Separation and growth monitoring of oyster mushroom spores using gravitational field-flow fractionation (GrFFF) (중력 장-흐름 분획법을 이용한 느타리버섯 포자의 분리 및 성장 모니터링)

  • Gravitational field-flow fractionation (GrFFF) is a separation technique that utilizes earth's gravity as the external field. GrFFF is a convenient tool for the size and/or density-based separation of micron-sized particles of various origins. In this study, GrFFF was employed for size-based separation of oyster mushroom spores. Oyster mushroom spores have smooth surface and are of cylindrical to narrow kidney-shapes with 5 to 12 im in longer dimension and 3 to 4 im in shorter dimension, as was confirmed by optical microscope (OM). GrFFF conditions were optimized for separation and characterization of spores by varying the channel flow rate from 0.5 to 1 mL/min. During the GrFFF elution of the spores, 3 fractions were collected to confirm the growth of oyster mushroom spore. The collected fractions were incubated for 30 days in water to examine the influence of the size on the growth of the spores. Results suggested that the oyster mushroom spores collected at the middle part of the GrFFF fractogram grew faster than those collected at the beginning or at the end of the fractogram.

  • This research is for dental hygienists to pursue affection and commitment in organization together with improvement of quality of working life by analyzing factors of working life affecting affective commitment in organization of dental hygienists, to find out plans to increase sense of accomplishments of working life and ultimate satisfaction of life, and to suggest direction for human resources management in management of dental clinics. Objects of investigation were dental hygienists working at dental medical institutions in Busan area with questionnaire survey and 197 sheets of questionnaires were used in the study. As the result of the study, high correlation was shown between composition elements of quality of working life and affective commitment per factor and as the result of quality factors of working life affecting affective commitment in organization, work environment, inter-personal relations, and social meaningfulness provided meaningful impacts on affective commitment in organization. It is believed to be important that dental medical institutions need to promote pride and love of companies in the future by providing regular social services or services for local residents so that employees can form smooth inter-personal relationships among employees with improvement of working environment and welfare in personnel management dimension. And for dental hygienists, they shall improve their work performances and enhance vocational values for development of their institutions and them with positive thinking through ceaseless efforts.

An Analysis of Factors Influencing the Dementia Patient Home Care Intention of Family Caregivers (치매가족의 치매대상자 재가돌봄의사에 대한 영향요인분석)

  • This study was conducted to find a practical implication regarding efficient and smooth service utilization including care burden reduction of family caregivers by empirically analyzing the factors influencing the dementia patient home care intention of family caregivers, based on Anderson's behavior model. For this study, the subjects and characteristics suitable for the study purpose were selected using the source data of "A Survey on the Dementia Recognition and Needs" targeting 26 cities and countries in Gyeonggi-do and then secondary analysis was conducted. This study targeted 539 dementia families, and based on Anderson's behavior model, variables related to predisposing factors, enabling factors and need factors were inserted, and hierarchical regression analysis was applied. The analysis result showed that at a significance level of 5%, the home care intention was high when family caregivers were not living in cities, they were spouses or children and their economic level was high as predisposing factors, and the dementia-related attitude was positive as a enabling factor, and the degree of dementia was mild and care burden was low as need factors. Moreover, need factors were relatively important variables among the three factors mentioned above. These findings demonstrate that for improving the home care intention, there is a need to construct a support strategy, which considers the degree of dementia and economic characteristics and develop diverse counseling and education programs for a positive attitude towards dementia and a differentiated strategy depending on regional and family characteristics.

Dispersion of Particle Pulse in Human Lung Airway (인체기관지내의 입자펄스 확산 실험)

  • In order to develop the aerosol bolus technique which is thought to be a potential tool for probing geometries or abnormalities of small airways, an experimental system of measuring fast time variations of particle concentration in the inhaled and exhaled breathing air was developed. The system generates monodisperse sebacic acrid particles of 1 micron size and 1.2 of geometric standard deviation in high concentration of $10^8$ particles/cc, delivers a short pulse of particles at the controlled instant during inhalation using a solenoid valve, and measures the fast change of particle concentration in using the laser light scattering. Successful operation of the generator and the measuring system was confirmed by smooth concentration profiles in inhalation. It was also confirmed that maintaining a constant breathing rate is essential to stable outputs and any disturbance in flow rate near the mode (maximum concentration) induces a large number of spurious peaks in the exhalation. Experimental data without strict control of breathing flow rate showed a substantial amount of scatter. The measured results showed an improvement in scatter over the existing results. When compared with theoretical predictions from 1-D convective diffusion equation and other experiments, general characteristics of dispersion for several penetration depths showed a good agreement, but there exists some difference in absolute values, which is attributed to the difference in body conditions. Improvements are needed in the theory, especially in relation to correcting for the effect of breathing flow rate.

Different Mechanisms for $K^+-Induced$ Relaxation in Various Arteries

  • $[K^+]_o$ can be increased under a variety of conditions including subarachnoid hemorrhage. The increase of $[K^+]_o$ in the range of $5{\sim}15$ mM may affect tensions of blood vessels and cause relaxation of agonist-induced precontracted vascular smooth muscle $(K^+-induced$ relaxation). In this study, effect of the increase in extracellular $K^+$ concentration on the agonist-induced contractions of various arteries including resistant arteries of rabbit was examined, using home-made Mulvany-type myograph. Extracellular $K^+$ was increased in three different ways; from initial 1 to 3 mM, from initial 3 to 6 mM, or from initial 6 to 12 mM. In superior mesenteric arteries, the relaxation induced by extracellular $K^+$ elevation from initial 6 to 12 mM was the most prominent among the relaxations induced by the elevations in three different ways. In cerebral arteries, the most prominent relaxation was produced by the elevation of extracellular $K^+$ from initial 1 to 3 mM and a slight relaxation was provoked by the elevation from initial 6 to 12 mM. In superior mesenteric arteries, $K^+-induced$ relaxation by the elevation from initial 6 to 12 mM was blocked by $Ba^{2+}\;(30\;{\mu}M)$ and the relaxation by the elevation from 1 to 3 mM or from 3 to 6 mM was not blocked by $Ba^{2+}.$ In cerebral arteries, however, $K^+-induced$ relaxation by the elevation from initial 3 to 6 mM was blocked by $Ba^{2+},$ whereas the relaxation by the elevation from 1 to 3 mM was not blocked by $Ba^{2+}.$ Ouabain inhibited all of the relaxations induced by the extracellular $K^+$ elevations in three different ways. In cerebral arteries, when extracellular $K^+$ was increased to 14 mM with 2 or 3 mM increments, almost complete relaxation was induced at 1 or 3 mM of initial $K^+$ concentration and slight relaxation occurred at 6 mM. TEA did not inhibit $Ba^{2+}-sensitive$ relaxation at all and NMMA or endothelial removal did not inhibit $K^+-induced$ relaxation. Most conduit arteries such as aorta, carotid artery, and renal artery were not relaxed by the elevation of extracellular $K^+.$ Among conduit arteries, trunk of superior mesenteric artery and basilar artery were relaxed by the elevations of $[K^+]_o.$ These data suggest that $K^+-induced$ relaxation has two independent components, $Ba^{2+}-sensitive$ and $Ba^{2+}-insensitive$ one and there are different mechanisms for $K^+-induced$ relaxation in various arteries.

Effects of Cyclobuxine D on the Derangement Induced by Ischemia and Reperfusion in the Isolated Rat Heart (Ischemia에 의해 유발된 흰쥐의 적출 심장 손상에 대한 Cyclobuxine D의 보호효과)

  • Cyclobuxine D is a steroidal alkaloid, which was extracted from Buxus microphylla var. koreana Nakai. In our previous studies, we clarified several pharmacological actions of cyclobuxine D: an antiinflammatory action, hypotensive and bradycardiac effects, negative inotropic effects on the several smooth muscles and cardiac muscle. The present study was undertaken to elucidate possible mechanisms by protection of myocardial tells from ischemia and reperfusion induced derangement in cardiac function and metabolism by cyclobuxine D. For this purpose, the isolated rat heart was used. Rat hearts were perfused for 60 min under ischemia conditions in the presence and absence of cyclobuxine D and verapamil, and for 30 min under reperfusion conditions. Ischemia produced a marked decline in contractile force, an increase of resting tension, an immediate release of ATP metabolites and an accumulation of calcium in the left ventricle. Cyclobuxine D (100ng/ml) ameliorated the myocardial injury produced by ischemia.

  • Objective: To improve customer satisfaction through accurate information using computerized automation of the test reservation system and by performing all fasting tests on the same day with one fasting. Methods: Quality improvement activities of the test reservation system of Seoul National University Bundang Hospital improves internal and external customer satisfaction at the time of test scheduling by having the reservation system reflect test order and test time automatically and making the system to increase the same day test reservation rate. Results: Test process was smooth due to accurate test reservation and information based on test priority due to the introduction of the computerized test reservation system. Internal and external customer satisfaction improved due to the marked reduction in time required for making a reservation thanks to the improvement of work process. Conclusion: Building a computerized test reservation system was an effective measure to improve external and internal customer satisfaction.

A Study of Characteristics of Business Cycle in the Jeju Region (제주지역 경기변동의 특성 연구)

  • The purpose of this paper is to examine the business cycle in the Jeju region and what differences exist in relation to the nation as a whole, to calculate the relative importance between the random walk stochastic trend and cyclical factor, and to find out its causes and implications. Results of empirical analysis found that the characteristics of the business cycle in the Jeju region were as follows: First, the Jeju region, which is likely to have a growth component of the economy such as technological development and the accumulation of capital, was projected to have a possibility of high growth due to a greater proportion of the stochastic trend factor(46.8%) than the entire country(27.8%). Secondly, employment fluctuation in Jeju, which varies from 0.007 to 0.058 depending on the model, was lowest compared to the fluctuation of other indicators. The employment market in Jeju remained firm, showing that it is not smooth enough to create new jobs despite the production growth in industry. Third, the tourism industry was acting as a stabilizing factor, whereas the mining and manufacturing production was the opposite of tourism industry. This implies that the mining and manufacturing production was based on a weak foundation.

The Impact of Social Support for and Self-esteem of North Korean Adolescent defectors on their Problem Solving Ability (탈북청소년의 사회적지지 및 자아존중감이 문제해결능력에 미치는 영향)

  • Given that the ratio of adolescents among North Korean refugees is increasing, it is the right time to study the present situation of juvenile North Korean defectors. North Korean adolescents tend to find themselves in an uncomfortable situation as they experience unfamiliar lives in South Korea, and act passively in various situations. A proper resolution of such problems in this situation will help them adapt smoothly to South Korean society. This study investigates the relationship between social support, self-esteem, and problem-solving abilities of North Korean adolescent refugees based on a survey on 213 North Korean teens which was performed for 24 days beginning on June 14 2017. The study results show that emotional, material, informational, and evaluative support for North Korean adolescent defectors, which is a type of social support, had a positive effect on their self-esteem, and self-esteem in turn had a positive effect on their problem solving ability. In addition, self-esteem had a mediating effect on the relationship between the four types of social support and problem solving ability of North Korean adolescent defectors. This study suggested a practical plan to improve the problem solving ability of North Korean adolescent defectors for their smooth adaptation to South Korean society, through social support and improved self-esteem of North Korean adolescents, such as program development or self - development training.

THE EFFECTS OF DIAZEPAM ON THE CARBACHOL INDUCED CONTRACTION OF THE ISOLATED RAT ILEUM (Diazepam이 흰쥐 회장 평활근의 Carbachol 유발 수축에 미치는 영향)

  • To investigate the effect of diazepam on the contractility of the intestinal smooth muscle, longitudinal muscle strip isolated from rat ileum was prepared for myography in isolated organ bath. 1) Basal tone of ileal muscle was reduced by diazepam concentration-dependently. 2) Higher concentrations(30 and 100 microM) of diazepam inhibited(p<0.05, p<0.001) the carbachol-induced contraction in a concentration-dependent manner ; but lower concentration of diazepam(10 microM) enhanced(p<0.05). 3) Histamine-induced contraction was inhibited by pretreatment with diazepam in a concentration-dependent manner. 4) $Ca^{++}$-induced tension recovery in calcium-free solution was inhibited in the presence of diazepam concentration-dependently. These results suggest diazepam reduces the contractility of the longitudinal muscle isolated from rat ileum via interference with influx of calcium into the muscle cells.
