Bioactivity of precalcified nanotubular $TiO_2$ layer on Ti-6Al-7Nb alloy (나노튜브 $TiO_2$ 층 생성 후 전석회화 처리한 Ti-6Al-7Nb 합금의 생체활성도)

  • Purpose: The purpose of this study was to investigate the bioactivity of precalcified nanotubular $TiO_2$ layer on Ti-6Al-7Nb alloy. Materials and methods: Anodic oxidation was carried out at a potential of 20 V and current density of 20 mA/$cm^2$ for 1 hour. The glycerol solution containing 1 wt% $NH_4F$ and 20 wt% deionized water was used as an electrolyte. Precalcification treatment was obtained by soaking in $Na_2HPO_4$ solution at $80^{\circ}C$ for 30 minutes followed by soaking in saturated $Ca(OH)_2$ solution at $100^{\circ}C$ for 30 minutes, followed by heat treatment at $500^{\circ}C$ for 2 hours. To evaluate the activity of precalcified nanotubular $TiO_2$ layer, specimens were immersed in a simulated body fluid with pH 7.4 at $36.5^{\circ}C$ for 10 days. Results: 1. Nanotubular $TiO_2$ layer showed the highly ordered dense structure by interposing small diameter nanotubes between large ones, the shape of nanotubes was enlarged as going down. 2. The mean length of nanotubes was $517.0{\pm}23.2\;nm$ innm glycerol solution containing 1 wt% $NH_4F$ and 20 wt% $H_2O$ at 20 V for 1 hour. 3. The bioactivity of Ti-6Al-7Nb alloy was improved with formation of nanotubular $TiO_2$ layer and precalcification treatment in $80^{\circ}C$ 0.5 M $Na_2HPO_4$ and saturated $100^{\circ}C$ $Ca(OH)_2$ solution. Conclusion: Bioactivity of precalcified nanotubular $TiO_2$ layer on Ti-6Al-7Nb alloy was improved.

Browning Inhibition of Paeonia lactiflora Root during Hot Air Dehydration (작약근(芍藥根)의 열풍건조시(熱風乾燥時) 갈변(褐變)의 효과적(效果的) 억제(抑制))

  • This research was carried out to investigate the effective methods of browning inhibition on Paeonia lactiflra Pall during hot air dehydratin. After drying for 36 hrs without pretreatment and with briquet fumigation moisture contents of Paeonia lactiflora Pall. was 16.0% and 16.2%, respectively, while with acidic solution, sulfite salt solution and blanching it was lower of 13.5, 12.9 and 14.8%, respectively. Using freeze drying moisture content was highest of 18.8%. The Hunter values of dried Paeonia lactiflora Pall., L, a, band ${\triangle} E$ showed that non-treatment had the most browning with 61.60, 1.89, 10.20 and 39.78, respectively, while briquet fumigation and sulfite salt solution were excellent in reducing browning. During freeze drying browning didn't occur. Paeoniflorin content was 2.41 and 2.51 %, respectively, in briquet fumigation and sulfite salt solution and was the highest (2.70%) in freeze drying. The content of $SO_2$, was 0.63% both in briquet fumigation and sulfite salts. It was 0.15% and 0.17% higher than nontreatment and freeze drying. The most effective pretreatment to inhibit browning of Paeonia lactiflora Pall. was soaking in sulfite salt solution $(0.5% NaHSO_4-0.5% Na_2S_2O_3)$ considering many things such as manpower, economical efficiency, drying time, commodity, etc. but safety should be evaluated to treat sulfite salts solution on peony roots.

Characteristics of Soybean Soaking Water after Heat Treatment (대두 열처리 담금 용액의 특성)

  • Soybeans released proteins when immersed in water at $50{\sim}60^{\sim}C$. We investigated the changes in the characteristics of soybean when soaked in water at different temperatures and studied the electrophoretic properties of soy proteins in recommended Korean soybean varieties after heat treatment. Soybean seeds were heated in soaking water at temperatures of 30, 40, 50, 60, $70^{\circ}C$ for 90 min, and also from 10 to 150min at $60^{\circ}C$. The pH value of the water decreased with heating time at $60^{\circ}C$, and the amount of soluble solids increased with temperature and heating time. The protein concentration of the solution increased with temperature and time. From SDS-PAGE of the proteins in soaking water, we detected two new bands of 16 kDa- and 31 kDa-proteins from the Korean soybean varieties on heat treatment.

Effect of Rice Seed Disinfection of Loess-sulfur on the Suppression of Bakanae disease caused by Fusarium fujikuroi (벼 키다리병 방제에 관한 황토유황의 종자소독 효과)

  • This study was conducted to evaluate rice seed disinfection efficacy of loess-sulfur for the suppression of Bakanae disease caused by Fusarium fujikuroi. Rice seeds were treated at different concentrations of loess-sulfur, soaking time and temperature, and combination of hot-water treatment. Rice cultivar, Shindongjin harvested from Bakanae disease-infested area in 2015, was used. Loess-sulfur was treated as follows; concentration of undiluted solution, 2%, 1% and 0.5%; soaking time of 24 and 48 hours; treatment temperature of $20^{\circ}C$ and $30^{\circ}C$; hot water treatment or not. Optimal conditions of rice seed disinfection were selected soaking time of 48 hours and the suspension of 0.5% and 1% loess-sulfur by investigating seed germination and isolation frequency of Fusarium spp. on Komada agar medium in vitro, and were established 3 disinfection conditions as hot water ($60^{\circ}C$, 10 min.) + 1% loess-sulfur ($20^{\circ}C$, 48 hours), 1% loess-sulfur only ($30^{\circ}C$, 48 hours) and 1% loess-sulfur only ($20^{\circ}C$, 48 hours) through additional test in greenhouse. Above 3 conditions were verified by rice seedling box and paddy field test in the way of investigating Bakanae diseased plants (%) and healthy plants (%). Consequently, most effective rice seed disinfection conditions on Bakanae disease were combination of hot water and 1% loess-sulfur and loess-sulfur only at $30^{\circ}C$. Furthermore, treatments with these conditions showed control value of 100% were maintained from seedling to the heading stage in the field. However, treatment of 1% loess-sulfur only at $20^{\circ}C$ showed low control value of 78.2% in paddy field. Hot water only treatment turned out to be an effective disinfection method when conducted thoroughly with $60^{\circ}C$, 10 min. However, it was thought additional soaking process with loess-sulfur after hot water treatment served more high control effect against Bakanae disease when rice seeds were disinfected on a large scale. This results expected rice seed disinfection with loess-sulfur were effectively and easily usable method if farmers had only one of either hot water-disinfector or seed-disinfector. In addition, loess-sulfur is well-known to farmers, simple to manufacture method and cheap.

  • Problems with maintaining service and equipment in some developing countries suggest that the rumen bag technique may be more appropriate for the determination of plant dry matter digestibility. The technique has been adapted for use in goats in the 16-25 kg liveweight range. Reliable results were obtained for animals maintained under shelter in cages and fed on a mixed legume/grass diet. The results showed that up to 7 bags containing dried and ground (2 mm screen) plant samples (1-3 g) could be satisfactorily used in each goat. The digestibility of the legumes studied did not increase with incubation times over 48 hours, but there was an increase in the digestibility of grasses. However an incubation time of 48 hours was adapted for both legumes and grasses as it allowed more efficient work scheduling for large numbers of samples while still giving acceptable comparisons between species. Losses of material from the bags during a 6 hour soaking in water were 2-9% as fine solids and 14-21% in solution. In the method finally adapted the disappearance was measured for plant samples that were placed in Dacron mesh bags ($7{\times}14cm$, 44 micron) and 6 bags suspended in the rumen of each sheep for 48 hours.

Changes in Chemical Components of Soybean Cheese Making from Cow한s Milk Added Soybean Curd (우유 첨가두부를 이용한 대두 치이즈 제조 중 화학성분의 변화)

  • In order to making the good quality soybean cheese, it is prepared byover growing cow's milk added soybean curd with Actinomucor elegans and the nold-overgrown curd soked in salt-brine/ethanol mixture. The physicochemical changes and sensory evaluation were investigated during the aging period. Crude protein, carbohydrate, crude fat ash contents of the pehtze were increased by elapsing the fermentation time, whereas moisture's decreased . Amino-N and ammonia -N contents of cow's milk added pehtze and soybean phetze were increased 17.25%, 7.23% and 16.16%, 8.42% respectively. Total nitrogen content of the pehtze was decreased by elaping the aging time but soaking solution's increased. Free amino acid content of soybean cheese was increased as a result of the proteolytic action of molds. As a result, sulfur containing amino acid such as methionine and cysteine of the cow's milk added soybean cheese were enriched 1.3 times more than the soybean cheese. Flavor, taste and texture of the cow's milk added soybean cheese were higher than soybean cheese.

  • Mung bean sprout has been grown for 96 hours by soaking mung bean in such eight test divisions as Gibberellin 5-50ppm solution, well-water, and so on. Study on the charge of vitamin C an reducing sugar during its growth can be summarized as follows. 1. During the growth of mung bean sprout, the length and weight in the sample treated with Gibberellin 20ppm are highest of all Gibberellin divisions. 2. When mung bean sprout has grown for 24 hours, its vitamin C content reaches maximum in each sample. 3. There is little dehydro asscorbic acid in each sample during the growth period of 24-96 hours. 4. During the period of growth, the content of vitamin C in the sample treated with Gibberellin 5ppm are highest of all. 5. When mung bean sprout has been grown enough for cooking, in 48 hours, the content of vitamin C in the sample treated with Gibberellin 10ppm is highest but a significant difference is not recognized. 6. After 24 hours growth, vitamin C decreases but reducing sugar increases.

  • This experiment has been carried out for the purpose of investigating the effect of additives on densification and grain growth in magnesium oxide by a two-step process; hot pressing and heat treating. MgO powder has been obtained by calcining extra reagent grade MgCO3 at 90$0^{\circ}C$ for 30 minutes, and additives have been added to $MgCO_3$ in the form of soluble salts-Al$(NO_3)_3$$. $9H_2O$ and $Cr(NO_3)_3$.9H_2O$. The hot pressing has been carried out with changes of soaking time at 125$0^{\circ}C$ under the pressure of 250kg/$\textrm{cm}^2$, and the heat treating also at same temperature. The initial particle size of MgO measured by particle size analyzer was 0.86 microns. Densification rate obeyed the equation D=K lnt + C, and grain growth rate obeyed the equation G-G0=kt1/2. It was vaporization of some $Cr_2O_3$ and formation of solid solution that had an influence on desification of MgO containing $Cr_2O_3$. Activation energy for grain growth of pure MgO was 62.4 kcal/mole, therefore grain growth was supposed to be diffusioncontrolled process. But after heat treatmeat, excess additives were expected to slow down the grain growth by the formation of second phase or the solute atoms at grainboundary.

A Study on the Silk Dyeing With Natural indigo Extracted from Polygoum tinctorium -On the fermentation dyeing- (쪽 色素에 의한 絹纖維 染色에 관한 硏究 -발효 염색에 대하여-)

  • Colorants were prepared by extraction of natural indigo which was harvested just in the blooming season(in the late of July). 100 g of fresh leaves soaking in 1 ιwater was kept at 3$0^{\circ}C$, 30 hours. A solution of 3g/l calcium hydroxide was added into it to precipitate dye substance and it was freezing-dried into powder form. The fermentation and dyeing conditions were investigated. The results obtained are summarized as follows; K/S value of dyed silk fabrics of fermentation conditions was higher at 95$^{\circ}C$ for 20 min. than at 4$0^{\circ}C$ for 20 hours. Furthermore, K/S value of dyed silk fabric was raised by the addition of 5g/l of glucose and 5g/l of NaOH. K/S value of dyed silk fabric was raised by the addition of 5g/l of glucose and 5g/l of NaOH. K/S value increased as extending of dyeing time when dyed till 2 hours at 3$0^{\circ}C$. K/S value decreased in order of 3$0^{\circ}C$, 4$0^{\circ}C$ and 5$0^{\circ}C$, at the various dyeing temperatures and dyeing concentrations, and colour fastness ranged from 4 to 5 grade in terms of washing, perspiration and light fastness.

Butyric Acid Fermentation of Sodium Hydroxide Pretreated Rice Straw with Undefined Mixed Culture

  • This study describes an alternative mixed culture fermentation technology to anaerobically convert lignocellulosic biomass into butyric acid, a valuable product with wide application, without supplementary cellulolytic enzymes. Rice straw was soaked in 1% NaOH solution to increase digestibility. Among the tested pretreatment conditions, soaking rice straw at $50^{\circ}C$ for 72 h removed ~66% of the lignin, but retained ~84% of the cellulose and ~71% of the hemicellulose. By using an undefined cellulose-degrading butyrate-producing microbial community as butyric acid producer in batch fermentation, about 6 g/l of butyric acid was produced from the pretreated rice straw, which accounted for ~76% of the total volatile fatty acids. In the repeated-batch operation, the butyric acid production declined batch by batch, which was most possibly caused by the shift of microbial community structure monitored by denaturing gradient gel electrophoresis. In this study, batch operation was observed to be more suitable for butyric acid production.
