Effects of Sodium Polyacrylate and Phytase-Supplemented Diet on Performance and Phosphorus Retention in Chicks

  • Two experiments were conducted to evaluate the effects of addition of sodium polyacrylate (SPA) to a phytasesupplemented diet on the performance and phosphorus (P) retention of chicks. In experiment 1, chicks were randomly allocated to four dietary treatments which were fed from 7 to 21 days of age: i) basal diet (low nonphytate phosphorus (0.23% NPP)); ii) basal with 250 U/kg diet of phytase; iii) as (ii) with 2.5 g/kg diet of SPA; and iv) as (ii) with 5.0 g/kg diet of SPA. In experiment 2, three replicates, each with three chicks, were fed from 7 to 28 days of age the basal diet (0.23% NPP) with supplementation of phytase (0, 300, 600, 900 U/kg diet) and SPA (0, 2.5 g/kg diet) in a $4{\times}2$ factorial arrangement. In Experiment 1, feed efficiency was improved and excreted P was 10% less with phytase supplementation. However, the addition of SPA did not affect performance or P excretion. Dietary SPA supplementation to the diets showed significantly higher amounts of P retention, and highest values were observed in chicks fed 2.5 g/kg of the SPA-supplemented diet. In Experiment 2, feed efficiency was improved with phytase supplementation, and the addition of SPA showed significant improvement in feed efficiency. Excreted P was significantly lower in chicks fed SPA-supplemented diets, and the retained P coefficient improved with SPA supplementation. In conclusion, the increased transit time of digesta with suitable supplementation levels of SPA may allow phytase activity to be more effective in the degradation of phytate, and improve P retention.

식육에서 피틴산염의 Salmonella typhimurium균에 대한 항균효과 (Antibacterial Activity of Sodium Phytate Against Salmonella typhimurium in Meats)

  • 피틴산(phytic acid)은 inositol hexaphosphate로서 식물성식품 및 씨 중에 1-5%가량 존재하는 자연 항산화 물질로서 소화효소에 의해 쉽게 분해되지 않는다. 한편 인산염은 식육에서 품질개량제로 식육에서의 항균 효과에 대해서는 이미 여러 보고가 있으나 그 결과는 다양하다. 일반적으로 생육에서는 그 효능이 낮은데 이것은 생육에 존재하는 phosphatase의 작용에 의해 인산염들이 가수분해되기 때문이다. 한편 피틴산염은 열에 강하고 쉽게 분해되지 않으므로 생육이나 멸균 처리된 식육에서 공히 인산염들이 가지고 있는 항균효과를 기대할 수 있다. 본 실험에서는 pH가 다른 선택배지에서 Salmonella typhimurium에 대한 피틴산의 항균효과를 평가하였고, 더불어 생육에서 Salmonella typhimurium의 접종 후에 항균 효과를 평가하였다. 선택된 배지인 tryptic soy broth에서 Salmonella typhimurium에 대한 항균 효과는 0.1, 0.5 및 1%의 세 가지 농도에서 농도에 의존적으로 유의성 있게 나타났다. 생육인 닭고기, 돼지고기 및 소고기에 피틴산염을 각각 0.1, 0.5, 그리고 1%의 농도별로 첨가했을 때 Salmonella typhimurium의 증식이 모두 유의성 있게 감소하였다(p<0.01). 또한 피틴산염의 첨가는 모든 식육에서 pH를 0.5-0.6단위만큼 증가시켰다. 이러한 결과로부터 피틴산염은 배지 및 식육에서 항균효과가 뛰어나며 따라서 축산식품의 첨가제로서 사용시 식품의 기능성 향상과 더불어 매우 유용하게 사용될 수 있을 것이다.

Zinc Deficiency Elevates Fecal Protein, But Not Electrolyte and Short-Chain Fatty Acid, Levels in Enterotoxigenic Escherichia coli-Induced Diarrhea in Rats

  • Purpose: To determine the effect of zinc deficiency on fecal protein, electrolyte, and short-chain fatty acid levels in both heat-stable (ST) and heat-labile (LT) enterotoxigenic Escherichia coli (ETEC)-induced diarrhea in rats. Methods: Albino rats, weighing 100 to 150 g, were divided into 2 groups, with 15 animals each: non-zinc and zinc-deficient. These two groups were sub-divided into three sub-groups with five rats each: control (saline); LT-ETEC; and ST-ETEC. Sodium phytate (30 mmol/L) was added to the animals' water to induce zinc deficiency, while diarrhea was induced using 5×109 ETEC cells/mL. Fecal protein levels were estimated using the Bradford method, while sodium and potassium levels were determined using atomic absorption spectrophotometry. Short-chain fatty acids were measured using gas chromatography-mass spectrometry. Results: Among the non-zinc and zinc-deficient groups, there were significant increases (p=0.04), (p=0.03) in fecal protein concentrations (mg/mL) in the LT-ETEC- (4.50±0.33), (6.50±0.26) and ST-ETEC- (3.85±0.19), (5.98±0.32) induced groups compared to the control groups (2.60±0.52), (3.50±0.11) respectively. Fecal sodium and potassium levels (mg/L) were significantly (p=0.029) increased in non-zinc-deficient rats induced with LT-ETEC (9.35±0.95, 1.05±0.48), and ST-ETEC (9.96±1.02, 1.21±0.45) compared with the control group (8.07±0.44, 0.47±0.17) but the increase were not statistically significant (p=0.059) in the zinc deficient rat groups. Fecal acetate and propionate levels (mg/g) significantly (p=0.032) increased when induced with LT-ETEC and ST-ETEC in non-zinc and zinc-deficient groups compared with the control groups. Conclusion: Zinc deficiency among rats with ETEC-induced diarrhea elevated fecal protein loss but may not have an effect on fecal sodium, potassium and short-chain fatty acid levels.

Bacillus coagulans IDCC 1201이 생산하는 Phytase의 특성 (Characterization of Phytase from Bacillus coagulans IDCC 1201)

  • 현재 probiotics로 상업화되어 있는 B. coagulans IDCC 1201(상업용 명칭 : Lactobacillus sporogenes)이 acid phytase 및 $Co^{2+}$를 cofactor로 갖는 metalloenzyme 특성을 가진 phytase를 생산하고, cofactor로 사용되는 $Co^{2+}$ ion이 B. coagulans IDCC 1201 유래 phytase의 열 안정성에 기여하였다. 또한 B. coagulans IDCC 1201의 phytase 유전자 서열을 분석한 결과 B. subtilis 168 유래의 enzyme 서열과 높은 상동성을 나타내었다. 본 연구결과를 토대로 B. coagulans IDCC 1201 장내 균총의 정상화를 가져와, 질병 예방과 면역력 향상을 구현함과 동시에 가축이 곡물에 함유된 phytic acid의 섭취로 인한 항영양인자, 환경오염등의 문제점을 해결할 수 있는 사료 첨가제로써의 산업적 효용가치가 풍부할 것으로 전망된다.

Graded levels of phytase on performance, bone mineralization and carcass traits of broiler fed reduced dicalcium phosphate

  • Objective: This study was conducted to assess increasing doses of phytase added to broiler diets formulated with different levels of available phosphorus (avP), calcium (Ca), and sodium (Na), and the respective effects on performance parameters, quantitative carcass characteristics, ash and phosphorus deposition in tibia and weight of organs. Methods: Three different matrices were assumed for phytase with the following nutritional values: matrix A (MT A): 0.165% Ca, 0.150% avP, and 0.035% Na; matrix B (MT B): 0.215% Ca, 0.195% avP, and 0.045% Na; matrix C (MT C): 0.245% Ca, 0.225% avP, and 0.053% Na. There were six different diets: No phytase (formulated to meet the nutritional requirements); phytase 500 FTU/kg+MT A; phytase 1,000 FTU/kg+MT A; phytase 1,500 FTU/kg+MT A; phytase 1,000 FTU/kg+MT B and phytase 1,500 FTU/kg+MT C. Results: There was no significant phytase influence on performance, quantitative carcass characteristics, ash and phosphorus deposition in tibia and weight of the organ throughout the study period, however, it was possible to observe a tendency of improvement in body weight corrected feed conversion for broilers fed the phytase 1,500+MT C diet, where potentially these birds were more efficient on utilize phytic phosphorus and other nutrients bounded to phytate molecule, translating into improvement in performance, and there was also a non significant numerical improvement in body weight corrected feed conversion of broilers fed this diet. Conclusion: Broilers fed with diets formulated with different levels of avP, Ca, and Na and increasing doses of phytase have shown no change on performance, quantitative carcass characteristics, ash and phosphorus deposition in tibia and weight of organs.
