The Factors on Somatic Embryogenesis of Soybean [Glycine max. (L.) Merrill]

  • To enhance in vitro plantlet regeneration efficiency of soybean through embryogenesis, the culture conditions such as material part and size of immature seed, 2,4-D, pH and solidifying agents for somatic embryogenesis were investigated. Somatic embryogenesis was induced from the immature embryo, immature cotyledon and embryonic axis explants of the immature seed on MS medium supplemented with 2.0 mg/L 2,4-D. The highest rate (up to 22.9%) of somatic embryogenesis was obtained from the immature cotyledon, following embryonic axis and the immature embryo. The rate varied with the developmental stages of seed. The maximum rate (25.4%) of embryogenesis was obtained from 3-4 mm length of the seed (after 25 days of flowering). The optimum concentration of 2,4-D for embryogenesis was 10 mg/L. The optimum pH was at 5.8 and solidifying agent for medium was better with 0.4% gelrite than with agar. For rapid multiplication of shoot tips from the germinating somatic embryos, they were cultured on MS medium containing 2 mg/L indole-3-butyyic acid (IBA) and 1 mg/L 6-benzyladenine (BA). After then somatic embryos with one and three cotyledons were transferred to the growth regulator free medium. The medium exhibited the higher rate (ca. 50%) of development than the multiplication medium.

전남지역 논 조건에서 무경운 재배가 콩의 생육, 수량 및 잡초 발생에 미치는 영향 (Effect of No-Tillage on Soybean Yield and Weed Emergence in Drained Paddy Field Condition in Jeonnam Province)

  • This study was conducted to compare soybean growth and yield and the degree of weed emergence according to no-tillage and conventional tillage system in two different drained paddy fields, loam of Chilgok series and silty clay loam of Deokpyeong series. In both soil conditions, the maturing time of the soybeans by the no-tillage system was two days earlier than that by the conventional tillage system. In the loam of Chilgok series, the stem length of the soybeans in the no-tillage system was 5.7 cm longer than that in the conventional tillage system. The miss-planted rate and diseased plants of black root rot (Calonectria iliacola) in the no-tillage system were 9.2% and 2.8% lower, respectively than those in the conventional tillage system. Also, the nodulation and seed yield in the no-tillage system were 32% and 13% more, respectively, than those in the conventional tillage system. In the silty clay loam of Deokpyeong series, the stem length of the soybeans in the no-tillage system was 4.6cm shorter than in the conventional tillage system. The diseased plants of black root rot (Calonectria iliacola) in the no-tillage system were 4.2% lower than those in the conventional tillage system. Also, no significant difference in the seed yield between the no-tillage and conventional tillage systems was observed. On the other hand, there was a lower occurrence of weed in the no-tillage system than in the conventional tillage system, and the income increased by 19% owing to yield increase and reduced management costs.

대두 (Glycine max) 부배유 유래 칼루스의 원형질체 분리 및 배양 (Isolation and Culture of Protoplasts from Hypocotyl-derived Callus of Soybean (Glycine max))

  • The isolation and culture of protoplasts from hypocotyl-derived calluses of Glycine max (L.) Merr. cv. Jangyeop were obtained by digestion for 6 hrs in an enzyme solution containing 3.5% cellulase, 1.5% macerozyme, 10% sorbitol and 0.1% CaCl2.2H2O at pH 5.8. Newly formed cell wall of protoplasts cultured in MS agar medium containing 10 $\mu$M $\alpha$-naphthaleneacetic acid (NAA) and 32 $\mu$M N6-benzylaminopurine (BAP) could be observed after 24 hrs culture. The first cell division of the protoplasts was observed after 3 days of culture; cell clusters after 2 weeks of culture. When transferred to solid media, the protoplasts formed cell clusters gave rise to proliferating calluses.

Phytotoxic Effects of Xanthium occidentale Extracts and Residues on Seedling Growth of Several Plant Species

  • Compositae plants are known to contain biologically active substances that are allelopathic to agricultural crops as well as weed species. Aqueous or methanol extracts and plant residues from leaves of Xanthium occidentale were assayed against alfalfa (Medicago sativa) to determine its allelopathic effects, and the results showed that the extracts applied onto filter paper significantly inhibited seed germination as well as root growth of alfalfa. Aqueous leaf extracts of $40g\;L^{-1}$ were completely inhibitory on root growth of alfalfa, while root growths of barley (Hordeum vulgare L.) and soybean (Glycine max L.) were less sensitive. Leaf residue incorporation at $100g\;kg^{-1}$ into soil on seedling growth of barnyard grass (Echinochloa crus-galli Beauv. var. oryzicola Ohwi) inhibited both shoot and root fresh weights of barnyard grass by 94 and $96\%$, respectively. Methanol extracts from BuOn and EtOAc fractions of X. occidentale reduced alfalfa root growth more than did those from hexane and water fractions. The results based on bioassay of extracts and residues show that X. occidentale had potent an allelopathic activity against other plant species.

  • Human and monogastric animals can not synthesize 10 out of the 20 amino asids and therefor need to obtain these from their diet. The plant seed is a major source of dietary protein. It is particular important in their study to increase nutritional quality of the seed storage proteins. The low contents of lysine, asparagine and threonenein various cereal seeds and of cystein and methionine. In legume seeds is due to the low proportions of these amino acids in the major storage proteins, we have tried to apply the three strategies; (1) mutagenesis and selection of specific amino acid analogue resistance, (2) cloning and expression study of lysine biosynthesis related gene, (3) transfomation of lysine rich soybean glycinin gene. The 5-methyltryptophan (5MT) resistant cell lines, SAR1, SAR2 and SAR3 were selected from anther derived callus of rice (Oryza sativa L. "Sasanishiki"). Among these selected cell lines, two (SAR1 and SAR3) were able to grow stably at 200 mg/L of 5MT. Analysis of the freed amino acids in callus shows that 5MT resistant cells (SAR3) accumulated free tryptophan at least up to 50 times higher than those that of the higher than of SAS. These results indicated that the 5MT resistant cell lines are useful in studies of amino acid biosynthesis. Tr75, a rice (Oryza sativa L., var. Sasanishiki) mutant resistant to 5MT was segregated from the progenies of its initial mutant line, TR1. The 5MT resistant of TR75 was inherited in the M8 generations as a single dominant nuclear gene. The content of free amino acids in the TR75 homozygous seeds increased approximately 1.5 to 2.0 fold compared to wild-type seeds. Especially, the contents of tryptophan, phenylalanine and aspartic acid were 5.0, 5.3 and 2.7 times higher than those of wild-type seeds, respectively. The content of lysine is significantly low in rice. The lysine is synthesized by a complex pathway that is predominantly regulated by feedback inhibition of several enzymes including asparginase, aspatate kinase, dihydrodipicolinat synthase, etc. For understanding the regulation mechanism of lysine synthesis in rice, we try to clone the lysine biosynthetic metabolism related gene, DHPS and asparaginase, from rice. We have isolated a rice DHPS genomic clone which contains an ORF of 1044 nucleotides (347 amino acids, Mr. 38, 381 daltons), an intron of 587 nucleotides and 5'and 3'-flanking regions by screening of rice genomic DNA library. Deduced amino acid sequence of mature peptide domain of GDHPS clone is highly conserved in monocot and dicot plants whereas that of transit peptide domain is extremely different depending on plant specie. Southern blot analysis indicated that GDHPS is located two copy gene in rice genome. The transcripts of a rice GDHPS were expressed in leaves and roots but not detected in callus tissues. The transcription level of GDHPS is much higher in leaves indicating enormous chloroplast development than roots. Genomic DNA clones for asparaginase genes were screened from the rice genomic library by using plaque hybridization technique. Twelve different genomic clones were isolated from first and second screening, and 8 of 12 clones were analyzed by restriction patterns and identified by Southern Blotting, Restriction enzyme digestion patterns and Southern blot analysis of 8 clones show the different pattern for asparaginase gene. Genomic Southern blot analysis from rice were done. It is estimated that rice has at least 2-3 copy of asparaginase gene. One of 8 positive clones was subcloned into the pBluescript SK(+) vector, and was constructed the physical map. For transformation of lysine rich storage protein into tobacco, soybean glycinin genes are transformed into tobacco. To examine whether glycinin could be stably accumulated in endosperm tissue, the glycinin cDNA was transcriptionally fused to an endosperm-specific promotor of the rice storage protein glutelin gene and then introduced into tobacco genomic via Agrobacterium-mediated transformation. Consequently the glycinin gene was expressed in a seed-and developmentally-specific manner in transgenic tobacco seeds. Glycinin were targeted to vacuole-derived protein bodies in the endosperm tissue and highly accumulated in the matrix region of many transgenic plant (1-4% of total seed proteins). Synthesized glycinin was processed into mature form, and assembled into a hexamer in a similar manner as the glycinin in soybean seed. Modified glycinin, in which 4 contiguous methionine residues were inserted at the variable regions corresponding to the C - teminal regions of the acidic and basic polypeptides, were also found to be accumulated similarly as in the normal glycinin. There was no apparent difference in the expression level, processing and targeting to protein bodies, or accumulation level between normal and modified glycinin. glycinin.

대두조직배양세포(大豆組織培養細胞) - Rhizobium에 의(依)한 질소고정력(窒素固定力) (The Establishment of Nitrogen Fixation by Cultured Cell-Rhizobium Association Through Tissue Culture Technique in Soybean)

  • Rhizobium의 Nitrogenase 생성요인(生成要因)과 감염기작(感染機作)을 구명(究明)하고 배양세포(培養細胞)와 Rhizobia의 혼합배양(混合培養)에서 질양고정계(窒養固定係)를 확립(確立)하기 위(爲)하여 황금(黃金), 남천(南川), D68-0099등(等) 세 품종(品種)을 조직배양(組織培養)한 결과(結果)는 다음과 같다. Callus 형성능(形成能)은 배(胚)와 유근(幼根)에서는 양호(良好)하나 배축(胚軸)에서는 전혀 없었으며 2mg/${\ell}$ 2,4-D, 4mg/${\ell}$ NAA에서 가장 양호(良好)하고 2,4-D/Kinetin 조합농도(組合濃度)에서는 0.2mg(2,4-D)/${\ell}$과 0.05mg(Kinetin)/${\ell}$에서 가장 이상적(理想的)이었다. 배양세포(培養細胞)의 성장(成長)에는 2,4-D 2mg/${\ell}$와 2,4-D (0.2mg/${\ell}$)/Kinetin(0.05mg/${\ell}$)일 때가 가장 양호(良好)하며 R. japonicum 019, 011을 접종(接種)하였을때 배양세포(培養細胞)의 성장(成長)은 상당히 둔화되었다. 단일(單一) 아미노산은 배양세포(培養細胞)의 성장(成長)을 저해(沮害) 하였는데 황금(黃金)의 경우 Methionine, Leucine에서 저해(沮害)가 가장 컷으며 다른 아미노산의 첨가(添加)로 저해작용(沮害作用)이 상당히 회복되었다. 부정근(不定根)의 생성(生成)은 2,4-D 2.0mg/${\ell}$에서나 0.2mg/${\ell}$ 2, 4-D/0.05mg/${\ell}$ Kinetin에서 양호(良好)하였다. 배양세포(培養細胞)-Rhizobium의 친화(親和)에 의(依)한 질소고정력(窒素固定力)은 25개(個) 사용(使用) 균주중(菌株中)에서 황금(黃金)에서는 10개(個) 균주(菌株), 남천(南川)에서는 7개(個) 균주(菌株)에서 나타났으며 D68-0099에서는 전혀 나타나지 않았으며 황금(黃金)의 경우(境遇) 85-HG-1 019, 007, 남천(南川)에서는 007, 119등(等)이 높은 활성(活性)을 나타내었다.

국내 콩(Glycine max) 품종 형질전환 초기조건 확립 (Optimization of Genetic Transformation Conditions for Korean Soybean Cultivars)

  • 경제적으로 중요한 식용작물인 콩(Glycine max)의 초기형 질전환 효율증대를 위하여 연구를 수행하였다. 높은 초기형 질전환 조건 확립을 위하여 기초실험으로 Agrobacterium 감 염에 순응적인 국내 콩 품종을 스크린 하였으며, 적정한 agar 농도와 선발항생제 hygromycin 농도 규명, 상처방법, explant의 치상방향, 호르몬 전처리, 액체배지 선발 등 형질 전환 초기효율에 미치는 영향을 transient GUS 분석을 통하여 알아보았다. Agrobacterium 감염에 순응적인 품종을 선발하기 위하여 32개의 국내 콩 장려품종을 스크린한 결과, 일품검정콩, 은하콩, 만리콩, 대원콩 등 14개의 순응형 품종을 선발하였으며, 효율적인 제균제로는 cefotaxime이, 효율적인 선발항생제로서는 hygromycin이 선택되었다. Hygromycin 농도는 10-15 ppm이 효율적이었고, agar 농도 0.6-0.8%, explant의 치상방향은 향배축(adaxial side)을 down하여 치상하였을 때가 높은 GUS ($\beta$-glucuronidase)발현빈도를 나타냈고 상처방법은 scalpel보다. 동양침을 사용하였을 때 높게 나타났다. 형질전환 체계확립을 위하여 호르몬 전처리를 한 결과, BA 5 ppm 과 10 ppm을 처리하였을 때 후기 선발에서 높은 생존율을 보였고, 전처리 후에 액체배지에서 선발을 하였을 때 비형질 전환체(escape)의 빈도를 크게 낮출 수 있었다. 또한 여러 조건의 결과를 비교하여 볼 때, 국내 콩 품종 중에서 형질전환을 위한 재료로써는 은하콩이 가장 적합한 것으로 생각된다.

남부지방에서 조생종 콩 품종의 특성과 조기수확 한계기 (Characteristics and Possible Early Harvesting Time of Early Maturing Soybean Cultivars in Southern Korea)

  • 본 연구는 주요 조생종 콩의 생육과 종실품질 및 수량을 비교하여 남부지방 다모작 재배농가의 조생종 콩 품종선택에 활용하고자 전남 나주(위도 35^{\circ}04'N, 경도 126^{\circ}54'E)에서 6월 15일에 파종하여 수행하였다. 큰올콩과 화성풋콩은 성숙기가 9월 12일로 빨라 작부체계 도입에 유리할 뿐만 아니라 종실품질과 수량이 양호하였다. 그리고 새올콩과 신록콩은 성숙기가 9월 20일 전.후로 조금 늦으나 종실품질이 좋고 수량이 많았다. 따라서 콩 후작물을 9월 중순에 파종하고자 할 때는 큰올콩과 화성풋콩이, 9월 하순에 파종하고자 할 때는 새올콩과 신록콩이 유리하였다. 그리고 주요 조생종 콩의 조기수확에 따른 종실품질과 수량성 등을 검토하여 조기수확 한계기를 구명하고자 수행하였다. 큰올콩은 수확적기(9월 12일)보다 6일전(9월 6일)에 수확할 때 입중, 수량, 종실품질 등이 적기수확과 차이가 없었다. 한편 새올콩은 수확적기(9월 24일)보다 6일전(9월 18일)에 수확할 때 입중, 수량, 종실품질 등이 적기수확과 차이가 없다. 따라서 큰올콩과 새올콩의 안정적인 조기수확 한계기는 수확적기 6일 전이다.

한국 품종 대두(Glycine max L) 미숙배로부터 체세포배발생과 배지의 2,4-Dichlorophenoxyacetic Acid 농도가 체세포배의 형태에 미치는 영향 (Somatic Embryogenesis in Immature Zygotic Embryo Cultures of Korean Soybean (Glycine max L.) Culitivars and Effect of 2,4-Dichlorophenoxyacetic Acid on Somatic Embryo Morphology)

  • 국내 5가지 품종의 대두 미숙배로부터 발생되는 체세포배 형성빈도와 자엽변이에 대한 2,4-D 농도의 영향을 알아보였다. 여러가지 농도의 2,4-D가 첨가된 MS 기달배지에 각 품종의 미숙배를 6주동안 암소에서 배양한 결과 '울산콩', '금산콩', '뱀콩' 및 '반청두'의 경우 1-2 mg/L 2,4-D를 첨가한 배지에서, '아주까리'는 20 mg/L 2,4-D를 첨가한 배지에서 가장 높은 체세포배 형성빈도를 보였다. 또한 횡으로 반분한 체세포배를 0.1-1 mg/L 2,4-D가 첨가된 배지에서 배양하였을 때 최고 70%의 빈도로 이차배를 형성하였다. 미숙배(접합자배)에서 유도된 체세포배의 형태는 2,4-D 농도에 따라 다르게 나타났는데 즉, 0.2 mg/L 이하의 농도에서는 하나의 자엽을 갖는 배와 나팔형보다는 두개의 자엽을 갖는 정상적인 형태가, 0.5-4 mgh 농도에서는 정상적인 형태보다는 나팔형의 체세포배 형성빈도가 높게 나타났다. 또한 체세포배의 자엽형태에 따라 식물체 재생률을 조사하기 위하여 ME 기본배지와 0.1 mg/L IAA, 1 mg/L BA, 1mg/L Gh₃ 및 0.5% 활성탄을 각각 U 기본배지에 첨가하여 16시간 광주기하에서 4주동안 배양한 결과, 식물체 재생률은 일반적으로 두개의 자엽을 갖는 정상적인 배가 하나의 자엽을 갖는 배, 나팔형 및 구형기의 체세포배에 비하여 높았으며, 특히 1 mg/L GA3를 첨가한 배지에서 가장 높은 재생률(25%)을 보였다.

생분해성 생리활성물질 5-aminolevulinic acid의 제초 및 살충활성 (Herbicidal and Insecticidal Potentials of 5-Aminolevulinic acid, a Biodegradable Substance)

  • 5-Aminolevulinic acid (ALA)는 protoporphyrinogen IX oxidase(Protox IX)의 작용기작에 의해 tetrapyrrole 의존형 광활성 제초제 또는 살충제로서 제안되어 왔다. 본 연구는 생분해성 생물농약 물질로서 ALA에 대한 식물과 해충의 생육반응을 검토하기 위해 수행되었다. 수도적 조건에서 ALA는 벼와 피 두 초종에 대해 독성을 보였으며 벼보다는 피의 초장과 지상부 생체중을 더 억제하였다. 밭 조건에서 두과작물 콩과 화본과 잡초 바랭이에 5 mM ALA를 처리한 결과 바랭이가 더 민감한 생육반응을 보였다. ALA 10 mM(10배액)로 파밤나방에 대한 살충효과는 살포법으로 처리한 결과 단제 및 합성 살충제 lufenuron과의 조합처리에서 다소 높게 나타났다. leaf disk법으로 응애에 대한 살비효과를 검정한 결과 10 mM ALA 처리 후 72시간에 가장 높게 나타났다. 이상의 결과로 볼 때 ALA는 비록 그 활성이 기존의 합성농약보다 낮을지라도 농업유해생물에 대해 잠재적인 제초 및 살충활성을 갖고 있는 것으로 나타났다.
