용매증발법에 의한 부피바카인 마이크로스피어의 제조 및 평가 (II) (Preparation and Evaluation of Bupivacaine Microspheres by a Solvent Evaporation Method (II))

  • Various bupivacaine-loaded microspheres were prepared using poly(d,1-lactide) (PLA) and poly(d,1-lactic-co-glycolide) (PLGA) by a solvent evaporation method for the sustained release of drug. The effects of process conditions such as drug loading, polymer type and solvent type on the characteristics of microspheres were investigated. The prepared microspheres were characterized for their drug loading, size distribution, surface morphology and release kinetics. Drug loading efficiency and yield of PLGA micro- spheres were higher than those of PLA microspheres. The prepared microspheres had an average particle size below 5${\mu}{\textrm}{m}$. The particle size range of microspheres was 1.65~2.24${\mu}{\textrm}{m}$. As a result of SEM, the particle size of PLA microspheres was smaller than that of PLGA microspheres. In morphology studies, microspheres showed a spherical shape and smooth surface in all process conditions. In thermal analysis, bupivacaine-loaded microspheres showed no peaks originating from bupivacaine. This suggested that bupivacaine base was molecular-dispersed in the polymer matrix of microspheres. The release pattern of the drug from microspheres was evaluated for 96 hours. The initial burst release of bupivacaine base decreased with increasing the molecular weight of PLGA, and the drug from microspheres released slowly. In conclusion, bupivacaine-loaded microspheres were successfully prepared from poly(d,1-lactide) and poly (d,1- lactic-co-glycolide) polymers with different molecular weights allowing control of the release rate.

Correlation of the Rates of Solvolysis of Methyl Fluoroformate Using the Extended Grunwald-Winstein Equation

  • The specific rates of solvolysis of methyl fluoroformate have been measured at $40.0\;{^{\circ}C}$ in several hydroxylic solvents. Analysis with the extended Grunwald-Winstein equation leads to sensitivities toward changes in solvent nucleophilicity (l) of $1.33\;{\pm}\;0.10$ and toward changes in solvent ionizing power (m) $0.73\;{\pm}\;0.06$. For methanolysis, a solvent deuterium isotope effect of 3.98 is compatible with the incorporation of general-base catalysis into the substitution process. For four representative solvents, studies were made at several temperatures and activation parameters determined. These observations are also compared with those previously reported for alkyl halogenoformate esters and mechanistic conclusions are drawn.

Selectivity of between K+ and Na+ Ions to 12-Crown-4: QSPR Analysis by a Monte Carlo Simulation Study

  • The solvent effects on the relative free energies of binding of K+ and Na+ ions to 12-crown-4 and Dlog Ks (the difference of stability constant of binding) have been investigated by a Monte Carlo simulation of statistical perturbation theory (SPT) in several solvents. Comparing the relative free energies of binding of K+ and Na+ ions to 12-crown-4, in CH3OH of this study with experimental works, there is a good agreement among the studies. We have reported here the quantitative solvent-polarity relationships (QSPR) studied on the solvent effects the relative free energies of binding of K+ and Na+ ions to 12-crown-4. We noted that DN(donor number) dominates the differences in relative solvation Gibbs free energies of K+ and Na+ ions and DN dominates the negative values in differences in the stability constant (Dlog Ks) as well as the relative free energies of binding of K+ and Na+ ions to 12-crown-4 and p* (Kamlet-Tafts solvatochromic parameters) dominates the positive values in differences in the stability constant (Dlog Ks) as well as the relative free energies of binding of K+ and Na+ ions to 12-crown-4.

Analysis of Flavor Composition of Coriander Seeds by Headspace Mulberry Paper Bag Micro-Solid Phase Extraction

  • This paper reports the example of headspace mulberry paper bag micro solid phase extraction (HS-MPB-$\mu$-SPE) as a new sampling method for the determination of volatile flavor composition of coriander seeds. Adsorption efficiencies between two configurations of mulberry paper bag were compared, and several parameters affecting the HS-MPB-$\mu$-SPE were investigated and optimized. The optimized technique uses an adsorbent (Tenax TA, 0.1 mg) contained in a mulberry paper bag of front configuration where fine surface was outside, and minimal amount of organic solvent (0.6 mL). Linalool and $\gamma$-terpinene were found as abundant flavor compounds from coriander seeds. The limit of detection (LOD) and the limit of quantitation (LOQ) for linalool of major flavor in coriander seeds were 10.3 ng/mL and 34.4 ng/mL, respectively. The proposed method showed good reproducibility and good recovery. The HS-MPB-$\mu$-SPE is very simple to use, inexpensive, requires small sample amounts and solvent consumption. Because the solvent for extraction is reduced to only a very small volume, there is minimal waste or exposure to toxic organic solvent and no further concentration step.

  • This experiment was carried out to determine the separation condition of 1-dimethylaminonaphthalene-5-sulfony(Dansyl) derivatives of amino acids by reverse-phase high performance liquid chromatography with Nova-Pak C18 column. Determined solvent system was solvent A(200mA phosphate buffer pH 6.8 15%, acetonitrile 11%, water 74%) and solvent B(acetonitrile 65%, methanol 28%, water 7%). Linear gradient of solvent B was applied from 12% to 80% for 50min. Complete separation of 20 amino acids including asparagine and glutamine which constitute protein was achieved within 50min. As the detection limit was the range of picomole, the resolution power was excellent. Reproducibility of the retention time was less than mean $\pm$0.05min. According to the above optimum chromatographic conditions, the amino acid composition of some food and human blood was examined. The most affluent amino acid was alanine in human blood, aspartic acid and glutamic acid in soy sauce, alanine and threonine in soy milk and proline in milk and yoghurt.

Solvent-Induced Photoemissions of High-Energy Chromophores of Conjugated Polymer MEH-PPV: Role of Conformational Disorder

  • This study examined the photoemission behaviors of isolated chains of poly[2-methoxy, 5-(2'-ethylhexyloxy)-1,4-phenylenevinylene](MEH-PPV) dispersed in various solvents including dichloromethane, chloroform and tetrahydrofuran(THF). A change in polymer-solvent interactions in these solutions caused the MEH-PPV chains to adopt different local conformations, which in turn affected their radiative de-excitation pathways. For the polymer in dichloromethane and chloroform, in which the conjugated chains are relatively extended, photoemission occurs mostly at the long chromophores with lowest HOMO-LUMO energy gap. Their emission spectra showed a main peak at ${\sim}560\;nm$. Dual photoemission of high- and low-energy chromophores was observed when the conjugated chains were forced to partially collapse in a poor solvent THF. Novel high-energy peaks and a typical low-energy peak were detected at ${\sim}414\;nm$ and ${\sim}554\;nm$, respectively. The observation of the high-energy peaks indicates significant suppression of the intrachain energy transfer process, which was attributed to the increase in conformational disorder in the partially collapsed coils. An analysis of the excitation spectra suggests that the high-energy peaks belong to short chromophores constituting of one or two repeat units. This study systematically investigated the effects of polymer concentration, temperature and single bond defects along the backbone on the photoemission of the high-energy chromophores.

비수적정법을 이용한 약품분석 연구 (제1보) 약염기성혼합약품의 분리정량법에 대하여 (Studies on Pharmaceutical Quantitative Analysis by means of Non-aqueous Titration Method (I) Isolative Determination of Mixed Weak Basic Chemicals.)

  • The study of the isolative determination of the mixed weak bases of INAH and NA-Pas potentimetrically considering the properties of solvents for the INAH and Na-PAS, dielectric constant and solvative properties of solvents are described. The methanol: dioxan (4:1) and glacial acetic acid: dioxan (4:1) are studied first as the mixed solvent, using the N/10 perchloric acid and glacial acetic acid solution as the titrant. The authors found that there is no inflection on INAH with the methanol: dioxan solvent system and on Na-PAS at glacial acetic acid dioxan solvent system. By applying methanol glacial acetic acid dioxan (1:1:1) solvent system, Na-PAS and INAH were successfully determined isolatively from the mixed sample, showing the distinguished inflections respectively as shown in the titration curves in figures 3 and 4. It is found that this method could save considerable time for the isolative determination of the mixed sample of week bases as Na PAS and INHA which were quite difficult to be determined by the present routine at control laboratory.

Solvothermal synthesis and characterization of silver nanoparticles

  • Among the various nanoparticles reported for commercial applications, considerable interest has been generated by the use of silver nanoparticles. Owning to extremely small size, silver nanoparticles exhibit enhanced properties when compared with the bulk material. In this research work, silver nanoparticles were prepared by the reduction of silver salt with a reducing agent by a solvothermal method using different solvent mediums such as ethanol, hexane, toluene and acetone with water. The prepared silver nanoparticles were characterized systematically by X-ray diffraction (XRD), particle size analysis and scanning electron microscope (SEM). The results revealed the formation of pure silver phase and nano-sized particles. Among the different solvent mediums used, the silver nanoparticles prepared by hexane and water as solvent mixture resulted in very low particle size.

하천저질중의 유기물질에 관한 연구 (Studies on the Organic Matters in River Sediments)

  • The writer measured methemoglobin formation with solvent (n-hexane, ethylacetate and methanol) organic extracts from the sediments of the Wonju stream run through Wonju city on Octover 2nd, 1985. Hemoglobin was converted to methemoglobin with the n-hexane extract obtained from the sediment of the St. C and St. D was polluted industrial wastewater, but not with that from the St. B was polluted with the urban wastewater. The formation of methemoglobin was remarkable with the all solvent extracts from the sediment of the uper stream of the Wonju stream. Doses of the solvent extracts for 20% methemoglobin formation in the researched Wonju streams were as follows; 0.095~0.28 mg/m.$\ell$ in n-hexane extract. 0.85-1.3 mg/ m$\ell$ in methanol extract and 1.95~2.80 mg/m$\ell$ in ethylacetate extract.

An Analysis on Remediation of Soil Contaminted with Cobalt by Solvent Flushing

  • A soil whose texture is silt loam was collected for the study from an area around a nuclear facility in Korea. The equilibrium sorption coefficient between Co$^{2+}$in water and the soil was 1.51/kg, on the other hand, that between Co$^{2+}$ in EDTA and the soil was 0.21/kg. The values calculated by the developed nonequilibrium sorption code corresponded to the experimental values better than those calculated by the existing equilibrium sorption code. When an EDTA solution was used as a solvent to decontaminate Co$^{2+}$ in the soil column, the relative Co$^{2+}$ concentrations of the effluent were higher at 2~10 pore volumes than those of the case using water. The soil in the column was decontaminated by 95.5% of the total amount of Co$^{2+}$ after being flushed with EDTA solution of 20 pore volumes.e volumes.
