Production of $\beta$-Mannanase and $\beta$-Mannosidase from Sporolactobacillus sp. M201. (Sporolactobacillus sp. M201 균주에 의한 $\beta$-Mannanase와 $\beta$-Mannosidase의 생산)

  • A bacterial strain producing high levels of an extracellular ${eta}$-mannanase and intracellular ${eta}$-mannosidase and ${alpha}$-galactosidase was isolated from soil. The strain isolated was identified as a strain of Sporolactobacillus sp. and designated as Sporolactobacillus sp. M20l. Synthesis of ${eta}$-mannanase by Sporolactobacillus sp. M20l was induced by sucrose, maltose, or locust bean gum. The highest induction rate was obtained with 2% locust bean gum added to the culture medium as a sole carbon source. On the other hand, induction of ${eta}$-mannosidase was observed only with locust bean gum. The optimal media for the enzyme production were established as follows: for ${eta}$-mannanase; 2% locust bean gum, 0.5% peptone, 0.2% KH$_2$PO$_4$, 80 mg/l MgSO$_4$, and 8 mg/l ZnSO$_4$ (pH 6.0), and for ${eta}$-mannosidase; 2% locust bean gum, 0.5% yeast extract, 0.2% KH$_2$PO$_4$, 80 mg/l MgSO$_4$, and 8 mg/l ZnSO$_4$ (pH 5.0). The optimal culture temperatures for production of ${eta}$-mannanase and ${eta}$-mannosidase were found to be 37$^{\circ}C$ and 3$0^{\circ}C$, respectively. Under the optimal culture conditions, the production of ${eta}$-mannanase and ${eta}$-mannosidase reached the highest levels of 10.6 units/ml and 1.35 units/ml after 30 h and 24 h cultivation, respectively.

Phosphate Uptake by Acinetobacter lwoffi PO8 and Accumulation (Acinetobacter lwoffi PO8에 의한 인산흡수 및 축적)

  • To remove phosphate accumulated in the soil and water, Acinetobacter lwoffi PO8 possessing a high ability to accumulate phosphate was isolated from a active sludge. Bacterium was cultured in the liquid medium containing $150\;{\mu}g/mL$ of phosphate at $30^{\circ}C$ in different culture conditions to examine intracellular phosphate uptake. The initial pH in the range of $7.5{\sim}8.5$ was effective on the growth and phosphate uptake of the strain. Glycerol and arabinose used as a carbon sources showed 93 and 91% the phsphate uptake, respectively. Among the nitrogen sources, ammonium salt such as $NH_4NO_3$ and $(NH_4)_2SO_4$ was effectively utilized on the phosphate uptake compared with amino compounds. The rate of phosphate uptake of $NH_4NO_3$, and $(NH_4)_2SO_4$, was 95 and 96%, respectively The growth and Phosphate uptake ability in the strain were significantly promoted when metal ions were added in the medium; $Co^{2+}$, however, was not utilized by the strain. The capacity of phosphate uptake was enhanced to $10{\sim}20%$ when arginine, methionine, or lysine was added. Using $^{32}P$ to examine the uptake Pattern of intracellular phosphate, experiment result showed that polyphosphate was largely found in the fraction of intracellular inorganic phosphate of Acinetobacter lwoffi PO8.

Constant Rate of Strain Consolidation Test with Rowe Cell on the Clay with Sand Seam (샌드심이 존재하는 점토에 Rowe Cell를 이용한 일정변형률 압밀시험)

  • The sand layer deposited in clay is called a sand seam, which is formed by inflow of sands due to river flooding or slope failure in the middle of sinking and sediment of clay. When the sand seam exists in clay layer, the drainage direction changes from one way to both ways, and the time of consolidation may be reduced. However, it is not clearly proved due to lack of studies of sand seam and currently is not reflected in the design of soft soil improvement. As a fundamental study about sand seam, the oedometer tests and constant rate of strain tests with Rowe cell were conducted on clay specimens with sand seam. For tests, a frozen method was specially designed for making the sand seam. It was concluded that the test results showed the sand seam affects the coefficient of consolidation of clay. If the thickness of sand seam exceeds 0.05 times of specimen height, the sand seam works as drainage layer of pore water horizontally as well as vertically, and consequently the consolidation is accelerated.

Mass Cultivation of Rhodococcus sp. 3-2, a Carbendazim-Degrading Microorganism, and Development of Microbial Agents (카벤다짐 분해 미생물인 Rhodococcus sp. 3-2의 대량 배양 및 미생물 제제 개발)

  • Rhodococcus sp. 3-2 strain has been reported to degrade benzimidazole-based pesticides, such as benomyl and carbendazim. Therefore, this study aimed to optimize culture medium composition and culture conditions to achieve cost-effective and efficient large-scale production of the Rhodococcus sp. 3-2 strain. The study identified that the optimal media composition for mass culture comprised 0.5% glucose, 0.5% yeast extract, 0.15% NaCl, 0.5% K2HPO4, 0.5% sodium succinate, and 0.1% MgSO4. Additionally, a microbial agent was developed using a 1.5-ton fermenter, with skim milk (20%), monosodium glutamate (15%), and vitamin C (2%) as key components. The storage stability of the microbial agent has been confirmed, with advantages of low temperature conservation, which helps to sustain efficacy for at least six months. We also assessed the benomyl degradation activity of the microbial agent within field soil. The results revealed an over 90% degradation rate when the concentration of viable cells exceeded 2.65 × 106 CFU/g after a minimum of five weeks had elapsed. Based on these findings, Rhodococcus sp. 3-2 strain can be considered a cost-effective microbial agent with diverse agricultural applications.

Isolation of Synthetic Detergent Decomposing Microorganisms in Wastewater and Synthetic Detergent Decomposition Characterization of the Microorganisms (폐하수중 합성세제분해균의 분리(分離) 및 합성세제 (ABS) 분해특성(分解特性))

  • A bacterium which degrades efficiently synthetic detergents was isolated from the polluted waters, activated sludge of wastewater treatment plants or polluted soil. This bacterium showed considerably higher growth rate in the agar plate containing $2,000{\mu}g/ml$ of synthetic detergents than any other isolated strains, was identified as a Pseudomonas fluorescens or strains similar to it. The strain was named as a Pseudomonas fluorescens S1. Optimum pH and temperature for the growth of the Pseudomonas fluorescens S1 were pH 7.0 and $30^{\circ}C$, respectively. The strain was resistant to streptomycin and gentamycin, but sensitive to kanamycin. The strain was greatly resistant to zinc chloride, lead nitrate and copper sulfate, but unable to grow in the presence of relatively low concentrations of mercury chloride and silver nitrate. This strain utilized benzene, catechol, cyclohexane and xylene as a sole carbon source. The strain was well grown in the medium containing ABS 10,000${\mu}g$/ml. Degradation of ABS was 55% and 60% at 20${\mu}g$/ml and 100${\mu}g$/ml of ABS, respectively. Benzene ring was degraded 45% in 100${\mu}g$/ml of ABS. During the incubation of the strain in the medium containing ABS 100${\mu}g$/ml and COD 10,000${\mu}g$/ml for 4 days, degradation of ABS and COD were reduced to 40${\mu}g$/ml and 3,200${\mu}g$/ml, respectively. Total amino acid content of the Pseudomonas fluorescens S1 grown with 1,000${\mu}g$/ml of ABS was 115mg/g cell, whereas its content was decreased in the bacterium grown without synthetic detergent by 9.4%.

  • Cyclic simple shear tests were performed to find out the effect of preshear on dynamic strength of the sandy soil. Tests were performed for the specimens with 40% and 60% of relative density, under three different effective vertical stress of 50, 100 and 200kPa. For 50 and 100kPa, preshear ratios 0.00, 0.08, 0.12 and 0.16 were given, respectively, For low and high relative densities, two different results are shown in dynamic tests. Under the dense conditions, the maximum shear stress ratio($\tau$$\_$cyc//$\sigma$$\_$vo/) and the cyclic shear stress ratio($\tau$$\_$cyc//$\sigma$$\_$vo/) causing a certain shear strain increase with augmenting preshear ratio(${\alpha}$). However, the maximum shear stress ratio and the cyclic shear stress ratio increase or decrease with increasing preshear ratio under the loose conditions. Correction factor(K$\_$${\alpha}$/) for preshear increases at an early stage and then decreases with increasing preshear ratio at loose condition and increase with increasing preshear ratio at dense condition. Correction factor (K$\_$${\alpha}$,Max/) for preshear increases with the increasing preshear ratio irrespective of relative density, and the value of has same behavior as K$\_$${\alpha}$/.

  • Reactive oxygen species (ROS) are very harmful to living organisms due to the potential oxidation of membrane lipids, DNA, proteins, and carbohydrates. transformed E.coli strain QC 871, superoxide dismutase (SOD) double-mutant, with three sequence variant MnSOD1, MnSOD2, and MnSOD3 manganese superoxide dismutase (MnSOD) gene isolated from wheat. Although all QC 871 transformants grown at $37^{\circ}C$ expressed mRNA of MnSOD variants, only MnSOD2 transformant had functional SOD activity. MnSOD3 expressed active protein when grown at $22^{\circ}C$, however, MnSOD1 did not express functional protein at any growing and induction conditions. The sequence comparison of the wheat MnSOD variants revealed that the only amino acid difference between the sequence MnSOD2 and sequences MnSOD1 and 3 is phenylalanine/serine at position 58 amino acid. We made MnSOD2S58F gene, which was made by altering the phenylalaine to serine at position 58 in MnSOD2. The expressed MnSOD2S58F protein had functional SOD activity, even at higher levels than the original MnSOD2 at all observed temperatures. These data suggest that amino acid variation can result in highly active forms of MnSOD and the MnSOD2S58F gene can be an ideal target used for transforming crops to increase tolerance to environmental stresses.

Behaviour of geocell reinforced soft clay bed subjected to incremental cyclic loading

  • The paper deals with the results of the laboratory cyclic plate load tests performed on the reinforced soft clay beds. The performances of the clay bed reinforced with geocells and geocells with additional basal geogrid cases are compared with the performance of the unreinforced clay beds. From the cyclic plate load test results, the coefficient of elastic uniform compression ($C_u$) was calculated for the different cases. The $C_u$ value was found to increase in the presence of geocell reinforcement. The maximum increase in the $C_u$ value was observed in the case of the clay bed reinforced with the combination of geocell and geogrid. In addition, 3 times increase in the strain modulus, 10 times increase in the bearing capacity, 8 times increase in the stiffness and 90% reduction in the settlement was observed in the presence of the geocell and geogrid. Based on the laboratory test results, a hypothetical case of a prototype foundation subjected to cyclic load was analyzed. The results revealed that the natural frequency of the foundation-soil system increases by 4 times and the amplitude of the vibration reduces by 92% in the presence of the geocells and the geogrids.

  • In this paper, the comparisons between field measurments and numerical results ware performed for the settlements, lateral displacement in Jinwol interchange works on the Honam express way whose site was improved by sand drain for the constructions of over bridges, piers and abutments. The computer program was developed by coupling Biot's equation with Sekiguchi's elasto-viscoplastic model under plane strain conditions. Steel pipe piles for piers were replaced into the equivalent steel sheet pile wall. The characteristics of behavior for both the soil foundations and the sheet piles wall were investigated with the variation of axial force on the wall, rigidity of the wall, supported condition of sheet pile into hard strata and the location of anchored point.

Degradation of Phytate Pentamagnesium Salt by Bacillus sp. T4 Phytase as a Potential Eco-friendly Feed Additive

  • A bacterial isolate derived from soil samples near a cattle farm was found to display extracellular phytase activity. Based on 16S rRNA sequence analysis, the strain was named Bacillus sp. T4. The optimum temperature for the phytase activity toward magnesium phytate (Mg-$InsP_6$) was $40^{\circ}C$ without 5 mM $Ca^{2+}$ and $50^{\circ}C$ with 5 mM $Ca^{2+}$. T4 phytase had a characteristic bi-hump two pH optima of 6.0 to 6.5 and 7.4 for Mg-$InsP_6$. The enzyme showed higher specificity for Mg-$InsP_6$ than sodium phytate (Na-$InsP_6$). Its activity was fairly inhibited by EDTA, $Cu^{2+}$, $Mn^{2+}$, $Co^{2+}$, $Ba^{2+}$ and $Zn^{2+}$. T4 phytase may have great potential for use as an eco-friendly feed additive to enhance the nutritive quality of phytate and reduce phosphorus pollution.
