체세포 수에 의한 유방염의 재 정의 (Redefining Mastitis Based on Somatic Cell Count)

  • 체세포 수(SCC = somatic cell count)는 유방염과 밀접한 관계가 있는 생리적 반응의 표시이다. 그래서 유질과 유방염의 관리에 대단히 중요한 영향인자로 중요하게 사용된다. 현재 그 수에 의한 세계적인 평가는 나날이 연구의 성과가 보고되고 있다. 우리는 체세포 수로 많은 애로를 가지고 있으면서도 감소대책과 수에 대한 국제적 흐름에 민감하지 못한 것이 현실로 보인다. 이상과 같은 것을 감안하여 근래의 체세포 수와 유방염에 관한 정보를 제공하려고 한다.

사료내 ${\beta}$-glucanase 활성 강화 고역가 복합효소제 첨가급여가 착유우의 유생산 및 체세포수 변화에 미치는 영향 (The Effects of Dietary Enzyme Mixture Reinforced with ${\beta}$-Glucanase Activity on Mini Production and the Change of Somatic Cell Count in Lactating Dairy Cows)

  • In recent years, many researches are actively undertaken for environmental-friendly animal production according to the increased understanding about food safety because of the outbreak of various diseases such as mad cow disease, Foot and mouth disease and Poultry Influenza virus. However, high quality(higher safety)- animal production may not be successful without increasing of disease resistance of animal and the improvement of feeding environment. To increase the disease resistance is able to be accomplished by stimulating the immune function. The present study was undertaken to investigate the effects of enzyme mixture reinforced with ${\beta}$-glucanase activity which degrade polysaccharide to release ${\beta}$-glucan known as stimulator of immune function on the change of milk production and somatic cell count. After 12weeks of experimental feeding, milk production tended to be increased and somatic cell count was decreased from average $227{\times}10^4$ to $37.1{\times}10^4$. Milk protein and solid-fat content were tended to increase but milk fat showed decreasing tendency by the feeding of enzyme mixture. All together, it has been suggest6d that the improvement of high quality milk production may be possible through the dietary addition of immune modulating enzyme mixture in lactating dairy cows.

체세포수 감소를 위한 우유여과시스템 개발 (Development of a Milk Filtering System for Decreasing Somatic Cell Count)

  • The present study was conducted to develop a milk filtering system for decreasing somatic cell count (SCC) in bulk tank milk. The pore sizes of the filter were 0.1, 0.3, 0.4, 0.5 and $0.8{\mu}m$. The rate of SCC reduction of $1^{st}$ grade milk on $0.1{\mu}m$ filter was 76% and significantly higher than other treatments. The rates of SCC reduction for 0.3, 0.4, 0.5 and $0.8{\mu}m$ were 35, 32, 18 and 6.4%, respectively. The effects of the milk filtering system on bacterial count and milk fat content were minimal. The milk flow rates per minute between the filter sizes were similar. But discharge pressures were increased as the pore size of the filter decreased. In conclusion, Considering the rate of SCC reduction, discharge pressure and cost, $0.4{\mu}m$ filter could be recommended.

Immunomodulatory and Therapeutic Potential of Enrofloxacin in Bovine Sub Clinical Mastitis

  • Immunomodulatory and therapeutic potential of Enrofloxacin was studied in bovine sub clinical mastitis (SCM). The therapeutic efficacy was adjudged by Somatic cell count and Total bacterial count of the milk, whereas, the immuno modulatory potential of the drug was assessed by measuring myeloperoxidase (MPO) and acid phosphates (ACP) enzyme level in the milk leukocytes. Forty-five cows were divided into three equal groups. Gr I consisting 15 cows served as healthy control, whereas, 30 cows (SCM), Gr II and Gr III, selected on the basis of California Mastitis Test (CMT) positive reaction. Gr II cows received 150 mg of Enrofloxacin, once a day for three days and Gr.III received sterile 5 ml PBS (pH 7.4) for 7days, both the treatment were given by intramammary route. The observation was made up to 30 days post-treatment (PT). The CMT of the healthy milk was negative (0), whereas, it ranged between 1 point score and 2 point score in SCM. The Somatic cell count (SCC) and Total bacterial count (TBC) decreased significantly (p<0.05) on day 3 PT in GrII cows in Enrofloxacin treated group, however, such changes were insignificant in PBS treated group. Traces of MPO and ACP enzyme were found in the healthy milk. The mean ACP level enhanced by 70% on day 3 PT in GrII and only 18.7% in Gr. III cows. The mean MPO level enhanced to 32% in Gr. II and 18 % in Gr. III cows on day 3 PT. Concomitant use of Enrofloxacin in SCM at sub optimal dose was found to reduce the bacterial load by increasing the bactericidal enzyme level in the milk polymorphonuclear cells (PMNs) in bovine SCM, which indicates its immunomodulatory potential in mastitis.

  • Mastitis is characterized by physical, chemical and bacteriological changes in the milk and pathological changes in the glandular tissue of the udder and affects the quality and quantity of milk. The bacterial contamination of milk from the affected cows render it unfit for human consumption and provides a mechanism of spread of diseases like tuberculosis, sore-throat, Q-fever, brucellosis, leptospirosis etc. and has zoonotic importance. Somatic cell count (SCC) is a useful predictor of intramammary infection (IMI) that includes leucocytes (75%) i.e. neutrophils, macrophages, lymphocytes, erythrocytes and epithelial cells (25%). Leucocytes increase in response to bacterial infection, tissue injury and stress. Somatic cells are protective for the animal body and fight infectious organisms. An elevated SCC in milk has a negative influence on the quality of raw milk. Subclinical mastitis is always related to low milk production, changes to milk consistency (density), reduced possibility of adequate milk processing, low protein and high risk for milk hygiene since it may even contain pathogenic organisms. This review collects and collates relevant publications on the subject.

분광분석법을 이용한 우유의 체세포수 측정기술 개발 (Development of Measuring Technique for Somatic Cell Count in Raw Milk by Spectroscopy)

  • The objective of this study was to develop models to predict SCC (somatic cell count) in unhomogenized milk by visible and near-infrared (NIR) spectroscopic technique. Total of 100 milk samples were collected from dairy farms and preserved to minimize propagation of bacteria cells during transportation. Reductive reagents such as methyl red, methylene blue, bromcresol purple, phenol red and resazurin were added to milk samples, and then colors of milk were changed based on SCC of milk. For optimal reductive reagents, reaction time was controlled at 3 level of reaction time. A spectrophotometer was used to measure reflectance spectra from milk samples. The partial least square (PLS) models were developed to predict SCC of unhomogenized milk. The PLS results showed that milk samples with reductive reagents had a good correlation between predicted and measured SCC at 5 minutes of reaction time in the visible range. The PLS models with resazurin reagent had the best performance in $400{\sim}600\;nm$. The prediction results of milk samples with resazurin had 0.86 of correlation coefficient and 14,184 cell/mL of SEP.

산양유의 체세포수 등급 설정에 관한 연구 (Study of the Somatic Cell Count Grade of Goat Milk in Korea)

  • 본 연구는 최근 웰빙식품으로 소비가 증가하고 있는 산양유의 체세포수 등급제정을 위해 (주)엠젠에 납유하고 있는 12개 목장에서 2006년 12월부터 2007년 11월까지 납유한 산양유의 체세포수를 분석하였다. 프랑스와 노르웨이의 체세포 등급체계 및 우리나라 원유의 체세포 등급체계를 고려할 때, 산양유의 체세포수 등급체계는 1급이 mL당 100만 미만, 2급이 100-150만 미만, 3급이 150-200만 미만, 4급이 200-250만 미만, 5급이 250만 초과로 규정하는 것이 바람직할 것으로 생각된다. 이러한 체계를 가정할 때 우리나라 산양유의 1등급 비율이 26.2%로 다소 낮고, 5등급 비율이 11.8%로 다소 높으나 이를 통해 산양유의 체세포수가 개선될 것으로 기대된다.

Relationship of Somatic Cell Count, Physical, Chemical and Enzymatic Properties to the Bacterial Standard Plate Count in Different Breeds of Dairy Goats

  • The objective of the present study was to investigate the accuracy of mastitis diagnostic indicators for different dairy goat breeds. Biweekly milk samples were collected from individual half mammary gland of seven Saanen and seven Alpine dairy goats in the period of 40 to 120 days in milk. With threshold value set at 2.8 and 3.1 for Alpine and Saanen dairy goats, respectively, log (SPC) offered good sensitivity (0.89, 0.93), specificity (0.88, 0.95), positive predictive value (0.75, 0.85) and negative predictive value (0.95, 0.98) as a mastitis diagnostic tool. The correlations of log (SPC) with milk yield, log (SCC), ALP, LDH, $Na^{+}$, $K^{+}$ and EC were significant in Saanen dairy goats (p<0.05), with the highest correlation coefficient (0.653) existing between log (SPC) and log (SCC). The correlations of log (SPC) with milk yield, milk fat, milk protein, log (SCC), $Na^{+}$, $K^{+}$, EC were significant in Alpine dairy goats (p<0.05), with the highest correlation coefficient (0.416) existing between log (SPC) and log (SCC). There were different best-fit regression equations with different multiple diagnostic indicators for Saanen and Alpine dairy goats. In conclusion, different breeds of dairy goats may have to adapt different mastitis diagnostic parameters for a better diagnosis.

Effect of Subclinical Mastitis on Milk of Cross Bred Sahiwal × Jersey Cows: A Biochemical Study

  • The experiment was undertaken to study the effect of subclinical mastitis on the biochemical parameters of milk in crossbred (Sahiwal $\times$ Jersey) cows. Subclincial mastitis was judged using California Mastitis Test and graded on a scale of 0 to 2. Twenty six milk samples of each grade were subjected to somatic cell count. Separated milk whey was analysed for total protein, immunoglobulin and trace mineral content. Results indicate linear increases in somatic cell count, total protein and immunoglobulin concentrations in milk with increase in grade of subclinical mastitis. However, copper, zinc, manganese and iron concentration in milk was not affected by grade of mastitis.
