Studies on the Conditions of Glucose Oxidase Production by Aspergillus niger KUF-04 (Aspergillus niger KUF-04에 의한 Glucose Oxidase 생산조건에 관한 연구)

  • To maximize the production of glucose oxidase by Aspergillus niger KUF-04 isolated from a soil, the cultivation conditions and nutrient sources for the enzyme production were studied. The results obtained were as fellows: 1. The optimum temperature, pH of the medium, and cultivation time for the enzyme formation were found to be 28-34$^{\circ}C$, 7.0-8.0 and 40 hours, respectively. 2. The best carbon source was proved to be glucose and its most effective concentration was 15 percent. 3. Ammonium sulfate was the best nitrogen source as compared with the other inorganic and organic nitrogen sources tested. Its optimum concentration for the glucose oxidase production was 0.02 percent. 4. As mineral sources, 0.05% of Mag cesium sulfate 7-hydrate and 0.02% of Potassium phosphate, monobasic seemed to be necessary to further increase the level of the enzyme production.

  • The objectives of this study were to isolate and culture methanotrophs and to apply them for biological removal of nitrogen and phosphorous. Methanotrophs (dominant species: Methylomonas methanica) were isolated from a landfill cover soil, cultured in a NMS medium, and analyzed to reveal their characteristics of growth and nutrient removal. The methanotrophs themselves can produce substantial amount of organic substances(as COD) including methanol, formaldehyde, and formate, as carbon sources required for denitrification. For instance, the production rate for methanol was $8\;mg/L{\cdot}hr$. Moreover, the analysis of nitrogen and phosphorous in the sludge suggested that the methanotrophs assimilate nitrogen and phosphorous as growth substances.

  • A bacterial strain, producing extracellular inulin fructotransferase which converts inulin into di-D-fructofuranose 1,2':2,3' dianhydride(DFA III), was isolated from soil and presumed as Bacillus sp.. The highest production of the enzyme was obtained by using medium containing Jerusalem artichoke extract as carbon source, peptone as organic nitrogen source, and $NH_4H_2PO_4$, as inorganic source. Under optimum condition, the enzyme activity of the culture broth supernatant reached maximal 2.61 units/ml after cultivation for 45 hrs.

  • Retention ponds have been dug along some of the motorways in France to minimize environmental pollution by keeping pollutants from spreading over the surrounding area. In order to study heavy metal pollution and diagenetic behaviour of sediments, eight core samples were collected from the bottom of a retention pond located along the A-71 motorway in Sologne. The metal concentrations in interstitial waters and extractable metal concentrations in sediment layers using sequential chemical extraction method were determined. The depth distributions of Pb, Zn and Cd concentrations in interstitial water and particulate sediments were studied, and distribution coefficients (KD) were also determined to investigate the environmental mobility of these elements. In addition. the index of geoaccumulation and the Fe-normalized enrichment factor were calculated to differentiate the natural accumulation from the anthropogenic pollution. The vertical concentration profiles of heavy metals in core sediments indicate that surface enrichments (0~2 cm) of Pb, Zn, Cd and organic carbon were always observed at each core sample, due to the early diagenesis. However, the major factor contributing to the accumulation of Cd at the sediment surface is attributed to the dissolution of Cd from polluted roadside soil during periods of rainstorms and its subsequent redeposition on the sediment surface after being carried to the retention pond. A comparison of the KD values indiactes that a decrease in the KD values for Pb and Zn was observed with depth while KD values for Cd increase. According to the KD values. the relative mobility of studied metals was determined as following: Mn>Zn>Cd>Pb, for the upper layer, and Mn>Cd>Zn>Pb, for the lower layers.

EFFECTS OF UREA NITROGEN ON THE METABOLISM OF PLANTS (1) Studies on Nitrogen Absorption and Metabolism in Sunflower Leavessprayed with Urea Solution

  • In order to detect the way of absorption and metaboism of the urea it is sprayed on the surface of the leaves of sunflower. The sunflowers used in this study are grown in different conditions such that the one in nittogen aboundant and the other in nitrogen deficient soil, respectively. The urea-N, ammonia-N, amide-N, and 80% alcohol soluble-N in the leaves were quantitatively determined. All of the nitrogenous components measured are generally tended to increased with rising the concentration of urea except only amide-N at 24 hours after sprayed, and these were highly significances. It seemed that hydrolizing of urea into ammonia and carbon dixide and the assimilation of ammonia into other organic nitrogenous constituents were rapid in the young leaves than in the mature. It is interest that the amide content, in the young leaves and nitrogen defieient one were enhanced with the increasing concentration of urea, although in the mature leaves it did not show any change in the urea treatment. It is presumed that the assimilation rate of ammonia and the urease activity were lower in the matture leaves than in the young and nitrogen deficient leaves. No significance at 5% level showed all of the nitrogenous components except total nitrogen between nitrogen abundant and deficent leaves. Urea content was a high peak at first 12 hours, ammonia at 48 hours, and amide and alcohol soluble nitrogen at 96 hours, whence decrease4d the content of these constituents gradually. The total nitrogen content is not incrased obviously by only one time of urea spray in this study. When the concentration of urea was relatively high there appeared the wilting spots on t도 edge of leaves. As a whole, it seemed that sprayed urea was rapidly absorbed and taken part in nitrogen metabolism within relatively short period.

  • Since the radioactivity of U was discovered by Becquerel in 1896, a number of radiometric dating techniques based on radioactive decay have been developed with the help of precise and accurate instruments. Among them, radiocarbon dating was introduced as the most effective method in archeology in Korea. However, though the radiocarbon methods have contributed greatly to the development of archeology in Korea, it is limited to organics in a matrix of soil and sediment, including organic carbon, charcoal, bone and so on. If there are no organics in some paleolitic sites, other methods are required for dating. Therefore, we introduced OSL (Optically Stimulated Luminescence) dating method in this paper. The method is mainly based on quartz grains or other particles. The aim of this paper is to discuss about sampling method and the limitation of its application in archeology.

  • 최근 지구온난화로 인한 전 세계적 기후변화가 일어나고 있으며, 이러한 지구온난화 방지 대책으로 탄소의 중요성과 탄소중립을 선언하는 국가가 증가하고 있다. 탄소의 중요성이 증가함에 따라 유역 내의 탄소 중립이 중요 이슈로 떠오르고 있다. 유역 내 탄소 저장원으로는 숲, 하천, 토양 등이 존재하나 하천의 경우 탄소의 저장이 곧 수질 오염과 연결이 되기에 바람직한 방안이 될 수 없다. 그러나 토양의 경우 방대한 양의 탄소를 저장하기에 적합한 기능을 하기에 다른 저장원들에 비해 중요한 저장원으로 대두되고 있다. 토양탄소의 경우 일반적으로 유기물과 무기물의 형태로 토양에 저장된다. 이중 토양유기탄소는 장기간 토양 속에서 대기와의 탄소 조절 역할을 하기에 중요 요인으로 대두되고 있다. 하지만 기후변화로 인한 국지성 호우 및 무분별한 개발 등이 증가함에 따라 유역 내 토양 생태계의 변화가 일어나고 있으며, 이에 따른 유역 내 토양유기탄소 또한 손실이 일어나고 있다. 따라서 본 연구에서는 토양의 특성과 모델을 활용하여 유역단위 토양유기탄소량의 변화량을 산정하여 비교 및 분석을 하고자 한다. 이를 위해서 토양유기탄소의 모의가 가능한 APEX 모델을 활용하였으며, 선정된 연구 대상 지역의 토양 특성 자료를 활용하여 입력자료 전처리를 진행 후 모의를 진행하였다. 이후 선행연구 및 보고서를 통한 실측자료를 기반으로 모델 매개변수 보정을 진행하였으며, 보정된 결과를 통해 유역에 대한 토양유기탄소를 산정을 진행하였고 기간별 변화의 차이를 분석하였다. 해당 연구를 통해 유역 내 잠재되어있는 토양유기탄소량 정량화 등의 연구에 활용될 수 있을 것으로 기대한다.

Application of LCA on Lettuce Cropping System by Bottom-up Methodology in Protected Cultivation (시설상추 농가를 대상으로 하는 bottom-up 방식 LCA 방법론의 농업적 적용)

  • This study was conducted to apply LCA (Life cycle assessment) methodology to lettuce (Lactuca sativa L.) production systems in Namyang-ju as a case study. Five lettuce growing farms with three different farming systems (two farms with organic farming system, one farm with a system without agricultural chemicals and two farms with conventional farming system) were selected at Namyangju city of Gyeonggi-province in Korea. The input data for LCA were collected by interviewing with the farmers. The system boundary was set at a cropping season without heating and cooling system for reducing uncertainties in data collection and calculation. Sensitivity analysis was carried out to find out the effect of type and amount of fertilizer and energy use on GHG (Greenhouse Gas) emission. The results of establishing GTG (Gate-to-Gate) inventory revealed that the quantity of fertilizer and energy input had the largest value in producing 1 kg lettuce, the amount of pesticide input the smallest. The amount of electricity input was the largest in all farms except farm 1 which purchased seedlings from outside. The quantity of direct field emission of $CO_2$, $CH_4$ and $N_2O$ from farm 1 to farm 5 were 6.79E-03 (farm 1), 8.10E-03 (farm 2), 1.82E-02 (farm 3), 7.51E-02 (farm 4) and 1.61E-02 (farm 5) kg $kg^{-1}$ lettuce, respectively. According to the result of LCI analysis focused on GHG, it was observed that $CO_2$ emission was 2.92E-01 (farm 1), 3.76E-01 (farm 2), 4.11E-01 (farm 3), 9.40E-01 (farm 4) and $5.37E-01kg\;CO_2\;kg^{-1}\;lettuce$ (farm 5), respectively. Carbon dioxide contribute to the most GHG emission. Carbon dioxide was mainly emitted in the process of energy production, which occupied 67~91% of $CO_2$ emission from every production process from 5 farms. Due to higher proportion of $CO_2$ emission from production of compound fertilizer in conventional crop system, conventional crop system had lower proportion of $CO_2$ emission from energy production than organic crop system did. With increasing inorganic fertilizer input, the process of lettuce cultivation covered higher proportion in $N_2O$ emission. Therefore, farms 1 and 2 covered 87% of total $N_2O$ emission; and farm 3 covered 64%. The carbon footprints from farm 1 to farm 5 were 3.40E-01 (farm 1), 4.31E-01 (farm 2), 5.32E-01 (farm 3), 1.08E+00 (farm 4) and 6.14E-01 (farm 5) kg $CO_2$-eq. $kg^{-1}$ lettuce, respectively. Results of sensitivity analysis revealed the soybean meal was the most sensitive among 4 types of fertilizer. The value of compound fertilizer was the least sensitive among every fertilizer imput. Electricity showed the largest sensitivity on $CO_2$ emission. However, the value of $N_2O$ variation was almost zero.

Soil Classification of Paddy Soils by Soil Taxonomy (미국신분류법(美國新分類法)에 의(依)한 답토양의 분류(分類)에 관한 연구)

  • According to Soil Taxonomy which has been developed over the past 20 years in the soil conservation service of the U. S. D. A, Soils in Korea are classified. This system is well suited for the classification of the most of soils. But paddy field soils have some difficulties in classification because Soil Taxonomy states no proposals have yet been developed for classifying artificially irrigated soils. This paper discusses some problems in the application of Taxonomy and suggestes the classification of paddy field soils in Korea. Following is the summary of the paper. 1. Anthro aquic, Aquic Udipsamments : The top soils of these soils are saturated with irrigated water at some time of year and have mottles of low chroma(2 or less) more than 50cm of the soil surface. (Ex. Sadu, Geumcheon series) 2. Anthroaquic Udipsamments : These sails are like Anthroaquic, Aquic Udipsamments except for the mottles of low chroma within 50cm of the soil surface. (Ex. Baegsu series) 3. Halic Psammaquents : These soils contain enough salts as distributed in the profile that they interfere with the growth of most crop plants and located on the coastal dunes. The water table fluctuates with the tides. (Ex. Nagcheon series) 4. Anthroaquic, Aquic Udifluvents : They have some mottles that have chroma of 2 or less in more than 50cm of the surface. The upper horizon is saturated with irrigated water at sometime. (Ex. Maryeong series) 5. Anthro aquic Udifluvents : These soils are saturated with irrigated water at some time of year and have mottles of low chroma(2 or less) within 50cm of the surface soils. (Ex. Haenggog series) 6. Fluventic Haplaquepts : These soils have a content of organic carbon that decreases irregularly with depth and do not have an argillic horizon in any part of the pedon. Since ground water occur on the surface or near the surface, they are dominantly gray soils in a thick mineral regolith. (Ex Baeggu, Hagseong series) 7. Fluventic Thapto-Histic Haplaquepts : These soils have a buried organic matter layer and the upper boundary is within 1m of the surface. Other properties are same as Fluventic Haplaquepts. (Ex. Gongdeog, Seotan series) 8. Fluventic Aeric Haplaquepts : These soils have a horizon that has chroma too high for Fluventic Haplaquepts. The higher chroma is thought to indicate either a shorter period of saturation of the whole soils with water or some what deeper ground water than in the Fluventic Haplaquepts. The correlation of color with soil drainage classes is imperfect. (Ex. Mangyeong, Jeonbug series) 9. Fluventic Thapto-Histic Aeric Haplaquepts : These soils are similar to Fluventic Thapto Histic Haplaquepts except for the deeper ground water. (Ex. Bongnam series) 10. Fluventic Aeric Sulfic Haplaquepts : These soils are similar to Fluventic Aeric Haplaquepts except for the yellow mottles and low pH (<4.0) in some part between 50 and 150cm of the surface. (Ex. Deunggu series) 11. Fluventic Sulfaquepts : These soils are extremely acid and toxic to most plant. Their horizons are mostly dark gray and have yellow mottles of iron sulfate with in 50cm of the soil surface. They occur mainly in coastal marshes near the mouth of rivers. (Ex. Bongrim, Haecheog series) 12. Fluventic Aeric Sulfaquepts : They have a horizon that has chroma too high for Fluventic Sulfaquepts. Other properties are same as Fluventic Sulfaquepts. (Ex. Gimhae series) 13. Anthroaquic Fluvaquentic Eutrochrepts : These soils have mottles of low chroma in more than 50cm of the surface due to irrigated water. The base saturation is 60 percent or more in some subhroizon that is between depth of 25 and 75cm below the surface. (Ex. Jangyu, Chilgog series) 14. Anthroaquic Dystric Fluventic Eutrochrepts : These soils are similar to Anthroaquic Fluvaquentic Eutrochrepts except for the low chroma within 50cm of the surface. (Ex. Weolgog, Gyeongsan series) 15. Anthroaquic Fluventic Dystrochrepts : These soils have mottles that have chroma of 2 or less within 50cm of the soil surface due to artificial irrigation. They have lower base saturation (<60 percert) in all subhorizons between depths of 25 and 75cm below the soil surface. (Ex. Gocheon, Bigog series) 16. Anthro aquic Eutrandepts : These soils are similar to Anthroaquic Dystric Fluventic Eutrochrepts except for lower bulk density in the horizon. (Ex. Daejeong series) 17. Anthroaquic Hapludalfs : These soils' have a surface that is saturated with irrigated water at some time and have chroma of 2 or less in the matrix and higher chroma of mottles within 50cm of the surface. (Ex. Hwadong, Yongsu series) 18. Anthro aquic, Aquic Hapludalfs : These soils are similar to Anthro aquic Hapludalfs except for the matrix that has chroma 2 or less and higher chroma of mottles in more than 50cm of the surface. (Ex. Geugrag, Deogpyeong se ries)

  • A series of experiments was conducted to study the behavior of the phosphorus added to the soils having the high phorphorus fixing capacity derived from volcanic ash in Cheju Island. Soil samples were taken from different depths of 0-10, 10-30, and 30-50cm in six citrus orchards where heavy application of phosphate fertilizer has been practised.
