Fluorine-18 is considered to be the radionuclide of choice for positron emission tomography (PET). Thus, the development of small molecule-based radiopharmaceuticals for use in diagnostic imaging relies heavily on efficient radiofluorination techniques. Until the early 2000s, diaryliodonium salts and aryliodonium ylides were widely employed as labeling precursors to yield aromatic PET radiotracers with cyclotron-produced [18F]fluoride ion. Rapid recent progress in the development of efficient borylation methods has led to a paradigm shift in 18F-labeling methods. In addition, deoxyfluorination has attracted a great deal of interest as an alternative approach to aryl ring activation with 18F-. In this review, methods for radiolabel development are discussed with a specific focus on the progress made in the last 5 years. Other interesting 18F-based protocols are also briefly introduced. New methods for exploiting 18F- are expected to increase the number of 18F-labeling methods, to allow applications in a range of chemical environments.
Clinical and preclinical in vivo immune cell imaging approaches have been used to study immune cell proliferation, apoptosis and interaction at the microscopic (intra-vital imaging) and macroscopic (whole-body imaging) level by use of ex vivo or in vivo labeling method. A series of imaging techniques ranging from non-radiation based techniques such as optical imaging, MRI, and ultrasound to radiation based CT/nuclear imaging can be used for in vivo immune cell tracking. These imaging modalities highlight the intrinsic behavior of different immune cell populations in physiological context. Fluorescent, radioactive or paramagnetic probes can be used in direct labeling protocols to monitor the specific cell population. Reporter genes can also be used for genetic, indirect labeling protocols to track the fate of a given cell subpopulation in vivo. In this review, we summarized several methods dealing with dendritic cell, macrophage, and T lymphocyte specifically labeled for different macroscopic whole-body imaging techniques both for the study of their physiological function and in the context of immunotherapy to exploit imaging-derived information and immune-based treatments.

The rotation of the flagellar motor is powered by the electrochemical gradient of specific ions across the cytoplasmic membrane. Recently, the gents of the Na'-driven motor have been cloned from marine bacterium of Vibrio sp. and some of the motor proteins have been purified and characterized. Also, motx gene encoding a channel component of the sodium type flagellar motor was identified from Vibrio Huuiaiis (KTCC 2473). The amino acid sequence of MotX protein from V, Huvialis shared 90, 85, $85\%$ identity with V, cholerae, V. alginolyticus, V parahaemolyticus, respectively. We have studied the localization of the expressed MotX protein in Escherichia coli by immune-gold labeling of ultra-thin frozen section. Our observation of the expressed protein indicated that MotX protein could be existed as attachment to inner membrane in E. coli.
A robot usually adopts ANN (artificial neural network)-based object detection and instance segmentation algorithms to recognize objects but creating datasets for these algorithms requires high labeling costs because the dataset should be manually labeled. In order to lower the labeling cost, a new scheme is proposed that can automatically generate a training images and label them for specific objects. This scheme uses an instance segmentation algorithm trained to give the masks of unknown objects, so that they can be obtained in a simple environment. The RGB images of objects can be obtained by using these masks, and it is necessary to label the classes of objects through a human supervision. After obtaining object images, they are synthesized with various background images to create new images. Labeling the synthesized images is performed automatically using the masks and previously input object classes. In addition, human intervention is further reduced by using the robot arm to collect object images. The experiments show that the performance of instance segmentation trained through the proposed method is equivalent to that of the real dataset and that the time required to generate the dataset can be significantly reduced.

The classical ABC (avidin-biotin peroxidase complex) method for immunohistochemistry in the paraffin-embedded tissues bring into being disadvantage such as low sensitivity of antigen detection and highly background. The biotinyl-tyramide conjugation recently introduced for sensitive immunohistochemistry was applied to light microscopy in paraffin-embedded pancreatic and liver tissues. The protocol consists of an indirect method in which 4-5㎛ tissue sections are reacted successively within a specific primary antibody, followed by a biotinylated secondary antibody, streptavidin-horseradich peroxidase (HRP), and then finally with biotinyl-tyramide. The labeling obtained for insulin and collagen antigen tested in pancreatic and liver tissues, respectively, was found to be highly specific with the labeling for each antigen confined to its particular cellular compartment. In this study, fish (flounder) serum was specially applied to remove nonspecific binding. Background levels and nospecific deposition of the staining were negligible. This results suggest that super-signal induction method by biotinyl-tyramide conjugate can readily applied to antigen detection of the paraffin-embedded tissues.
The authentication and implications of misleading labeling in milk and dairy products is important to protect against cheating consumers from adulteration and to alert sensitive consumers to any undeclared potential allergens. This need to support milk and dairy products labeling has led to the development of specific analytical techniques for the analysis of milk and dairy products ingredients. Recently, several methods based on polymerase chain reaction (PCR), including restriction fragment length polymorphism (PCR-RFLP), multiplex PCR, species-specific PCR, and real-time PCR, have been proposed as useful means for identifying species of origin in milk and dairy products, as well as quantifying and detecting any adulteration. These methods have particular advantages owing to their high specificity and sensitivity, as well as rapid processing time. In this review, we provide an updated and extensive overview of the PCR-based methods used for milk and dairy products authentication with a particular focus on the application of PCR methods to detect adulteration.
${\alpha}$-Expansins are bound to the cell wall of plants and can be solubilized with an extraction buffer containing 1 M NaCl. Localization of ${\alpha}$-expansins in the cell wall was confirmed by immunogold labeling and electron microscopy. The subcellular localization of vegetative ${\beta}$-expansins has not yet been studied. Using antibodies specific for OsEXPB3, a vegetative ${\beta}$-expansin of rice (Oryza sativa L.), we found that OsEXPB3 is tightly bound to the cell wall and, unlike ${\alpha}$-expansins, cannot be solubilized with extraction buffer containing 1 M NaCl. OsEXPB3 protein could only be extracted with buffer containing SDS. The subcellular localization of the OsEXPB3 protein was confirmed by immunogold labeling and electron microscopy. Gold particles were mainly distributed over the primary cell walls. Immunohistochemistry showed that OsEXPB3 is present in all regions of the coleoptile and root tissues tested.
Based on the specificty of recognition of the vacuolar arginine transporter, N-p-nitrobenzoxycarbonyl (NBZ)-L-arginyl diazomethane was synthesized and used as an affinity label specific for the arginine transporter. This arginyl derivative ingibited both ATP-dependent and independent L-arginine transport into vacuolar membrane vesicles. When vacuolar proteins were labeled with radioactive NBZ arginyl diazomethane, the binding was irreversible, detached by treatment with base and blocked by treatment with cysteinyl blocking groups suggesting cysteine as a labeling site.
This paper examines the categorical labeling of lexical pitch accent contrasts in North Kyungsang and South Cholla Korean listeners. It focuses specifically on investigating whether the pitch accent perception of adults and children has a dialect-specific effect. To evaluate the development of perceptual identification, slopes, intercepts, and positions at categorical boundaries were computed using a logistic regression function. The results showed that differences in slopes and intercepts were significant between North Kyungsang child and adult listeners, but the same was not the case for the positions at boundaries. As far as South Cholla child and adult listeners were concerned, there was a significant difference in slopes, but not intercepts and positions at boundaries. In the present study, the comparison of intercepts and slopes at the boundaries indicated developmental differences between North Kyungsang adult and child listeners. This improvement in categorical proportion seems to be a result of developmental changes in categorical perception. For South Cholla adult and child listeners, however, perception of the non-native contrast becomes less categorical.
Bone is an active tissue, in which bone formation by osteoblast is followed by bone resorption by osteoclasts, in a repeating cycle. Proteomics approaches may allow the detection of changes in cell signal transduction, and the regulatory mechanism of cell differentiation. LC-MS/MS-based quantitative methods can be used with labeling strategies, such as SILAC, iTRAQ, TMT and enzymatic labeling. When used in combination with specific protein enrichment strategies, quantitative proteomics methods can identify various signaling molecules and modulators, and their interacting proteins in bone metabolism, to elucidate biological functions for the newly identified proteins in the cellular context. In this article, we will briefly review recent major advances in the application of proteomics for bone biology, especially from the aspect of cellular signaling.
