Plasmid-Mediated Aniline Assimilation by Pseudomonas sp. B10

  • An aniline-utilizing microorganism identified as a species of Pseudomonas was isolated from soil contaminated highly with aniline and urea-herbicide. This strain was able to utilize aniline as the sole source of carbon and energy, and was shown to harbor a single large plasmid mediating the aniline assimilation. Subsequent plasmid-curing of this bacterium resulted in the abolishment of the aniline utilizing phenotype and the loss of catechol-C2,3O-oxygenase. The reestablishment of the plasmid, denoted pB10, in cured Pseudomonas sp. via filter surface mating, resulted in restoration of the aniline assimilation abilities and enzyme activity.

  • For a studyon the production of ${\gamma}$-linolenic acid(GLA) by fungi 3 fungal strains were isolated from soil. Their cell growth lipid content and fatty acid composition were compared in shake flask culture. Among these fungi the fungus designated as FA-007 has high lipid content(21.1%) and GLA content(15.6% of total fatty acids) The fungal strain FA-007 was tentatively identified as Mucor sp. on the basis of morphological characteristics, Fungal oil produced by this fungus was composed of 75.2% neutral lipid 5.3% glycolipid and 19.5% phospholipid. Although the GLA content in phospholipid was higher than it in neutral lipid the GLA content in neurtal lipid was high as 15.5%.

  • The aim of the present study was to produce low molecular weight cathepsin B inhibitor. A strain of Streptomyces aburabiensis isolated from soil in Korea was selected and the optimum condition for the production of the inhibitor was evaluated. Glucose and soytone were selected as best carbon and nitrogen sources, respectively. From the kinetic analysis in batch fermentation, it was found that the specific cathepsin B inhibitor production rate (q$_{p}$) was linearly related to specific growh rate ($\mu$). The inhibitor in culture filtrate was purified by adsorption on activated charcoal, butanot extraction, silica gel chromatography, ion exchange chromatography using Dowex-1 (Cl form) and Amberlite IRC-50 (H$^{+}$ form), and preparative TLC. From the UV, IR, Mass spectroscopy and $^{1}$H-NMR, the inhibitor was thought to be a new inhibitor of which molecular weight was 199.

  • For soils with high void ratios, the inverse method of utilizing results obtained from centrifuge model test was used to find the constitutive relation of effective stress - void ratio - permeability whereas conventional oedometer test and constant rate of strain consolidation(CRS) test about settlement of interface and pore pressure and distribution with time were compared with numerically estimated values to confirm such a constitutive relation as obtained from the inverse method. As results of numerical method, the volumetric ratio and reclamation velocity were obtained for the reclamation condition.

  • Many electric poles in the softground have been collapsed due to external load. In this study, 10 types of tests were performed with variation of location, numbers and depths of anchor blocks as well as depth of poles to find stresses acting on concrete electric poles. The stresses of concrete poles are relaxed at 600~700[kg] of tensile load, and stresses are concentrated at top of pole, and spread to lower part of pole. In the concrete pole collapse test, tensile load at failure was approximately 1,400[kg], which is twice of design load. As passive zone in the soil increases, the stresses acting on concrete pole are concentrated at lower part of pole based on moment arm earth pressure distribution.

  • Three dimensional (3D) non-linear finite element analysis of offshore pipeline is investigated in this work with the help of general purpose software Abaqus. The general algorithm for the finite element approach is introduced. The 3D seabed mesh, limited to a corridor along the pipeline, is extracted from survey data via Fledermous software. Moreover soil bearing capacity and coefficient of frictions, obtained from the field survey report, and are introduced into the finite element model through the interaction module. For a case of study, a 32inch pipeline with API 5L X65 material grade subjected to high pressure and high temperature loading is investigated in more details.

  • Bacillus thuringiensis 2385-1, which showed toxicity to lepidopteran but not to dipteran was isolated from Korean soil sample and characterized. The H-serotype of 2385-1 was identical to that of serovar kenyae (H4a4c), and its crystal toxin was bipyramidal-shaped with a molecular weight of 130 kDa. However, the plasmid profile of 2385-1 was different from that of serovar kenyae. (omitted)

  • In order to obtain Ti plasmid which may be used in genetic engineering of higher plants, virulent Agrobacterium tumefaciens were isolated from soil with selective medium. For their identification, morphological and cultural characteristics were investigated in parallel with physiological and turnorigenesis test. 14 strains were isolated from various regions in Korea. 3 of them were identified as biotype 1 and the others were so heterogeneous that they are not yet conformed to any biotypes. However, Ti plasmid was isolated from A. tumefaciens KU 14 strain, one of the above mentioned 14 strains.

  • The alkalophilic bacterium, Xanthomonas sp. JK311, producing extracellular proteases, was isolated from soil. Xanthomonas sp. JK311 produced five extracellular proteases that are all metalloproteases. Four of them were resistant against 1% SDS. Chromosomal DNA of the Xanthomonas sp. JK311 was digested with BamHI and cloned into PUC19. Among E. coli strain HB101 transformants, a clone secreting the proteases was screened through halo formation on skim-milk agar plate and by Southern blot analysis. It had the recombinant plasmid pXEP-1 containing the 7.5 kb-BamHI DNA fragment and produced three extacellular proteases. Their protease properties corresponded to those of Xanthomonas sp. JK311.

  • In this study, slope stability analysis of LCRS(Leachate Collection Removel System) in waste landfill was peformed by large scale field test. Geocomat is new type of geocomposite product. Gecomat is a sort of Geocomposite product. It is composed of nonwoven geotextiles, woven geotextile, and geonet. Large scale field tests were performed on the slope of different two LCRSsections with static loading condition. One is LCRS section witch consist of GCL, HDPE and Geocomat, another is GCL, HDPE, and woven type geocomposite. The behavior of geosynthetics lined slope was monitored by incorporating instrumentation including vertical soil pressure meter, settlement plate, strain gauges, potential meter, displacement pin.. Based on the field monitoring, the Geocomat LCRS section is less sliding than the conventional geocomposite LCRS section.
