해양균류의 항균활성 검색 (Screening of Antimicrobial Activity from the Marine-Derived Fungus)

  • Acetone extracts of 301 strains of marine-derived fungus were tested for antimicrobial activity against three strains of bacteria. The bacteria consisted of three pathogens, Staphylococcus aureus, methicillin-resistant S. Aureus, and multidrug-resistant S. aureus. The acetone extracts of 10 strains (MFA117, MFA130, MFA134, MFA206, MFA217, MFA268, MFA277, MFA291, MFA292, MFA301) showed strong activity, inhibiting 100% of the bacterial growth. These antimicrobial active strains were cultlued in SWS medium on a 1 L scale and the resulting broth and mycelium were extracted to afford mycelium extract (000M) and broth extract (000B), respectively. Antimicrobial activity for all extracts has been tested as the results, the mycelium extract of one strain (217M) and the broth extracts of 9 strains (117B,130B, 134B, 206B, 268B, 277B, 291B, 292B, 301B) exhibited relatively high levels of activity at minimal inhibitory concentration (MIC) values of $500-125\;{\mu}g/mL$ range. Among them, the extracts, 277B, 291B, 292B and 301B showed the most significant antimicrobial activity with $IC_{50}$ values of $125\;{\mu}g/mL$.

  • Antibacterial activity is an important feature for the development of antibiotics alternatives. Plant extract is considered as a promising alternative for organic farming. In this study, a total of 11 plants were extracted using ethanol to determine their antibacterial activities against to Staphylococcus aureus, Listeria monocytogens, Mannheimia haemolytica and Salmonella gallinarum. The synergistic interaction among plant extracts was also investigated. Plants used in this study were Carthamus nctoricus L. (pA), Poncirus trifollata Raf. (pB), Scutellaria balcalensis Georgi (pC) Prunus sargentii (pD), Cucurbita moschata $D_{UCH}$ Leaf (pE), Allium cepa L. peel (pF) Portulaca oleracea L. (pG), Xanthium strumarium L. (pH), Duchesnea chrysantha (pI), Cudrania tricuspidata (pJ) and Juniperus chinensis L. (pK). The pB and pA had the most broad antibacterial spectrum and the highest activity against to Staph. aureus among plant extract, respectively. In the synergistic interaction, the mixtures of pA and pC as well as pA and pF had batter antibacterial activity against to Staph. Aureus compared with other mixtures.

황색포도상구균 감염에 의한 어린 말티스의 자궁축농증 증례 (Spontaneous pyometra induced by Staphylococcus aureus in a Maltese)

  • An 8-months-old young Maltese bitch was presented for investigation of anorexia and occasional vomiting. The body weight was 2 kg. Abdominal sonography demonstrated abnormal enlargement of uterus. The dog had been clinically and pathologically examined. Also, ovariohystectomy was performed. Staphylococcus aureus was identified in the pus-like materials of the uterine lumen. This case was confirmed as pyometra induced by Staphylococcus aureus. Gross examination of the dissected uterus revealed enlarged uterine horn, which had yellowish or brownish pus-like materials in the lumen. Also, there was several segments formation between the enlarged areas of left and right uterine horns. Histopathologically, endometritis and necrosis were present in uterine wall. The bitch made a complete recovery following an ovariohystectomy. This case was spontaneous pyometra induced by Staphylococcus species. On our knowledge, this is a very rare report of pyometra occurrence in young dog and Staphylococcus species-induced pyometra. This study may provide a valuable data on the study of canine pyometra.

Synergistic Inhibition by Bacteriocin and Bacteriophage against Staphylococcus aureus

  • Staphylococcus aureus is a representative pathogenic bacterium carefully controlled in the dairy industry because it causes bovine mastitis and thus, can enter the dairy chain. Furthermore, the emergence of multi-drug resistant S. aureus is a big problem. We previously isolated a Lactococcus lactis strain producing a bacteriocin that exhibited strong antimicrobial activity against S. aureus. In this study, we investigated the synergistic inhibition of S. aureus by the bacteriocin and a bacteriophage (SAP84) which is specific to the organism. The bacteriocin (12.5-100 AU/mL) inhibited the growth of S. aureus KCTC 3881 in a dose-dependent manner, as did the bacteriophage SAP84 (0.001-1 MOI; multiplicity of infection). Co-treatment with the bacteriocin (100 AU/mL) and the bacteriophage (0.1 MOI) significantly inhibited the growth of S. aureus compared to each treatment alone (bacteriocin or bacteriophage), indicating the two components showed synergistic inhibition of S. aureus. Therefore, the bacteriocin and bacteriophage combination can be used as a good strategy for controlling pathogenic bacteria.

Staphylococcus aureus에 대한 한약재의 항균활성 (Antimicrobial Activity of Medicinal Herbs against Staphylococcus aureus)

  • 18종의 한약재 MeOH 추출물과 EtOH 추출물의 Staphylococcus aureus에 대한 항균력을 조사한 결과 하고초, 소목, 오배자 및 향유는 5mg/ml 수준에서 항균활성을 나타내었으며 지실, 금은화 및 황금 등은 10mg/ml, 오미자, 초두구 및 희렴 등은 30mg/ml 이상에서 항균성을 보였다. 최소저해농도는 MeOH추출물에서는 $1{\sim}34mg/ml$, EtOH추출물에서는 $1{\sim}46mg/ml$ 농도에서 항균활성을 보였으며 분획 층 중 EtOAc층에서 성장억제효과가 가장 높게 나타났다. 항균력이 가장 우수한 것으로 확인된 소목, 오배자 및 황금추출물의 미생물 증식억제 효과를 조사하기 위해 증식배지에 각각의 추출물을 0, 100, 300 및 500ppm의 농도로 첨가하여 균주의 증식을 조사한 결과 배양후 24시간에 소목 추출물 무첨가구의 $OD_{620}$값이 0.4인 반면 100ppm 이상의 추출물 첨가 시 $0.02{\sim}0.03$정도로 균증식이 현저히 억제되었고, 오배자 추출물 무첨가구의 $OD_{620}$값이 0.4인 반면 300ppm 이상의 추출물 첨가 시 $0.02{\sim}0.03$정도로 균증식이 현저히 억제되었으며 황금 추출물 무첨가구의 $OD_{620}$값이 0.4인 반면 300ppm 이상의 추출물 첨가 시 $0.02{\sim}0.07$ 정도로 균증식이 현저히 억제되었다. 이상의 결과로 볼 때 제대염, 부종성 피부염 및 관절염 등의 다양한 가축질병을 일으키는 Staphylococcus aureusi의 감염을 예방하거나 또는 치료할 수 있는 한약재 추출물 사료첨가제의 개발 가능성을 확인 할 수 있었다.

영상의학과 촬영실 장비와 방사선사의 손 오염의 세균학적 모니터링 (Bacteriological Monitoring of Radiology Room Apparatus in the Department of Radiological Technology and Contamination on Hands of Radiological Technologists)

  • 대구시내 중 소형병원의 영상의학과 촬영실의 촬영테이블, 에어프론, 각종 손잡이, 이동형 촬영장치의 손잡이 및 방사선사의 손 등에서의 미생물의 분포를 조사하였다. 그 결과 이동형 촬영장치의 손잡이와 에어프론에서 비교적 많은 세균이 검출되었으며, 분리된 세균 중 Acinetobacter baumanni (7.3%), Klebsiella pneumoniae (6.7%), Staphylococcus aureus (3.9%), Serratia liquefaciens (1.7%), Enterobacter cloaceae (0.6%), Providenica rettgeri (0.6%)는 원내감염의 원인균으로 알려져 있다. 그리고 방사선사 손에서도 위와 유사한 집락들이 나타났는데, Klebsiella pneumoniae (8.4%), Staphylococcus aureus (6.6%), Yersinia enterocolotica (5.4%), Acinetobacter baumanni (4.2%), Enterobacter cloaceae (2.4%), Serratia liquefaciens (1.8%), Yersinia pseuotuberculosis (1.8%), Enterobacter sakazakii (1.2%), Escherichia coli (0.6%) 등의 세균이 분리되었으며, 특히 Staphylococcus aureus 와 Escherichia coli 는 강력한 병원성을 나타내기에 임상적 의의가 크다고 할 수 있다. 따라서 방사선사에게 구체적인 원내 감염에 관한 지속적인 예방교육이 필요할 것으로 사료된다.

편백나무 잎 추출액을 이용한 천연염색포의 항생제 내성균주에 대한 항균성 (Antibacterial Function of Fabrics Dyed with Extract from Chamaecyparis obtusa Leaves against Methicillin-resistant Staphylococcus aureus)

  • Bacteria exist everywhere and continuously come into contact with daily surroundings and humans. Super bacterium methicillin-resistant Staphylococcus aureus, resistant to methicillin, has recently appeared. The morbidity and rate of death associated with super bacteria infection has increased. This study investigated the antibacterial activity of fabrics naturally dyed with Chamaecyparis obtusa leaves extract against methicillin-resistant Staphylococcus aureus. Fabrics were left for 15 min in a natural dyeing solution prepared by extraction from C. obtusa leaves using 11.3% (o.w.f) with a fixed liquor ratio of 1:22 at $40^{\circ}C$. The dyeing process was conducted using three different mordants; subsequently, the K/S value of the dyed fabrics increased in the order of None < Cu < Fe < Al. The color fastness property of the fabrics to washing, dry-cleaning, and rubbing was found to be excellent and ranked in the 4-5 grade. The color fastness to light of natural dyeing is low in most cases and has the problem that the dye color soon becomes bleached. Yet, in most cases cloth dyed with retinispora leaves, the color fastnezz to light was good with a third to fourth grade. Non-mordant fabrics, aluminum mordants, and copper mordants also showed better antibacterial properties (99.9% reduction) against methicillin-resistant Staphylococcus aureus, compared to the control fabrics. The dyed fabrics showed the same antibacterial activity even after three washes. The results highlight the strong potential of fabrics naturally dyed with C. obtusa-extract as a medicinal material with excellent antibacterial function against methicillin-resistant Staphylococcus aureus.

Flavonoid Inhibitors of β-Ketoacyl Acyl Carrier Protein Synthase III against Methicillin-Resistant Staphylococcus aureus

  • ${\beta}$ Ketoacyl acyl carrier protein synthase III (KAS III) initiates fatty acid synthesis in bacteria and is a key target enzyme to overcome the antibiotic resistance problem. In our previous study, we found flavonoid inhibitors of Enterococcus faecalis KAS III and proposed three potent antimicrobial flavonoids against Enterococcus faecalis and Vancomycin-resistant Enterococcus faecalis with MIC values in the range of 128-512 ${\mu}g/mL$ as well as high binding affinities on the order from $10^6$ to $10^7\;M^{-1}$. Using these series of flavonoids, we conducted biological assays as well as docking study to find potent flavonoids inhibitors of Staphylococcus aureus KAS III with specificities against Staphylococcus aureus and Methicillin-resistant Staphylococcus aureus. Here, we propose that naringenin (5,7,4'-trihydroxyflavanone) and eriodictyol (5,7,3',4'-tetrahydroxyflavanone) are potent antimicrobial inhibitors of Staphylococcus aureus KAS III with binding affinity of $3.35{\times}10^5$ and $2.01{\times}10^5\;M^{-1}$, respectively. Since Arg38 in efKAS III is replaced with Met36 in saKAS III, this key difference caused one hydrogen bond missing in saKAS III compared with efKAS III, resulting in slight discrepancy in their binding interactions as well as decrease in binding affinities. 4'-OH and 7-OH of these flavonoids participated in hydrogen bonding interactions with backbone carbonyl of Phe298 and Ser152, respectively. In particular, these flavonoids display potent antimicrobial activities against various MRSA strains in the range of 64 to 128 ${\mu}M$ with good binding affinities.

Effect of the Ag3PO4 on Staphylococcus aureus Growth and Human Immunity

  • Silver (Ag) has been widely used in commercial products and medical fields since ancient times because of its antibacterial effect. It is harmless and non-toxic to the human body. For this reason, recent research has actively evaluated antimicrobial activity using silver (Ag). In this study, we investigated the inhibitory effect of a silver-based compound, silver phosphate ($Ag_3PO_4$) on the growth of Staphylococcus aureus and the activation of human immunity. First, the inhibitory effect of $Ag_3PO_4$ on the growth of Staphylococcus aureus was confirmed by a growth curve and a colonyounting method. As a result, the growth inhibitory effect increased as the concentration of $Ag_3PO_4$ increased. Specifically, treatment with $5{\mu}g/mL$ of $Ag_3PO_4$ resulted in no bacteria growth, and the colony-counting method showed a remarkable inhibition. In addition, the expression of cytokine IL-8 by $Ag_3PO_4$ was examined to investigate the cellular immune system activation by $Ag_3PO_4$. After pretreatment of Staphylococcus aureus for 1 hour with $50{\mu}g/mL$ $Ag_3PO_4$, an increased IL-8 mRNA expression resulted. In cells treated with $Ag_3PO_4$, we found that the expression of IL-8 was enhanced in a time-dependent fashion compared to non-treated cells. These results indicate that $Ag_3PO_4$ induces antimicrobial activity against Staphylococcus aureus and activates human immunity. These results are expected to contribute to the future study of the mechanism of silver (Ag) and silver-based compounds in relation to antibacterial activity.

농산물 생산 환경에서 분리된 Staphylococcus aureus의 항생제 내성 및 독소 유전자 확인 (Identification of Toxin Gene and Antibiotic Resistance of Staphylococcus Aureus Isolated from Agricultural Product Cultivation Environments)

  • Objectives: This study was undertaken to analyze Staphylococcus aureus from cultivation environments for agricultural products and to confirm antibiotic resistance and enterotoxin genes for the isolated S. aureus. Methods: A total of 648 samples were collected from apple, peach, ginseng and balloon flower farms. S. aureus was isolated from soil, agricultural water, personal hygiene elements (hands, gloves and clothes) and work utensils (boxes). Results: S. aureus was detected in a total of 25 samples and 72 strains were isolated. The resistance rate of the isolated S. aureus strains was confirmed at 33.3%, with 24 resistant strains among the total of 72. Fourteen different patterns types were found, and three pattern types (NV, OX, VA) were confirmed most frequently. As result of the detection of enterotoxin gene type, four gene types (sea: 1, sed: 4, seg: all isolated S. aureus, sei: all isolated S. aureus) were analyzed among a total of nine types. Conclusions: This study demonstrates that personal hygiene techniques should be properly managed, such as washing and sterilization before or after work, because agricultural contamination by S. aureus frequently developed through improper management.
