Hydrogen Production by Purple Sulfur Bacteria, Thiocapsa roseopersicina in Photoheterotrophic Culture Condition (홍색유황세균, Thiocapsa roseopersicina의 photoheterotrophic 조건에서의 수소생산)

  • The purple sulfur phototrophic bacterium, Thiocapsa roseopersicina NCIB 8347 has been studied on hydrogen production and cell growth under different culture conditions, such as light source, light intensity, and growth temperature. T. roseopersicina showed maximum cell growth of 1.38 and 1.42 g-DCW/L under 7.5-10 klux of halogen and fluorescent light, respectively, and produced maximum amount of hydrogen with values of 0.90 and 0.48 $mL-H_2/mg$-DCW under the irradiation of 10 klux of halogen and fluorescent light, respectively. The optimum growth temperature for hydrogen production was $26^{\circ}C$, and hydrogen production rate was lowered over $30^{\circ}C$. When T. roseopersicina was grown photoheterotrophically under irradiation of 8-9 klux of halogen lamp, the generation time was 4.2 hr. The strains started producing hydrgen from the middle of the logarithmic growth phase and continued until succinate concentration leveled out.

  • Non-point pollutants from surface runoff during rainfall exert adverse effects on urban river water quality management. In particular, the first flush effect during the initial phase of rainfall can deliver significant amounts of pollutant loads to surface waters with extremely high concentrations. In this study, a sustainable first flush effect management system was developed by using settling and filtration that require no additional power or chemicals. A pilot scale experiment has shown that the removal of total suspended solid (TSS), total nitrogen (TN) and total phosphorus (TP) are in ranges of 84 - 95%, 31 - 46%, and 42 - 86%, respectively. An Integrated Stormwater Runoff Management System (ISTORMS) was also developed to efficiently manage the developed system by linking weather forecast, flow rate and water quality modeling of surface runoff and automatic monitoring systems in fields and in the system. This study can provide effective solutions for the management of urban river in terms of both quantity and quality.

  • The stormwater runoff from paved area are highly polluted because of particulate materials as well as metals from various vehicular activities. The Division of Road Maintenance in Ministry of Construction and Transportation was recently developed the Guidelines of Environment-kindly Road Maintenance. It is actually requiring the BMP construction to control the nonpoint source pollution as based on the TMDL program. This research is carried out in order to define the characteristics of stormwater runoff from the toll-gate of highways since 2006, which is actually one of the main pollutant sources of paved areas. This monitoring is the first phase work for establishing the treatment facilities in the toll-gates. The one of the main characteristics from toll-gate runoff is the first flush phenomenon containing lots of sediments and metal compounds at the beginning of a storm event. Usually it is used to determine the size of treatment facilities and to calculate the reduced pollutant mass in the facility. The research results shows that the mean EMC vaules for heavy metals are determined to $274.3{\mu}g/L$ for Cd, $1,273.4{\mu}g/L$ for Cr, $1,822.0{\mu}g/L$ for Cu, $6,504.9{\mu}g/L$ for Fe, $14,930.3{\mu}g/L$ for Pb, and $714.1{\mu}g/L$ for Zn. Also the metal mass loadings from the toll-gates are calculated using EMC, watershed area and storm duration.

  • HBsAg. was identified in the urine of the patients positive for serum HBsAg. by Tripatzis in 1970. In 1977, Hourani et al reported the incidence of HBsAg. in urine was about 52% in the patients positive for serum HBsAg. with hemodialysis treatment due to chronic renal failure. A series of studies on the HBsAg. in urine has revealed the urine of the patients positive for serum HBsAg. to be important source of infection. But there's much room to debate on the relationship of HBsAg. in urine with infectivity and the exact mechanism of urinary emergence of HBsAg. The authors detected HBsAg. in serum and urine by employing sandwitch solid-phase rad ioimmunoassay, and performed urinalysis, liver function test and renal function evaluation. Percutanous liver and/or kidney biopsis were done. Among 38 renal disease patients, 9 cases (23.4%) were shown to be positive for serum HBsAg. and 5 cases (55.5%) among above 9 patients positive for urine HBsAg.. 56 cases (67.4%) of 83 liver disease patients revealed positive for serum HBsAg. but only 11 cases (13.2%) among the 56 cases positive fo urine HBsAg. All 10 renal and liver disease patients revealed positive serum HBsAg., and among the 9 cases (90%) positive for urine HBsAg.. In the 25 patients positive for urine HBsAg. all of 5 renal patients and 9 renal and liver patients had hematuria or/and proteinuria above 2 positive for albumin. But in the 11 liver patients 6 cases (55.1%) were normal findings. And there's no significant difference in cpm of urine HBsAg. between the patient positive for serum HBsAg. and negative, and in cpm of serum HBsAg. between liver and renal disease patients. But there's statistical significance in cm of urine HBsAg. between renal and liver diseases.

Change of Fractured Rock Permeability due to Thermo-Mechanical Loading of a Deep Geological Repository for Nuclear Waste - a Study on a Candidate Site in Forsmark, Sweden

  • Opening of fractures induced by shear dilation or normal deformation can be a significant source of fracture permeability change in fractured rock, which is important for the performance assessment of geological repositories for spent nuclear fuel. As the repository generates heat and later cools the fluid-carrying ability of the rocks becomes a dynamic variable during the lifespan of the repository. Heating causes expansion of the rock close to the repository and, at the same time, contraction close to the surface. During the cooling phase of the repository, the opposite takes place. Heating and cooling together with the, virgin stress can induce shear dilation of fractures and deformation zones and change the flow field around the repository. The objectives of this work are to examine the contribution of thermal stress to the shear slip of fracture in mid- and far-field around a KBS-3 type of repository and to investigate the effect of evolution of stress on the rock mass permeability. In the first part of this study, zones of fracture shear slip were examined by conducting a three-dimensional, thermo-mechanical analysis of a spent fuel repository model in the size of 2 km $\times$ 2 km $\times$ 800 m. Stress evolutions of importance for fracture shear slip are: (1) comparatively high horizontal compressive thermal stress at the repository level, (2) generation of vertical tensile thermal stress right above the repository, (3) horizontal tensile stress near the surface, which can induce tensile failure, and generation of shear stresses at the comers of the repository. In the second part of the study, fracture data from Forsmark, Sweden is used to establish fracture network models (DFN). Stress paths obtained from the thermo-mechanical analysis were used as boundary conditions in DFN-DEM (Discrete Element Method) analysis of six DFN models at the repository level. Increases of permeability up to a factor of four were observed during thermal loading history and shear dilation of fractures was not recovered after cooling of the repository. An understanding of the stress path and potential areas of slip induced shear dilation and related permeability changes during the lifetime of a repository for spent nuclear fuel is of utmost importance for analysing long-term safety. The result of this study will assist in identifying critical areas around a repository where fracture shear slip is likely to develop. The presentation also includes a brief introduction to the ongoing site investigation on two candidate sites for geological repository in Sweden.

Improved Physical Properties of Ni-doped $BiFeO_3$ Ceramic

  • Recently, multiferroic materials have attracted much attention due to their fascinating fundamental physical properties and potential technological applications in magnetic/ferroelectric data storage systems, quantum electromagnets, spintronics, and sensor devices. Among single-phase multiferroic materials, $BiFeO_3$, in particular, has received considerable attention because of its very interesting magnetoelectric properties for application to spintronics. Enhanced ferromagnetism was found by Fe-site ion substitution with magnetic ions. In this study, $BiFe_{1-x}Ni_xO_3$ (x=0 and 0.05) bulk ceramic compounds were prepared by solid-state reaction and rapid sintering. High-purity $Bi_2O_3$, $Fe_3O_4$ and NiO powders were mixed with the stoichiometric proportions, and calcined at $450^{\circ}C$ for 24 h to produce $BiFe_{1-x}Ni_xO_3$. Then, the samples were directly put into the oven, which was heated up to $800^{\circ}C$ and sintered in air for 20 min. The crystalline structure of samples was investigated at room temperature by using a Rigaku Miniflex powder diffractometer. The Raman measurements were carried out with a Raman spectrometer with 514.5-nm-excitation Ar+-laser source under air ambient condition on a focused area of $1-{\mu}m$ diameter. The field-dependent magnetization and the temperature-dependent magnetization measurements were performed with a vibrating-sample magnetometer. The x-ray diffraction study demonstrates the compressive stress due to Ni substitution at the Fe site. $BiFe_{0.95}Ni_{0.05}O_3$ exhibits the rhombohedral perovskite structure R3c, similar to $BiFeO_3$. The lattice constant of $BiFe_{0.95}Ni_{0.05}O_3$ is smaller than of $BiFeO_3$ because of the smaller ionic radius of Ni3+ than that of Fe3+. The field-dependent magnetization of $BiFe_{0.95}Ni_{0.05}O_3$ exhibits a clear hysteresis loop at 300 K. The magnetic properties of $BiFe_{0.95}Ni_{0.05}O_3$ were improved at room temperature because of the existence of structurally compressive stress.

  • The screening methodology modeling, dispersion modeling procedures for continuous and instantaneous releases of the gas phase flow from the storage tank and pressure relief valve were considered. This study was performed to develop the screening methodology for prediction and control of hazardous/toxic gas releases by estimating the 1-hr average maximum ground-level concentration of $Cl_2$ gas vs. downwind distance by incorporating source term model including the general/physical properties of released material and release mode of the $Cl_2$ storage tank of the chemical plant facilities, dispersion model, and meteorological/topographical data into the TSCREEN model. As the results of the study, it was found that dispersion modes of the dense gas were affected by the state of the released material, the released conditions, physical-chemical properties of released material, and the released modes (continuous and instantaneous releases), and especially largely affected by initial (depressurized) density of the released material and release emission rate as well as the wind velocity. Especially, this study was considered to release hazardous material as meteorological data. It was thought that this screening methodology can be useful as a preliminary guideline for application of the refined analysis model by developing the generic sliding scale methodology for various senarios selected.

Identification of process generating formaldehyde in a furniture manufacturer (특정 가구 제조 공장의 포름알데히드 발생 공정 노출 평가)

  • Formaldehyde is defined as carcinogen causing leukaemia, lymphoma or nasopharyngeal carcinoma at high level of exposure. Furniture-manufacturing workers can be exposed to formaldehyde, which implies serious impact on health of the workers. The authors carried out ambient monitoring of formaldehyde in the field, and identified the source of formaldehyde generated during the working process by testing the condition in the laboratory settings. After sampling formaldehyde in the air with 2,4-DNPH (2,4-dinitrophenylhydrazine) coated silica gel, we extracted formaldehyde derivative with acetonitrile and analyzed the extract using HPLC with UV detector at 360 nm. Formaldehyde was separated by ACQUITY UPLC BEH $C_{18}$ column at a flow rate of 0.5 mL/min using 45% acetonitrile as mobile phase. The workers were exposed to higher level of formaldehyde than normal air. Formaldehyde up to 0.31 ppm was detected in the process of veneer attachment, which exceeded 0.3 ppm, the ceiling value of ACGIH standard. The laboratory test of measuring formaldehyde generated from the glue and veneer used in the attachment process resulted in more formaldehyde generation as the temperature increased, and more from the veneer. Heating the veneer to $100-150^{\circ}C$ following the real condition of the manufacturing site generated 1.14-2.70 ppm of formaldehyde from the sample, which was 2-5 times higher level than Korean limit of exposure (0.5 ppm). As the workers handling and processing the veneer which was produced by wet process had high possibility to be exposed to formaldehyde, urgent improvement and management of working environment of furniture manufacturer is demanded.

Isolation of Pseudoalteromonas sp. HJ 47 from Deep Sea Water of East Sea and Characterization of its Extracellular Protease (동해 심층수로부터 Pseudoalteromonas sp. HJ 47의 분리 및 체외단백질분해효소 특성)

  • Proteases are enzymes that break peptide bonds between amino acids of other proteins and occupy a crucial position with respect to their applications in both physiological and commercial fields. In order to screen new source of protease, bacteria producing extracellular proteases at low temperature were isolated from deep sea water of East Sea, Korea. A bacterium showing the best growth rate and production of an extracellular protease at low temperature was designated HJ 47. The DNA sequence analysis of the 16S rRNA gene, phenotypic tests and morphology led to the placement of this organism in the genus Pseudoalteromonas. Although maximal growth was observed at $37^{\circ}C$, enzyme production per culture time was maximum at $20^{\circ}C$. At this temperature, extracellluar protease production was detected from the end of the exponential phage to stationary phase, and maximal at 15 hours after initial production. The optimum temperature and pH of the protease were found to be $35^{\circ}C$ and 8.

Nano-safety Management and Exposure Assessment of Nanomaterials Producing Facilities (나노물질 생산시설의 환경노출 평가와 안전관리)

  • With the development of nanotechnology, nano-consumer products have been popularized. For the past 10 years, potential risk of nanomaterials to human and environment have been raised carefully. Especially, workers, who directly handle nanomaterials in laboratories and manfacturers, will lead to direct exposure of nanomaterials. Therefore, direct exposure assessment and field monitoring of nanomaterials are required to assess and manage the nanomaterial exposure to human and environment. In this work, two nano-manufacturing companies, which had plasma and sol-gel processes, were selected to analyze the main exposure source and process with in-situ SMPS (scanning mobility particle sizer) and ex-situ TEM (transmission electron microscopy). The results showed that the colloidal nanoparticle in liquid phase was easily evaporated and monitored by SMPS. Most serious thing is that the workers does not know about the potential risk of nanomaterials, and thus they are not taking proper protection activities, such as PPE (personal protective equipment). Therefore, exposure assessment for nanomaterial handling facilities should be additionally carried out, and nano-safety management protocols are also provided.
