Ecological control can contribute to the sustainibility of vegetation management systems by reducing the input currently derived from non-renewable fossil energy sources. The use of turfgrass mixtures is an important tool in turf management. Turfgrass mixtures of two or more compatible and adapted species provide improved tolerance to pest and environmental stress, more so than monostands. The objectives of this study were to evaluated turf insects, pests and zoysia large patch control by turgrass mixtures. In April 2001 and 2002, plots were inoculated with 50g of Rhizoctonia solani AG2-2LP inoculum. Inoculum were treated within a 29cm diamater circle at Zoysia japonica, Zoysia japonica, Poa pratenis, or Festuca arundinacea mixtures. After four weeks, disease severity in each plot was determined. plot area visual ratings were assessed visually on a linera 0 to 100%. In August 2001 and October 2002, turf insects and pests in each plot were investigated in 10cm deep soil cores with 8cm diameters using hole cut. Zoysia large patch affected zoysiagrass monostands more severly than zoysiagrass and cool-season turfgrasses mixtures. It was suggested that the barrier effect of cool-season turfgrass suppressed zoysia large patch in the mixture of zoysiagrass and cool-season turfgrasses. Also, warm-season and cool-season turfgrasses mixtures suppressed insect populations more efficiently than warm-season turfgrass monostands.
The optimal conditions for cellulase and xylanase production by Trichoderma reesei 28217 were studied for enzymatic deinking of old newspaper. The amounts of cellulase and xylanase from the strain was varied by initial medium pH, Tween 80, inoculum size of spore suspension, and carbon and nitrogen sources. The optimal conditions for cellulase production were pH 5.0-6.5, 0.02% of Tween 80, 0.5-1.0% of inoculum size of spore suspension ($1{\times}10^{7}$/ml). cottonseed meal as nitrogen source, and corn flour as carbon source. On the other hand, the optimal conditions for xylanase production were pH 6.5, 0.01% of Tween 80, corn steep liquor as nitrogen source, and disintegrated old newspaper as carbon source. The inoculum size for xylanase production was the same as for cellulase production. The concomitant production of cellulase and xylanase in shake flask culture was efficiently induced in the medium containing 0.5% cottonseed meal as nitrogen source and 1.0% old newspaper and 2.0% corn flour as carbon sources. In this case the activities of cellulase and xylanase produced were 6.11-7.22 IU/mJ and 97.7 IU/ml. respectively. However, the cellulase production in $5{\ell}$ fermentor scale was slightly decreased compared with that in flask scale. Moreover, xylanase production was severely reduced in a fermentor scale. The study for the reason of decreased enzyme production in fermentor is further needed.
Deterioration of pod and seed quality by pod and stem blight and purple blotch is a serious problem in the production of vegetable soybean. Major inoculum sources for the causal pathogens, Phomopsis spp. and Cercospora kikuchii, are soybean straw and debris. Phomopsis spp. have been known to be either limited for hyphal growth or latent in immature soybean tissues. In this experiment, cultural practices using these ecological traits of the pathogens were applied as a control measure. In plastic mulching, seed infection was remark ably reduced by drip irrigation as compared to overhead sprinkling, but not reduced in no mulching cultivation. Control value of plastic mulching was 28.0% for pod and stem blight and 29.3% for purple blotch, which was lower than that of benomyl application. By the first week in June, 78% of overwintered soybean straw examined contained matured alpha spores in pycnidia, acting as primary inoculum. Secondary inoculum of phomopsix spp. was observed abundantly throughout from mid June to September on fallen cotyledons from current crops and subsequently was found on petiole debris. Consequently, both plastic mulching and drip irrigation were effective to some extent for controlling pod and stem blight and purple blotch without significant reduction in soybean yield, and the cultural practices in combination with field sanitation resulted in removing more the secondary inoculum.
Under standard condition (Han, et al., 1990: glucose 1%-nitrate 0.1% minimal medium, 30 ml in 9 cm plate, $10^6$ cells of inoculum per plate), wild type of Aspergillus nidulans developed both sexual and asexual organs in ballance, while velA1 mutant developed asexual ones preferentially. Increase of glucose concentration did not significantly affect the asexual sporulation. However, development of sexual organs were largely affected. It was greatly enhanced when favorable nitrogen source, for example, casein hydrolysate was added, which is contrary to the case of Neurospora or Saccharomyces where limitation of N source induces sexual development. On most of moderate C sources asexual development in $velA^+$ strain was largely inhibited except acetate on which only asexual spores were produced, while that in velA1 mutant strain was not affected. Lactose promoted the sexual development even in velA1 mutant indicating that lactose itself or its metabolic intermediate may induce sexual development independent of allelic state of velA gene. On other moderate favorable C sources, glycerol, galactose and ethanol, asexual development was largely inhibited in $velA^+$ strain but not in velA1 mutant strain. Sexual organs were, however, never produced on acetate. These results suggested that asexual development of wild type is largely dependent on C sources and the velA gene is involved in the repression of asexual development in not-enough-grown (non-competent) thalli resulting in preferential progression of sexual development.

This work aims at investigating the production of yellow, orange, and red natural colorants in a submerged culture of Penicillium purpurogenum DPUA 1275. For this purpose, different experimental conditions evaluating the effect of incubation time, type and size of inoculum, and different carbon and nitrogen sources were performed. Furthermore, the growth kinetics were obtained in the conditions of $10^8$ spores/ml and 5 mycelia agar discs during 360 h. These experiments showed that 5 mycelia agar discs and 336 h promoted the highest yellow (3.08 $UA_{400nm}$), orange (1.44 $UA_{470nm}$), and red (2.27 $UA_{490nm}$) colorants production. Moreover, sucrose and yeast extract were the most suitable carbon and nitrogen sources for natural colorants production. Thus, the present study shows a new source of natural colorants, which can be used as an alternative to others available in the market after toxicological studies.

Dispersal of Xanthomonas axonopodis pv. citri, causing citrus bacterial canker disease on Unshiu orange was investigated at previously infested plots at Seogwipo in Jeju island of Korea. The bacterial pathogen overwintered in lesions started to multiply at tate May, and disease firstly observed one month after detection of phage from lesions. The disease gradually increased, however, it dispersed non-directionally to nearby plants from inoculum sources. Diseased plants were aggregated to form a cluster throughout the experiment. Population dynamics of phage on symtomless leaf surface and the disease severity were compared in the nursery, Increase of phage population on symptomless leaf surface preceded one month to that of the disease severity Population of phage increased constantly from late July to October, however, the disease severity decreased from late August to late October. It was assumed that the decrease of disease severity might be due to disease-induced defoliation.
A suitable packing material for biofiltration of monoaromatic solvent vapors was selected among various types of packing materials such as peat, bark chips, vermiculite, and Hydroballs. A previously isolated strain, Pseudomonas pseudoalcaligenes BTXO2, which could utilize toluene, m-and p-xylene as carbon and energy sources was used as a biofilter inoculum. Four glass biofilters (6 cm dia. x 60 cm) were individually packed with each of the packing materials and solvent vapors were passed through the columns. During three weeks of peat biofilter operation, average removal efficiencies of toluene, m-and p-xylene were 90.4%, 95.3%, and 82.1%, respectively. With the other packings, the efficiencies were in the range of 10.1 to 58.6% which were significantly lower than those of the peat biofilter. The peat biofilter was continually operated for approximately nine months and the biofilter sustained its degradation activity during the operation period with minimal maintenance. At steady state, average removal rates of toluene, m- and p-xylene vapors were estimated as 14.2, 5.5, and 8.1 g m$\^$-3/ packing h$\^$-1/, respectively.
Sooty blotch and flyspeck (SBFS), a disease caused by a complex of fungi, results in substantial economic losses for commercial growers of sweet persimmon (Diospyros kaki L.) in Korea. However, many species causing SBFS in Korea have not been identified and sources of inoculum are uncertain. Based on mycological characteristics, pathogenicity, and molecular data, the causal fungi were identified as Dissoconium sp. and Zygophiala wisconsinensis. This is the first report of SBFS of sweet persimmon in Korea.
The effect of fermentation parameters and medium composition on the simultaneous mycelial growth and exo-polymer production from submerged cultures of Ganoderma applanatum was investigated in shake-flask cultures. The optimum initial pH for mycelial growth and exo-polymer production was 5.0 and 6.0, respectively. The optimum temperature was $25^{\circ}C$ and the optimum inoculum content was 3.0% (v/v). The optimal carbon and nitrogen sources were glucose and corn steep powder, respectively. After 12 days fermentation under these conditions, the highest mycelial growth was 18.0 g/l and the highest exo-polymer production was 3.9 g/l.
As a basic studies for the laboratory scale production of alfalfa inoculum, Rhizobium meliloti M14 was characterized for its carbon and nitrogen sources, and some parameters for broth cultivation in a chemostat were studied by semi-continuous operation. The result s obtained were as follows. 1. Growth rate of the strain was increased by disaccharides than by monosaccharides tested, and pentoses resulted in poor growth than hexoses. Sugar alcohols including inositol supported the best growth among sugars. 2. Mannitol in the yeast-mannitol-broth was substituted by natural carbon sources such as malt extract or molasses. 3. Ten per cent of fresh yeast water appeared to supply enough amount of growth factor s for the strain, and the effect was equivalent to 0.24 percent of the commercial yeast extract powder. 4. Batch growth of the stain in a chemostat, New Brunswick Micro Ferm 28L, reached in the early stationary growth phase of $5{\sim}7{\times}10^9cells/ml$ after 36 hours of incubation. The culture at this stage was switched to semi-continuous cultivation, and the culture broth of four-fifth of the working volume was recovered every 24 hours when the maximal count was obtained.
