Formation of Oxidative Cleavage Products from Lycopene (Lycopene으로부터 산화개열산물의 생성)

  • Lycopene was subjected to ozonolysis in ice-cold dichloromethane. The ozonolysis products were fractionated with a silica column and the carbonyl fraction was analyzed by ODS-HPLC with a photodiode array detector and by LC-MS. UV-vis spectra and $[M+H]^+$ of the carbonyl compound peaks showed clearly that acycloretinal, apo-14'-lycopenal, apo-12'-lycopenal, apo-10'-lycopenal, apo-8'-lycopenal and apo-6'-lycopenal were formed by ozonolysis of lycopene. Lycopene was solubilized in toluene and aqueous Tween 40, and then oxidized by incubating at $37^{\circ}C$ under atmospheric oxygen. Carbonyl compounds were produced. In comparison with autoxidation and ozonolysis, each compound showed the same retention time and UV-vis spectra are identical to the reference cleavage products prepared by ozonolysis of lycopene. Thus, eccentric cleavage of lycopene was confirmed to occur in vitro under oxidation condition.

  • In the course of screening for new bioactive compounds from oligotrophs in soil, a microorganism, designated as SA-246 and now identified as Micromonospora sp., has been shown to produce a strong antibacterial compound. The active compound was purified from broth filtrate by ethylacetate extraction, silica gel column chromatography, preparative TLC and HPLC, and was identified as crisamicin A based on mass and NMR spectral data. The compound SA- 246 exhibited not only strong antibacterial activity against Gram-positive bacteria but also cytotoxicity against cancer cell lines such as A549 (lung), SK-OV-3 (ovarian), SK-MEL-2 (melanoma), XF498 (central nervous system) and HCT15 (colon).

Antibacterial Activity and Identification of the Active Compound from Tumeric Extract (울금색소 추출물의 항균성과 물질분리 동정)

  • To identify the antimicrobial activity, of tumeric and its active compound tumeric was fractionated into four groups dichloromethane extract, hexane fraction, methanol soluble fraction and residue's extract. They were tested for antibacterial activity against E. coil and S. aureus and the methanol soluble fraction was found lo be the most active fraction. Compound I, II and III were isolated from TLC and silica gel column chromatography in the methanol soluble fraction. These compounds were analyzed by $^1H-NMR\;and\;^{13}C-NMR$ spectra and identified as curcumin I, II and III. They were also tested for antimicrobial activity against E. coli and S. aureus. Curcumin I was the must active curcuminoids due to the phenolic and methoxyl$(OCH_3)$ moiety in the same molecular structure.

  • Arils of Euporia longana L. was extracted with 80% aqueous MeOH and partitioned successively with EtOAc, n-BuOH and $H_2O$. From the n-BuOH fraction, furan compound was isolated through silica gel column chromatography. The results of physico-chemical data including NMR, MS and IR revealed the compound to be 5-(hydroxymethyl)-2-furfuraldehyde. This compound stimulated GDH I activity by $19.2{\pm}0.6$, $41.2{\pm}0.9$, $68.4{\pm}1.1$, $80.3{\pm}0.9$ and $85.9{\pm}1.6%$ at in vitro concentrations of 0.005, 0.008, 0.02 and 0.03 %, respectively.

Purification and Production Conditions of Antimicrobial Compound from Methylotrophic Actinomycetes MO-16 (Methanol 자화방선균 MO-16으로부터 항균성 물질의 정제 및 생산조건)

  • A methylotrophic actinomycetes strain MO-16, which produce the antimicrobial compound, was isolated from soil and supposed as Amycolatopsis sp. based on taxonomic studies. The cell-free extract of methanol-grown strain MO-16 showed dehydrogenase activity for methanol and formaldehyde when various electron acceptors were added for oxidation. On the other hand, methanol did not affect the production of antimicrobial compounds, and organic nitrogen sources such as corn steep liquor and peptone were better than inorganic nitrogen sources. These compounds showed broad antimicrobial spectrum to the tested strains such as bacteria and yeast. The antimicrobial comounds were very stable under heat(121$^{\circ}C$), acid(pH2.0), alkali(pH11.0) treatments. These compounds were isolated by ethylacetate extract, silica gel column chromatography and reverse phase HPLC. Two compounds(peak 1 and 2) were detected as antimicrobial compounds through the HPLC analysis. The peak 2 was purified as a single compound and revealed a 98% purity.

Antinocicepetive Effects of 3,4-Dicaffeoyl Quinic Acid of Ligularia fischeri var. spiciformis

  • The plant Ligularia fischeri var. spiciformis (Compositae) is a candidate for available functional foods. It has been used to treat diabetes mellitus and rheumatoid arthritis. We have reported the isolation of a new eremophilanolide named 6-oxoeremophilenolide and cytotoxic intermedeol together with the isolation of hydrophilic constituents, chlorogenic acid, 3,4-di-O-caffeoylquinic acie (3), and 5-O-[1-butyl]-3,4-di-O-caffeoylquinic acid. Compound 3 was again isolated by combination of silica gel- and ODS column chromatography for the anti-nociceptive action. Compound 3 and 4 were assayed in hot plate- and writhing tests in the rat. Although the three derivatives of caffeic acid exhibited significant anti-nociceptive effects at 10 mg/kg dose (i.p.),(activity potency: 4>3). These results suggest that compound 3 is responsible for at least rheumatoid arthritis, and caffeic acid moiety is the active moiety of dicaffeoylquinic acid.

PAF Antagonistic Activity of 2-hydroxy-3-methoxybenzoic Acid Glucose Ester from Gentiana scabra

  • In order to find out anti-platelet activating factor (PAF) from natural resources, Korean medicinal plants used for the treatments of peripheral circulation disorders were tested for their possible protective effects on PAF-induced anaphylactic shock. From the above screening, the methanol extract of Gentiana scabra showed a potent antagonistic activity against PAF. Water suspension of the extract was partitioned with $CH_2$$CI_2$ and EtOAc, successively. The EtOAc fraction which showed the highest activity was chromatographed on silica gel to yield 6 fractions. From the fraction which showed higher PAF-antagonistic activity than the other fractions, compound 1 was isolated by recrystallization. On the basis of spectral data, compound 1 was identified as 2-hydroxy-3-methoxybenzoic acid glucose ester. The compound prevented the mice from the PAF-induced death at a dose of 300 $\mu\textrm{g}$/mouse.

Isolation of 20(S)-Ginsenoside Rg3 and Rg5 from the Puffed Red Ginseng (팽화 홍삼으로부터 20(S)-Ginsenoside Rg3와 Rg5의 분리 및 구조동정)

  • Red ginseng tail roots (9.8 g water/100 g sample) were puffed at 7, 8, 9, and 10 $kg_{f}/cm^{2}$ using a rotational puffing gun. Puffed red ginseng was extracted with 70% ethanol, and the concentrated extract was successively partitioned with diethyl ether, n-butanol and $H_{2}O$. Two unknown ginsenosides from puffed red ginseng were found at 63 and 65 min of retention time in HPLC chromatogram suggesting that chemical structure of some ginsenosides might be altered during the puffing process. Identification of two unknown compounds was carried out using TLC, HPLC and NMR. Two major compounds were isolated from TLC. According to TLC result, compound I was expected to be the mixture of ginsenosides Rk1 and Rg5, and compound II was expected to be a 20(S)-ginsenoside $Rg_{3}$. Three compounds were isolated from n-butanol fraction through repeated silica gel and octadecyl silica gel column chromatographies. From the result of $^{1}H$- and $^{13}C$-NMR data, the chemical structures of unknown compounds were determined as ginsenoside $Rg_{5}$ and 20(S)-ginsenoside $Rg_{3}$. Unfortunately, ginsenoside $Rk_{1}$ could not be separated from ginsenoside-$Rg_{5}$ in the compound I. It was carefully reexamined using HPLC and confirmed that the last unknown compound was ginsenoside-$Rk_{1}$.

  • The gasket materials of for the lithium ion battery requires chemical resistance to electrolyte, electrical insulating, compression set, anti-contamination and low temperature property. To check the special characteristics of fillers which are applied to rubber for gasket, compound of EPDM, NBR and FKM (fluoro elastomer) were made by adjusting weights of carbon black and silica additive. Using these compounds, we had done tests of long-term stability against electrolyte, compression set and low-temperature property with considering operating condition of the lithium ion battery. From this test, we investigated the physical and chemical characteristics of rubber with using of carbon black and silica additive in each.

Isolation and Identification of Antimicrobial Compound from Mokdan Bark (Paeonia suffruticosa ANDR) (목단피로부터 식품부패 미생물에 대한 항균성 물질의 분리 및 동정)

  • Antimicrobial activity of Mokdan bark (Paeonia suffruticosa $A_{NDR}$) was investigated. Methanol extract of dried Mokdan was fractionated to hexane, chloroform, ethylacetate, butanol and aqueous fraction. Ethylacetate fraction among these fractions showed the highest inhibitory effect on the microorganisms such as L. monocytogenes, and E. coli at 500 $\mu\textrm{g}$/disc. Ethylacetate fraction was further fractionated into 3 fractions by silica gel column and thin layer chromatography (TLC). The results showed that ethylacetate fractions No. 1 and 2 had the highest antimicrobial activity. They were mixed again, reseparated, and 3 fractions were obtained. Among them, No. 1 had the highest inhibitory effect on the microorganisms, No. 1 fraction was identified as isobutyl isopentanoate by HPLC, and GC-MS.
