  • Acid dairy drinks(ADD) are a worldwide product existing in many variations: fruit milk drinks, yogurt drinks, soy milk, butter milk, whey drinks and kefir etc. These drinks are marketed with different shelf lives depending on processing: -Short shelf life(maximum 3 weeks, cold storage) - Long shelf life(2 to 9 months, pasteurized, sterilized or retorted) Acidic protein drinks tend to a separation or destabilization process in the absence of stabilizing system during the shelf life of the ADD. A phase separation results in sedimentation of large particles at the bottom of the package and/or the formation of a serum layer at the top(whey off). These beverages are usually composed of an acid dairy phase (fermented base)or a natural base(milk, soymilk etc.)with an acidic medium (fruit phase: pulp, fruit concentrate etc.) which can be flavored. Sugar and stabilizers are added. It has been proved since the late 1950's that adding high methoxy pectin (HM pectin)to acid milk drinks is the best way to prevent the formation of a sediment and/or the whey off. In this presentation, we explain about stabilization mechanism of ADD induced by pectin. Applications and market trend of ADD in Asia and Europe are explained.

Effects of Acupotomy Treatment on Relapsed Lateral Malleolar Bursitis: A Case Report (재발성 족외과 점액낭염에 대한 도침요법: 증례 보고)

  • Objectives : This study details on a case of acupotomy for relapsed lateral malleolar bursitis. Methods : A 71-year-old woman with lateral malleolar bursitis was treated with acupotomy twice in an outpatient setting. A sterilized, disposable, blade width 0.5 mm × needle length 50 mm sized acupotomy was administered on GB40 and BL62 on each visit. Pain intensity assessment using Numeric Rating Scale (NRS) and the response evaluation criteria was conducted right after each treatment. Results : Pain on NRS declined from 5 to less than 1 after twice of acupotomy treatment. This result was regarded as 'Complete Response' using the response evaluation criteria. There were no side effects during whole treatment procedure at all. Conclusions : It is turned out that acupotomy can have a positive clinical effect on lateral malleolar bursitis in this case without any adverse effect. Further controlled studies on acupotomy for lateral malleolar bursitis are required for clinical application.

Efficient Plant Regeneration from Shoot Tip and Young Leaf in Rhodiola sachalinensis A. Bor.

  • The shoot tip and young leaf of Rhodiola sachalinensis were cultured to invest the plant growth regulator condition for callus induction, shoot and root regeneration. When the shoot tip was sterilized in 2.0% of NaOCl for 20min., the contamination rate was the lowest. And the survival rate of the culture material was good in carbenicillin 500mg/L treatment group. Callus was obtained from shoot tip and young leaf segments. NAA 0.1-1.0mg/L and 2,4-D 0.1-0.5mg/L alone treatment were shown to have a good response on callus induction from shoot tip culture. In the case of young leaf culture, NAA and 2,4-D 0.1-0.5mg/L alone treatment were good in callus induction. In culturing shoot tip NAA 0.5mg/L and BA 0.5mg/L, NAA1.0mg/L and BA 0.lmg/L combination treatment was good in shoot regeneration. The regenerated shoots were rooted on MS medium supplemented with NAA and BA combination treatment. Especially, NAA 1.0mg/L and BA 0.1mg/L combination treatment was effective for root regeneration.

  • It has been known that periods of absorption varies allografts or xenografts of transplantations of freeze-dried cortical bone(FDCB). In this study changes of absorption of FDCB in xenograft transplantations were evaluated according to extracted time with chloroform-methanol solution(CM sol.). The FDCB from pig was removed soft tissue by surgical knife. Fat of the FDCB was removed with treatments of CM sol. for 2, 6, and 10 days, then the treated FDCB was freeze-dried at $-80^{\circ}C$ and sterilized with ethylene oxide gas. The FDCB was transplanted to fifteen millimeter artificial-defected regions of 6 dogs on fibular diaphyses. This was biweekly examined by radiograph for 18 weeks. In result new bone formation with FDCB treated for 6 days was higher than the other bones treated for 2 and 10 days. Duration of absorption with FDCB treated for 6 days was longer than the others. The remain with FDCB treated for 10 days was more than the others.

Quality and antioxidant activity of ginseng seed processed by fermentation strains

  • Background: Fermentation technology is widely used to alter the effective components of ginseng. This study was carried out to analyze the characteristics and antioxidant activity of ginseng seeds fermented by Bacillus, Lactobacillus, and Pediococcus strains. Methods: For ginseng seed fermentation, 1% of each strainwas inoculated on sterilized ginseng seeds and then incubated at $30^{\circ}C$ for 24 h in an incubator. Results: The total sugar content, acidic polysaccharides, and phenolic compounds, including p-coumaric acid, were higher in extracts of fermented ginseng seeds compared to a nonfermented control, and highest in extracts fermented with B. subtilis KFRI 1127. Fermentation led to higher antioxidant activity. The 2,2'-azine-bis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity was higher in ginseng seeds fermented by Bacillus subtilis than by Lactobacillus and Pediococcus, but Superoxide dismutase (SOD) enzyme activity was higher in ginseng seeds fermented by Lactobacillus and Pediococcus. Conclusion: Antioxidant activities measured by ABTS and SOD were higher in fermented ginseng seeds compared to nonfermented ginseng seeds. These results may contribute to improving the antioxidant activity and quality of ginseng subjected to fermentation treatments.

Preconditioning for Cryopreservation of in vitro Grown Bulblets of Lily using Droplet-Vitrification

  • This study was conducted to improve and supplement the system of cryopreservation for adventitious bulbs induced by tissue cultured bulb-scales of lily (Lilium spp.) cvs. 'Milky way'. The explants, bulblets and bulb-scale-bulblets, were treated to low temperature (4℃) for 7 days prior to the pre-culture. The adventitious bulbs were pre-cultured in Murashige and Skoog (MS) liquid medium supplemented with sucrose (0.3 and 0.7M). The pre-cultured adventitious bulbs were treated to loading solution (LS1 or LS2, C4 or C6) containing 35% of PVS3 (LS1, C4) or 40% of PVS3 (LS2, C6) for 40 min and exposed to dehydration solution (PVS3, B1) containing 50% glycerol and 50% sucrose for 60 min at 25℃. The adventitious bulbs were moved onto droplets containing 3 µl PVS3 on sterilized aluminum foils, and then soaked into liquid nitrogen (LN) for 60 min. The result of highest regrowth rate as 65.7% was obtained in cold treatment (4℃), osmoprotected with LS1 solution, and cultured in PCM3 medium by using bulb-scale-bulblet for cryopreservation. This result shows that droplet-vitrification could be used as a promising method for long-term storage of lily genetic resource.

Sterilization and storage of dried fishes by irradiation (방사선에 의한 건어물의 살균 및 저장에 관한 연구)

  • The effect of irradiation at dose of 3~10 kGy on the extension of the shelf-life of sliced dried squid and dried file and on its quality were investigated during nine months of storage at room temperature. The number of microorganisms, TBA value and TMA-N content were used as objective indices concerned to the sensory evaluation. In the number of initial microbial loading, total bacterial count of sliced dried squid and dried file fish were $2.7{\times}10^{8}\;and\;2.0{\times}10^{5}$, yeast and mold were $5.7{\times}10^{5}\;and\;6.2{\times}10^{5}$ and coliform group were $2.1{\times}10^{5}$ and above 10 per gram of the samples, respectively. The number of total bacteria was sterilized by 99% with irradiation of 3~10 kGy and irradiation of above 7 kGy was shown to be effective for the radurization of yeast, mold and coliform group. TBA value of irradiated groups were higher than storage time. The TMA-N content of nonirradiated group was markedly increased by microbial spoilage during storage, however above 8 kGy irradiated group were indicated about 10 mg% after nine months storage. In the sensory evaluation, the nonirradiated group was off favor after three months storage and was changed in color of dried fishes but 7~10 kGy irradiated group were maintained good qualit compared with those of fresh samples after nine months storage at room temperature.

Fungal Endophytes from Three Cultivars of Panax ginseng Meyer Cultivated in Korea

  • In order to investigate the diversity of endophytes, fungal endophytes in Panax ginseng Meyer cultivated in Korea were isolated and identified using internal transcribed spacer (ITS) sequences of ribosomal DNA. Three cultivars of 3-year-old ginseng roots (Chunpoong, Yunpoong, and Gumpoong) were used to isolate fungal endophytes. Surface sterilized ginseng roots were placed on potato dextrose agar plates supplemented with ampicilin and streptomycin to inhibit bacterial growth. Overall, 38 fungal endophytes were isolated from 12 ginseng roots. According to the sequence analysis of the ITS1-5.8S-ITS2, 38 fungal isolates were classified into 4 different fungal species, which were Phoma radicina, Fusarium oxysporum, Setophoma terrestris and Ascomycota sp. 2-RNK. The most dominant fungal endophyte was P. radicina in 3 cultivars. The percentage of dominant endophytes of P. radicina was 65.8%. The percentage of colonization frequency of P. radicina was 80%, 52.9%, and 75% in Chunpoong, Yunpoong, and Gumpoong, respectively. The second most dominant fungal endophyte was F. oxysporum. The diversity of the fungal endophytes was low and no ginseng cultivar specificity among endophytes was detected in this study. The identified endophytes can be potential fungi for the production of bioactive compounds and control against ginseng pathogens.

  • Human Dura mater treated by various processes was used to restore small periodontal defects, large bony defects for improvement of new bone formation, and soft tissue defects and replace the disc of TMJ, etc.. Tutoplast Dura is the solvent-preserved Human Dura mater and sterilized by gamma radiation. In our department, Tutoplast Dura was implanted in 32 patients, from 1994, 6 to 1995, 7. We implanted the Tutoplast Dura at 11 various cysts, 6 implantations, 4 fractures, 3 clefts, 2 TMJ disease, 2 maxillary sinusitis, etc.. We performed the retrospective study about the purpose of Tutoplast Dura implantation, postoperative complication and histologic examination of biopsy specimen at implant second surgery.

  • Purpose: The purpose of this research is to study about initial adhesion, proliferation and activation of osteoblast to titanium surface treated with machined, hydroxyapatite coating, resorbable blast material blasting and anodizing method. Material and Methods: After treating the titanium surface of each block with machined, impurities were removed and sterilized. The number of cells attached from cultured osteoblast of respective experimental groups were measured at 1, 4, 7, and 14day and alkaline phosphatase, calcium, and inorganic phosphate concentration of cultured solution was measured. Result: Anodizing group showed the highest rate of cell attachment and proliferation activity. RBM treated group showed the highest increasing on their alkaline phosphatase activity, on the calcium apposition, on inorganic phosphate apposition of 1 and 4 days in cultured osteoblast to compare with other groups. Conclusion: On the basis of these findings, we conclude that surface modification of titanium was profoundly effected on the attachment, proliferation and activation of osteoblast in initial stage osseointegration.
