자외선분광기를 이용한 미생물 세포 생촉매의 에폭사이드 가수분해효소 활성평가 (UV Spectrometric Assay of Epoxide Hydrolase Activity of Microbial Cell Biocatalysts)

  • 미생물 세포 유래의 에폭사이드 가수분해 효소 활성을 효율적으로 분석하기 위하여 UV 분광기 측정법을 최적화하였다. 세포 생촉매의 에폭사이드 가수분해 활성에 의해 p-nitrostyrene oxide (pNSO) 기질이 p-nitrostyrene diol로 전환된 양을 측정함으로써 에폭사이드 가수분해 활성을 평가하였다. Rhodosporidium toruloides를 생촉매로 사용하고 pNSO에 대한 입체선택적 가수분해 동력학을 UV 분광기 측정법으로 분석한 결과, $K_m$$V_m$ 값을 $2.457nmol/min{\cdot}mg$ 및 1.078 mM로 각각 결정할 수 있었다.

  • 시금공정의 시료중의 금의 정성 및 정량을 열중성자에 의한 방사화분석으로 조사하였다. 회수률은 81.0∼93.6%이고 결과는 재현성이 좋았다. 정양적 결과는 ${\gamma}$-Spectrometric photopeak-area counting으로 얻었으며 속중성자반응으로 인한 간섭은 무시할 수 있었다.

  • 다양한 라세믹 에폭사이드 기질에 대한 입체선택적 가수분해 반응을 촉매하는 epoxide hydrolase 활성을 측정할 수 있는 미생물 세포 기반의 자외선 활성 분석법을 최적화하였다. 2.5 mg/ml의 세포 농도에서도 비교적 쉽게 흡광도 변화량을 인식할 수 있는 흡광도 범위인 0.5 이상을 얻을 수 있고, 반응 동력학 분석에도 응용할 수 있었다. 기존의 GC, HPLC 분석 법 보다 분석 시간을 줄일 수 있으며, 효소를 별도로 분리$\cdot$정제하지 않고 미생물 세포 자체의 epoxide hydrolase활성 분석이 가능하므로 상업적 특성이 우수한 epoxide hydrolase을 가진 미생물을 효율적으로 선별하는데 응용될 수 있을 것으로 기대된다.

Simultaneous Determination of Benzidine, Acetylbenzidine and di-Acetylbenzidine in Rat Urine

  • A gas chromatography/mass spectrometric assay method has been developed for the simultaneous determination of benzidine (BZ), N-acetyl benzidine (ABZ) and N,N-diacetyl benzidine (DABZ) in rat urine. BZ, ABZ and DABZ were extracted from urine at pH 8 with ethyl ether. Conjugated urinary metabolites were extracted at pH 8 after hydrolysis with 1 M HCl for 30 min at 100 $^{\circ}C.$ The dried extract was dissolved in 100 ${\mu}{\ell}$ of ethylacetate and then injected in gas chromatography-mass spectrometric (GC-MS) system without further purification or modification. BZ, ABZ and DABZ have good chromatographic properties and offer very sensitive response for the EI-MS (SIM) without any derivatization. The recoveries for BZ, ABZ and DABZ were about 98.0, 81.8 and 71.4%, respectively, at pH 8.0 and the concentration of 5.0 ng/mL. The coefficients of variation of BZ and ABZ were less than 9.5% from 0.1 to 100 ng/mL and that of DABZ was less than 13% in the same concentration range. The detection limits of the assay were 0.01 ng/mL for both BZ and ABZ, and 0.05 ng/mL for DABZ in urine or plasma 1.0 mL.

Determination of Methyl Bromide Used for the Preservation of Cultural Materials from Insects

  • A thermal desorption-gas chromatography/mass spectrometric method was developed for the determination of methyl bromide in fumes formed during preservation of cultural materials from insects. Methyl bromide in lunes was collected by drawing 10ι of air through the adsorbent tube filled with a solid adsorbent (Chromosorb). The air samples were analyzed by using a special thermal desorption device and GC/MS determination. The recovery of methyl bromide by air sampling was 88% and the detection limit of the assay was 0.1 pptv based upon assayed air of 10ι. The method was applied to the determination of fumed methyl bromide used for the preservation of papers in a library. The mean concentration of methyl bromide determined in a library was 280.2 $\pm$ 25.4 pptv.

Determination of MTBE, TBA and BTEX in Soil by Headspace Gas Chromatography-Mass Spectrometry

  • A headspace gas chromatographic mass spectrometric (GC-MS) assay method was developed for the simultaneous determination of methyl tertiary butyl ether (MTBE), $tert$-butyl alcohol (TBA) and benzene, toluene, ethyl benzene and xylene (BTEX) in soil contaminated with gasoline. 2 g of soil sample were placed in a 10 mL headspace vial filled with 5 mL of phosphoric acid solution (pH 3) saturated with NaCl, and the solution was spiked with fluorobenzene as an internal standard and sealed with a cap. The vial was heated in a heating block for 40 min at $80^{\circ}C$. The detection limits of the assay were 0.08-0.12 ${\mu}g$/kg for the analytes. For five independent determinations at 10 and 50 ${\mu}g$/kg, the relative standard deviations were less than 10%. The method was used to analyze fifty six soil samples collected from various regions contaminated with gasoline in Korea. The developed method may be valuable for the monitoring of the analytes in soil.
