수소 생산 균주 ES392sms 부산 소재 동의대학교에 위치한 연못 담수에서 분리하였다. 세포는 직경 $0.6\;{\mu}m$, 길이 $1.4\;{\mu}m$의 간균이고 편모와 포자를 형성하지 않았다. 분리된 균주의 16s rRNA 염기서열과 생화학적 특성을 바탕으로 계통학적으로 분류한 결과, ES392 균주는 Enterobacter sp.에 속하는 것으로 동정되었다. 수소생산을 위한 생육최적 pH와 온도는 각각 7.5와 $35^{\circ}C$이었다. 분리한 Enterobacter sp. ES392 균주의 수소생산을 최대화 하기 위해 배지성분을 최적화하였다. 그 결과 4%(w/v) sucrose, 0.5%(w/v) yeast extract, 50 mM potassium phosphate를 첨가한 배지 조건에서 최대수소생산량을 나타내었다. 회분식 배양 조건에서 최대 수소 생산량은 3481 mL/L이었고, 수소생산수율은 1.33mol/mol sucrose를 나타내었다.
A novel ${\beta}$-proteobacterium, designated BXN5-$27^T$, was isolated from soil of a ginseng field of Baekdu Mountain in China, and was characterized using a polyphasic approach. The strain was Gram-staining-negative, aerobic, motile, non-spore-forming, and rod shaped. Strain BXN5-$27^T$ exhibited ${\beta}$-glucosidase activity that was responsible for its ability to transform ginsenoside $Rb_1$ (one of the dominant active components of ginseng) to compound Rd. Phylogenetic analysis based on 16S rRNA gene sequences showed that this strain belonged to the family Comamonadaceae; it was most closely related to Ramlibacter henchirensis $TMB834^T$ and Ramlibacter tataouinensis$TTB310^T$ (96.4% and 96.3% similarity, respectively). The G+C content of the genomic DNA was 68.1%. The major menaquinone was Q-8. The major fatty acids were $C_{16:0}$, summed feature 4 (comprising $C_{16:1}$${\omega}7c$ and/or iso-$C_{15:0}$ 2OH), and $C_{17:0}$ cyclo. Genomic and chemotaxonomic data supported the affiliation of strain BXN5-$27^T$ to the genus Ramlibacter. However, physiological and biochemical tests differentiated it phenotypically from the other established species of Ramlibacter. Therefore, the isolate represents a novel species, for which the name Ramlibacter ginsenosidimutans sp. nov. is proposed, with the type strain being BXN5-$27^T$ (=DSM $23480^T$ = LMG $24525^T$ = KCTC $22276^T$).

A strain of bacterium producing antifungal antibiotic was isolated and identification of the strain was attempted. We could identify the bacterium as being a Bacillus sp., based on morphological observation, physiological characteristics, and 16S rDNA sequence analysis, thus leading us to designate the strain as Bacillus sp. AH-E-1. The strain showed potent antibiotic activity against phytopathogenic and human pathogenic fungi by inducing mycelial distortion and swelling and inhibiting spore germination. The antibiotic metabolite produced by the strain demonstrated excellent thermal and pH (2-11) stability, but was labile to autoclaving. From these results, we could find a broader antifungal activity of Bacillus genus. Isolation and characterization of the active agent produced by the strain are under progress.
2013년 8월, 경기도 화성시에 위치한 과수원에서 체리 과실에 회색의 포자 덩어리가 발생하는 잿빛무늬병 증상이 관찰되었다. 과실에 갈색의 수침상 원형 병반이 나타나고, 진전되면 과실 전체가 부패하였다. 부패된 과실에 회갈색 내지 연회색의 포자덩어리가 관찰되었다. 병든 체리 과실에서 2개의 Monilinia균을 분리하여 균학적 특성을 조사한 결과 M. fructicola로 동정되었다. 이러한 결과를 확인하기 위하여 ITS 영역의 염기서열을 분석하였다. 그 결과, 분리균은 모두 M. fructicola로 확인되었다. 체리 과실을 상처구와 무상처구로 구분하고, 포자현탁액을 접종하여 병원성을 확인한 결과, 접종 7일 후 상처의 유무에 관계없이 분리균을 접종한 부위에서 수침상의 둥근 병반이 관찰되었다. 따라서 본 병을 Monilinia fructicola에 의한 체리잿빛무늬병으로 명명하며, 이 균에 의한 잿빛무늬병의 발생을 국내 최초로 보고한다.
Perilla rust is a damaging disease in perilla cultivation in Korea. Its causal agent was identified as Coleosporium plectranthi based on descriptions of morphological characteristics of spores and spore-producing fruiting structures(in uredinial and telial stages from perilla and in aecial stage from the alternate host pine) collected in 15 locations in Korea during the disease survey from 2004 to 2006. These characteristics were yellow or orange uredinium; globose or ellipsoid urediniospore of $20.8{\mu}m{\times}18{\mu}m$ in size; verruca of $0.3mm{\times}1.2mm$; orange telium; one-celled, oblong ellipsoid teliospore of $63.1{\mu}m{\times}19.7{\mu}m$ with one-layered crusts or four-celled(when mature), internal basidium of $64.2{\mu}m{\times}19.7{\mu}m$; ellipsoid to globoid basidiospore of $20.3{\mu}m{\times}12{\mu}m$; type 2 spermogonium; yellow, broadly ellipsoid peridial cell of $35.6{\mu}m{\times}23.1{\mu}m$; and broadly ellipsoidal or subglobose aeciospore of $25.9{\mu}m{\times}18.8{\mu}m$. Phylogenetic analysis of 28S rDNA sequences revealed the closest relatedness to those of the genus Coleosporium, a monophyletic group distinguished from other rust fungi and divided into two main lineages, one of which was C. plectranthi grouped with high bootstrap value(96%). In pathogenicity test, both aeciospores and urediniospores caused rust development on perilla leaves. This is the first description of C. plectranthi causing perilla rust with the first findings of its telial stage on perilla and the first rust disease on the aecial host in Pinus densiflora. These aspects would provide basic information for the development of control measures of the disease.
We have detected the slime mold, Diachea leucopodia (GNU06-10) in a strawberry greenhouse located in Sancheong-gun, Gyeongnam. Typical fruiting bodies had developed gregariously on the strawberry leaves, petioles, and plant debris on ground soil habitat, and also surprisingly on plastic pipes and a vinyl covering. Field samples were examined via stereomicroscopy, light microscopy, and SEM for the determination of morphological characteristics. Dark-brown to black spores formed gregariously within the stipitate cylindrical sporangium, and were covered by an iridescent peridium, which may be intact at maturity, or may have disintegrated. The upper portion of the peridium generally breaks up to expose the spores, whereas the lower portion was usually persistent. The results of energy dispersive X-ray spectrometer (EDS) analysis showed that lime was present in the stalk and columella but absent from the spores, capillitium, and peridium. The above characteristics confirm its taxonomic position in the genus Diachea. However, this genus is intermediate in character between the Physarales and Stemonitales of the Myxogastromycetidae. Hence, this genus had been classified as a member of the Stemonitales until the mid-1970's, on the basis of its iridescent peridium and noncalcareous capillitial system, similar to Comatricha of the Stemonitaceae. By way of contrast, emphasis on morphological characteristics, most notably the calcareous stalk and typical columella, places Diachea within the order Physarales. The presence of a phaneroplasmodium during the trophic stage and lime deposition in its sporophores, as was confirmed in this work, supported the inclusion of Diachea in the Physarales, and the noncalcareous capillitial system verified its identification as a member of the Didymiaceae. Further characteristics of the species D. leucopodia include the following: phaneroplasmodium, spore globose 7.5 ${\mu}m$ in diameter, very minutely roughened; sporangia $500{\mu}m\times1mm$, more or less cylindrical, gregarious, stalked 1.2mm; stalk and columella white.
호열성 미생물을 검토하기 위하여 전국 각지로부터 토양과 두엄을 채취하여 그로부터 호열성 미생물을 분리하였다. 토양과 두엄으로부터 분리된 호열성 미생물 1250여 균주를 선별하였고, 이들을 대상으로 미생물이 생산하는 효소 활성을 검토하여 호열성 전분 분해 효소를 생산하는 1주의 미생물을 확인하였다. 확인된 1주의 미생물을 strain 2719라 명명하였다. Strain 2719 균주는 형태학적으로 gram 양성 간균의 특징을 나타냈고, 균주의 표면은 매끄럽지 않았으며, 비교적 다양한 길이를 가지고 있었다. 또한 다른 gram 양성 간균들에 비해서 많은 수의 균사들이 각 균주들 사이에 복잡하게 얽혀있었다. 생화학적 특성을 확인한 결과 catalase 양성, glucose 발효, arabinose 발효, mannitol 발효, casein gelatin starch 가수분해의 특징을 가지고 있었으며, 이는 Bacillus sp.로 추정되었다. 생육 pH의 범위는 pH 6-pH 8범위에서 생육이 가능했으며, 생육 온도의 범위는 50-70${^{\circ}C}$였다. 16S rDNA sequence 분석결과 Bacillus thermoglucosidasius의 16S rDNA와 99.52%가 일치하였으나, sequence의 일부분이 다른 부분이 있고, 생육 특성에서 약간의 차이를 보였다. 또한 gene bank에 등록되어 있는 균주들의 16S rDNA sequence들과 비교하여도 일치하는 균주는 확인되지 않았다. 이와 같은 실험결과에 따라 2719 균주는 기존에 발표되지는 않았으나, Bacillus thermoglucosidasius와 매우 유사한 균주로 판단되어 Bacillus thermoglucosidasius 2719로 명명하였다.
Background: The microsporidian parasite Nosema ceranae is a global problem in honeybee populations and is known to cause winter mortality. A sensitive and rapid tool for stable quantitative detection is necessary to establish further research related to the diagnosis, prevention, and treatment of this pathogen. Objectives: The present study aimed to develop a quantitative method that incorporates ultra-rapid real-time quantitative polymerase chain reaction (UR-qPCR) for the rapid enumeration of N. ceranae in infected bees. Methods: A procedure for UR-qPCR detection of N. ceranae was developed, and the advantages of molecular detection were evaluated in comparison with microscopic enumeration. Results: UR-qPCR was more sensitive than microscopic enumeration for detecting two copies of N. ceranae DNA and 24 spores per bee. Meanwhile, the limit of detection by microscopy was 2.40 × 104 spores/bee, and the stable detection level was ≥ 2.40 × 105 spores/bee. The results of N. ceranae calculations from the infected honeybees and purified spores by UR-qPCR showed that the DNA copy number was approximately 8-fold higher than the spore count. Additionally, honeybees infected with N. ceranae with 2.74 × 104 copies of N. ceranae DNA were incapable of detection by microscopy. The results of quantitative analysis using UR-qPCR were accomplished within 20 min. Conclusions: UR-qPCR is expected to be the most rapid molecular method for Nosema detection and has been developed for diagnosing nosemosis at low levels of infection.
In recent years, myxosporean infection from the cultured olive flounders Paralichthys olivaceus, have been frequently observed in Jeju island, South Korea. This study aimed to compare histopathological and molecular-biological methods of examining myxosporean infection from these flounders. Samples were obtained from affected individuals exhibiting emaciation or abdominal distention and a polymerase chain reaction (PCR) indicative of Parvicapsular anisocaudata, Enteromyxum leei and Kudoa septempunctata were initiated. Histopathological examination were conducted with H&E stained tissue sections, and then in-situ hybridization (ISH) reaction were processed with selected sections using P. anisocaudata, E. leei, K. septempunctata and Scuticociliate probes. Renal and intestinal tissue degeneration were common symptoms associated with all samples. Sever glomerular and renal tubular degeneration were evident, as were intestinal epithelial desquamation and spore formation in the epithelial cells. The results of conventional PCR analysis and ISH reactions revealed differences, and we suspect that various microparasites may have been associated with the symptoms manifested.
