makgeollis were 0.412~0.467% and decreased to 0.157~0.365% after filtration and 0.143~0.280% after $CO_2$ removal (P<0.05). TAs of Makgeollis were compared by three methods using different indicators. The TAs of sterilized and unsterilized Makgeollis were 0.105~0.123% and 0.105~0.200%, respectively, by bromthymol blue+neutral red (light green), 0.129~0.154% and 0.130~0.255%, respectively, by phenolphthalein (faint pink), and 0.120~0.146% and 0.130~0.232%, respectively, by bromthymol (blue). Nowadays, Makgeolli is commercialized with various distinct colors, and thus it is important to select appropriate indicators for proper titration endpoint identification for TA measurement. The compositions of organic acids profiles varied depending on sterilized or unsterilized Makgeollis, in which oxalic acid (0.108~0.329 mg/mL), malic acid (ND to 0.134 mg/mL), lactic acid (0.127~0.776 mg/mL), and citric acid (ND to 1.159 mg/mL) were found, and lactic acid was in unsterilized more than sterilized Makgeollis.

Pre-treatment conditions on the powder of Tenebrio molitor for using as a novel food ingredient (갈색거저리의 식품 원료화를 위한 분말제조 조건 확립)

  • Although the mealworm larva (Tenebrio molitor) is high protein source, aversion feature of the larva made it difficult for consuming as a food. In this study, we established optimal powder manufacturing process for T. molitor larva for using as a novel food. For this purpose, it should be feed with the bran sterilized by water vapor for 3-5 days, starved without water or food for 3 days, and then the larvae were sterilized before freeze-drying. The sterilized T. molitor was lyophilzed and grinded by a blender. A safety of the powder as a food was validated by evaluation of Raw 264.7 macrophage cytotoxicity using MTS assay. As above results, we propose that optimal powder manufacturing process established in this study can be used in industrial production of T. molitor as a novel food.

On the preparation of new formulation of Diazinon and its residual pattern in submerged soil (새로운 Diazinon입제의 제조 및 담수토양중의 잔류특성에 관하여)

  • New formulation of diazinon as granule preparing with piperonyl butoxide(PBO) and triphenyl phosphate(TPP) known as the inhibitor of monooxygenase(mo) and esterase was tested with the mortality of rice leaf hopper. Also, the mixed formulation of diazinon with tricyclazole, carbofuran and EPN were investigated about the residual pattern of diazinon in submerged soil condition at $30{\pm}2^{\circ}C$. Although the mortality about rice leaf hopper was higher only 10% on new formulation of diazinon than commercial at recommended dose but was 30-60% at half dose. Half life of diazinon in the formulation with 0.1% of PBO+TPP was 4.53 days in sterilized soil and 2.33 days in non-sterilized soil, and it was delayed about 0.74 and 0.45 respectively in contrast with the commercial granule. Also, half life of the new formulation with 1% PBO and PBO+TPP was delayed 0.44(PBO) and 0.65day(PBO+TPP) in non-sterilized soil condition, respectively. Half life of mixed formulation with tricyclazole, carbofuran, EPN or EPN+Carbofuran was delayed 2.61, 1.04, 0.43 or 2.7 days, respectively. As a result, the persistence rate of new formulation and mixed formulation was increased by inhibition of two enzymes affecting to the dgradation of diazinon in submerged soil.
