Effect of Organic Matter on the Occurrence of Fusarium Wilt in Cucumber (Fusarium oxysporum f. sp. cucumerinum에 의한 오이덩굴쪼김병의 발생에 미치는 유기물 시용의 효과)

  • Fusarium wilt caused by Fusarium oxysporum f. sp. cucumerinum has caused high damage in cucumber under greenhouse condition. This disease was suppressed $30\~55\%$ by application of organic matters compared with natural cropping soils. The suppression effect was the highest in the mushroom humic compost and fowl excretion matter among the various organic matters, varying with kinds of organic matters and degrees of humic resolvability. There was a slight difference in severity of fusarium wilt between sterilized organic matters and soils. The disease occurrence was delayed more in the nonsterilized organic matters and soils than in the sterilized ones. At 30 days after inoculation of F. oxysporum, numbers of Actinomycetes, fungi and bacteria were considerably increased, whereas F. oxysporum was decreased in the organic matter amended-soils compared with natural control soils.

On-Line Measurement of Biochemical Oxygen Demand of livestock Wastewater by Multi-Biosensor System (Multi-Biosensor를 이용한 축산폐수의 생물화학적 산소요구량 실시간 측정방법 연구)

  • The present study was intended to examine a basic scheme to determine the biochemical oxygen demand(BOD) of livestock wastewater by means of six individual dissolved oxygen(DO) sensors and its multi-measurable meter. Maximal point of the first order time derivative of the DO difference between DO distribution of sterilized livestock wastewater and that of non-sterilized livestock wastewater, was considered as the oxygen uptake rate(OUR) of microorganisms in livestock wastewater, as determined to be 0.00074 mg $O_2/{\ell}{\cdot}sec$. The present study showed that there was a fair linear relationship(97.72%) between maximal OUR and BOD values of livestock wastewater, the latter being determined by classical Winkler azide method. It was thus concluded that the present multi-biosensor system might be applicable to an on-line system for measurement of BOD of livestock wastewater.

Optimum Sterilization Conditions and Quality Characteristics of the Retort-Sterilized Crab Analog (레토르트 Crab Analog의 살균 최적화 및 품질 특성)

  • This study developed a normal temperature-circulatable retort-sterilized crab analog (RSCA) with a long shelf-life by retort sterilization. We examined the optimum sterilization conditions and quality characteristics of the RSCA. A central composite design for response surface methodology (RSM) was adopted to optimize of the retort sterilization. The central composite design consisted of 11 samples: four factorial points, four star points, and three central points. The sterilization temperature and $F_0$-value for retort sterilization were the independent variables, and the shearing force, whiteness, and sensory score for texture were the dependent variable. The RSM analysis of the multiple response optimization for the RSCA using the Minitap statistical programing gave an $F_0$-value of 3.3 min. at $117.5^{\circ}C$. The RSM analysis also indicated that the sterilization temperature during retort sterilization was the most influential factor, while the $F_0$-value had little effect on the quality of the RSCA. The moisture, pH and volatile basic nitrogen contents of the RSCA product were 73.1%, 7.3 and 17.2 mg/100 g, respectively, and viable cells count was >18 CFU/g. And sensory quality characteristics of this RSCA was similar to those of a commercial crab analog.

Effect of supplementary glycerin on milk composition and heat stability in dairy goats

  • Objective: This experiment was studied the effects of various levels of crude glycerin (CG) in dairy goat diet on daily intake, milk yield, milk composition, some physical properties and some quality changes of goat milk after sterilization. Methods: Twelve 75% Saanen dairy goats (body weight = $49{\pm}3kg$; days in milk = $60{\pm}12d$) were randomly assigned in a completely randomized design to evaluate the effects of three experimental diets consisting of 0%, 5%, and 10% CG (dry matter basis) which were formulated to meet or exceed the nutrient requirements of goats. Experimental dairy goats were evaluated for feed and milk yield. Milk samples were analyzed for their composition, including fatty acids, casein profile, fat globule size, and color, and were sterilized to evaluate milk heat stability. Results: There were no significant differences between 0% and 5% CG treatments infeed. Increasing CG supplementation from 0% to 5% increased milk yield from $2.38{\pm}0.12$ to $2.64{\pm}0.23kg/goat/d$. In addition, milk samples from 5% CG treatment had the highest total solids, fat content and lactose content, and largest fat globule size. Increasing CG to 10% resulted in a decrease in milk fat. After sterilizing at $116^{\circ}C$, $F_0=3min$, goat milk samples from 5% CG treatment had slightly higher sediment content and comparatively higher degree of browning. Conclusion: Considering milk yield, milk fat content and quality of sterilized milk, 5% CG supplementation in a total mixed ration has a potential for implementation in dairy goats.

Salivary Bacterial Counts on Application Time of Oral Antiseptic Agents and Mechanical Irrigation (구강소독제의 적용 시간 및 기계적 세척 여부에 따른 타액 내 세균 수의 감소 효과)

  • Purpose: The purpose of this study is to establish the acceptable intraoral application time of antiseptic agents and evaluate the effect of mechanical irrigation. Methods: A total of 80 subjects were selected for this study. Saliva secreted at the resting state was taken. The subjects were divided into 8 experimental groups, and kept 10% povidone-iodine (PVI) or 0.2% chlorhexidine gluconate (CHX) for 20 or 40 seconds in their oral cavity with/without irrigation of the oral cavity with sterilized normal saline, respectively. Then, the saliva was taken and diluted with phosphate buffered saline and then plated onto 5% sheep blood agar plates, which were incubated. Colony forming unit (CFU) was measured for the salivary bacterial counts. Results: After application of PVI and CHX, all the experimental groups showed statistically significant decrease in CFU (P<0.01). Group 2 (PVI, 40 s) showed more significant reduction rate in CFU than group 4 (CHX, 40 s; P<0.01). Group 6 (PVI, 40 s, irrigated) showed more significant reduction rate than group 2 (PVI, 40 s; P<0.01). Group 2 (PVI, 40 s) showed more significant reduction rate than group 1 (PVI, 20 s; P<0.01). Conclusion: Application of PVI for 40 seconds and mechanical irrigation with sterilized normal saline showed the best result among the 8 groups in terms of the reduction rate of salivary bacterial counts.

Effects of Irradiation Temperature on the Sensory Quality Improvement of Gamma-irradiated Ganjang-gejang, Korean Traditional Marinated Raw Crab Portunus trituberculatus in Soybean Sauce

  • This study was conducted to confirm quality properties of sterilized Ganjang-gejang (marinated crab Portunus trituberculatus) with Korean soy sauce using by gamma irradiation and to improve quality of sterilized Ganjang-gejang. The Ganjang-gejang was irradiated at dose of 3, 6, 9, 12, and 15 kGy by gamma irradiation and there was evaluated in microbiological, physicochemical, and sensory properties. Total aerobic bacteria and fungi contents of non-irradiated samples were about 6 and 4 log CFU/g level, respectively. Gamma-irradiated samples at above 9 kGy did not contain aerobic bacteria or fungi at detection limit less than 2 log CFU/g, but sensory scores were significantly decreased depending on the irradiation dose. To improve the sensory qualities of gamma-irradiated Ganjang-gejang, the temperature was adjusted during sample irradiation. When samples were irradiated under freezing temperatures, especially on dry ice, the TBARS and the deterioration of sensory qualities of Ganjan-Gejang were reduced. Different odor patterns were observed among samples, as observed using electronic nose analysis system. The results of this study indicated that treatment with irradiation under low temperatures may help to preparing high-quality Ganjang-gejang.

Success Rate of Tubal Sterilization Reversal (정부지원 난관복원수술의 성공율에 관한 연구)

  • Five hundred and fory-four women were provided government funded sterilization reversal services with technique of microsurgery at 15 reversal institutions designated by the Korean Association for Voluntary Sterilization since the inception of Female Reversal Program in 1981. A large majority of reasons for requesting reversal surgery was a loss of children, comprising 87.3%, and the mean interval between sterilization and reversal was 33.6 months. Two hundred and ninty-one of 418 women who were post-operatively followed up with the length of 11 months to 11 years, have experienced term dilivery or ,intra-uterine pregnancy, represented 69.6%. Eight cases have experienced ectopic pregnancy and 9 cases spontaneous abortion. The largest number of reversal clients were sterilized by the laparoscopic unipolar coagulation technique and the next largest group was sterilized by the laparoscopic banding technique, representing 59.8% and 28.9% respectively. The highest pregnancy rate, 80.9%, was shown in clients who had undergone laparoscopic banding technique while the lowest, 61.8%, was the group of laparoscopic bipolar coagulation. The most common site of the anastomosis was isthmic-ampullary portion and the next was isthmic-isthmic portion. The highest success rate, 77.8 %, was marked in the isthmic-isthmic anastomosis and the lowest was in the ampullary-ampullary anastomosis, representing 50.0%. A more than 60% of the clients became pregnant within 6 months of their reversal surgery, with the shortest interval being 1 month, the longest 39 months, and the mean 7.6 months. A large majority of the successful cases were pregnancy within 1 year of their reversal surgery, representing 82.1 %. The higher rate of pregnancy, 73.5%, was in the clients undergone reversal surgery within 36 months of their sterilization and the lower pregnancy rate, 64.1 %, was in the clients undergone reversal surgery longer than 37 months of their sterilization.

The Effect of Seed-borne Mycoflora from Sorghum and Foxtail Millet Seeds on Germination and Disease Transmission

  • The seed-borne mycoflora of sorghum and foxtail millet collected from different growing areas in South Korea were isolated and taxonomically identified using dry inspection, standard blotter and the agar plate method. We investigated the in vitro and in vitro germination rates of disinfected and non-disinfected seeds of sorghum and foxtail millet using sterilized and unsterilized soil. The percent recovery of seed-borne mycoflora from the seed components of sorghum and foxtail millet seeds was determined and an infection experiment using the dominant species was evaluated for seedling emergence and mortality. A higher number of seed-borne fungi was observed in sorghum compared to that of foxtail millet. Eighteen fungal genera with 34 fungal species were identified from the seeds of sorghum and 13 genera with 22 species were identified from the seeds of foxtail millet. Five dominant species such as Alternaria alternata, Aspergillus flavus, Curvularia lunata, Fusarium moniliforme and Phoma sp. were recorded as seed-borne mycoflora in sorghum and 4 dominant species (Alternaria alternata, Aspergillus flavus, Curvularia lunata, Fusarium moniliforme) were observed in foxtail millet. The in vitro and in vitro germination rates were higher using disinfected seeds and sterilized soil. More seed-borne fungi were recovered from the pericarp compared to the endosperm and seed embryo. The percent recovery of seed-borne fungi ranged from 2.22% to 60.0%, and Alternaria alternata, Curvularia lunata and 4 species of Fusarium were isolated from the endosperm and embryo of sorghum and foxtail millet. Inoculation of the dominant seed-borne fungi showed considerable mortality of seedlings. All the transmitted seed-borne fungi might well be a primary source of infection of sorghum and foxtail millet crops.

Soil Chemical Properties, Microbial Community and Ginseng Root Rot in Suppressive and Conducive Soil Related Injury to Continuously Cropped Ginseng (인삼 연작장해 유발토양과 억제토양의 화학성, 미생물상 및 뿌리썩음병 발생 특성)

  • Background: Suppressive soil inhibits soil-borne diseases if pathogens are present, and ginseng does not show injury even if replanted in the same field. Methods and Results: Soil chemical properties and microbial community of soil were investigated in soil suppressive and conducive to ginseng root rot. Root rot disease in 2-year-old ginseng was tested by mixing conducive soil, with suppressive or sterilized suppressive soil. The root rot ratio in suppressive soil was 43.3% compared to 96.7% in conducive soil. Biological factors acted to inhibit the root rot because disease ratio was increased in the sterilized suppressive soil compared to that in non-suppressive soil. The suppressive soil had lower pH, nitrate nitrogen and sodium than the conducive soil. Dominat bacteria and fungi (more than 1.0%) were 3 and 17 species in conducive soil and 7 and 23 species in suppressive soil, respectively. The most predominant fungi were Pseudaleuria sp. HG936843 (28.70%) in conducive soil and Pseudogymnoascus roseus (7.52%) in suppressive soil. Conclusion: Microbial diversity was more abundant in the suppressive soil than in the conducive soil, and the proportion of pathogens (Nectriaceae sp.) causing root rot was significantly lower in the suppressive soil than in the conducive soil.

  • This study was conducted to obtain basic data which can be applied to process of canned boiled Kwamaegi. Commercial Kwamaegi was cut into $2{\times}3$ cm lengths, filled 90 g into can (301-3) and added with 60 g water and then precooked for 10 min. at $100^{\circ}C$. And water layer was drained. The precooked Kwamaegi was packed into the can, and added with 60 g of mixed salt solution, which is mixed with salt 0.5% and bamboo salt 0.7%. The cans were seamed using a vacuum seamer, and then sterilized for various Fo values (Fo 8~12 min.) in a steam system retort at $121^{\circ}C$. pH, VBN, amino-N, total amino acid, free amino acid, color value (L, a, b), texture profile, TBA value, mineral, sensory evaluation and viable bacterial count of the canned boiled Kwamaegi produced with various sterilization condition(Fo 8~12 min.) were measured. There was no remarkable difference between sterilization conditions and sensual characteristics. The results showed that the product sterilized at Fo 8 min. was the most desirable because this condition is the most economical and tasty.
