Analysis of Quantitative Trait Loci (QTLs) for Unsaturated Fatty Acid Contents in Soybean Seed Using Recombinant Inbred Lines (콩에서 microsatellite marker를 이용한 불포화지방산 함량의 양적형질 유전자좌의 분석)

  • Soybean oil is an important source of vegetable oil for human food and nonfood applications and accounts for approximately 22% of the world's total edible oil production. Improvement of the quality and quantity of soybean seed oil constituents is one of the most important objectives in soybean breeding. The objective of this study was to identify quantitative trait loci (QTLs) that control oleic, linoleic, and linolenic acid contents in soybean. The 117 $F_{2:10}$ recombinant inbred lines (RIL) developed from a cross of 'Keunolkong' and 'Shinpaldalkong' were used. Narrow-sense heritability estimates based on a plot mean on seed weight, protein and oil content were 0.85, 0.82 and 0.81, respectively. Eight independent QTLs for oleic acid content were identified from linkage group (LG) A2, C1, D2, F, G, L, and O. Seven QTLs for linoleic acid content were located on LG D1b, E, H, I and L. Oil content was related with five QTLs located on LG C1, H, J, K, and L. Oleic, linoleic, and linolenic acid have two common QTLs on LG C1 and L. Thus, we identified major loci improving soybean oil quality.

Chemical Components of Black Soybean Seeds Collected in Korea (수집 재래 검정콩의 화학적 성분)

  • The 1,081 soybean genotypes including 972 black soybeans and 109 other colored soybeans were collected in 197 locations from January to April, 1991. Seed chemical components of soybean collections in relation to growth characters were evaluated to survey germplasms for black soybean breeding for seed quality. Seeds of 1,081 genotypes collected were analyzed for crude protein, crude lipid, and total sugar contents. The crude protein content was averaged to 39.8% and ranged from 34.1% to 48.0%. The average crude lipid and total sugar contents were 20.1% and 10.1%, and the ranges of those were 14.1% to 23.8% and 8.3% to 12.1%, respectively. Higher crude protein content was shown in early soybean maturity group, whereas higher total sugar content was in late group. Black soybean collections with green seed embryo tended to be higher in total sugar content than those with yellow one. Black soybean oil is generally composed of about 22% oleic, 55% linoleic, 7% linolenic acid, and 16% the others.

De novo Genome Assembly and Single Nucleotide Variations for Soybean Mosaic Virus Using Soybean Seed Transcriptome Data

  • Soybean is the most important legume crop in the world. Several diseases in soybean lead to serious yield losses in major soybean-producing countries. Moreover, soybean can be infected by diverse viruses. Recently, we carried out a large-scale screening to identify viruses infecting soybean using available soybean transcriptome data. Of the screened transcriptomes, a soybean transcriptome for soybean seed development analysis contains several virus-associated sequences. In this study, we identified five viruses, including soybean mosaic virus (SMV), infecting soybean by de novo transcriptome assembly followed by blast search. We assembled a nearly complete consensus genome sequence of SMV China using transcriptome data. Based on phylogenetic analysis, the consensus genome sequence of SMV China was closely related to SMV isolates from South Korea. We examined single nucleotide variations (SNVs) for SMVs in the soybean seed transcriptome revealing 780 SNVs, which were evenly distributed on the SMV genome. Four SNVs, C-U, U-C, A-G, and G-A, were frequently identified. This result demonstrated the quasispecies variation of the SMV genome. Taken together, this study carried out bioinformatics analyses to identify viruses using soybean transcriptome data. In addition, we demonstrated the application of soybean transcriptome data for virus genome assembly and SNV analysis.

Breeding of Green Soybean Strain with Green Cotyledon and Tetra Null Genotype (Tetra null 유전자형과 녹색종피 및 자엽을 가진 콩 계통 육종)

  • A soybean cultivar with a green seed coat and cotyledon contains high levels of lutein, which is beneficial for eye health. Plus, antinutritional components such as lipoxygenase, Kunitz trypsin inhibitor (KTI), lectin and stachyose exist in the mature seed. The genetic elimination of these antinutritional factors is a necessary step in green soybean breeding. This research was conducted to improve a new green soybean line with the green cotyledon and tetra null genotype (lox1lox2lox3tilers2) in terms of lipoxygenase, KTI, lectin and stachyose. We used five germplasms to develop a breeding population. A total of 69 F2 seeds were obtained from the cross of parent 1 and parent 2, and from those, 21 F2 seeds were selected that had the green seed coat color, and which were free of lectin protein. Next, four F2 plants with the green seed coat and tetra null genotype were selected from the breeding population derived from four genotypes. The absence of lipoxygenase, KTI and lectin proteins was confirmed in the F5 strain. The breeding line has a green seed coat, green cotyledon and white hilum color. The 100-seed weight and stachyose content for the breeding line were 30.7 g and 2.40 g/kg, respectively. The line selected in this study could be used as a cultivar or parent to improve colored soybean cultivars through the removal of antinutritional components such as lip- oxygenase, KTI, lectin and stachyose.

Optimal Extraction Condition of Anthocyanins in Soybean (Glycine max) with Black Seed Coats (검정콩 종피 함유 안토시아닌의 최적 추출조건)

  • Soybeans (Glycine max) with a black seed coat have been widely utilized as food source and as a medicinal herbs in Korea. The pigmentation in the seed coat of black soybean is due to accumulate anthocyanins in the epidermis palisade layer. The anthocyanin content and composition of the black soybean seed coat are considered as a standard. of quality evaluation of black soybean. The main objective of this study was to investigate the optimal condition for an extraction method of anthocyanins and compare anthocyanin quantity and composition within black soybean varieties and germplasms. In the test of extraction solvent, absorbance at 530 nm and Hunter's a value were increased as increasing the concentration of MeOH, but Hunter's Land b values were the exact opposite of absorbance and Hunter's a values. There was no significant difference for anthocyanin contents from 1% HCl - $H_2O$ to 1% HCl - 80% MeOH. In the aspects of anthocyanin contents and HPLC peak resolution, 1% HCl - 20% MeOH extraction solution was the most suitable solvent. Among the 5 kinds of extraction method using 1% HCl - 20% MeOH solution, the anthocyanin contents of room temperature extraction at 72 h was the highest among the methods. High extraction temperature, sonication and reflux method influenced on the decrease of anthocyanin contents because of breakdown of anthocyanins. There was no significant difference for extraction time between 12 h and 24 h. However, the optimal extraction condition were at room temperature for 12 h. The anthocyanin contents in seed coats of black soybean were determined on the basis of HPLC peak area at 530 nm. Ten black soybean varieties and germplasms were tested with optimal conditions founded in this study. On the basis of antocyanin component, these can be classified into three groups; C3G, C3G + D3G and C3G + D3G + Pt3G. The total anthocyanin content in seed coats ranged from 1.58 to 10.62 mg/g of seed. The total anthocyanin content of the variety "Geomjeongol" was about 7 times higher than that of variety "Heugchong". Information for extraction method and diversity in antocyanin of soybean seed coats can be used for future research for germplasm evaluation and development of high quality black soybean varieties.

Nitrogen Balance and Biological Nitrogen Fixation of Soybean in Soybean-Barley Cropping System

  • This experiment was conducted to investigate the soil nitrogen credit of biological nitrogen fixation (BNF) and the nitrogen balance of soybean in soybean-barley cropping systems. Soybean cultivar, Shinpaldalkong2 and barley cultivar, Olbori, were used in soybean mono-cropping (SM), barley monocropping (BM), and barley­soybean double cropping system. The barley-soybean double cropping system was treated with two different levels of nitrogen fertilizers, 0 nitrogen fertilizer (BS-F0), and standard nitrogen fertilizer (BS-F1). Nitrogen and organic matter concentrations in soil of BS-F1 plot on October, 2001 were increased $4.8\%\;and\;5.9\%$, respectively, compared with those on October, 2000. The ranges of BNF rate in soybean were $69.1\~ 88.2\%$ in two years, and the rate was the highest in BS-F0 plot and the lowest in SM plot. The ranges of nitrogen harvest index (NHI) in all treatments were $83.9\~86.7\%$. The yield was 270 kg/10a in BS­F1 plot and 215 kg/10a in BS-F0 plot. However, the nitrogen balances were +0.6 kg/10a of gain of soil nitrogen in BS-F0 plot and -0.4 kg/10a of loss of soil nitrogen in BS-F1 plot. In comparisons of SM and BS-F1 plots, although the seed yields were similar in two plots, the loss of soil nitrogen was higher in SM than BS-F1 plot. Overall, our results suggest that barley-soybean double cropping system was more effective in respect to seed productivity and soil nitrogen conservation than soybean monocropping system, and the N credit to following crops by soybean cultivation was identified in soybean double cropping system.

Early Germination Response of Soybean Seed to Accelerated Aging and Low Dose Gamma Irradiation

  • The responses of soybean seeds were evaluated to accelerated aging and gamma irradiation with regard to germination, seed leakage, seed leachate component and dry weight of hypocotyl and primary root of the germinating seed. Accelerated aging significantly reduced the final germination rate while gamma irradiation increased the final germination rate. Furthermore, the interactive effects occurred that the final germination rate of 5-day aged seeds increased considerably in response to 4 Gy of gamma irradiation. The extent to which the electrolyte was leaked from the seeds (conductivity) was significantly affected by accelerated aging and showed a close negative correlation with the germination rate. Gamma irradiation, however, did not significantly affect the electrical conductivity of seed leachate. The accelerated aging significantly increased the concentrations of the particular electrolytes leaked from the seeds while the gamma irradiation did not affect those concentrations. Of the electrolytes leaked from the seeds, Ca and Mg showed relatively lower concentrations while K showed greater concentrations than others. Moreover, N and P showed similar responses to aging treatment. Aging treatment significantly affected dry weight (DW) of hypocotyls and primary root. Also, gamma irradiation decreased DW of hypocotyls and primary root, particularly for 8 Gy associated with 5 days aging treatment. The data were discussed in terms of the relationships of seed vigor with aging treatment and gamma irradiation.

The Relation of Soybean Seedcoat Mottling with Podding Location and Seed Transmissibility in Soybean Mosaic Virus (대두모자이크바이러스에 의한 결협절위별 갈반립의 형성과 바이러의 종자전염)

  • Association of SMV infection in soybean plants with seedcoat mottling and with seed transmission of SMV was examined. Seedcoat mottling was increased positively in proportion to the SMV severity in variety Clark. No correlation was found between the incidence of mottled seed and the seed's position on the plant. Susceptible variety with virus symptoms gave rise to mottled seeds but only a small percentage of these mottled seeds could transmit the virus. SMV could be transmitted even by nonmottled seeds harvested from SMV infected plants in similar rate with mottled seed. It seemed that the amount of mottled seed could be used as an indicator of the amounts of SMV infection in a seed production field.

Screening for Resistance to Purple Seed Stain 'Cercospora kikuchii' by Seed Inoculation Technique. (종자접종법에 의한 대두 자반병 저항성 검정에 관한 연구)

  • An attempt was made to establish a mass-screening technique for resistance to purple seed stain .disease in soybean. Seeds sterilized in 1 : 10000 'mercuric chloride for 1 minute and transffered to Petridishes containing 20ml water agar plus 50ppm of the sodium salt of 2,4-D were inoculated by placing a small agar cube containing fungal mycelium on the seed coat of each seed. A positive correlation between natural infection of purple seed :stain and purple discoloration by seed inoculation technique was highly significant and by this technique, some native soybean collections and introduced varieties were tested for resistance to the disease. Most of the soybean varieties tested were susceptible except for the varieties Hill, Harosoy and Sac, resistant comparatively.

Yield Loss Associated with Disease Severity of Soybean Black Root Rot by Cylindrocladium crotalariae (콩 흑색(黑色) 뿌리 썩음병(病)의 이병(罹病) 정도(程度)에 따른 수량(收量) 감소(減少))

  • Black root rot caused by Cylindrocladium crotalariae is one of the most serious soilborne disease in soybean. Plant height reduction of Hwangkeum Kong was 38% by the rotting of the whole main root and 9% by rotting of the half of the main root as compared with partial discoloration in the main root. Pod number per plant and seed weight were significantly reduced by the increase of the disease severity. Important yield components such as number of pods per plant and seed weight were negatively correlated with the soybean black root rot severity.
