The preparation method of a soluble dietary fiber from oak wood (Quercus mongolica) and the effect of the soluble dietary fiber on physiological function in rat fed high cholesterol diets was investigated. The best condition for steam explosion method was 25 kgf/㎤ pressure for 6 min. The exploded samples were delignified by the filtration treatment with 1% NaOH for several times, which is the best condition. The enzymatic hydrolysis of Cellusoft cellulase was more effective than Onozuka R-10 cellulase. The manufactured soluble dietary fiber was assayed using gel permeation chromatography (GPC) and it was dissolved in water. Average molecular weight distribution of manufactured soluble dietary fiber was about 348-1,200 and it was assumed the oligomer form fraction. In order to compare the manufactured soluble dietary fiber with commercial soluble dietary fiber (pectin) on the physiological function, Sprague-Dawley male rats weighing 100$\pm$10 g were randomly assigned to one normal diet and five high cholesterol diet containing 1% cholesterol. The high cholesterol diet groups were classified to fiber free diet (FF group), 5% pectin (5P group), 10% pectin (l0P group), 5% manufactured soluble dietary fiber (5M group) and 10% manufactured soluble dietary fiber (10M group). Body weight gains in all soluble dietary fiber groups were lower than FF group. Food intakes were increased in all soluble dietary fiber groups than that of FF group. Food efficiency ratio (FER) was significantly decreased in all soluble dietary fiber groups than that of the FF group, and it was especially was highest in 10% supplemented soluble dietary fiber group. The weight of liver of the soluble dietary fiber supplemented groups were lower than those of the FF group, but weights of cecum and small intestine of all supplemented soluble dietary fiber groups were significantly increased, compared with that of FF group. The weights and water contents in feces were significantly increased by the soluble dietary fiber. The activity of the glutamic oxaloacetic transaminase in soluble dietary fiber groups were significantly decreased than those of FF group. The hepatic glutathione S-transferase activity in all soluble dietary fiber supplemented groups were higher than that of FF group. The physiological effects of the manufactured soluble dietary fiber are the same as the commercial soluble dietary fiber (pectin). Sample preparation is significantly important to the high-resolution transmission electron microscopy (HRTEM) characterization of nanomaterials. However, many general organic solvents can dissolve the necessary organic polymer support layer in TEM grid, which causes it difficult to obtain high-quality samples of oil-soluble nanomaterials. In this study, a new sample preparation method for oil-soluble nanomaterials has been developed by using the sublimable material as a transition layer. Experiments also show that there is no damage to TEM grids and high-quality HRTEM images can be obtained via this method. This approach paves the way to applicable HRTEM sample preparation of oil-soluble nanomaterials.
Water-soluble browning pigments were artily purified from Korean red ginseng through the several procedures such as fractionation by n-butanol, precipitation by ethanol, dialysis and gel filtration. At least four kinds of water-soluble browning pigment were separated from each other, two kinds of low-molecular-weight and two kinds of high-molecular-weight pigments.
Enzymatic properties of immobilized transglucosidase (TG) from Aspergillus niger was characterized and compared with soluble TG. Michaelis-Menten constant $(K_m)$ and maximum velocity $(V_{max})$ of immobilized TG were $122\;mM,\;3.9{\times}10^{-2}\;mM/min$ and in case of soluble TG, 21 mM, 0.4 mM/min. The optimum pH of immobilized TG was pH 5.0 like soluble TG but immobilized TG showed 16% and 45% higher activity than soluble TG at pH 5.0 and pH 6.0. Both of immobilized TG and soluble TG were stable from pH 2.0 to pH 9.0, and therefore their activities in these pH ranges were remained more than 90%. The temperature was optimal at $60{\sim}70^{\circ}C\;and\;70{\sim}80^{\circ}C$ for soluble TG and immobilized TG, respectively. The thermal stability of immobilized TG was significantly improved than that of soluble TG, and immobilized TG retained $32{\sim}40%$ higher activity than soluble TG. D-values from thermal inactivation of immobilized TG were 7690 sec at $65^{\circ}C$, 83 sec at $75^{\circ}C$, 7.2 sec at $80^{\circ}C$. Z-values of soluble and immobilized TG were $6.4^{\circ}C\;and\;5.3^{\circ}C$, respectively. The little difference of activation energies of soluble TG and immobilized TG supposed that there was little difference in mass transfer limitation during the reaction of soluble TG and immobilized TG.
The enzyme invertase contributes to sugar unloading, pathogen defense, differentiation and development in plants. We cloned the complete cDNA of a soluble acid invertase from pea seedlings (Pisum sativum) via RT-PCR and the rapid amplification of the cDNA end (RACE) technique. The full-length cDNA of the soluble pea invertase comprised 2237 bp and contained a complete open reading frame encoding 647 amino acids. The deduced amino acid sequence showed high homology to soluble acid invertases from various plants. Northern blot analysis demonstrated the soluble acid invertase gene of P. sativum was strongly expressed in sink organs such as shoot tips and root tips, and induced by abscisic acid, gibberellic acid and jasmonic acid in shoots. Especially, gibberellic acid enhanced the gene expression of the soluble acid invertase in a time-dependent manner. This study presents that the gene expression patterns of a soluble acid invertase from pea are strongly consistent with the suggestion that individual invertase gene product has different functions in the growing plant.
In order to investigate the deposition chara.cteristics of water-soluble ionic components in dustfall in Kwangju city, dustfall samples were collected by deposit jar for one year from December 1992 to November 1993. The depositjon amount of dustfall and water-soluble ionic components ($SO_4^{2-}, NO_3^-, Cl^-, NH_4^+, Na^+, Ca^{2+}, Mg^{2+}, K^+$) were measured. The total deposition amount of dustfall was 10.0 ton/$km^2$/month and showed seasonal trend of Summer and Spring > Fall and Winter. The total deposition amounts of water-soluble components showed 2.41 ton/$km^2$/month and seasonal trend of Summer > Fall > Spring > Winter. Deposition amount of $SO_4^{2-}$ was 0.99 ton/$km^2$/month which makes up 41% of water-soluble components. The deposition amounts of dustfall and watersoluble components according to the sampling points were approximately similar to each other. From this result, it can be estimated that the deposition amounts of dustfall and water-soluble components in dustfall were more influenced by the seasonal variation than the regional emission characteristics of pollution source. The content of each ionic component to the deposition amount of water-soluble components showed in order of $SO_4^{2-} > Cl^- > NH_4^+ > Na^+ > Ca^{2+} = K^+ > NO_3^- > Mg^{2+}$ respectively.

This paper presents an experimental study to ivestigate mechanical property of concretes according to addition of urea and urea-water soluble sulfur contents. Urea was added at 5~20% replacement by weight of water, and water soluble sulfur was used at 2%, 4% replacement by weight of cement. The setting times, the hydration heat, the compressive strength, and the drying shrinkage, were measured on concretes with single and binary admixtures. From the test result, it was confirmed that the hydration heat of urea-water soluble sulfur was lower than that of normal concrete by $10.1^{\circ}C$, and the drying shrinkage of urea-water soluble sulfur concrete was more excellent than normal concrete. In the case of urea of 5%, Compressive strength were improved with an increase of water soluble sulfur contents. The urea-water soluble sulfur used in this research can be used as improvement materials for drying shrinkage and compressive strength.
This study was performed to investigate the air pollution levels in Daegu area titrough measuring of contents of water, chlorophyll, water soluble sulfur and lead metal in the leaves of roadside trees and chemical properties of soil under the urban trees. The results can be summarized as follows 1. The range of water content was from 60.4% to 74.6%. The comparisons of chlorophyll, water soluble sulfur and water content were regression equation, chlorophyll=0.1981+ 0.0040 water content (July), water soluble sulfur=3,139-0.0416 water content (July). and correlation coefficient, r=0.561 and r=0.549 respectively 2. Average contents of chlorophyll, water soluble sulfur and lead metal in the leaves of roadside tree at sampling sites were chlorophyll 0.050, 0.072mg/cm$^2$, water soluble sulfur 0.244, 0.333%, and lead metal 12.25, 12.68ppm in Oct. and Jul. respectively. 3. Correlation between chlorophyll and contents of water soluble sulfur and lead metal in the leaves of roadside tree at sampling sites were water soluble sulfur r=-0.564, -0.613 and lead metal r=-0.693, -0.699 in October and July, respectively. 4. Correlation between chlorophyll and water content, water soluble sulfur and lead metal in the leaves of tree showed positive significance.
Ceramics from Taean shipwreck were studied to investigate the deterioration by soluble salts and desalination pattern according to each ceramic characterization. NaCl crystals were found as a crystal growth on the ceramic microstructure and deteriorated to push off the glaze layer of some Puncheong ware sample. As a result of monitoring the desalination pattern of ceramics for 74days, the earthenware sample with big pores and no glaze removed soluble salts so fast, but Puncheong ware and Whiteware samples removed lots of soluble salts slowly and continuously. Generally, it takes long time to remove soluble salts from porcelains, due to small pores and glaze, but the long-term desalination probably can cause the damage on the surface layer such like glaze. As some soluble salts still remains in the ceramic microstructures after desalination, it is very important to control and keep the relative humidity stable through the proper storage. There are various ceramic deterioration patterns by soluble salts. Therefore, it needs to select the desalination method of ceramics from shipwreck, considering the material characteristics and the production techniques.
The objectives of this study were to examine the ability to predict soluble solid and firmness in intact apples based on the visible/near-infrared spectroscopic technique. Two cultivars of apples, Delicious and Gala, were handled, tested and analyzed separately. Reflectance spectra, Magness-Tayor (MT) firmness, and soluble solids in apples were measured sequentially. Maximum and minimum diameters, height, and weight of apples were recorded before the MT firmness tests. A spectrophotometer was used to collect reflectance spectra of intact apples over a wavelength range of 400 to 2, 498 nm. The W firmness tests were conducted using a standard 11.1mm (7/16 in.) MT probe mounted in an Instron universal testing machine. A digital refractormeter was used to measure soluble solid contents in the apples. Apple samples were divided into a calibration set and a prediction set. The calibration set was used during model development, and the prediction set was used to predict soluble solids and firmness from unknown spectra. The method of partial least square (PLS) analysis was used. An unique set of PLS loading vectors (factors) was developed for soluble solid content and firmness. The PLS model showed good correlations between predicted and measured soluble solids of intact apples in 860~1078 nm of the wavelengths. However, the PLS analysis was not good enough to predict the apple firmness.
