Isolation and Characterization of Biopolymer Producing Alkali-Tolerant Bacterial Strain (생물고분자 생산 알칼리 내성 균주의 분리 및 특성)

  • For the production of useful products from microorganism, a bacterial strain producing the biopolymer was isolated from soil. The bacteriological characteristics of the strain were examined and some chemical properties of the biopolymer produced were investigated. The bacterial strain was identified as an alkali-tolerant Bacillus sp. The results of chemical composition, various color reactions and I.R. spectrum revealed that the biopolymer contained high protein content, low amino sugar and no uronic acid. However, the biopolymer was precipitated by treating with cetylpiridinium chloride and was found to be acidic.

  • Conventional RC/TS(resonant column and torsional shear) device usesa specimen with an aspect ratio(height-to-diameter) of 2:1 and this generates a maximum shear strain in the sample of about 1.5% at the maximum rotation of the drives system. The objective of this study is to modify RC/TS device to generate higher strain amplitude. The modifications include a new base pedestal to overcome the limitations in the travel of the drive system and modification of coil wiring to increase torque. The effects of the new coil wire on torque in the electro magnetic drive system were evaluated and the application of modified device was illustrated using sand soil.

Optimal Conditions for Antimicrobial Metabolites Production from a New Streptomyces sp. RUPA-08PR Isolated from Bangladeshi Soil

  • An actinomycete strain was isolated from northern part of Bangladesh and identified as a new Streptomyces species on the basis of its morphological, biochemical, cultural characteristics and 16S rRNA data. Attempts were made to optimize the culture conditions for the production of antimicrobial metabolites by this strain. Antimicrobial metabolites production was started after 7 days of incubation of culture broth and reached its maximum levels after 10 days and thereafter gradually decreased. The maximum production of antimicrobial metabolites was obtained when the culture medium pH was adjusted to 8. The optimum temperature for antimicrobial metabolites production was $39^{\circ}C$, indicated the new strain as mesophilic organism. Basel medium supplemented with glucose and yeast extract as carbon and nitrogen sources, respectively, was proved to be the best for the production of bioactive metabolites. Maximum production of bioactive metabolites was when NaCl concentration was 1% and among different minerals tested, $K_2HPO_4$ and NaCl showed positive influence on antibiotic production by the strain.

Production, Purification and Antifungal Activity of Antibiotic Substances Produced by Pseudomonas aeruginosa Strain B5

  • Pseudomonas aeruginosa strain B5 with antagonistic activity against Phytophthora capsici and Magnaporthe grisea, was isolated from pepper-growing soil. From the culture of P. aeruginosa strain B5 grown on King's medium B, antibiotic substances were purified using XAD-2 column chromatography. XAD-2 eluates inhibited not only the mycelial growth of P. capsid and M. grisea, but also the development of Phytophthora blight on pepper plants. The crude antibiotic substances were further purified by using silica gel column chromatography, Sephadex LH-20 column chromatography, thin layer chromatography on silica gel plates, and high performance liquid chromatography. Silica gel column chromatogrphy gave good separation of the four antibiotic substances. The pure antibiotics P1, P2, and P3 finally purified by preparative HPLC inhibited the mycelial growth of P. capsici, at concentrations from 7 to 10 $\mu g/ml$. Only P1 and P2 had antifungal activity against M. grisea at 8 $\mu g/ml$. P1 and P3 were highly inhibitory to the mycelial growth of Botryosphaeria dothidea and Botrytis cinerea at relatively low concentrations. However, the three antibiotics had no antifungal activity against Rhizoctonia solani. The chemical structures of these antibiotics are being identified.

Evaluation of Dynamic Properties of Trackbed Foundation Soil Using Mid-size Resonant Column Test

  • A mid-size RC test apparatus (MRCA) equipped with a program is developed that can test samples up to D=10 cm diameter and H=20 cm height which are larger than usual samples used in practice. Using the developed RC test apparatus, two types of crushed trackbed foundation materials were tested in order to get the shear modulus reduction curves of the materials with changing of shear strain levels. For comparison purpose, large repetitive triaxial compression tests (LRT) with samples of height H=60cm and diameter D=30 cm were performed also. Resilient modulus obtained from the LRT was converted to shear modulus by considering elastic theory and strain level conversion and were compared to shear modulus values from the MRCA. It is found from this study that the MRCA can be used to test the trackbed foundation materials properly. It is found also that strain levels of $E_{v2}$ mostly used in the field should be verified considering the shear modulus reduction curves and proper values of $E_{v2}$ of trackbed foundation must be used considering the strain level verified.

  • A bacterial strain BS-214 producing extracellular pectinase was isolated from soil. The isolated bacterium was identified as a strain of Bacillus so. based on the morphological, biochemical, and physiological characteristics. Cell growth and pectinase activity of Bacillus sp. BS-214 were reached to a mixium in the culture condition of pH 8.5 at 4$0^{\circ}C$. Production of pectinase by the strain was the highest when polygalacturonic acid was added to culture medium as a carbon source, and its optimal concentration was 1%. Also, yeast extract was used as the best nitrogen source for the production of pectinase by the concentration of 0.25%. Decomposition of a constituent of Edzeworthia papyrifera by the strain was observed by scanning electron microscope.

  • The taxonomical properties of strain J-275L isolated from soil as a microorganism which produces extracellular adenine deaminase and cultural conditions for the enxyme production were studied. The hyphae of strain J-275L is fragmented into rod-or coccus-like elements. The elements of fragmented aerkal hyphae has smooth surfaces. The cell wall of the organism contains LL-diaminopimelic acid. Mycolic acid are not produced. As a result of taxonomical studies, strain J-275L is designated as Nocardioides sp. J-275L. The optimum medium for the enzyme production from Nocardioides sp.J-275L wascomposed of 0.5% peptone, 0.5% dextrin, 1% yeast extract, and 0.2% $K_{2}HPO_{4}$. The optimum initial pH of the medium was pH 7.5.

  • The appropriate description of the stress-anisotropy and time-dependent behavior of cohesive soils is very important in representing the real soil behavior. In this study, two constitutive relations have been incorporated based on the generalized viscous theory: one is the plastic constitutive relation adopted to capture the stress-anisotropy with a few model parameters; the other is the rate-dependent constitutive relation adopted to describe the strain rate-dependent behavior, an important time-dependent behavior in cohesive soils. The incorporated and proposed constitutive model has relatively a few model parameters and their values need not to be re-evaluated at different strain rates. The proposed model has been verified and investigated with the anisotropic triaxial test results obtained by using the artificial homogeneous specimens.

A Peptide Antibiotic AMRSA1 Active against Multidrug-resistant Staphylococcus aureus Produced by Streptomyces sp. HW-003

  • The antibiotic-producing strain HW-003 was screened from soil and found to be effective against the multidrug-resistant Staphylococcus aureus. The spore chain of HW-003 was retinaculiaperti, and the spore surface was spiny. Strain HW-003 has a LL-diaminopimelic acid isoform in the cell wall. The aerial mass color of the strain was gray, and the reverse side was yellow-brown. The strain produced melanin, but did not produce soluble pigments. According to the Taxon program, HW-003 showed best match with Streptomyces cyaneus. Antibiotic production reached a maximum after 72-h cultivation. The antibiotic was purified with silica gel column chromatography, octadecylsilyl column chromatography, and HPLC. The purified antibiotic, AMRSA1, showed strong inhibitory activity against multidrug-resistant Staphylococcus aureus and gram-positive bacteria. The molecular weight of AMRSA1 was about 1, 100. AMRSA1 was a peptide antibiotic containing alanine and serine.

  • A strain producing an extracellular cytosine deaminase was isolated from soil samples. The enzyme obtained from the strain possessed the substrate specificity to both cytosine and 5-fluorocytosine. From the results of its morphological, cultural, physiological, and biochemical properties, the strain was thought to be the genus Arthrobacter. Therefore, it was named as Arthrobacter sp. JH-13. The composition of optimum medium for the enzyme formation was 0.5% of peptone, 0.5% of meat extract, 0.5% of soluble starch, and 0.1% of KCl. The optimum pH for the enzyme formation was 8.0. When the microoganism was cultured aerobically in the above medium, enzyme production reached at maximum in 54 hours at $30^{\circ}C$.
