  • The soybean necrotic disease has been shown to be caused by a virulent strain or strains of soybean mosaic virus (SMV) in soybean cultivar Kwnaggyo. However, the disease was found in soybean cultivar Hwanggeum which was released as a leading and mosaic resistant soybean cultivar in Korea. The strain SMV-G5H appeared to an isolate showing similar characteristics with the strain SMV-G7, although there were some variations in reactions of soybean differentials used.

Differential Gene Expression of Soybean[Glycine max(L.) Merr.] in Response to Xanthomonas axonopodis pv. glycines by Using Oligonulceotide Macroarray

  • Xanthomonas axonopodis pv. glycines(Xag) is a pathogen that causes bacterial leaf pustule(BLP) disease in soybeans grown in Korea and the southern United States. Typical and early symptoms of the disease are small, yellow to brown lesions with raised pustules that develop into large necrotic lesions leading to a substantial loss in yield due to premature defoliation. After Xag infects PI 96188, only pustules without chlorotic haloes were observed, indicating the different response to Xag. To identify differentially expressed genes prior to and 24 hr after Xag inoculation to PI 96188 and BLP-resistant SS2-2, an oligonucleotide macroarray was constructed with 100 genes related to disease resistance and metabolism from soybean and Arabidopsis. After cDNAs from each genotype were applied on the oligonucleotide macroarrays with three replicates and dye swapping, 36 and 81 genes were expressed as significantly different between 0 hr and 24 hr in PI 96188 and SS2-2, respectively. Six UniGenes, such as the leucine-rich repeat protein precursor or 14-3-3-like protein, were selected because they down-regulated in PI 96188 and up-regulated in SS2-2 after Xag infection, simultaneously. Using tubulin and cDNA of Jangyeobkong(BLP-susceptible) as controls, the oligonucleotide macroarray data concurred with quantitative real-time RT-PCR(QRT RT-PCR) results in most cases, supporting the accuracy of the oligonucleotide macroarray experiments. Also, QRT RT-PCR data suggested six candidate genes that might be involved in a necrotic response to Xag in PI 96188.

Characterization of a Novel Necrotic Response of Glycine max Line 'PI96188' to Xanthomonas axonopodis pv. glycines

  • Typical susceptible symptoms of the bacterial pustule disease caused by Xanthomonas axonopodis pv. glycines are pustule and chlorotic haloes that usually occur in leaves of Glycine max plants. The soybean genotype 'PI96188' showed an atypical response to all tested races X. axonopodis pv. glycines, accompanied with necrosis without chlorotic haloes on the underside of the necrotic symptoms. X. axonopodis pv. glycines 8ra grew to levels from 10 to 100 fold lower on PI96188 than on susceptible cultivar Jinjoo1, but 10-fold higher than on the resistant cultivar CNS. The chlorophyll content in PI96188 leaves remained unchanged until 12 days after bacterial infection. Ultrastructural observation showed that the infected leaf cells of PI96188 had intact normal chloroplasts compared to those of the susceptible cultivar Jinjoo1. Chloroplast degradation or the absence of chloroplasts in cells of the infected tissues was observed in Jinjoo1. Senescence-related ACS7 gene was significantly induced in PI96188 compared to those in Jinjoo1 at 2 days after inoculation. While photosynthesis-related rbcS gene showed the dramatic change in Jinjoo1, this gene was constitutively expressed in PI96188. However, expression of the defense-related genes, such as peroxidase and isoflavone synthase in the infected PI96188 leaves was similar to that in Jinjoo1. Together, these results suggest that the novel necrotic symptom in PI96188 is a kind of resistant response different from a typical hypersensitive response in the resistant genotypes.

Epidemiology of Soybean Mosaic Virus Diseases (콩모자이크바이러스병의 역학적 연구)

  • Two soybean cultivars Hanan and Kwanggyo were used to monitor SMV spread in the field. The Haman producing mosaic symptoms by infection with SMV strain G7 was planted in $2.2m^2$ at the coater of $320.7m^2$ field and Kwanggyo producing necrosis by infection with SMV-G7 was planted around the Haman one day after inoculation of Heman with SMV-G7. The most severe incidence of the necrotic disease in soybean cultivar Kwanggyo occurred on 43 days after planting(July 13) whereas aphid population reached peak around 22 days after planting(June 22). Total incidence of the necrotic disease was $20.4\%$, in the whole field and $56\%$, in eight $mall plots around the spreader of SMV. The spread of S MV outward from the spreader source was greater downwind than it was upwind. Spread also showed a signigicant gradieut pattern leeward from the infection focus.

Growth Characteristics and Yield Potentials of Soybeans in Upland and Paddy Field (전과 답에서의 콩 생육특성과 수량성)

  • To obtain the basic information necessary for the development of soybean varieties well adaptable to upland-paddy field rotational croppings, the difference of growth characteristics between upland and paddy-field including yield potentials of current recommending soybean varieties were evaluated. The growth characteristics, both above and under-ground, which were measured at flowering stage were generally greater in paddy-field but the number of root nodules was much greater in upland, thus the artificial inoculation was practically recommended for soybean growing in paddy-fields. Mean seed yield was generally higher in paddy-fields than in upland. All soybean varieties showed higher seed yield in the early planting date, April 20, were somewhat susceptible to soybean mosaic virus(SMV), thus they could be escaped from the disasterous endemic necrotic soybean mosaic virus(SMV-N). Soybean varieties showed over 4.0 tons/ha seed yield in the paddy-field were Williams 79, Union, SS77053, and Namhaekong. At the same time, Jangyeobkong and Danyeobkong were the most stable soybean varieties among the tested soybean varieties with less than 10% of coefficient variation values in all planting dates in paddy fields. Compared with Hwangkeumkong which is most widely being cultivated on farmer's fields, soybean varieties showed high yields in paddy-field were higher in plant height, less in the number of branches, and more in the number of nodes on main stem. At the same time, they had medium seed size which would bring the good germination and stands. Disease resistance especially for necrotic soybean mosaic virus was also one of the most decisive factors in seed yields for the early planted soybeans.

  • "Wonkwang" is a new sprout-soybean cultivar developed from the cross between Danyeobkong and MS91001 at the Honam Agricultural Research Institute (HARI) and National Institute of Crop Science (NICS), RDA, in 2007. The preliminary, advanced and regional yield trials to evaluate the performance of Iksan 56 were carried out from 2003 to 2007. This cultivar has a determinate growth habit, purple flower, grayish brown pubescence, yellow seed coat, Grayish brown hilum, lanceolate leaflet shape and small seed size (10.9 g/100 seeds). The maturity date of "Wonkwang" is three days later than the check variety, "Pungsan". It has a good seed quality for soybean-sprout and resistance to lodging. The soybean-sprouts grown from "Wonkwang" have high isoflavone ($3,481{\mu}g/g$)contents. This cultivar has resistance to soybean mosaic virus (SMV) and necrotic symptom (SMV-N) and other most troublesome soybean diseases which are bacterial pustule and black root rot. The grain yield of "Wonkwang"in the regional yield trials (RYT) for 3 consecutive years was averaged 3.05 ton per hectare, which was 8% higher than that of the check cultivar "Pungsan".

A New Soybean Cultivar, "Sohwang" for Sprout with Disease Resistance, Small Seed Size and High Sprout yielding (내병·소립 고수율 나물콩 신품종 "소황")

  • A new sprout-soybean cultivar, "Sohwang" was developed at the National Institute of Crop Science (NICS) in 2008. "Sohwang" was selected from a cross between Pungsan and Pungsan/Nattosan. The preliminary, advanced, and regional yield trials to evaluate the performance of Iksan 58 were carried out from 2004 to 2008. This cultivar has a determinate growth habit with purple flower, grayish brown pubescence, yellow seed coat, Yellow hilum, lanceolate leaflet shape and small seed size (8.5 grams per 100 seeds). The maturity date of "Sohwang" is 10 days earlier than the check variety, "Pungsan". It has a good seed quality for soybean-sprout. The soybean-sprouts grown from "Sohwang" have high isoflavone ($3,041{\mu}g/g$)contents. This cultivar has resistance to soybean mosaic virus (SMV) and necrotic symptom (SMV-N), and other most troublesome soybean diseases which are bacterial pustule and black root rot. Especially, "Sohwang" has useful characteristics, such as lodging and shattering resistance. The average yield of "Sohwang" was 2.69 ton per hectare in the carried out for three years from 2006 to 2008 regional yield trials(RYT) for double cropping.
