Variation of Anthocyanin Contents according to Collection Site and Maturity in Black Soybean (검정콩 수집지역과 성숙기에 따른 안토시아닌 함량 변이)

  • Anthocyanin contents of black soybean were analyzed for development of superior breeding lines with high anthocyanin contents. Total 292 genotypes of black soybean collected through the whole country were analyzed by HPLC in which C3G (Cyanidin-3-Glucoside), D3G (Delphinidin-3-glucoside), and Pt3G (Petunidin-3-Glucoside) were detected main anthocyanin pigments and each pigment contents were significantly different according to genotypes. C3G content showed the highest value in all materials and its variation was also wide, whereas D3G and Pt3G were not detected in 4 and 24 genotypes. Mean value of C3G, D3G, and Pt3G contents were $8.05{\pm}4.225$, $1.80{\pm}0.854$, and $1.15{\pm}0.781\;mg/g$, respectively. In case collected sites, genotypes collected in Chungnam region were higher the anthocyanin contents than other collections, which was $13.75{\pm}3.861\;mg/g$. It might be concluded that it takes more than 36 days for anthocyanin accumulation since beginning of seed-coat pigments formation, in that case it showed $13.09{\pm}4.190\;mg/g.$. Also total anthocyanin contents were present higher concentration in seed coat as maturation period was longer from flowering stage.

Identification of a Locus Associated with Resistance to Phytophthora sojae in the Soybean Elite Line 'CheonAl' (콩 우수 계통 '천알'에서 발견한 역병 저항성 유전자좌)

  • Phytophthora root rot (PRR) is a major soybean disease caused by an oomycete, Phytophthora sojae. PRR can be severe in poorly drained fields or wet soils. The disease management primarily relies on resistance genes called Rps (resistance to P. sojae). This study aimed to identify resistance loci associated with resistance to P. sojae isolate 40468 in Daepung × CheonAl recombinant inbred line (RIL) population. CheonAl is resistant to the isolate, while Daepung is generally susceptible. We genotyped the parents and RIL population via high-throughput single nucleotide polymorphism genotyping and constructed a set of genetic maps. The presence or absence of resistance to P. sojae was evaluated via hypocotyl inoculation technique, and phenotypic distribution fit to a ratio of 1:1 (R:S) (χ2 = 0.57, p = 0.75), indicating single gene mediated inheritance. Single-marker association and the linkage analysis identified a highly significant genomic region of 55.9~56.4 megabase pairs on chromosome 18 that explained ~98% of phenotypic variance. Many previous studies have reported several Rps genes in this region, and also it contains nine genes that are annotated to code leucine-rich repeat or serine/threonine kinase within the approximate 500 kilobase pairs interval based on the reference genome database. CheonAl is the first domestic soybean genotype characterized for resistance against P. sojae isolate 40468. Therefore, CheonAl could be a valuable genetic source for breeding resistance to P. sojae.

Effects of Alfalfa Meal on Growth Performance and Gastrointestinal Tract Development of Growing Ducks

  • A study was conducted to evaluate effects of alfalfa meal on growth performance and gastrointestinal tract development of growing layer ducks to provide evidence for application of alfalfa meal in the duck industry. Two hundred and fifty-six healthy Shaoxing 7-wk old growing layer ducks were selected and randomly allocated to 1 of 4 dietary treatments based on corn and soybean meal and containing 0, 3, 6, and 9% of alfalfa meal for 8 wks. Each treatment consisted of 4 replicates of 16 ducks each. Briefly, birds were raised in separate compartments, and each compartment consisted of three parts: indoor floor house, adjacent open area and a connecting water area. The results showed: i) Growing ducks fed alfalfa meal diet were not significantly different in average daily gain, feed intake and gain-to-feed ratio from those fed no alfalfa diet (p>0.05). ii) Alfalfa meal increased the ratio crop, gizzard to live weight, caecum to live weight, the caecum index of growing ducks (p<0.05). iii) Villus height in duodenum and jejunum of growing ducks increased significantly with the increase of alfalfa meal levels (p<0.05). Crypt depth in duodenum and jejunum of growing ducks decreased significantly with the increase of alfalfa meal levels (p<0.05). This experiment showed that feeding of alfalfa meal to growing layer ducks could improve gastrointestinal tract growth and small intestinal morphology without effect on performance. This experiment provides evidence that alfalfa meal is a very valuable feedstuff for growing layer ducks.

Breeding of Aspergillus oryzae for the Alkaline Pretense Overproducing Strain. (재조합 Alkaline Protease를 대량 생산하는 Aspergillus oryzae 균주개발)

  • Aspergillus oryzae M-2-3 strain (argB$\^$-/) was transformed with pTAalp plasmid which was constructed for expression of the alkaline pretense gene, alpA, and 16 transformants were selected on arginine minus medium. When these transformants were tested for productivity of alkaline proteases using agar plate containing skim milk, the halo was observed around each colony of transformants, but not observed around the host strain in this condition. Southern analysis showed that the pTAalp plasmid having alpA gene was integrated into the chromosome of the host strain. The highest level of alkaline protease production was obtained in the culture filtrate of the transformant No. 14, which was estimated to 80-90% of total secreted proteins, and the enzyme activity was 64-450 times higher than those of host strain and industrial strain. Total nitrogen content and the digestion rate in soybean Koji extracts were also increased to 1.5 times in Aspergillus oryzae transformant No. 14.

Influence of insect pollinators on gene transfer from GM to non-GM soybeans (GM 콩의 도입유전자 이동에 미치는 화분 매개충의 영향)

  • The cultivation area and use of genetically modified (GM) crops have been increased continuously over the world and concerns about the potential risks of GM crops are also increasing. One of the major concern in risk assessment is the possible development of hybrids through interspecific and intergeneric crosses with related species. This study was conducted to investigate the pollinator have an influence on insect-mediated gene transfer from GM soybeans. Hybrid was induced from GM soybeans by honeybee and western flower thrips, and non-GM soybeans were used as pollen receptor. The analysis for gene-flow was conducted by herbicide selection, immunostrip test, and PCR analysis. In the result of the analysis, three hybrids were detected on the distance 15, 75, 105 cm from pollen source in western flower thrips treatment. In honeybee treatment, one hybrid was detected in the farthest distance (300 cm). These results suggested honeybee and western flower thrips have a possibility they can transfer the introduced gene from GM soybeans to non-GM soybeans.

  • New breeding method by genetic engineering is expected as a key technology to solve food shortage due to the growing world population in the year 2000s. Many genetically modified organisms (GMOs) were already developed and the commercial cultivation had started. The first GMO, Flavr Savr tomato, which rotted at a much slower pace than ordinary ones, was developed in US in 1994. Since then, over than 70 different agricultural products including corn, cotton, soybean, papaya, potato, and squash made with genetically modified plants are reportedly on sale worldwide. Supporters favor the GMOs because they have greater yields, longer shelf lives and stronger resistance to disease and insects. On the other hand, opponents say that the supporters ignore a potential danger that they may damage the environment as well as human beings. To assure the safe development and use of GMOs as food and other biotech products, the possible risks on biological environment and human health should be throughly examined and regulated by developer and government. Because the biosafety problem is a global, environmental, and trade issue, a new international treaty is under development. The Cartagena Protocol on Biosafety was adopted at the 1 st Extraordinary Conference of Parties of the Convention on Biological Diversity which was held at Mont-real, Canada, Jan. 29th, 2000. The adoption of the Protocol is seen as a breakthrough in that it is based on the" Precautionary Principle" despite scientific uncertainties surrounding potential risks that GMOs may inflict on human health and the environment and that it has laid the ground for introduction of specific steps to handle international trading of GMOs. In this paper, the authors would like to introduce the current status and perspective of environmental and human risk assessment of GMOs.t of GMOs.

Effect of graded levels of soapnut (Sapindus mukorossi) shell powder on reproductive performance in broiler breeders

  • Objective: This study investigated the effects of soapnut (Sapindus mukorossi) shell powder (SSP) on serum hormone level, egg quality, semen characteristics and reproductive performance of broiler breeders fed with a maize-soybean meal based diet. Methods: Ninety six female and twenty four male CARIBRO-VISHAL broiler breeders, 38-week old, were individually caged and randomly allocated to four treatment groups (24 female breeders/treatment and 6 male breeders/treatment): an un-supplemented control (T1) and three groups with 0.0176% SSP (group T2), 0.026% SSP (group T3) and 0.0528% SSP (group T4), to have supplementary saponin at 0, 50, 75, and 150 ppm, respectively, for 42 days. Results: The results indicated that serum (p<0.001) and seminal plasma (p<0.05) testosterone level, semen volume (p<0.001), mass motility (p<0.001), and live spermatozoa count (p<0.001) was increased in groups T3 and T4 compared to T2 and control groups. Compared with control group, total sperm count was increased (p<0.001) and dead spermatozoa count was decreased (p<0.001) in SSP supplemented groups. Supplementation of SSP did not affected the quality of egg lay. Compared with control group, fertility (p<0.01) and hatchability (total eggs set and fertile eggs set) (p<0.001) were significantly improved in SSP supplemented groups with the highest improvement in T3 treatment group. Embryonic death was decreased (p<0.001) in SSP supplemented groups compared to control; lowest embryonic death was recorded in T3 treatment group. Conclusion: Thus, it was concluded that dietary supplementation of 0.026% SSP (saponin equivalent 75 ppm) improved the reproductive performance of broiler breeders.

Variation of Contents and Color Difference of Anthocyanin by Different Cultivation Year in Black Soybean Seed (재배연도에 따른 검정콩 종자의 안토시아닌 함량 및 색차변이)

  • This research was carried out to offer the basic informations about new varietal breeding for specific use and physiological characteristics through investigation of detection, content variation and color difference of anthocyanin individual pigments within seed coats in domestic black soybean. The seed of thirteen cultivars such as Geomjeongkong 1, Geomjeongkong 2, Seonheugkong, Tawonkong, Ilpumgeomjeongkong, Geomjeongolkong, Cheongjakong, Jinju 1, Heugcheongkong, Juinunikong-Y, Juinunikong-G, Geomjeongkong 3, Geomjeongkong 4 was tested. C3G (cyanidin-3-glucoside) was detected in only Geomjeongkong 1 and Seonheugkong but D3G (delphinidin-3-glucoside) and C3G were found in Heugcheongkong. The rest cultivars that there were three anthocyanins such as D3G, C3G, and Pt3G (petunidin-3-glucoside). Anthocyanin content of tested cultivars showed a high variation. The ranges of D3G, C3G, Pt3G, and TA (total anthocyanin) contents were $0.55\~2.63mg/g,\;2.77\~8.38mg/g,\;0.38\~5.65mg/g,\;and\;3.32\~16.92mg/g$, respectively. These contents showed variation among cultivars as well as variation between two years, 2001-2002. As a result of variation of anthocyanin color difference, the ranges of L (lightness), a (redness), and b (yellowness) as Hunter's value were $34.09\~42.89,\;12.77\!22.85,\;and\;5.36\~12.10$, respectively, and these color differences showed variation among cultivars and also variation between two years, 2001-2002. D3G, C3G, Pt3G, and TA showed reciprocally a positive correlation being representive of high significance.

Variation of Saponin Content in Korean Native Soybean Landraces Reintroduced from USA to Korea (미국으로부터 재도입된 한국 원산 콩 재래종의 Saponin 함량 변이)

  • This study was carried out to assess the variation of saponin contents in 293 Korean native soybean landraces reintroduced from USA to Korea in 2007. Group A saponin content ranged from 149.8 ${\mu}g\;g^{-1}$ to 1279.0 ${\mu}g\;g^{-1}$ with an average of 484.9 ${\mu}g\;g^{-1}$. Group B saponin content ranged from 2160.1 ${\mu}g\;g^{-1}$ to 7868.6 ${\mu}g\;g^{-1}$ with an average of 3670.0 ${\mu}g\;g^{-1}$. Total saponin content including both group A and B contents ranged from 2502.8 to 8764.0 ${\mu}g\;g^{-1}$ with an average of 4154.9 ${\mu}g\;g^{-1}$. Five promising landraces for use as breeding materials that showed higher than any other landraces in group B and total saponin content were IT226841, IT226761, IT226841, IT226828 and IT228534. IT226841 showed the highest saponin content with group B content of 7868.5 ${\mu}g\;g^{-1}$ and total saponin content of 8764.0 ${\mu}g\;g^{-1}$ among landraces. In the relationship among each components, group A saponin content showed a positive correlation with group B saponin content (r=0.3708) and total saponin content (r=0.5119). Group B saponin content showed a very high positive correlation (r=0.9876) with total saponin content. When landraces were compared for the total saponin content based on collected location, landraces from North Korea showed the highest, Gyeonggi-do showed second highest content, followed by Chungcheongbuk-do, Gangwon-do, Gyongsangbuk-do, Chungcheongnam-do, Gyongsangnam-do, Jeollabuk-do and Jeollanam-do. Landraces were also grouped according to seed size and seed coat color. Small seed group was higher than medium and large seed groups that showed no significant difference in total saponin content. Seed coat colors showed no significant difference in total saponin content.

Effects of Growth Period and Cumulative Temperature on Flowering, Ripening and Yield of Soybean by Sowing Times (파종시기별 생육일수 및 적산온도 변화가 콩의 개화, 등숙 및 수량에 미치는 영향)

  • The purpose of this research was to analyze the effect of different sowing times on the flowering and maturing of major soybean cultivars by varying day length and temperature in the central plain region. The average of growth period and cumulative temperature in five test cultivars by sowing times were 121 days and 2,972℃ on June 1, respectively and gradually decreased to 85 days, 2,042℃, respectively on July 20. Analysis of the flowering response according to the sowing times showed that flowering was greatly influenced by the decrease of photoperiod until the sowing on July 10, and the minimum number of days for flowering were 27 days, 36 days, respectively in early and mid-rate maturing type in the central plain region. Daepung 2 is classified to the same ecotype with Daewonkong, the total number of growing days was not different between two cultivars, but ripening period (R2-R6) was longer by 5 days and yield was higher by 11% in Daepung 2. The maturity rate was also high and safe enough to maintain more than 90% through the entire sowing times. This ecological characteristic can be usefully applied as a section index for breeding environmental stress resistant and high yielding soybean varieties. The yield of 4 domestic cultivars (except TI196944) sowing on July 20 were 85~92% levels compared to sowing on June 20.
