  • Bacillus stearothermophilus No.239 isolated from soil was mutated with N-methyl-N'nitro-N-nitrosoguanidine (MNNG) to yield a series of mutants with increasing levels of cyclomalto-dextrin glucanotransferase (EC 2.4.1.19` CGTase) production. After five consecutive mautation steps, a mutant MNNG 8 with about 14 times of CGTase activity than the parent strain was obtained.

  • Bacterium strain, K-173-10, which was isolated from waste soil of Korean brewing factories, could grow on acetate as the sole carbone source and accumulate a considerable amount of L-glutamic acid (24g/l) in the liguid culture medium. This strain was named by Brevibacterium ammoniagenes sp. by the standard method of taxonomy procedures given in the Manual of Microbiogical Methods.

  • The aim of the present study was to develop strains of actinomycetes producing low molecular weight cathepsin B inhibitor. Among 700 isolates from soil samples, a strain of Streptomyces sp. SMF30 producing cathepsin B inhibitor showing specificity and heat stability was selected by an economical and effective screening method. 50 units characteristics for major cluster analysis and 34 units characteristics for minor cluster were tested and the data were analyzed numerically using the TAXON program. The Isolate SMF30 was identified as a strain of Streptomyces aburabiensis

  • Underground flexible pipes for electric cables are subject to external loads and surrounding soil pressure. Particularly, strain of flexible pipes is of great concern in terms of safety and maintenance for electric cables. In this study, the actual size tests were carried out to investigate strain-deformation relation for underground flexible pipes subject to vehicle loads with various depths and degree of compaction.

  • Burkholderia cepacia strain G4 (pHG-2) containing toluene 2-monooxygenase and toluene dioxygenase, was able to grow on toluene and accumulate cis-3-methyl-3,5-cyclohexadien-1,2-diol (cis-toluene dihydrodiol) in the liquid culture. The cis-toluene dihydrodiol produced was identical to the authentic compound, as judged through mass spectrometry and nuclear magnetic resonance analysis. Our results indicate that pHG-2 provides an economical means to produce chemically-important chiral synthons while growing on toluene.

  • This research presents the whole-genome sequence of Enterobacter asburiae strain IK3, which was isolated from the rhizosphere soil of soybean (Glycine max). The genome of the strain is composed of a single chromosome with 4 plasmids, total size of 5,084,040 bp, and the GC content is 55.5%.

  • This study presents the complete genome sequence of Cytobacillus firmus strain T8, which was obtained from the rhizosphere soil of pepper (Capsicum annuum L.). The genome of the strain consists of a single chromosome with a total size of 4,383,751 bp and the GC content of 42%.

  • The stress transfer mechanism between soil and grid reinforcements involves two basic mechanism : frictional soil resistance and passive soil resistance. However the mechanism of the passive soil resistance is very complex to understand. To study the failure mechanism of ribbed reinforcement, the direct shear tests which are dominated by passive soil resistance are analyzed by using the finite element method. The finite element method is used to examine the effects of ribs on this passive soil resistance development and the met hanism of failure. The calculated behavior of the ribbed reinforcement is compared with the measured behavi or. Comparisons between the measured and the simulated strain pat terns, failure modes and load displacement relationship are presented. The behavior of the ribbed reinforcements in a cohesive soil is predicted on the basis of a good agreement between the measured and the Predicted behavior of the Ottawa sand.

Monitoring Expression of bphC Gene from Ralstonia eutropha H85O Induced by Plant Terpenes in Soil

  • A PCB degrader, Ralstonia eutropha H850 was shown to induce bphC gene encoding 2,3-dihydroxy-biphenyl-1,2-dioxygenase in a carvone-amended pure culture in our previous study (Park et al.,1999). The present study was carried out to examine how plant terpenes, as natural substrates, would cause an expression of a PCB degradative gene in soil that was amended with terpenes. The population of Ralstonia eutropha H850 was maintained at least around 10$\^$8/ (CFU/g fresh soil) in the soil amended with carvone or limonene in the presence of succinate as a growth substrate at 50 th day. The gene expression was monitored by RT-PCR using total RNA directly extracted from each soil and bphC gene primers. The bphC gene expression of the seeded strain H850 was observed in the soil amended with biphenyl (4 days) but not with succinate, carvone and limonene. These results indicate that terpenes widely distributed in nature could be a potential inducing substrate for effective PCB biodegration in the soil but their bioavailability and specific induction behavior should be taken into account before PCB bioremediation implementation.
