Various kinds of positive effects of green tea extracts had been studied for long time which included anti-inflammation, anti-aging, and cardiometabolic effects. Although topical steroid and non-steroidal calcineurin inhibitors may control clinical symptoms of allergic contact dermatitis, some of patients also present allergic reaction to these topical agents. Therefore, we have tried green tea extracts for managing this skin disorder with expectation of anti-inflammatory effect without potential side effects including skin irritation and toxic responses. The toxicity test of green tea extract also did not show any sign of irritation in the skin throughout the test period. Moderate severity of allergic contact dermatitis presented satisfactory clinical outcome at second week follow-up which was final visit of outpatient. This result mean that green tea extract has a positive effect for managing allergic contact dermatitis but its potency and efficacy seem to be so not strong enough to control moderate severity allergy skin lesion. In this pilot study, we were able to conclude that green tea cell extracts might be applied for potential anti-inflammatory soaking without skin toxicity.
Chitosan cross-linked collagen-glycosaminoglyan (CCGM) is an artificial skin substitute made to form a sponge like dimensional matrix. It can be used to facilitate reconstruction of dermal tissue when applied on large wounds such as severe burns. In order to study the toxicological effects of CCGM the cytotoxicity, local irritation and skin sensitization test were carried out according to the standards of ISO 10993. In the cytotoxicity test utilizing LDH and MTT test, both the CCGM and its extract had no toxicity of Balb/c 3T3 cells. The local irritatioin test on rabbit skin demonstrated that CCGM did not promote any harmful when directly applied on skin. In addition, it did not elicit any allergic reaction in the guinea pig maximization test. Based on these results, it is suggested that CCGM is a material without cytotoxicity, local irritation and allergenicity.
In vitro cell culture system has been proposed as a promising alternative model to in vivo skin irritation test. These studies were performed to screen the cytotoxicity effects of surfactants using normal human skin fibroblasts. Cell membrane integrity assessed by the leakage of lactate dehydrogenase (LDH) and mitochondrial integrity by MTT [3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromides reduction test were affected in a dose dependent manner. The irritation potential of surfactants to human skin patch test, and the changes of capillary permeability by rabbit intradermal safety test were assessed as in vivo methods. Our results suggest that LDH leakage assay and MTT reduction test using cultured human fibroblasts could be predictive for the irritancy of various surfactants in human, and LDH assay is superior correlated with in vivo test (r=0.886) to MTT test with in vivotest (r=0.757).

Objectives: In this study, we analyze statistics on industrial accidents caused by chemical skin contact and provide skin toxicity hazard information on the related domestic system and circulation volumes. Methods and Results: We analyzed occupational fatalities and skin diseases caused by chemical leaks and contact from 2007 to 2016(10 years) and surveyed data on occupational skin diseases using the 2014 work environment survey data. The NIOSH Skin Notation Profiles for 57 chemical substances, which are provided to prevent occupational skin diseases, were searched and hazard information on skin contact with chemical substances was classified. In order to identify skin toxicity information among domestically distributed and legally regulated substances and to investigate skin-toxic substances, MSDS basic data on 19,740 chemical substances provided on the homepage of Korea Occupational Safety & Health Agency were searched. Acute toxicity(dermal) category 1-4 substances totaled 1,020, and the number of chemical substances classified as category 1 and 2 substances were 135 and 137, respectively. In the chemical substances prescribed by the Ministry of Employment and Labor, 173 substances were classified into acute toxicity(dermal) categories 1-4, 58 of which correspond to category 1 or 2. Conclusions: Within the present range of industrial accidents, the proportion of skin diseases due to contact with chemicals is not high. However, there is always a risk of occupational skin diseases due to increasing chemicals and due to the use of new chemicals. It is hoped that this information will be used by workplace safety and health officials and health and safety experts to prevent acute toxity(dermal) due to chemical skin contact.

Oral mucous membrane test using Syrian hamsters was performed to evaluate the reliability as a model system for the assessment of the potentially irritating substances intended for the mucous membranes, and to determine the irritating potential of a new emulsion-type formulated toothpaste. After test substances were implanted into the cheek pouches of hamsters with diluents (20 mg/kg) under pento-barbital sodium anesthesia, we made the comparison in irritation between emulsion-type and dispersion-type of triclosan (TCS) formulations in the range of 0.2% to 0.3%. The emulsion-type formulations using non-ionic surfactant showed less mucosal lesion than other commercial toothpastes with 0.3% TCS, or dispersion-type ones. However, no significant difference in irritation was detected between 0.2% and 0.3% TCS. We report that this hamster cheek pouch method could be a reliable approach for the evaluation slight difference in the irritating potentials of cosmetics and hygiene products intended for the lips or other mucous membranes, and this method showed that the new emulsion-type formulation significantly lowered the TCS-induced toxicity, compared with other commercial toothpastes.

As research results showed that ozonated olive oil has excellent therapeutic effects on skin diseases, attempts were made to develop cosmetics manufacturing technology using ozone. Ozone is harmful to the human body, so a separate facility must be prepared to manufacture ozonated olive oil as a cosmetic raw material. The manufacturing device developed in this study was designed as an optimized process for manufacturing cosmetics while ensuring the safety of workers involved in production. To verify the ozonated olive oil produced from the implemented device, a toxicity test was conducted on animals. After applying ozonated oil (high concentration) to the rabbit's back for 24 hours, mortality, general symptoms and symptoms were measured. A skin irritation evaluation was performed. As a result of the experiment, as a result of evaluating the test substance treatment area after a certain period of time, it was confirmed that no skin irritation was observed in all animals, confirming the safety of the ozonated oil production process and the safety of the product.
Chlorpyrifos is an organophosphate insecticide and one of the most commonly and widely used insecticide. However, a little known about the dermal risk of chlorpyrifos on human being. Therefore, this study was conducted for the dermal risk assessment after exposure to chlorpyrifos in Korean farmers. First, skin irritation by chlorpyrifos (10 mg/$\textrm{cm}^2$, 50 mg/$\textrm{cm}^2$, 100 mg/$\textrm{cm}^2$, 250 mg/$\textrm{cm}^2$ in acetone) was determined in rabbits for 5 days considering the usage of chlorpyrifos short term highly exposure. The index of skin irritation by chlorpyrifos was increased in each dose and length of exposure dependent manners. Next, using benchmark dose (BMD$_{5}$) approach, the dose-response relationship was assessed to calculate the reference dose (RfD). The value of RfD was 2.84 $\mu\textrm{g}$/kg/day from 142.16 $\mu\textrm{g}$/kg/day BMD5 value divided uncertainty factor 50. Finally, we assessed human dermal risk of chlorpyrifos with exposure level and RfD. Skin absorbed levels were assumed with several exposure scenarios encounting the circumstances of exposure that application method, protection equipment and cloth, exposure time and exposure frequency during chlorpyrifos spraying. By the comparison of skin absorbed dose with the reference dose, it was identified that risk values (risk index) to skin chlorpyrifos exposure were 0.958 from the point of above results and it was recommended that the occurrence of hazard effect (skin irritation toxicity) of chlorpyrifos would not be expected. Risk index was smaller than 1 in the case of spraying vehicle mounted application, 1hour exposure time and wearing protective cloth exposure. Whereas, risk index was above 1 in the case of hand-held application, 2hour exposure time and wearing common cloth. Comparing two kinds of application method, total risk index of the hand held application (1.67) was higher than vehicle mounted (0.27). Therefore, chlorpyrifos skin exposure was mainly affected by application equipment and applied form. The results of risk assessment on the human dermal toxicity of chlorpyrifos should be required to control in keeping safety rules, skin surface area available for contact, spraying time ,and spraying frequency.y.
This study was investigated to review the potentials of irritation and skin sensitization of active ingredients and end-use products of pesticides for pesticide registration, prediction of irritation and sensitization, and data requirements for other purposes. Pesticide irritation and sensitization referred to the Pesticide Manual(14th edition), while toxicity was evaluated based on the data submitted by the pesticide registrant. Totally 148 active ingredients and 149 end-use products were analyzed to compare the positive response, formulation type, and correlation between active ingredients and products. Among active ingredients, ratio of positive response to skin irritation, eye irritation, and skin sensitization were 18.8, 47.0 and 20.6% respectively. While, positive response to skin irritation, eye irritation, and skin sensitization of the products were 14.9, 38.9 and 23.6%, respectively. Emulsifiable concentrate showed the highest positive response among formulation types showing skin irritation 31.3%, eye irritation 81.3%, and skin sensitization 31.3%. On the other hand granule type showed the lowest response with skin 4.8, eye 14.3 and sensitization 14.8%. There was no correlation by active ingredient content on irritation and sensitization of products. However, both active ingredients and products showed same positive response were skin 73.1%, skin sensitization 66.2%, and eye irritation 44.7%. By the these results, correlation between technical grade of active ingredients and end-use products would be use for pesticide management at the screening stage, especially skin irritation and skin sensitization.
It is common to use many experiment animals to evaluate the toxicity of chemicals including pesticides. For protecting animal, the concepts of 3R (Reduction, Replacement, Refinement) were introduced and in vitro alternatives methods actively have been developed all over the world. Many experimental animals for toxicological tests have been used, so that it is important to establish the alternative methods. In this study, the alternative method using reconstituted human skin model (Keraskin$^{TM}$) was conducted for classification of skin irritation on pesticides. Sixteen formulations selected on the basis of the degree of irritation were treated by Keraskin$^{TM}$ test. The percent of cell viability was measured into the culture medium collected after treatment of the pesticides for 24-72 hrs. The skin irritations of formulations were evaluated by the cell viability. In this study, The 4 formulations with mild irritation in rabbits were evaluated as nonirritant, the 6 formulations with moderate and severe irritation were evaluated as irritant in human skin model test. We suggest that the alternative test using Keraskin$^{TM}$ model could be used as toxicity evaluation for primary irritation index (P.I.I.) score of greater than or equal to 2.1 of pesticides. The further studies should be required to apply for hazardous assessment of pesticides on alternative skin irritation methods because of the interindividual variability of the sensitivity of skin irritation on pesticides.

We evaluated the anti-aging potential and safety of black garlic extract for cosmeceutical ingredient. Black garlic was made by spontaneous fermentation for 40 days at $60{\sim}70^{\circ}C$, 85~95% RH without any additives. The 10% black garlic extract had sweet odor, antioxidant activities and inhibitory activities of skin againg enzymes such as tyrosinase and elastase. The skin safety was performed to evaluate of potential toxicity using the primary irritation test and skin sensitization test. The black garlic extract did not show any adverse reactions such as erythema and edema on intact skin sites at primary irritation test, but on abraded sites, some experimental animals showed very slight erythema. So, the black garlic extract was classified as a practically non-irritating material based on the score 0.23 of primary irritation index. The skin sensitization study was tested by the guinea pig maximization test (GPMT) and Freund's complete adjuvant (FCA) with intradermal injection of 10% black garlic extract. The skin sensitization test showed no skin sensitization. The allergic sensitization depends on tumor necrosis factor-$\alpha$ (TNF-$\alpha$) and interleukin-6 (IL-6). The concentration of IL-6 on challenged tissue of treated with black garlic extract was not significantly different with negative control group (saline treated group). Based on this study, the potential for black garlic as a cosmeceutical ingredient was proven.
