A Thermodynamic Study on Thermochromism of Blue Dye Systems (Blue 계열 염료의 열변색 현상에 관한 열역학적 연구)

  • Two different dyes containing the same molecular weight but different chemical structure have been utilized for the study of thermodynamic parameters. In this study, {3-(4-(diethylamino)phenyl)-3-(1-ethyl-2-methyl-1H-indol-3-yl)isobenzofuran-1(3H)-one} (Blue 502) and {3-(4-(diethylamino)-2-methylphenyl)-3-(1,2-dimethyl-1H-indol-3-yl)isobenzofuran-1(3H)-one} (Blue 402) were used. It has been performed by measuring UV spectra of the two dyes. In general, the blue shift has been observed from both dyes in higher carbon number alcohol solvents. Interestingly, Blue 502 showed higher stability than Blue 402 in the same conditions used in this study. And, the equilibrium constants (0.9~1.0) of the dyes depending upon temperature change were also calculated using UV absorbance. The standard enthalpy calculated from equilibrium constants and molar absorptivity($\varepsilon$) are 10.94 kJ/mol in Blue 402 and 9.010 kJ/mol in Blue 502, respectively.

Establishment of a Tm-shift Method for Detection of Cat-Derived Hookworms

  • Melting temperature shift ($T_m-shift$) is a new detection method that analyze the melting curve on real-time PCR thermocycler using SYBR Green I fluorescent dye. To establish a $T_m-shift$ method for the detection of Ancylostoma ceylanicum and A. tubaeforme in cats, specific primers, with GC tail of unequal length attached to their 5' end, were designed based on 2 SNP loci (ITS101 and ITS296) of the internal transcribed spacer 1 (ITS1) sequences. The standard curve of $T_m-shift$ was established using the standard plasmids of A. ceylanicum (AceP) and A. tubaeforme (AtuP). The $T_m-shift$ method stability, sensitivity, and accuracy were tested with reference to the standard curve, and clinical fecal samples were also examined. The results demonstrated that the 2 sets of primers based on the 2 SNPs could accurately distinguish between A. ceylanicum and A. tubaeforme. The coefficient of variation (CV) of $T_m$- values of AceP and AtuP was 0.07% and 0.06% in ITS101 and was 0.06% and 0.08% in ITS296, respectively. The minimum detectable DNA concentration was $5.22{\times}10^{-6}$ and $5.28{\times}10^{-6}ng/{\mu}l$ samples of AceP and AtuP, respectively. The accuracy of $T_m-shift$ method reached 100% based on examination of 10 hookworm DNA samples with known species. In the clinical detection of hookworm in 69 stray cat fecal sample, the $T_m-shift$ detection results were consistent with the microscopic examination and successfully differentiated between the 2-hookworm species. In conclusion, the developed method is a rapid, sensitive and accurate technique and can provide a promising tool for clinical detection and epidemiological investigation of cat-derived hookworms.

Simple and Selective Flow Injection Catalytic Determination of Ruthenium with Spectrophotometric Detection (분광광도 검출법으로 간단하고 선택적인 흐름주입촉매법에의한 루테늄 정량)

  • A simple, selective and highly sensitive flow injection catalytic method was presented for determination of ruthenium based on its catalytic effect on the oxidation of pyronin B by periodate in pH=1.0. The reaction rate is controlled specrophotometricaly by monitoring the dye absorbance at 555 nm. The optimized conditions make it possible to determine ruthenium in the ranges of 0.1-10.0 ng/mL (r2=0.9982) and 10.0-50.0 ng/mL (r2=0.9934) with a detection limit of 0.04 ng/mL and a sample rate of 30±5 samples/h. Relative standard deviation for the results of five replicate measurements does not exceed 1.44%. The proposed method has been successfully applied for quantitation of ultra trace amounts of ruthenium in some environmental and biological samples.

Microsatellite multiplex PCR method for selective breeding studies in Pacific abalone (Haliotis discus hannai) (북방전복 (Haliotis discus hannai)의 선발육종 연구를 위한 microsatellite multiplex PCR법 개발)

  • The multiplex PCR system including six microsatellites from Haliotis discus hannai, consisting of dinucleotide and trinucleotide repeat units, is developed. The six loci were coamplified in a single reaction employing dye-labeled primers. Alleles from these loci were sized using an internal standard by automated sample processing in an ABI3100 Genetic Analyser. Amplified alleles in profiles containing selected microsatellites were typed clearly, providing easily interpretable results. In this results suggest that the presented multiplex PCR system may be a useful tool in a selective breeding program of H. discus hannai in which genetic identification will allow different genotypes to be reared together from fertilization. This should have a great impact as it will make selective breeding more efficient. Moreover, it will be useful in a variety of applications, including strain and hybrid identification, parentage assignment, pedigree reconstruction, estimating genetic diversity and/or inbreeding.

Low-shrinking composites. Are they reliable for bonding orthodontic retainers?

  • Objective: To evaluate the shear bond strength (SBS), fracture mode, wire pull out (WPO) resistance and microleakage between low-shrinking and conventional composites used as a lingual retainer adhesive. Methods: A total of 120 human mandibular incisor teeth, extracted for periodontal reasons, were collected. Sixty of them were separated into two groups. To determine the SBS, either Transbond-LR (3M-Unitek) or Silorane (3M-Espe) was applied to the lingual surface of the teeth by packing the material into standard cylindrical plastic matrices (Ultradent) to simulate the lingual retainer bonding area. To test WPO resistance, 20 samples were prepared for each composite where the wire was embedded in the composite materialand cured. Then tensile stress was applied until failure of the composite occurred. The remaining 60 teeth were divided into two groups and multi-stranded 0.0215-inch diameter wire was bonded with the same composites. Microleakage was evaluated by the dye penetration method. Statistical analyses were performed by Wilcoxon, Pearson chi-square, and Mann-Whitney-U tests at p < 0.05 level. Results: The SBS and WPO results were not statistically significant between the two groups. Significant differences were found between the groups in terms of fracture mode (p < 0.001). Greater percentages of the fractures showed mix type failure (85%) for Silorane and adhesive (60%) for Transbond-LR. Microleakage values were lower in low-shrinking composite than the control and this difference was found to be statistically significant (p < 0.001). Conclusions: Low-shrinking composite produced sufficient SBS, WPO and microleakage values on the etched enamel surfaces, when used as a lingual retainer composite.

Sentinel Node Biopsy Examination for Breast Cancer in a Routine Laboratory Practice: Results of a Pilot Study

  • Background: Examination of sentinel lymph node (SLN) biopsies provides accurate nodal staging for breast cancer and plays a key role in patient management. Procurement of SLNs and the methods used to process specimens are equally important. Increasing the level of detail in histopathological examination of SLNs increases detection of metastatic tumours but will also increase the burden of busy laboratories and thus may not be carried out routinely. Recommendation of a reasonable standard in SLN examination is required to ensure high sensitivity of results while maintaining a manageable practice workload. Materials and Methods: Twenty-four patients with clinically node-negative breast cancer were recruited. Combined radiotracer and blue dye methods were used for identification of SLNs. The nodes were thinly sliced and embedded. Serial sectioning and immunohistochemical (IHC) staining against AE1/AE3 were performed if initial H&E sections of the blocks were negative. Results: SLNs were successfully identified in all patients. Ten cases had nodal metastases with 7 detected in SLNs and 3 detected only in axillary nodes (false negative rate, FNR=30%). Some 5 out of 7 metastatic lesions in the SLNs (71.4%) were detected in initial sections of the thinly sliced tissue. Serial sectioning detected the remaining two cases with either micrometastases or isolated tumour cells (ITC). Conclusions: Thin slicing of tissue to 3-5mm thickness and serial sectioning improved the detection of micro and macro-metastases but the additional burden of serial sectioning gave low yield of micrometastases or ITC and may not be cost effective. IHC validation did not further increase sensitivity of detection. Therefore its use should only be limited to confirmation of suspicious lesions. False negative cases where SLNs were not involved could be due to skipped metastases to non-sentinel nodes or poor technique during procurement, resulting in missed detection of actual SLNs.

  • Capillary malformation is common vascular malformation. In case of facial capillary malformation, patients' cosmetic and functional deficits are quite significant. The standard treatment which has been applied so far for capillary malformation is pulsed dye laser with 585nm. But in case of advanced capillary malformation, surgical interventions are inevitable. The problem of large size facial capillary malformation is how to cover the remnant defect, which occurs after resection. In this case, authors have experienced surgical treatment of large size facial capillary malformation and covered the large facial defect with free thoracodorsal artery perforator flap. The flap was thick, so facial asymmetry remained after the first surgery. But with the secondary procedure, authors have made more symmetric figures. The patient was satisfied with the result. Using free flap to replace the defect after resection due to capillary malformation is useful for these kinds of cases.

Studies on the Anti-Inflammatory Effects of Clerodendron trichotomum Thunberg Leaves

  • Clerodendron trichotomum Thunberg Leaves (CTL) have been used for centuries in Chinese folk medicine for their anti-inflammatory properties. We have studied the anti-inflammatory effects of CTL extracts in rats, mice and in Raw 264.7 cells. 1 mg/kg solutions of the 30% and 60% methanol extracts of CTL were used and a 1 mg/kg of indomethacin was used as a positive anti-inflammatory standard; these were then administrated to rats. Carrageenan was injected subcutaneously to induce hind paw edema in rats. The result of carrageenan-induced rat paw oedema showed that a 1 mg/kg of the 30%, and 60% methanol fraction of CTL and 1 mg/kg of indomethacin inhibited the hind paw edema by 19.5%, 23.0%, and 20.5% respectively. The effect of CTL on inflammation in mice by a capillary permeability assay was examined by detecting Evans blue leakage from capillaries after the intraperitoneal injection of acetic acid, a potent inflammatory stimulus. The 60% methanol fraction of CTL inhibited Evans blue dye leakage by 47.0%, which was 10% higher than that of the inhibition of 1 mg/kg of indomethacin. Also, the 60% methanol fraction of CTL suppressed the prostaglandin $E_2$ ($PGE_2$) generation in RAW 264.7 macrophage cells after treatment with lipopolysaccharide (LPS) by as much as the inhibition of 1 mg/kg of indomethacin and this led to the synthesis of $PGE_2$ by COX-2 induction. The inhibition of the carrageenan-induced rat paw oedema, vascular permeability and the $PGE_2$ generation demonstrates that the 60% methanol fraction of CTL contains a potent anti-inflammatory activity.

  • It is more necessary to consider the various factors for developmenting visible PV module of alternative window than traditional PV module. It must have sufficient performance which is Tvis, daylighting, daylight factor, glare index. so that more needs to consider suitable plan and total evaluated technology. Under the this background. For using commonly a combination BIPV module system and Daylinghting that can alternative architectural window, our goal on this study is drawing proper window area ratio as the window by analyzing lighting performance and glare index depending on transmittance of DSSC. On this study, we drew the result about window area ratio that can apply in the building when applying DSSC in the window. In situation that window is alternated as curtain wall in atrium that has big Widow area, if applying red 15.8% DSSC of low transmittance, it is expect to proper because it is suitable illumination standard and doesn't occur a discomfort glare. In case of office, we propose to apply red 33.2% or blue 35.2% DSSC of high transmittance for no affecting lighting load. we expect to contribute to select proper and effective window when applying the window in the building by drawing the window area ratio that can apply in thee building depending on transmittance of DSSC and offering the glare index data.

Native Plants of the Islands of Chungcheongnam-do (충남 도서지역의 민속식물)

  • This study was carried out in order to catalogue the native plants of 17 islands of Chungcheongnam-do Province from March to October, 2011. 905 pages of field notes relating to the native plants were collected from approximately 50 local inhabitants, and were subsequently analysed. The identified native plants were of 274 taxa, composed of 246 species, 3 subspecies, 22 varieties and 3 forms of 219 genera under 84 families. In accordance with their usage, they were classified into; 169, edible; 134, medicinal; 67, ornamental; 17, spice; 7, aroma; 7, fuel; 6, oil; 5, dye; 4, nectar; 3, fiber; 1, resin; 26, other plants. Coincidence degree dependent on age between the standard name and the local name was the highest at 50's, and the lowest at 60's.
