Butyric Acid Fermentation of Sodium Hydroxide Pretreated Rice Straw with Undefined Mixed Culture

  • This study describes an alternative mixed culture fermentation technology to anaerobically convert lignocellulosic biomass into butyric acid, a valuable product with wide application, without supplementary cellulolytic enzymes. Rice straw was soaked in 1% NaOH solution to increase digestibility. Among the tested pretreatment conditions, soaking rice straw at $50^{\circ}C$ for 72 h removed ~66% of the lignin, but retained ~84% of the cellulose and ~71% of the hemicellulose. By using an undefined cellulose-degrading butyrate-producing microbial community as butyric acid producer in batch fermentation, about 6 g/l of butyric acid was produced from the pretreated rice straw, which accounted for ~76% of the total volatile fatty acids. In the repeated-batch operation, the butyric acid production declined batch by batch, which was most possibly caused by the shift of microbial community structure monitored by denaturing gradient gel electrophoresis. In this study, batch operation was observed to be more suitable for butyric acid production.

Numerical and Experimental Investigation of the Heating Process of Glass Thermal Slumping

  • The glass thermal forming process provides a high volume, low cost approach to producing aspherical reflectors for x-ray optics. Thin glass sheets are shaped into mirror segments by replicating the mold shape at high temperature. Heating parameters in the glass thermal slumping process are crucial to improve surface quality of the formed glass. In this research, the heating process of a thermal slumping glass sheet on a concave parabolic mold was simulated with the finite-element method (FEM) to investigate the effects of heating rate and soaking temperature. Based on the optimized heating conditions, glass samples 0.5 mm thick were formed in a furnace with a steel concave parabolic mold. The figure errors of the formed glass were measured and discussed in detail. It was found that the formed glass was not fully slumped at the edges, and should be trimmed to achieve better surface deviation. The root-mean-square (RMS) deviation and peak-valley (PV) deviation between formed glass and mold along the axial direction were 2.3 μm and 4.7 μm respectively.

  • $\beta$-SiC bonded SiC bodies were prepared from various conditions such as several compositions of(Si+C)/$\alpha$ -SiC ratio and different firing schedules and were respectively investigated compressive strength MOR and mi-crostructure. One firing schedule which produced the specimens that had $\beta$-SiC neck form with the highest strength was selected and experimented by each firing temperature. results obtained are as follows : 1) The amount of (Si+C) for th highest MOR of SiC-(Si+C) sintered body is 20wt% 2) By adding 20wt% content of (Si+C) and heating up to 1, 500 with soaking 3hrs respectively at 1,150$^{\circ}C$ 1,250$^{\circ}C$ 1,350$^{\circ}C$ and 1,400$^{\circ}C$ the highest MOR of fired specimen was resulted and its microstructure of ma-trix was composed of close $\beta$-SiC neck. 3) Microstructure of $\beta$-SiC were different greatly from each other by firing time and/or quantity of adding mix-ture and it was confirmed that they were composed of neck particle-like and heterogeneous texture. 4)$\beta$-SiC synthesis proceed rapidly at the temperature between 1,250$^{\circ}C$ and 1,350$^{\circ}C$ 5) All of the properties of 85 SiC-20(Si+C) specimen improved according to increasing temperature above 1,350$^{\circ}C$.

  • A serious problem of the 21st century is the supply of energy resources. Reserves of fossil fuels are facing depletion: renewable energy resources must be developed in this era. Dye sensitized solar cells(DSC) have been very economical and easy method to convert solar energy to electricity. DSC can reach low costs in future outdoor power applications. However, to commercialize the DSC, there are still many shortages to overcome. When the DSC is commercialized in the near future, the productivity is an important factor. In the process of soaking in a dye, it usually takes 12${\sim}$24 hours. In this study, we varied the dye coloring temperature from 0$^{\circ}C$ to 60$^{\circ}C$. At the temperature of 40$^{\circ}C$, DSC cell showed the best performance. We also conducted the time variant experiment to reduce the manufacturing time. Counter electrode surface of DSC is deposited by RF magnetron sputtering under the conditions of Ar $2.8{\times}10^{-3}torr$, RF power of 120W and substrate temperature of 100$^{\circ}C$.

Phytotoxic Effect of 5-Aminolevulinic Acid, a Biodegradable Photodynamic Biomaterial, on Rice and Barnyardgrass

  • ALA (5-aminolevulinic acid) has been proposed as a tetrapyrrole-dependent photodynamic herbicide by the action of the protoporphyrinogen IX oxidase (Protox IX). A study was conducted to determine photodynamic herbicidal effect of ALA on seedling growth of rice (Oryza sativa L.) and barnyard grass (Echinochloa crus-galli Beauv. var. oryzicola Ohwi) under dry and wet conditions. ALA effect on early plant growth of rice and barnyardgrass was greatly concentration dependant, suggesting that it promotes plant growth at very low concentration and inhibits at high concentration. No significant difference in herbicidal activity of biologically and synthetically produced ALAs on plant lengths of test plants was observed ALA exhibited significant photodynamic activity regardless of PSDIP and its duration. Significant shoot growth inhibition by ALA soaking treatment exhibited apparently, indicating that ALA absorbed through root system was translocated into shoot part of plants. ALA reduced plant heights of rice and barnyardgrass seedlings by 6% and 27%, respectively, showing more tolerant to ALA in rice under wet condition. Leaf thickness was reduced markedly by ALA with increasing of ALA concentration, due to mainly membrane destruction and severe loss of turgidity in mesophyll cells, although the epidermal was little affected. It was observed that photodynamic herbicidal activity of ALA applied by pre-and post-emergence application exhibited differently on plant species, and that the activity of ALA against susceptible plants was highly correlated with growing condition.

Bioconversion of Straw into Improved Fodder: Preliminary Treatment of Rice Straw Using Mechanical, Chemical and/or Gamma Irradiation

  • Crude protein (CP) content of mechanically ground rice straw into small particles by an electric grinder and reducing value (RV) and soluble protein (SP) in the culture filtrate were lower than that of the chopped straw into $5{\sim}6\;cm$ lengths when both ground and chopped straws were fermented with Aspergillus ochraceus, A. terreus or Trichoderma koningii, at steady conditions. The reduction rate of RV, SP and CP was 22.2, 2.4, 7.3%; 9.1, 4.9, 8.5% or 0.0, 0.0, 3.6% for the three fungi, respectively. Chemical pretreatment of straw by soaking in $NH_{4}OH$ for a day caused significant increase in CP of the fermented straw than the other alkali and acidic pretreatments. Gamma irradiation pretreatment of dry and wet straw with water, specially at higher doses, 100, 200 or 500 kGy, caused significant increase in RV and SP as CP in the fermented straw by any of these fungi. Chemical-physical combination pretreatment of rice straw reduced the applied dose of gamma irradiation required for increasing fermentable ability of fungi from 500 kGy to 10 kGy with approximately the same results. Significant increases in RV and SP of fermented straw generally occurred as the dose of gamma irradiation for pretreated straw, which combined with $NH_{4}OH$, gradually rose. Whereas, the increase percentage in CP of fermented straw that was pretreated by $NH_{4}OH-10\;kGy$ was 12.4%, 15.4% or 8.6% for A. ochraceus, A. terreus or T. koningii, respectively.

Study on the Structure of Expanded Rice with Deep-Frying for Salyeotgangjung (유탕처리된 쌀엿강정용 팽화쌀의 조직학적 연구)

  • This study was carried out to observe the character change of expanded rice grain for Salyeotgangjung at various gelatinization methods. The expansion and structure change of rice grain was different at conditions of heating methods and gelatinization, respectively. The boiling showed good expansion compared to other heating methods by electric rice-cooker, pressure cooker, and steam cooker. There was not difference in expansion of rice grain between the boiling and the boiling after soaking. Rice grain did greatly expand as the boiling time was extended. But the quality for Salyeotgangjung would be not good because of crack of expanded rice grain surface when the boiling time was too long.

Development of an Automatic Brown Rice Germinating System of Air-Phase Type with Intermittent Water Spraying and Anion Radiation (간헐 분무와 음이온 방사를 가진 공기노출형 자동 현미발아시스템의 개발)

  • In this study an automatic germinating system for germinated brown rice (GBR) was developed for accelerating germination and sprouting of brown rice. With the developed germinating system, the effects of anion radiation treatment on germination ratio, sprout growth, color quality and ${\gamma}$-aminobutyric acid (GABA) content of brown rice were investigated. This system can fast produce GBR by controlling the environmental conditions such as ambient temperature, relative humidity, and proper anion radiation. Air-phase germination with intermittent water spraying improved both germination ratio and sprout growth by about 2.2 and 2.3 times, respectively, compared to the conventional water soaking method. Our results strongly suggest that this germinating system that provides air-phase germination with intermittent water spraying and anion radiation can produce germinated brown rice with good color quality, high GABA content, and fast germination ratio as well as sprout growth.

Changes in Chemical Components of Chungkugiang Prepared with Small Black Bean (소립검정콩 청국장의 화학성분 변화)

  • Changes in chemical components of small black bean chungkugjang(SBBC) added with kiwi and radish as foodstuffs to repress off-odor and enhance the quality of SBBC suring fermentation were investigated. Optimal pretreatment conditions of small black bean suitable to the fermentation of chungkugjang were 3 hrs of soaking time 1.5 times of ratio of water to black bean. 1.0 atm of high pressure, 20 min of heating time, cutting and crushing of heat-treated black bean. Moisture content of SBBC was remarkably lower than that of soybean chungkugjang(SBC) as control. Crude protein of SBBC was in the range 23.37∼25.71% and higher than that of SBC, Crude lipid of SBBC was lower than that of SBC. Crude lipid of SBBC added with kiwi and radish paste was decreased than that of SBBC without two foodstuffs. pH of SBBC were rapidly increased to 24 hrs of fermentation and gradually increased thereafter. Total acidity was shown to be reversely decreased as compared to pH tendency. Reducing sugar was increased to 24 hrs of fermentation and then decreased. In SBBC and SBC, potassium was the most abundant followed by phosphorus, magnesium and calcium.

In the external treatment of sprain and strain using the herb, Adhesive(貼敷) is used most. Because Adhesive(貼敷) brings fast reactions, has less side effects, and can control the processing time. 3. The effects of herbs used in this external treatment are almost the same as oral herb-medicines such as, promoting blood flow to remove blood stasis(活血化瘀), relieving rheumatic conditions(祛風濕), and removing obstruction in meridians and collaterals(通經絡).
