A Mycological Survey in Indoor Environments (건물(建物) 환경(環境)의 진균(眞菌) 분포(分布) 조사(調査))

  • This paper was concerned with the investigation on the species of fungi detected in indoor environments. Mycological survey on the wall of buildings and houses in Seoul was performed from December 26, 1983 to February 11, 1984. The collected fungi with the sterilized cotton rod were caltivated and isolated in the Potato-Detrose Agar medium with chloramphenicol. The fungi detected in hospital were Cladosporium sp., Penicillium sp., Alternaria sp., Chaetomium sp. and others. The fungi detected in household were Penicillium sp., Alternaria sp., Aspergillus sp., Cladosporium sp., Rhizopus sp., Trichoderma sp. and others. The kitchen of household was more contaminated by fungi than the living room and others. In hospital, it was much contaminated by the same species of the aboves on the wall of lobby and refectory. As the above results were recognized, the higher humidity and the more poor ventilation, the more species and amount of fungi were contaminated. Further studies on the identification of pathogenic fungi should be continued. The proper regulation for the hygienic maintenance of indoor environment in household, hospital and other public buildings should be recommended as well.

Inactivation of Infectious Microorganisms by Disinfection and Sterilization Processes for Human Amniotic Membrane Grafts (이식을 위한 사람 양막의 소독 및 멸균공정에 의한 감염성 위해인자 불활화 효과)

  • Viral, bacterial, and fungal infection can be transmitted from donor to recipient via transplantation of human amniotic membrane. Therefore human amniotic membrane for transplantation should be disinfected and sterilized before use. The purpose of this study was to examine the efficacy of the disinfection process and sterilization processes used at human tissue bank in the inactivation of viruses, bacteria, and fungi. A variety of experimental model viruses, bacteria, and fungus for human pathogens, including the human immunodeficiency virus type 1 (HIV-1), bovine herpes virus (BHV), bovine viral diarrhoea virus (BVDV), hepatitis A virus (HAV), porcine parvovirus (PPV), Escherichia coli, Bacillus subtilis, and Candida albicans were all selected for this study. Enveloped viruses such as HIV-1, BHV, and BVDV were effectively inactivated to undetectable levels by 70% ethanol treatment, gamma irradiation process, and ethylene oxide (EO) gas sterilization process. Also non-enveloped viruses such as HAV and PPV were effectively inactivated to undetectable levels by gamma irradiation and EO gas treatment. However HAV and PPV showed high resistance to 70% ethanol treatment. E. coli and C. albicans were effectively inactivated to undetectable levels by 70% ethanol treatment, gamma irradiation process, and EO gas treatment. Also B. subtilis was effectively inactivated to undetectable levels by gamma irradiation process and EO gas treatment. However it showed high resistance to 70% ethanol treatment.

Right Thoracotomy for Reoperation of Mitral Valve (우측 개흉을 통한 승모판 재수술)

  • A right thoracotomy was used for the reoperation or mitral valve of 15 patients who had previously undergone a cardiac operation through a median sternotomy. In our experience. this approach provided dn excellent exposure of the nlitral valve and easy cannulations of both cavie with minimal dissection, ilvoiding any damage of cardiac and major vessels during re-sternotomy Arterial cannulation was performed in the ascending aorta in 13 patients And in the femoral artery in 2 patients. In earlier cases, venous cannulation was done in the SVC And IVC through the right atrium and snared. In later cases, this could be done without snaginly of both cavae or by placing a silgle light-angled catheter into the right atrium. Crystalloid cardioplegic solution was infused for myocardial protection. Hypothermia was controlled at 20\ulcorner$25^{\circ}C$. For defibrillation, internal paddles were used In one patient while sterilized external paddles were used in 10 patients. In the remaining four patients. however. the heart beat spontaneously The respirator could be weaned within 48 hours alter the operation and no pulmonary complication was observed. One out of the 15 patients expired due to sudden attack of ventricular tarchycardid developed ten days after the operation, but the rest of the patients were discharged with good condition.

Adsorption, Movement and Decomposition of New Herbicide Bensulfuron-methyl in Soils (신규(新規) 제초제(除草劑) Bensulfuron-methyl 토양중(土壤中) 흡착(吸着), 이동(移動) 및 분해성(分解性))

  • This study was undertaken to elucidate the behavior of herbicide bensulfuron-methyl[methyl-2-[[[[[(4,6-dimethoxy pyrimidine-2yl)amino]carbonyl]amino]sulfonyl]methyl]benzoate]in soils. Adsorption of the herbicide in soils was mainly correlated with content of organic matter and clay, and canon exchange capacity. Adsorption distribution coefficient(Kd value) in clay loam soil was greater than those in loam and sandy loam soils. The Kd value decreased in the order of zeolite, bentonite, halloysite and laziolite clay minerals. Bensulfuron-methyl moved to 3cm deep in clay loam soil and 4cm deep in sandy loam and herbicide treated layer was 0 to 2cm profile in the two soils. The decomposition rate of bensulfuron methyl varied with the soil properties. The rate was slower in sterilized soil than in nonsterilized. Addition of organic matters to the soils accelerated the decomposotion. The degradation was more rapid in 30$^{\circ}C$ soil temperature than in 20$^{\circ}C$.

Identification and Characterization of an Endophytic Strain of Streptomyces from Rice Roots (Orysa sativa L.) (벼(Orysa sativa L.) 뿌리로부터 분리된 내생 Streptomyces 균주의 동정 및 특성)

  • We isolated an endophytic actionmycete from root tissues of rice plant collected from paddy field near Dankook University, Cheonan, Korea. Surface sterilized roots were laid on the selective agar plates and incubated. The powdery actinomycete colonies appeared on the root surface after four weeks incubation. We isolated a strain JK-5 among them and could determine its taxonomical position as Streptomyces diastaticus subsp. ardesiacus by using 16S ribosomal DNA sequencing. The chemotaxonomical and morphological studies confirmed the taxonomical position of the strain JK-5. The shape of aerial hyphae was flexible and they contained spore chains with more than 30 smooth spherical spores per chain. Cell walls contained LL-diaminopimelic acid. There was no characteristic sugar in whole-cell hydrolysates. The major fatty acids were anteiso-15:0, anteiso-17:0 and iso-16:0. The specific menaquinones, MK-9 ($H_6$), MK-9 ($H_8$), were detected. The GC content was 72%. Antifungal activities of the strain JK-5 were relatively strong against fungal plant pathogens. The endophytic growth of the strain JK-5 was confirmed by SEM observation of the root and stem of the infected rice plant.

Study on Antibacterial Activity of Ag Nanometal-deposited TiO2 Prepared by Sonochemical Reduction Method (초음파환원법에 의해 제조된 Ag-TiO2의 항균 활성도 고찰)

  • In this work, Ag-$TiO_2$ nanocomposites were prepared via the sonochemical deposition of Ag nanometals on $TiO_2$ nanoparticles. The size of deposited Ag nanometals was ranged in 1~3 nm and the number of Ag nanometals deposited on $TiO_2$ increased in proportion to the dosage amounts of Ag precursors. As-prepared Ag-$TiO_2$ was loaded on the sterilized agar plate together with an aliquot volume of diluted E-coli, followed by 30 min irradiation of the solar simulated light ($600{\sim}1800{\mu}w/cm^2$). Finally, the agar plate was incubated for 24 h at $37^{\circ}C$ and the number of survived colonies were counted. It was experimentally confirmed that Ag-$TiO_2$ exhibited the higher antimicrobial activity than that of pure $TiO_2$, based on measuring the colony number of control sample. The survived colony numbers on the agar plate decreased with the increase of dosage amounts of Ag-$TiO_2$ and the irradiated intensity of solar simulated light for 30 min before incubating. The increase of Ag nanometal doposition induced the progressive enhancement of antimicrobial activity, but rather reduced the photocatalytic activity of Ag-$TiO_2$ probably due to the excessive presence of Ag nanometals on $TiO_2$ matrix.

The prediction of shelf-life of Commercially Sterilized Korean Soups using accelerated experiment (가속실험을 이용한 상업적 멸균처리 한식 탕반류의 유통기한 평가)

  • The objective of this research was to estimate the shelf-life of 4 kinds of Korean soup (Yukkaejang, Sagol-woogeojitang, Kalbitang, and Seolleongtang), under an accelerated experiment, based on the changes in the selected physico-chemical, microbiological and sensory characteristics. The 4 kinds of Korean soup (Yukkaejang, Sagol-woogeojitang, Kalbitang and Seolleongtang) were 1.sated at $121^{\circ}C$ for 30 min. and the shelf life was evaluatedthrough accelerated experiment, based on tile changes in the physico-chemical, microbiological and sensory characteristics. No viable cells were detected in any of the treatments and no significant differences were shown in pH, TBA and Hunter's color values for 3 weeks at $37^{\circ}C$. The periods found to be acceptable by sensory evaluation were 1 days for Yukkaejang and Kalbitang, 14 days for Sagol-woogeojitang, and 21 days for Seolleongtang. For an assumed $Q_10$ value of 2.5, shelf-life was predicted as 109 days for Yukkaejang and Kalbitang, 219 days for Sagol-woogeojitang, an d 328 days for Seolleongtang at $5^{\circ}C$, suggesting that these 4 kinds of Korean soup are suitable for commercial distribution.

Sterilization of Yakju(Rice Wine) on a Serial Multiple Electrode Pulsed Electric Field Treatment System (직렬배열 다중전극 고전압 펄스 전기장 처리장치를 이용한 약주의 살균)

  • Yakju(rice wine) was sterilized with high-voltage square-wave pulses of $1\;{\mu}s$ duration at various electric field strengths and frequencies on a serial multiple electrode pulsed electric field(PEF) treatment system consisted of 7 electrodes connected in series. The initial microbial counts of Yakju were $1.88{\times}10^3{\sim}2.13{\times}10^4$ CFU/mL for total aerobes, $1.55{\times}10^3{\sim}2.85{\times}10^4$ CFU/mL for lactic acid bacteria and $1.72{\times}10^3{\sim}2.39{\times}10^4$ CFU/mL for yeasts. The sterilization of microorganisms in Yakju was a first order reaction and the sterilization effect increased as the field strength and the frequency increased. The $D_{Hz}-value$ and the $D_{PEF}-value$ decreased with the electric field strength. Yeast showed lower $D_{PEF}-value$ than bacteria. Lactic acid bacteria showed lower $D_{PEF}-value$ than general aerobic bacteria under the electric field strength below 30 kV/cm, but higher ones under that above 40 kV/cm. The $Z_{PEF}-value$ of general aerobic bacteria, lactic acid bacteria and yeast in Yakju were 39.4, 49.3 and 47.6 kV/cm, respectively. The PEF sterilization resulted in less changes in color and sensory properties than heat sterilization, and the PEF treated Yakju showed superior quality to the heat treated one. The commercial sterilization of Yakju was accomplished with 2-cycle treatment on the tested serial PEF treatment system.

Comparison of UV-Irradiation and High Hydrostatic Pressure Sterilization for Storage Stability of Carrot Juice (UV살균과 초고압살균에 의한 당근주스의 저장성 비교)

  • The objective of this study was to investigate the non-thermal sterilization effect of methods such as high hydrostatic pressure (HHP) and UV-irradiation (specifically with regard to the storage stability and shelf-life of carrot juice. The microbial counts of the non-sterilized product increased from 5.51 to 7.34 log CFU/mL up to the fifth day, and then decreased to 5.46 log CFU/mL at six days. UV-irradiation was increased from 2.37 to 4.92 log CFU/mL. HHP was maintained under the 3 log CFU/mL. The pH of UV-irradiation and HHP was maintained withing the range of 6.29~6.30 and 6.20~6.22 during storage. However, the pH arising from non-sterilization decreased from 6.31 to 4.49. The color of non-sterilization changed significantly during storage, but UV-irradiation and HHP were similar during storage. The ${\beta}$-carotene content of non-sterilization was noted to have decreased from 269.45 to $65.19{\mu}g/mL$ during storage. The UV-irradiation and HHP decreased from 263.46 and $268.35{\sim}281.16{\mu}g/mL$ to 243.42 and $244.09{\sim}269.29{\mu}g/mL$, respectively. In conclusion, these findings suggest that HHP can be used for the pasteurization, or sterilization, of carrot juice and the optimal condition is two minutes.

Proteolytic Yeasts Isolated from Mackerel (Scomber japonicus) (고등어에서 분리된 부패성 효모)

  • Microbiological spoilage of marine fish is complex process occurring by bacteria, yeasts and molds. There have been rare study for saprophytic yeasts although having enormous numbers of bacteriological studies on the spoilage of marine fish. The 14 genera of yeasts isolated from mackerel (Scomber japonicus) with high frequency of occurrence were Candida sp., Rhodotorula sp., Torulopsis sp., Cryptotoccus sp. and Tricosporon sp. Among these ones Candida lipolytica was identified as the strongest proteolytic yeast, then named Candida lipolytica FM5 (C. lipolytica FM5). C. lipolytica FM5 showed optimum growth at $25^{\circ}C$, pH 7.0 and could grow at $5^{\circ}C$ and in medium containing $10\%$ sodium chloride, To evaluate the saprophytic activity of the selected strain, C, lipolytica FM5 and Pseudomonas fluorescens ATCC 17571 which is one of representative spoilage bacteria were individually inoculated into the sterilized fish muscle homogenates, and then pH changes and volatile basic nitrogen (VBN) values were checked during the storage at various temperatures. According to the experimental results, the productions of VBN by C. lipolytica FM5 in the fish muscle homogenates were 50 mg-N/100 g at $5^{\circ}C$, 152 mg-N/100 g at $15^{\circ}C$ and 379 mg-N/100 g at $25^{\circ}C$ for 1 week storage, respectively. Above results were nearly same as in case of Ps. fluorescens ATCC 17571 inoculation. It suggest that sapyophytic yeasts also have important role in spoilage of marine fish.
