Objectives : Since ancient times, Koreans have applied medicinal spabaths for treatment of various diseases. The objective of this study was to investigate the effect of strontium, one of the common ingredients of such baths, experimentally on acute skin barrier damage. Materials and Methods : Male hairless mice, average weight 20g, were divided into six groups. Each group consisted of five mice. The first was the normal, non-treated group. The second was the control group with acute skin barrier damage intentionally induced by TS. The third was the Ba-Sr1 group bathed in 1mg/L strontium chloride before and after inducing acute skin barrier damage by TS. The fourth was the Ba-Sr7 group bathed in 7mg/L strontium chloride before and after inducing acute skin barrier damage by TS. The fifth was the Sr1 group bathed in 1mg/L of strontium chloride only after intentionally inducing acute skin barrier damage by TS. The sixth was the Sr7 group bathed in 7mg/L of strontium chloride only after intentionally inducing acute skin barrier damage by TS. External changes of skin, skin erythema level, transepidermal water loss level, and GOT and GPT level of each group were checked immediately before and after TS, 3 hrs, 5 hrs and 24 hrs after inducing acute skin barrier damage. Then, tissue samples were made and examined for damage to epithelial cells, stratum corneum, change of mucous polysaccharide in dermis and amount of mast cells. Statistical analysis was performed by one way-ANOVA, Scheffe and Duncan for a post hoc test and pairwise comparison for comparing for difference between each time. Statistical significance was achieved if the probability was less than 5% (p<0.05) Results : 1. From skin erythema and TEWL level indicating the function of the skin barrier, we can know that it is helpful to the skin barrier to bathe in a water solution including a low concentration of strontium. 2. In the control group with acute skin barrier damage induced by TS, skin barrier damage persisted until 3-5 hrs and recovered after 5-24 hrs. Differently from the control group, in the case of taking a bath in a water solution including strontium, skin barrier damage recovered after only 3-5 hrs. Therefore, the bath with a water solution including strontium can promote recovery of the skin barrier. 3. Bathing in water solution including a higher concentration of strontium was more beneficial to recovery of skin barrier damage. 4. There was no influence on serum GOT and GPT from bathing in a water solution including strontium. Conclusions : The strontium was effective for recovery and mitigation of acute skin barrier damage induced by tape stripping. I suggest that strontium (Sr) can be used as an external treatment medicine, addedinto bath water to treat acute skin barrier damage.
Background: Ginsenosides (GS) have potential value as cosmetic additives for prevention of skin photoaging. However, their protective mechanisms against skin barrier damage and their active monomeric constituents are unknown. Methods: GS monomer types and their relative proportions were identified. A UVB-irradiated BALB/c hairless mouse model was used to assess protective effects of GS components on skin epidermal thickness and transepidermal water loss (TEWL). Skin barrier function, reflected by filaggrin (FLG), involucrin (IVL), claudin-1 (Cldn-1), and aquaporin 3 (AQP3) levels and MAPK phosphorylation patterns, were analyzed in UVB-irradiated hairless mice or HaCaT cells. Results: Total GS monomeric content detected by UPLC was 85.45% and was largely attributed to 17 main monomers that included Re (16.73%), Rd (13.36%), and Rg1 (13.38%). In hairless mice, GS ameliorated UVB-induced epidermal barrier dysfunction manifesting as increased epidermal thickness, increased TEWL, and decreased stratum corneum water content without weight change. Furthermore, GS treatment of UVB-irradiated mice restored protein expression levels and epidermal tissue distributions of FLG, IVL, Cldn-1, and AQP3, with consistent mRNA and protein expression results obtained in UVB-irradiated HaCaT cells (except for unchanging Cldn-1 expression). Mechanistically, GS inhibited JNK, p38, and ERK phosphorylation in UVB-irradiated HaCaT cells, with a mixture of Rg2, Rg3, Rk3, F2, Rd, and Rb3 providing the same protective MAPK pathway inhibition-associated upregulation of IVL and AQP3 expression as provided by intact GS treatment. Conclusion: GS protection against UVB-irradiated skin barrier damage depends on activities of six ginsenoside monomeric constituents that inhibit the MAPK signaling pathway.
Objective : To study the effect of the Insamyangyoung-tang(ISYT) extract on the skin barrier function, the skin pH, skin humidity and transepidermal water loss(TEWL) were measured and histological changes were observed in DNCB(2,4-dinitrochloro-benzen)-induced contact dermatitis(CD) hairless mice. Methods : The male hairless mice were divided into three groups. Each group consisted of 15 mice. The normal group which had acetone- olive oil applied. The control group which had intentionally induced CD by DNCB and it was fed normal saline orally. The ISYT group which had intentionally induced CD by DNCB and it was fed ISYT extract orally for 7 days. The three groups were checked 24h, 48h and 72h later after inducing CD, and the skin pH, skin humidity and TEWL were observed. Tissue samples were taken, and damage to the epithelial cell was observed. Statistical analysis was performed by using one way-ANOVA: significance was set at p values less than 5% (p<0.05). Results : ISYTextract efficiently maintained the pH balance, it kept the skin humidity at a normal level, and it inhibited TEWL of the DNCB-induced CD hairless mouse. The damage to the epithelium was decreased and the regeneration power of the skin was increased in the ISYT group. Conclusion : Insamyangyoung-tang has a good effect on the skin barrier function of DNCB induced contact dermatitis hairless mice.
To investigate the alleviative effects of jujube water extract (JWE) on the inflammation and skin barrier damage, both the irradiation of UVB and the application of squalene monohydroperoxide (Sq-OOH) were applied to the back skin of experimental animals for 4 weeks. And at the same time experimental materials were applied topically. Six weeks female SKH-1 hairless mice were divided into five groups (five animals for each group) including normal (N; saline), control (C; UVB+Sq-OOH+saline), vehicle control (VC; UVB+Sq-OOH+vehicle), positive control (PC; UVB+Sq-OOH+0.01% retinoic acid) and experimental (E; UVB+Sq-OOH+JWE) groups. The skin erythema index in the E group was significantly low compared to the C group (p<0.05). Lipid (p<0.05) and water (p<0.01) capacities in the E group were significantly high compared to the C group. In comparison with the C group, E group showed a relatively well preserved lipid lamellae in the epidermis and a relatively much less infiltration of mast cells in the dermis or hypodermis. As for the both absolute and relative weights of the spleen, PC group were significantly higher than the other groups. These results suggest that JWE have a considerably inhibitory effect on the inflammation and the skin barrier damage induced by UVB irradiation and Sq-OOH application.
The purpose of this study is to establish experimental canine skin barrier disruption model in dog. The study was designed to investigate the predictive properties of acetone damage using as effect parameters transepidermal water loss (TEWL) and stratum corneum (SC) hydration. To compare the structures of SC intercellular lipids in normal and acetone damaged skin, TEM observations were performed. Six clinically normal, male Beagles without dermatological problems were chosen for this study. Acetone damage was performed at 48hrs after clipping. Efficacy measurements for TEWL and skin hydration were performed before ($t_1$) and after ($t_2$) damage in a temperature- and humidity-controlled room ($2{0\sim}22^{\circ}C$, 50-60%). TEWL and SC hydration values were decreased in the acetone damaged model compared with non damaged skin. In TEM observation of acetone damaged sample, the intercellular lipid lamellae exhibit abnormal and incomplete structure compared with those of normal skin. It seems that acetone damage would be one of canine skin epidermal barrier disruption model for the study of canine atopic dermatitis (AD) as well as dry skin in veterinary dermatology research.
Objective This study aimed to confirm the effect of Sabaek-san extract on the recovery of skin damage in atopic dermatitis-induced mice. Methods In this study, we used 4-week-old NC/Nga mice that were assigned to four groups: control (Ctrl), lipid barrier elimination (LBEG), dexamethasone (Dx) administration after lipid barrier elimination (DxAG), and Sabaek-san extract administration after lipid barrier elimination (SBAG). Ten rats were assigned to each treatment group. After drug administration for 3 days following lipid barrier elimination, ceramide kinase, caspase 14, sodium hydrogen antiporter (NHE), cathelicidin, claudin, and Toll-like receptor (TLR2) were observed to confirm restoration of skin moisturizer production, antimicrobial barriers, and tight junctions in the skin barrier. Results Ceramide kinase and caspase 14 positive reactions were significantly higher in the SBAG group than in the LBEG or DxAG groups. NHE and cathelicidin showed a higher positive reaction in the SBAG group than in the LBEG and DxAG groups. Claudins and TLR2 showed a higher positive reaction in the SBAG group than in the LBEG or DxAG groups. Conclusion It was confirmed that Sabaek-san extract may have the potential to restore damaged skin barrier in atopic dermatitis.
Objectives The purpose of this study is to confirm the effect of alleviating inflammation through creating skin fat barrier from Scutellaria baicalensis extract. Methods Four-week-old Balb/C mice were divided into four groups: control group (Ctrl), lipid barrier eliminated group (LBEG), dexamethasone (DM) treated group after lipid barrier elimination (DMTG), and Scutellaria baicalensis (SB) treated group after lipid barrier elimination (SBTG). Scutellaria baicalensis extract were administered for 5 days after removal of the fat barrier. Changes in skin condition, improvement of the fat barrier, and relief of inflammation were observed in each group. Results Compared to LBEG and DMTG, pathological skin damage and tissue changes were less in SBTG, and transepidermal water loss (TEWL) and pH were also significantly reduced. Filaggrin was also significantly increased in SBTG. KLK7, PAR-2, and TSLP in SBTG also showed significant reduction compared to the LBEG and DMTG. Conclusions Scutellaria baicalensis extract restores skin barrier and relieves inflammation through the creation of skin fat barrier. This means that the Scutellaria baicalensis extract can regulate Th2 differentiation through the creation of the epithelial fat barrier.
To investigate the alleviative effects of Peonia japonica water extract(PJWE) on inflammation and skin barrier damage, both the irradiation of UVB and the application of squalene monohydroperoxide (Sq-OOH) to the backs of hairless mice were performed for 4 weeks. And at the same time experimental materials were applied topically. The skin erythema indices for the positive control (PC, 0.01% retinoic acid) and experimental (E, PJWE) groups were lower than that of the control (C) group. Whereas both the lipid and water capacities for the PC and E groups were higher than those of the C group. Epidermis and dermis of the C group were remarkably thickened in comparison with the PC and E groups. Relatively much less number of inflammatory cells, including lymphocytes, neutrophils and macrophages were found in dermis of the PC and E groups compared with the C group. Lipid lamellae of the C group were broken severely showing an irregular arrangement and lipid content was much reduced. Whereas those of the PC and E groups were almost intact with a regular arrangement, which were similar to that of the N group. Taken the results all together, it was confirmed that PJWE could be effective natural herbal material for the alleviation of inflammation and skin barrier damage in hairless mice skin which were induced by UVB and Sq-OOH.
Objectives The purpose of this study is to confirm the regulate effect of T helper (Th) 2 differentiation that Baekho-tang extract may produce to improves skin lipid barrier damages. Methods Four-weeks-old NC/Nga mice were divided into four groups: control group (Ctrl, n=10), lipid barrier eliminated group (LBE, n=10), Dexamethasone treatment after lipid barrier elimination group (DxT, n=10), and Baekho-tang extract treatment group after lipid barrier elimination group (BHTT, n=10). Baeko-tang extract was administered for 3 days after removal of the skin fat barrier in BHTT group. Then, we identified changes in external symptoms of the skin, factors affecting skin barrier such as potential of hydrogen (pH), filaggrin (FLG), transepidermal water loss (TEWL) and Th2 differentiation factors like Interleukin (IL)-4, Kallikrein Related Peptidase 7 (KLK7) and protease activated receptor 2 (PAR-2) through our immunohistochemistry. Results After lipid barrier elimination, the reduction of morphological skin inflammations was less in BHTT group than in LBE group and DxT group. Also, pH and TEWL were significantly decreased with BHTT group. However, FLG was significantly increased in BHTT group compared to LBE, DxT, and Ctrl group. All kinds of Th2 differentiation factors (IL-4, KLK7 and PAR-2) were also decreased in BHTT compared to the LBE and DxT. Conclusions As a result of this study, BHT administration decreased pH, TEWL, and increased FLG, thus participating in recovering damaged skin barrier. Since Th2 differentiation factors were decreased as well, BHT's regulatory effect in sequential immune reactions may be a possible explanation of how it enhances recovery of the damaged lipid barrier.

Objectives : Atopic dermatitis has a close relationship with damage of skin barrier function. To investigate the effects of Bangpungtongsungsan(BT) extract to the skin damage on mice model after atopic dermatitis elicitation, this study was done through forcing injury to mice's skin. Methods : The BALB/c mice were distributed into three groups: control(CON) group, atopic dermatitis(AD)-elicited group, Bangpungtongsungsan(BT)-treated group. AD-elicited and BT-treated group were caused AD according to the method of Christophers E., Mrowietz and Minehiro. The BT extract was administered for 48 hours to BT-treated group. We observed changes of external dermal formation, eosinophils in vasculature, lipid formation in stratum corneum, distribution of ceramide, distribution of capillary, $I{\kappa}B$ kinase(IKK) and induce nitric oxide synthase(iNOS) mRNA expression. We used the statistical methods of student t-test(p<0.05). Results : After dispensing BT extract into the AD-elicited group, the number of eosinophil as an atopic index in mice noticeably decreased and dermal injury decreased. Also the decrease of hyperplasia, degranulated mast cells, angiogenesis and substance P were shown. The lipid lamellae, lipid protect formation, were repaired and the distribution of ceramide which inhibit protein kinase C(PKC) activation increased, and the PKC caused inhibition of nuclear $factor(NF)-{\kappa}B$ activation. As a result of inhibition of $NF-{\kappa}B$ activation, iNOS production were inhibited and apoptotic cell were increased. Moreover the decrease of IKK and iNOS mRNA expression in BT-treated RAW 264.7 cell were noted. Conclusion : BT mitigated skin damage on mice model after atopic dermatitis elicitation through recovering skin barrier function and inhibiting nuclear $factor(NF)-{\kappa}B$ activation.
