Effect of Dexamethasone on the Surface Expression of Marker Molecules and Differentiation of Murine B Cells (덱사메타손이 생쥐 B세포의 세포 표면 인식자와 분화에 미치는 영향)

  • Background: There are at least two different subsets of B cells, B-1 and B-2. The characteristic features and function of B-2 cells in addition to the effect of steroids on B-2 cells are well-known. Although B-1 cells have different features and functions from B-2 cells, the effect of steroids on B-1 cells is not completely understood. Therefore, this study examined the effects of dexamethasone on peritoneal (or B-1 cells) and splenic B cells (or B-2 cells). Methods: Purified B cells were obtained from the peritoneal fluid and the spleens of mice. The isolated B cells were cultured in a medium and after adding different concentrations of dexamaethasone. The cell survival rate was measured by flow cytometry using propidium iodide. The expression level of the B cell surface marker was analyzed by flow cytometry. During the culture of these cells, immunoglobulin secreted into the culture supernatants was evaluated by an enzyme-linked immunosorbent assay. Results: The survival rate of peritoneal and splenic B cells decreased with increasing dexamethasone concentration. However, the rate of peritofieal B cell apoptosis was lower than that of splenic B cells. CDS and B7.1 expression in peritoneal B cells and CD23 and sIgM expression in splenic B cells after the dexamethasone treatment were reduced. When B cells were treated with dexamethasone, the spontaneous IgM secretion decreased with increasing dexamethasone concentration. Conclusion: Dexamethasone induces apoptosis in peritoneal and splenic B cells. However, peritoneal B cells are less sensitive to dexamethasone. The dexamethasone suppressed expression of the surface markers in peritoneal B cells is different from those in splenic B cells.

Effect of T-2 Toxin on the Mitogen-Induced Blastogenesis in Chick Splenic Cell (T-2 Toxin이 병아리 비장세포의 유전질 발생에 미치는 영향)

  • The effects of T-2 toxin on mitogen-induced blastogenesis of chick splenic cells were investigated. The [$^3H$] thymidine incorporation in splenic cells stimulated by lipopolysaccharide and concanavalin A were equally inhibited as the concentration of T-2 toxin was increased. The effective dose of T-2 toxin causing a 50% reduction of [$^3H$] thymidine incorporation was inbetween 1.0 and 5.0 ng/ml for both mitogens. Mitogen-induced blastogenesis in chick splenic cells showed differences among experimental groups with different exposure time of T-2 toxin, exhibiting the most inhibition in the experimental group exposed to T-2 toxin at both embryonic and chick periods.

Splenic Mast Cell Tumors in Two Cats

  • Two 11-year-old cats, female Korean shorthair cat and male Siamese cat, with abdominal distention were presented to the local animal hospitals. Radiographic and ultrasonographic examinations revealed moderate to severe splenomegaly in both cats. In Korean shorthair cat, multiple masses were also existed on the anal and facial skin. Surgically excised whole spleens of two cats were requested for histopathologic examination. Histopathologically, numerous neoplastic round cells with cytoplasmic fine granules were widely infiltrated in the splenic parenchyma. The cytoplasmic granules were metachromatic on toluidine blue staining. These splenic masses were diagnosed as splenic mast cell tumors. Among them, Korean shorthair cat was remained healthy for at least 1 year after splenectomy. Because of no visiting of owner, we were only able to know the information for Siamese cat until 10 months after the splenectomy. To our best knowledge, this is the first detail case reports for splenic mast cell tumors in cats in Korea.

B-1 Cells Differ from Conventional B (B-2) Cells: Difference in Proliferation (B세포의 증식에 있어 B-1 임파구와 B-2 임파구의 차이점에 대한 연구)

  • Background: B-1 cells differ from conventional B-2 cells both phenotypically and functionally. The aim of this study was to investigate the difference between peritoneal B-1 cells and splenic B-2 cells in proliferation. Methods: We obtained sorted B-1 cells from peritoneal fluid and B-2 cells from spleens of mice. During the culture of these cells, immunoglobulin secreted into the culture supernatants was evaluated by enzymelinked immunosorbent assay. Entering of S phase in response to LPS-stimuli was measured by proliferative assay. Results: Spontaneous Immunoglobulin M production occurred in peritoneal B-1 cells but not in splenic B-2 cells. LPS stimulated peritoneal B-1 cells secreted IgM at day 1, but splenic B-2 cells at day 2. In thymidine incorporation, peritoneal B-1 cells entered actively S phase after 24hours LPS-stimulation but splenic B-2 cells entered actively S phase after 48 hours. Conclusion: IgM secretion and S phase entering occurred early in peritoneal B-1 cells compared to splenic B-2 cells.

  • Objective: Experimental studies were done to research the effects of Jahap-san(zige-san) on the allergic reaction Methods: For measuring the response about the antiallergic effect we investigated cytokines m-RNA expression of murine splenic B cell, production in anti-CD40 mAb-stimulated murine splenic B cells and the histamine release in IC-2 cells by anti-CD40 mAb-stimulated murine splenic B cells. Results: The extract of Jahap-san(zige-san) revealed significant decrease effect on cytokines m-RNA expression of murine splenic B cell, production in anti-CD40 mAb-stimulated murine splenic B cells and the histamine release in IC-2 cells by anti-CD40 mAb-stimulated murine splenic B cells. Conculusion : The extract of Jahap-san(zige-san) revealed significant effect on the antiallergy.

Radiation Protective Effect of vitamin C and Cysteine on DNA Damage in Mice Splenic Lymphocytes by Single Cell Gel Electrophoresis Assay (단세포 겔 전기영동법을 이용한 생쥐 비장 림프구 DNA 손상에 대한 비타민 C 및 시스테인의 방사선 방어효과)

  • The alkaline comet assay, employing a single-cell gel electrophoresis(SCGE), is a rapid, simple and sensitive technique for visualizing and measuring DNA damage leading to strand breakage in individual mammalian cells. The protecting effect of pretreatment with vitamin C and cysteine on the DNA damage of gamma ray was investigated in mice splenic lymphocytes. Vitamin C and cysteine were administered orally for five consecutive days before irradiation. Four week old ICR male mice were irradiated wish 3.5Gy of γ-radiation and were sacrificed 3 days later. Spleens were taken for DNA damage examination by Comet assay and the tail moments of DNA single -strand breaks in tole splenic lymphocytes were evaluated. The results show that pretreatment with vitamin C and cysteine were effective in protecting against DNA damage by gamma ray. Administration of antioxidants like vitamin C and cysteine to mice before irradiation was effective in reducing the tail moment of splenic lymphocytes DNA.

Studies on Kagamboatang(KGBT) on the Hematopoiesis and Proliferation of Immune Function in Mice (가감보아탕(加減補兒湯)의 조혈(造血) 및 면역증진(免疫增進)에 관한 연구(硏究))

  • The KGBT has been used to weak children with anorexia, fatigue, and growth retardation. This study was carried out to prove the effects of the hematopoiesis and the immune proliferation by KGBT. Previously, C57BL/6 mice was treated with cyclophosphamide(100mg/kg) for leukopenia, and then administered KGBT (concentration is 1.37 g/kg, 504 mg/kg, and 137 mg/kg) to the treated mice. The mice was analyzed expression of thrombopoietin(TPO), stem cell factor(SCF) and interleukin-3 from bone marrow cell, interleukin-10 (IL-10), and interferon-$ {\gamma}$(INF-${\gamma}$) from splenic cell, and NOSⅡ gene from macrophage using by RT-PCR. Also proliferation of immune cell was analyzed using 3H-thymidine uptake and flow cytometery in splenic cells. The results were obtained as follows ; 1. The total number of WBC, RBC and PLT was increased in the KGBT treated group than in the control group. 2. In vitro, the proliferation of splenic cells was increased in normal, control, and KGBT treated group. And Administration of KGBT was reduced the cytotoxicity by CTX. 3. In bone marrow cell, the gene expression of immune regulatory factor that associated with hematopoiesis, such as TPO, SCF, and IL-13 was increased in the KGBT treated group than control. 4 The titer of hemagglutinin and hemolysin was increased in the KGBT treated group than control. 5. In analysis of positive leucocytes from splenic cell of BALB/c mice, the subpopulation percent of CD4+, CD8+,and CD19+ was increased in the KGBT treated group than control.   • Objectives This experimental study was carried out to evaluate the effects of Phellinus Linteus on immune activation. Methods In order to investigate the effect of Phellinus Linteus, the following was performed: The fraction of $CD4^+$, $CD8^+$, $CD19^+$ in splenic cell, gene expression of IL-12 (p35), IL-12 (p40) , $IFN-{\gamma}$, and splenic cell proliferation by PL-E. Results PL-E helped $CD4^+$, $CD8^+$, $CD19^+$ expression more effectively compared with control group. PL-E helped IL-12 (p35), IL-12 (p40), $IFN-{\gamma}$ gene expression in splenic cells more effectively compared with control group. PL-E proliferated splenic cells more effectively compared with control group. Conclusions: It is suggested that PL-E is able to activate immune response system.

Pro-inflammatory Cytokine Expression of Spleen Dendritic Cells in Mouse Toxoplasmosis

  • Dendritic cells have been known as a member of strong innate immune cells against infectious organelles. In this study, we evaluated the cytokine expression of splenic dendritic cells in chronic mouse toxoplasmosis by tissue cyst-forming Me49 strain and demonstrated the distribution of lymphoid dendritic cells by fluorescence-activated cell sorter (FACS). Pro-inflammatory cytokines, such as IL-$1{\alpha}$, IL-$1{\beta}$, IL-6, and IL-10 increased rapidly at week 1 post-infection (PI) and peaked at week 3 PI. Serum IL-10 level followed the similar patterns. FACS analysis showed that the number of $CD8{\alpha}^+/CD11c^+$ splenic dendritic cells increased at week 1 and peaked at week 3 PI. In conclusion, mouse splenic dendritic cells showed early and rapid cytokine changes and may have important protective roles in early phases of murine toxoplasmosis.

Indolent B-Cell Lymphoid Malignancy in the Spleen of a Man Who Handled Benzene: Splenic Marginal Zone Lymphoma

  • We present the case of a 45-year-old man with a history of benzene exposure who developed splenic marginal zone lymphoma. For 6 years, he had worked in an enclosed space cleaning instruments with benzene. He was diagnosed with splenic marginal zone lymphoma 19 years after retirement. During his time of working in the laboratory in the 1980s, working environments were not monitored for hazardous materials. We indirectly estimated the cumulative level of past benzene exposure using job-exposure matrices and technical assumptions. Care must be taken in investigating the relevance of occupational benzene exposure in the occurrence of indolent B-cell lymphoma. Because of the long latency period and because occupational measurement data do not exist for the period during the patient's exposure, the epidemiological impact of benzene exposure may be underestimated.
