Serotyping and antibiotic susceptibility of Salmonella species from pigs with rectal stricture found in slaughterhouses (도축장 출하 직장협착 돼지에서 분리된 살모넬라균 혈청형 및 항생제 감수성)

  • From November 2021 to October 2022, 151 pigs were discarded due to the suspected salmonellosis, when assessed by antemortem inspection at slaughterhouses in Gwangju. Among them, 85 cases shown with the rectal stricture were confirmed to be infected with Salmonella spp, when assessed by postmortem inspection and Salmonella-oriented culture in the laboratory, accounting for 56.3% of the carcass discard cases. Salmonella spp. were isolated in 58 cases (68.2%) out of 85 total discard cases. As for serotyping of the Salmonella isolates, 28 cases (48.3%) belonged to serogroup B and 28 cases (48.3%) belonged to serogroup C, those were the most frequent isolates, and 2 cases (3.4%) belonged to serogroup E. The most prevalent serotype was Rissen (39.7%), followed by I4,[5],12:i:-(37.9%), Derby (8.6%), Bareilly (5.2%), Infantis (3.4%), Give (3.4%), and Typhimurium (1.7%). The 58 isolates of salmonellae showed resistance to tetracycline (63.8%), chloramphenicol (56.9%), ampicillin (53.4%), and ampicillin-sulbactam (51.7%). S. Rissen showed resistance to chloramphenicol (82.6%), ampicillin (52.2%), ampicillin-sulbactam (52.2%), and tetracyclin (47.8%). S. I4,[5],12:i:-was resistant to tetracycline (90.9%), ampicillin (81.8%), ampicillin-sulbactam (77.3%), and chloramphenicol (59.1%). Among the isoates, S. Bareilly (3 strains), S. Infantis (2 strains), S. Give (2 strains), S. Rissen (3 strains), and S. I4,[5],12:i:- (1 strain) showed no resistance to the tested antimicrobials. As for the prevalence of the antibiotic resistance pattern, AM-SAM-TE-C was the most common, which was shown in 15.5% of S. Rissen. At second, the resistance pattern was occurred as AM-SAM-TE (12.1%) and AM-SAM-CF-CZ-FOX-CTX-NA-TE-C (12.1%), respectively in the case of S. I4,[5],12:i:-.

  • HACCP is a scientific and systematic program that identifies specific hazards and gives measures for their control to ensure the safety of foods. Based on the Livestock Products Processing Act, the HACCP system is now being applied to Korean livestock products since December 1997, and Korea is accelerating its application from farm to table, including in farms, slaughterhouses, livestock product industries, retail markets, and transportation. The transport of livestock and its products is one of the vulnerable sectors in terms of food safety in Korea. Meats are transported in trucks in the form of carcasses or packaged meats in boxes. Carcasses may be exposed to microbiological, physical, and chemical hazards from the environment or through cross-contamination from other meats. Poor cleaning or maintenance of vehicles and tools may also raise the exposure of carcasses to microbiological or chemical hazards. HACCP application and its acceleration in distribution, particularly in transport, is regarded as critical to the provision to consumers of ultimately safe livestock products. To achieve this goal, steady efforts to develop practical tools for HACCP application should be carried out.

  • This study was carried out to investigate the possibility to utilize feather meal by bacterial strains. A bacterial strain SE-103 producing keratinolytic enzyme was isolated from the soil of the poultry slaughterhouses. It was identified as Bacillus sp. by judging from its morphological and physiological characteristics. Subsequently the optimal culture conditions for the production of keratinolytic protease by Bacillus sp. SE-103 were investigated. The composition of optimal medium was 3.0% glucose, 0.4% urea, 0.2% $NaNO_3$, and 0.15% KCl. In addition, optimal initial pH and temperature were 6.0 and $35^{\circ}C$, respectively.

Prevalence of Eimeria infection in commercial broilers in Gyeonggi province, Korea (경기지역 육계의 콕시듐 감염실태 조사)

  • Protozoan parasite, the genus Eimeria, causes an intestinal disease, coccidiosis, in young broilers. Coccidiosis induces significant economic loss in poultry production. This study was designed to identify the prevalence of Eimeria spp. in broilers in Gyeonggi province, Korea. Fecal samples from broilers at 94 farms were collected in two chicken slaughterhouses between March and June 2015. Eimeria infection was determined by microscopic examination and PCR using ITS-1 region. The prevalence of Eimeria was 58.5% (55 of 94). E. acervulina was identified in 96.4% of samples, E. tenella in 81.8%, E. maxima in 12.7%, E. praecox in 12.7%, E. brunetti in 5.5%, E. necatrix in 1.8%, and E. mitis in 1.8%. Body weight (BW) of broilers infected with both E. tenella and E. acervulina ($mean=1.53{\pm}0.19^Bkg$) was significantly lower than broilers with E. acervulina positive/E. tenalla negative ($mean=1.74{\pm}0.07^Akg$) or Eimeria negative ($mean=1.65{\pm}0.15^Akg$) (Duncan's multiple range test, P<0.01). High prevalence of Eimeria tenella and the impact on the broiler body weight shows the importance of this protozoa in broiler industry. Development of the periodic monitoring strategy and systemic management for the purpose of the prevention/eradication of Eimeria infection among broilers is required.

  • The occupations involved in food animal production have long been recognized to carry significant health risks for workers, with special attention to injuries. However, risk of pathogen exposure in these occupations has been less extensively considered. Pathogens are a food safety issue and are known to be present throughout the food animal production chain. Workers employed at farms and slaughterhouses are at risk of pathogen exposure and bacterial infections. The industrialization of animal farming and the use of antimicrobials in animal feed to promote growth have increased the development of antimicrobial resistance. The changed nature of these pathogens exposes workers in this industry to new strains, thus modifying the risks and health consequences for these workers. These risks are not yet recognized by any work-related health and safety agency in the world.

  • There were eight Korean indigenous cattles affected with bovine tuberculosis (BTB) detected by inspectors at slaughterhouses located in Chungbuk province from May 2006 through July 2007. Postmortem finding of BTB cases was characterized by the presence of several caseous or calcified nodules encapsulated by connective tissue from the pleural/peritoneal surface, livers, lungs and regional lymph nodes On micro-scopic examinations, the characteristic lesion of BTB was the formation granulomatous nodules, which contains central calcified necrotic zone surrounded by epithelioid cells, macrophages and a few Langhans' type giant cells. In addition, mononuclear cells and fibroblasts were also infiltrated. At the periphery, encapsulation was formed that protect the neighboring healthy tissues.

Comparison of histopathological and molecular diagnostic methods in Helicobacter suis infection in Korea (국내 도축돈의 위병변과 Helicobacter suis의 검출과의 상관관계)

  • Helicobacter suis is a gram negative bacterium and colonizes in porcine stomach. It causes gastric diseases in the stomach and plays a significant role in daily weight gains in pigs. Recent studies about one of potential sources of human gastric diseases. Therefore, this study was conducted to compare histopathological lesions and molecular detection of Helicobacter suis in the pyloric mucosa of porcine stomachs transferred from slaughterhouses, based on gross and histological examinations and a PCR assay. A total 90 stomach samples were investigated to record gastric lesion scores by characteristic gastric lesions, followed by routine H & E and Warthin-Starry silver staining to detect Helicobacter-like organisms. For PCR assay, H. suis specific primers and conditions are used. Sixty-one samples (67.8%) showed gross gastric lesions, of which 38 samples (40.2%) presented grade 1, 12 samples (13.3%) presented grade 2, and 11 samples (12.2%) presented grade 3, respectively. In Warthin-Starry silver stain, Helicobacter-like organisms were detected from 11 samples (12.2%) with 4 samples (4.4%) for grade 0, 5 samples (5.6%) for grade 1, 1 sample (1.1%) for grade 2 and 1 sample (1.1%) for grade 3, respectively. The PCR resulted positive in 37 samples (41.1%) with 14 samples (15.6%) for grade 0, 14 samples (15.6%) for grade 1, 3 samples (3.3%) for grade 2 and 6 samples (6.7%) for grade 3, respectively. Positive samples for both examinations were 5 samples (5.6%). The result suggested that it should be considered as one of factors causing a gastric disease in pigs. Also, it could be acknowledged to research fundamental aspects of Helicobacter-induced gastritis in human as an animal model.

A Study on the Sulfametazine residues in Swine Tissues (일부지역 돼지장기 및 근육내 잔류설파메타진에 대한 조사연구)

  • This study was carried out to determine the sulfametazine residues in swine tissues. For this experiment, 22 samples of porks were collected at meat markets in Seoul, 21 samples of swine muscles for export were collected at slaughterhouses in Gyunggi areas, and 18 samples of swine livers, kidneys, and muscles were collected from 6 swine at slaugterhouse in Seoul from the end of August to the early of October and were analyzed by High Performance Liquid Chromatography (HPLC). the results obtained were summarized as follows. 1. The sulfametazine residues were liver > kidney > muscle, and among liver, kidney and muscle were very significantly different(p<0.005). 2. The sulfametazine residue in liver was very significantly higher than that in muscle (p<0.005), the sulfametazine residue in kidney was significantly higher than that in muscle (p<0.005), but, the sulfametazine residue in liver was not significantly higher than that in kidney(p<0.05). 3. the sulfametazine residues in swine muscles for export wer exhibited a tendency to lower in small breeding size farms. 4. the sulfametazine fesidue in one of 22 samples of porks for domestic consumption was exceeded 0.1 ppm.

Seroepidemiological Studies on Bovine Respiratory Syncytial Virus of Cattle in Chonnam Province (전남지방의 소 합포체성 폐렴바이러스(Bovine Respiratory Syncytial Virus) 감염에 관한 혈청학적 연구)

  • Sera from 425 Korean native and 203 Holstein cattle were collected from October 1994 to September 1995 from dairy farms and slaughterhouses in Chonnam province to study the exposure rate to bovine respiratory syncytial virus(BRSV). Serum antibody titers against BRSV were measured by neutralization test, and results were as follows: Overall prevalence of seropositive cattle to BRSV were 74.5%, and the exposure rate to BRSV was higher among the Holstein (77.3%) than among the Korean native cattle(73.2%). The serum antibody titers against BRSV ranged from 1:2~$\geq$1:256 in both species. Among Korean native cattle, the most frequent serum antibody titer was 1:4 against BRSV(19.3%), while only 1.4% of seropositive cattle had serum titer of $\geq$1: 256. Among Holstein cattle, 22.7% of examined cattle contained serum titer of 1:8, while 1.5% of seropositive cattle showed $\geq$ 1:256. Antibody titers against BRSV were higher among males than females in both Holstein (82.1% vs. 73.1%) and Korean native (74.5% vs. 69.2%) cattle. Prevalence of seropositive cattle by age in both species were evenly distributed, although the highest number (76.9%) of seropositive were at the age of 3 in Korean native cattle, while 83.5% of seropositive Holstein cattle were of 2 years old. The lowest seropositive rate was observed in cattle of less than 1 year old(25.0%). Seasonal occurrunce of BRSV was the highest in spring season in both Holstein (86.6%) and Korean native (81.0%) cattle (P<0.05).

Genetic Diversity of Echinococcus granulosus Genotype G1 in Xinjiang, Northwest of China

  • Cystic echinococcosis (CE) caused by E. granulosus is a serious helminthic zoonosis in humans, livestock and wildlife. Xinjiang is one of high endemic province for CE in China. A total of 55 sheep and cattle livers containing echinococcal cysts were collected from slaughterhouses in Changji and Yining City, northern region of Xinjiang. PCR was employed for cloning 2 gene fragments, 12S rRNA and CO1 for analysis of phylogenetic diversity of E. granulosus. The results showed that all the samples collected were identified as G1 genotype of E. granulosus. Interestingly, YL5 and CJ75 strains were the older branches compared to those strains from France, Argentina, Australia. CO1 gene fragment showed 20 new genotype haploids and 5 new genotype haplogroups (H1-H5) by the analysis of Network 5.0 software, and the YLY17 strain was identified as the most ancestral haplotype. The major haplotypes, such as CJ75 and YL5 strains, showed identical to the isolates from Middle East. The international and domestic trade of livestock might contribute to the dispersal of different haplotypes for E. granulosus evolution.
