  • 1. 원유와 시유 40점에서 내열성균으로 생각되는 330균주의 세균이 분리되었다. 2. 현미경적 관찰, 생리적시험, 단백질분해성과 지방분해성으로 보아 Bacillus stearothermophilus 125균주, Bacillus coagulans 69균주, Bacillus subtilis 57균주, Bacillus cereus 76균주, Lactobacillus thermophilus 3균주로 동정되었다. 3. 이들 반이상의 균주들은 skim milk배지에서 $85^{\circ}C$, 15분간 처리에도 생존할 수 있었다.

Isolation and Morphological Characterization of Monilinia sp. KV-27 Associated with Apple Anthracnose of Fuji Apples in Korea

  • This study was undertaken to isolate and to identify a fungal pathogen Monilinia sp. KV-27 associated with apple anthracnose. Rotted Fuji apples were used for the isolation of the fungus. The infected tissues were sterilized with 70% ethanol, washed with sterilized distilled water and were transferred to 50 ml containing potato dextrose broth (PDB) flasks. The peripheral hyphae of the fungal colony which developed from the infected tissues were isolated on to potato dextrose agar (PDA). On PDA plates the fungus grew well at $25^{\circ}C$ and occupied more than half of a 9 cm petri dish within 5 days. The fungal cultures on PDA were used for morphological observation and identification of the fungus. Conidiophores were produced on the gray to whitish sporodochial structures scattered on PDA plates. These conidiophores gave rise to chains of conidia, which were branched and easily detached in water. These structures were dark brown to black and consisted of hyphal masses. Conidia produced on PDA plates were hyline or light colored, lemon shaped or ellipsoidal ($10-13{\times}8.5-11{\mu}m$) in size.

  • Sphingomonas chlorophenolica ATCC39723 was successfully labeled with the gfp (green fluorescent protein) gene inserted into the pcpB gene by homologous recombination. As the gfp recombinant was easily distinguished from other indigenous organisms, the population of gfp recombinant was monitored after being released into the soil microcosms. Their population density dropped from 10$\^$8/ to 10$\^$6/ (cfu/$m\ell$) in the non-sterilized soil microcosms during the first 6 days. Moreover, the gfp recombinant was not detected even at lower dilution rates after a certain time period. The recombinant, however, survived for at least 28 days in the sterilized soil microcosms. Although the gfp recombinant did not degrade pentachlorophenol (PCP), this experiment showed the possibility of using gfp as a monitoring reporter system for S. chlorophenolica ATCC39723 and potentially other species of Sphingomonas.

The structures of the conidia produced on PDA plates were subcylindrical to obovoid, fusoid, tapered and 4 to $6\;{\mu}m$ in size.

  • Comparison of ethylene oxide(E.O) fumigation and gamma irradiation on the sterilization, microbiological, physicochemcial and sensory quality of dried fishery products (shucked shellfish and mussel powder) were investigated. The viable cell count of mesophilic total bacterial was reduced by over 2 to 3 log cycles with irradiation of 3 to 5 kGy, and those samples were completely sterilized by irradiation dose of 7 to 10 kGy. In view of reduction of microorganisms, D10 value of each samples were ranged from 1. 36-1. 46 kGy. fungi, mesophilic spores, acid tolerant bacteria and coliforms were sterilized at 5 to 7 kGy irradiation but E.O. fumigation proved insulfficient to eliminate the mesophilic total bacteria and fungi. In the physicochemical and sensory quality of samples, such as the amino acid, TBA value, TMA-N, color difference and overall acceptability irradiated sample with optimum dose was similar to those of the nonirradiated sample, while E.O. fumigated sample was remarkably deteriorated in the physicochemical and sensory quality of samples.

  • For comparison of mycelial colonization of Phellinus linteus on logs, several techniques of inoculation were tested; sterilized short log inoculation, drilling inoculation and log-end sandwich inoculation. The mycelial colonization of P. linteus on logs was good in the treatment of sterilized short log inoculation, but poor in the traditional methods such as drilling inoculation and log-end sandwich inoculation. The initial mycelial growth and the full mycelial colonization of P. linteus in logs were the best in case of 20 cm logs under the condition of 42% moisture content. Also, the initial mycelial growth of P. linteus was accelerated over 12 hours of sterilization. Burying method of logs after $5{\sim}6$ months of incubation was the best for formation of basidiocarps of P. linteus. The formation of fruiting body of P. linteus was quite good in the cultivation house at $31{\sim}35^{\circ}C$ and over 96% of relative humidity.

콩 흑색뿌리썩음병의 발생과 Propagule의 형성 (The Outbreak and Propagule formation of black root rot caused by Calonectria crotalariae in Korea)

  • The infection rate of soybean black root rot disease caused by Calonectria crotalariae was about $14\%$. The isolated fungi from the infected soybean roots and stems were Calonectria crotalariae, Fusarium solani, F. roseum, Phomopsis sojae, Pythium aphanidermatum, Rhizoctonia solani and Macrophomina sp. Among them, C. crotalariae was the most virulent pathogen under the laboratory conditions. Mycelial growth and microsclerotial formation were good on PSA containing 1000cc of water, 100g of potato and 20g of sugar. Mycelial growth, sporulation and microsclerotial formation were good on sterilized root. Perithecial formation was better in the dark condition than in the light. Survival of macroconidia was not available between $0\~25\%$ soil water content. Microsclerotia and mycelium in infected plant debris were survived for 4 months at to $8\%\;50\%$ soil water content. The plant height, when inoculated with $1.2\%$ inoculum density, reached approximately half of uninoculated plants. Disease severity was much higher at nonsterilized soil than completely sterilized soil. It was determined that the host range of this pathogen includes soybean, peanut, green bean and red bean.

  • 복합레진을 광중합할 경우 광중합기의 광섬유말단은 환자의 구강점막과 직접 접촉하게 되어 광섬유말단의 오염이 불가피하다. 광섬유말단은 Centers for Disease Control and Prevention (CDC)에서 "semicritical category"로 분류되며 가압증기 멸균을 하거나, 화학 용액에 10시간 이상 잠기도록 넣어 멸균을 하거나 최소한 고도의 소독처리를 하도록 요구한다. 현재 광중합기의 광섬유말단을 멸균/소독하는 방법은 가압증기멸균이 가능한 광섬유말단을 사용하여 멸균하는 법, 매 환자마다 glutaraldehyde와 같은 화학용액으로 멸균/소독을 하는 법, 멸균되어 시판되는 일회용 플라스틱 광섬유말단을 사용하는 법, 그리고 투명 랩과 같은 일회용 차단막으로 광섬유말단을 감싸는 방법 등이 있다. 일회용 차단막을 사용할 경우 광섬유말단과 환자의 구강점막의 직접적인 접촉을 막아 비교적 간단하게 교차감염의 위험성을 줄일 수 있다.

인삼의 표면 세척시스템을 개발을 위한 공정처리기술에 관한 연구 (Study on Process Development of Ginseng's Surface Washing System)

  • This study was attempted to develop surface washing-system of ginseng. The effect of sterilization, washing and keeping freshness of ginseng through analyzing unit process were examined to establish optimal condition for washing system. Surface washing method of fresh ginseng used two way and full cone spray type. Sterilization was used at $2^{\circ}C$ water with electrolysis water of 50 and 80 ppm. Ginseng was sterilized with electrolysis water during 30 and 60 s, dehydrated during 1 min and dried during 1min at 30 and $50^{\circ}C$. Hardness of surface-washed ginseng showed good result on 1 min spraying time with 80 ppm electrolysis water at $10^{\circ}C$ storage. Ginseng with 80 ppm electrolysis water was sterilized better with $1.05{\times}103$. There are no changes with 0% on appearance quality at 80 ppm electrolysis.
