Genetic analysis of clubroot resistance in Chinese cabbage using single spore isolate of Plasmodiophora brassicae and development of RAPD marker linked to its resistance gene

  • To identify inheritance of clubroot disease resistance genes in Chinese cabbage, seedling tests of $BC_1P_1,\;BC_1P_2$, and $F_2$ populations derived from $F_1$ hybrid(var. CR Saerona) using single spore isolate(race 4 identified with William's differential host) from Plasmodiophora brassciae were conducted. Resistance(R) and susceptible(S) plants segregated to 1:0 in backcross to the resistant parent. The $F_2$ population segregated in a 3(R):1(S) ratio. This result implied that the resistance of clubroot disease is controlled by a single dominant gene to the race 4 of P. brassicae in CR Saerona. To develop DNA markers linked to clubroot resistance genes, 185 plants of CR Saerona among $F_2$ populations were used. A total of 300 arbitrary decamer was applied to $F_2$ population using BSARAPD(Bulked segregant analysis-Randomly amplified polymorphic DNA). One RAPD marker linked to clubroot resistance gene in CR Saerona($OPJ_{1100}$) was identified. This marker was 3.1 cM in distance from resistance gene in $F_2$ population. This marker may be useful for a marker-assisted selection(MAS) and gene pyramiding of the clubroot disease resistant gene in Chinese cabbage breeding programs.

  • This study is for the consideration of the existence tendency of Kudoa septempunctata in olive flounder. In general, muscle has shown a strong PCR positive reaction in spores containing tissues rather than non-containing tissues. However, blood PCR results showed opposed tendency. In various organs of the tested fish containing spores in muscle tissue, heart had shown positive reaction along with muscle at PCR analysis. Muscle fiber necrosis was observed at the histological observation, and this degeneration was common in both samples. The one sample was the PCR positive muscle containing spore and the other was the PCR positive muscle non-containing spore. Both of muscle tissues indicated a positive reaction at ISH (in-situ hybridization) against K. septempunctata.

Isolation and Identification of Streptomyces sp. Producing Anti-vancomycin Resistant Staphylococcus aureus Substance (반코마이신 내성 Staphylococcus aureus 억제 물질 생산 Streptomyces sp.의 분리 및 동정)

  • An Actinomycetes producing an anti-VRSA (vancomycin-resistant Staphylococcus aureus) substance was isolated from soil. The cultural, morphological, physiological and phylogenetic analyses of an isolated strain were investigated for identification. Cultural characteristics based on ISP (International Streptomyces Project) were as follows: white aerial mycelium, yellow reverse side, and good growth on various medium. Also, the isolate did not produce the soluble pigment. Morphological characteristics were showed cylindrical spore chain and smooth spore surface by SEM (Scanning Electron Microscope). Physiological characteristics were showed LL-type by DAP isomer analysis and detected glycine, glutamic acid and alanine. A phylogenetic analysis of the 16S rDNA provided a clue that the isolated strain was actually a member of the genus Streptomyces, because the determined sequence exhibited a higher homology with Streptomyces echinatus. The isolate was identified to be a genus of Streptomyces sp.. The optimal culture conditions for the maximum production of anti-VRSA substance by Streptomyces sp. were attained in a culture medium composed of $2.0\%$ (w/v) glucose, and $0.4\%$ (w/v) yeast extract. The anti-VRSA substance was highly produced after 5 days of culture. Optimal pH and temperature conditions for the production of anti-VRSA substance were pH 7.0 and $28^{\circ}C$, respectively.

  • The cellular fatty acids and quinones of streptomycetes isolated from volcanic soils were analysed. The strains contained fatty acids of 14 to 17 carbon chains, and 12-methyltetradecanoic acid and 14 methylpentadecanoic acid were dominant in most strains. The total profiles consisted of 74% branched fatty acid family, 16.8% linear family and 8.2% unsaturated family. The largest cluster of grey spore meases defined by numerical classification was separated from the remainders in the principal component analysis, but the other clusters were overlapped with one another. In the analysis of respiratory quinones, all of the strains contained either the menaquinone of 9 isoprene units with 6 hydrogenations of 8 hydrogenations as the major species. The distribution of menaquinones among the clusters could provide an important key in the chemotaxonomy of streptomycetes.

A Pollen Analysis on the Peat of Paengsung Area, Kyonggi-do (京畿道 彭城地域의 土炭의 花紛分析)

  • Pollen analysis on a 320cm core collected from paengsung area (36°56′50"N, 127°03′10"E) in southwesterm korea was carried out to investigate the changes in past climate and vegetation in that area. In this research it was found that pollen zones are divided into two layers of substage IIb and IIc. Pollen zone IIb, the lower layer, was dominated by high percentages of deciduous broadleaved trees such as alnus, Quercus, Corylus, Betula, Carpinus and Salix. Trapa, Typhaceae and Nymphaceae growing in wet habitat and Chenpodiaceae were pornounced amont the nonrboreal pollens. At that time, it seems that it was warmer and more humid than present. In pollen zone IIc, the upper layer, pollen of Pinun was dominant and those of Artemisia, Gramineae, Compositae, Cyperaceae and spore increased while of Quercus, Betula and Nymphaceae decreased. It is supposed that the climate of this zone was drier that the lower laywer.

  • Bacillus anthracis(Ba) is a Gram-positive spore-forming bacterium that causes the disease anthrax. The feature of Ba is the presence of two large virulence plasmids, pXO1 and pXO2. Molecular genotyping of Ba has been difficult to the lack of polymorphic DNA marker. Ba isolated from Korea has been genotyped using various nucleotide analysis methods, such as 16s rDNA sequencing and multiple-locus variable-number tandem repeat (MLVA) analysis. We identified genotypes that represent a genetic lineage in the B1 cluster. This study emphasized the need to perform molecular genotyping when attempting to verify a strain-specific Ba.

Modelling Algae Transport in Coastal Areas with Marine Afforestation (바다숲 조성해역의 해조류 포자 확산모델링)

  • To arrange effectively artificial reefs for marine afforestation, tidal currents were analyzed by numerical experiments, and particle tracking based upon tidal currents were carried out to clarify the path of algae spore. The experiments were carried out by EFDC (Environmental Fluid Dynamics Code), and water column was vertically divided 10 layers. Tidal current patterns showed to be affected by main current at outside of study area, and circle currents of two were observed from analysis of residual currents. Particle tracking were experimented for 15 days at 2 installation places in which artificial reefs for marine afforestation would be deployed. According to the results of particle tracking experiment, particle movement at St.1 showed belt type along coastal line, and St.2 showed ellipse type at 300~500 m distant from coastal line. It suggest that artificial reefs for marine afforestation should be installed belt zone at station of St.1 and ellipse zone at St.2. Modelling algae transport was also tested to account for local dispersion of algae spore due to the suspended materials.

Complete Genome Sequence of Enterococcus faecalis CAUM157 Isolated from Raw Cow's Milk

  • Enterococcus faecalis CAUM157, isolated from raw cow's milk, is a Gram-positive, facultatively anaerobic, and non-spore-forming bacterium capable of inhabiting a wide range of environmental niches. E. faecalis CAUM157 was observed to produce a two-peptide bacteriocin that had a wide range of activity against several pathogens, including Listeria monocytogenes, Staphylococcus aureus, and periodontitis-causing bacteria. The whole genome of E. faecalis CAUM157 was sequenced using the PacBio RS II platform, revealing a genome size of 2,972,812 bp with a G+C ratio of 37.44%, assembled into two contigs. Annotation analysis revealed 2,830 coding sequences, 12 rRNAs, and 61 tRNAs. Further, in silico analysis of the genome identified a single bacteriocin gene cluster.

  • The importance and diversity of the genus Stemphylium highlights the need for accurate identification of species. However, many Stemphylium isolates have been misidentified due to the use of spore size as the only identifying character. Molecular phylogenetic analyses were performed on fifty-four isolates covering 9 Stemphylium species collected in Korea. Phylogenetic analysis of the translation elongation factor -1 alpha (EF-1) and the calmodulin gene sequence data showed that Stemphylium species were segregated into seven distinct groups, most of w hichcorrelated with species identified by morphology. Analysis of EF-1 in particular was useful for establishing well- supported relationships among the species of Stemphylium.

  • Identification of the Actinomycetes isolate strain No. 497 producing an actinomycin antibiotic MT-497 was performed by ISP and chemotaxonomic methods. The strain Nu. 497 formed various shapes of sclerotia and smooth surface spore. Menaquinone MK-9 ($H_6, H_8$) and iso-, anteiso-branched $C_{15}C_{17}$ fatty acids were detected from whole cell extract. The wall chemotype of stram No. 497 was decided as wall chemotype I from the analysis of DAP isomer, peptidoglycan type and sugar pattern. From these morphological, chemotaxonomic characteristics and analysis of various physiological characteristics. the strain No. 497 was identified as Streptomyces nigrifaciens.
