Optimum Concentration of Supply Nutrient Solution in Hydroponics of Sweet Pepper using Coir Substrates (코이어 배지를 이용한 착색단고추 수경재배 시 적정 급액농도)

  • This experiment was carried out investigation of optimum concentration of supply nutrient solution in hydroponics of sweet pepper using coir substrates (coconut dust fiber=70% : 30%, v/v). During the growing period, it was found out that the electric conductivity (EC) would increase in proportion to the supply nutrient concentration but it was in inverse proportion to the moisture content. The pH of drainage was stable, while EC was high showing EC $7.3\;dS{\cdot}m^{-1}$ in EC $4.0\;dS{\cdot}m^{-1}$ of supply nutrient concentration. Also, standard deviation and coefficient of variation were high. Plant length was no difference by the supply nutrient concentration. Photosynthesis rate was generally high in supply nutrient concentration EC$4.0\;dS{\cdot}m^{-1}$. Fruit weight was heavy in supply nutrient concentration EC $4.0\;dS{\cdot}m^{-1}$, fruit shape was close to a regular square in supply nutrient concentration EC $3.5dS{\cdot}m^{-1}$.

Breeding of King Oyster Mushroom, Pleurotus eryngii with a High Yield and Earliness of Harvest Trait and Its Sensory Test (큰느타리(새송이)버섯 다수확 속성 품종 육종 및 관능평가)

  • Two strains Pleurotus eryngii 'Aeryni' and 'Na' carrying superior traits of a pileus and a earliness of harvest were selected to improve previously bred strains by single crosses. New hybrid, Aeryni 3 (Aeryni10 ${\times}$ Na5) showed superiority to other hybrids in yield, fruit body shape and days for harvest. The new strain, Aeryni 3 was harvested earlier than Keunneutari No. 2 by 2~3 days, and yielded 110.5 g/bottle (850 mL) which was 108% of that of Keunneutari No. 2. The ratio of diameter of pileus and stipe was 1.8 indicating that new stain will be likely low damage rate of fruit body during a distribution, and that was better than 2.1 of Keunneutari No. 2. A sensory test of taste of the new strain showed that 84.7% of evaluation panels selected "very good" while that of Keunneutari No. 2 was 55.5%. In purchasing intent test, 86.9% of panels will buy the new stain whereas 46.8% will buy Keunneutari No. 2 implicating that the new strain will likely be more marketability than previously bred strain.

Tree Growth, Productivity, and Management Efficiency of High-Density Apple Orchards according to Training Systems in Korea (한국 밀식사과원의 정지전정에 따른 수체생장과 생산성 및 경영효율 비교)

  • The present study conducted a comparison on tree growth and productivity of high-density apple orchards by several orchard management systems: making scaffolds by heading-back and thinning out pruning (T-1), maintaining scaffolds upward and bearing shoots downward (T-2), managing branches slightly upward without heading-back (T-3), keeping leaders downward and shoots pending (T-4), maintaining leaders high and branches horizontal with severe pinching (T-5), making leaders with lower branches vigorous and upper shoots pending (T-6), and controlling very high planting density with bending branches (T-7). In conclusion, the orchards of (T-5) and (T-6) management systems showed a superior performance in controlling tree growth, productivity, and quality of fruits. Also, superior management efficiency was obtained in the orchards of (T-5) and (T-6).

Characteristic of Progeny in Pepper Transformants (고추 유전자변형체 후대 생육특성 검정)

  • For the resistance test for Phytophthora blight of $T_1$ and $T_2$ transformants in pepper, Phytophthora blight fungus was inoculated to seedlings of the $T_1$ and $T_2$ transformants by concentration (density: zoospore $10^3/ml$). Occurrence rate of blight at 5days after inoculation was 4.0 % in T1-1 line and 10.0% in $T_1-2$ line, and its rate for 12 days after inoculation was 52.0% in $T_1-1$ line, 64.0% in $T_1-2$ line, respectively. Therefore, the lower occurrence rate to blight was enable to select resistant transformants in the some inoculation density (zoospore $10^3/ml$), meanwhile 'Kumtap' and 'Subicho' were 100% in highest occurrence rate to blight. For field test, in which blight was commonly occurred, of the Youngyang Pepper Experiment Station, the acquired transformant resisting to blight was similar to characteristics of domestic varieties, 'Subic ho' for fruit shape, but there are some differences in growth, days to flowering, fruit characteristics. Occurrence of blight in $T_2-1-6$, and $T_2-4-9$ lines was smaller approxmately 30% than commercial varieties, 'Kumtap', although occurrence of blight in field was showed higher difference among tested lines. In this study, we concluded that the transformants showing blight resistance selected from habitual field could be fixed at every generation, and the developed transformation system was also considered to develop transformants in pepper.

Characteristics of New Ever-bearing Strawberry 'Jangha' Bred for High Soluble Solids Contents (고당도 사계성 딸기 '장하' 육성)

  • 'Jangha' is a new strawberry (Fragaria ${\times}$ ananassa Duch.) cultivar, which was released by the Highland Agriculture Research Institute in 2014. The 'Jangha' cultivar originated in 2008 from a cross between 'Goha' and 'Elsinore' that showed excellent ever-bearing characteristics, including continuous flowering habit and high soluble-solids contents under long-day and high temperature conditions. This cultivar was initially named 'Saebong No. 6' after examining its characteristics and productivity in summer culture from 2010 to 2012. After two regional adaptability tests in 2013-2014, 'Jangha' was selected from Saebong No. 6 as an elite cultivar. The general characteristics of 'Jangha' include semi-spreading type, elliptical leaves, and moderately vigorous growth. The fruit are conical in shape, and red in color. 'Jangha' plants have 21.4 leaves, 55.8 fewer than 'Flamenco' plants. The soluble-solids content of 'Jangha' was 8.9%, which was 1.2% higher than that of 'Flamenco'. The average fruit weight of 'Jangha' was about 11.7g and the marketable yield was $19,013kg{\cdot}ha^{-1}$, 141% higher than 'Flamenco'. 'Jangha' is suitable for summer season cultivation as a high soluble solids contents cultivar, because it shows continuous flowering habit under long-day and high temperature conditions.

Mycelial growth and fruit body cultural characteristics of a new Grifola frondosa variety, 'Bakyeon' (잎새버섯 신품종 '백연' 균사배양 및 자실체 생육 특성)

  • This study was conducted to reduce the phenomenon of the biased cultivation of certain mushroom varieties and to develop a competitive variety of Grifola frondosa. We developed the first Korean white commercial mushroom strain, 'Bakyeon', by crossing monokaryons derived from brown strains. We have collected and tested the characteristics of mushrooms from domestic and international genetic resources since 2018. We bred the unique domestic variety, 'Bakyeon', which has the following characteristics. The optimal temperature for mycelial growth was 25~28℃ and the optimal temperature for fruit body growth was 16~18℃. The new variety was similar to the control variety (Daebak) in terms of the pileus, which formed a pine cone shape, and the number of days of cultivation. The yield was 94.1 g/bottle, which was 23% lower than the 108.5 g/bottle yield of the control variety. When incubating the parent and control varieties, the replacement line was clear. Moreover, polymerase chain reaction analysis of mycelial DNA resulted in different band patterns between the parent and control varieties, confirming the hybrid species.

Construction of a DNA Profile Database for Commercial Cucumber (Cucumis sativus L.) Cultivars Using Microsatellite Marker (Microsatellite 마커를 이용한 오이 유통품종 DNA Profile Data Base 구축)

  • Microsatellite is one of the most suitable marker for cultivar identification as it has great discrimination power for cultivars with narrow genetic variation. The polymorphism level between 358 microsatellite primer pairs and 11 commercial cucumber cultivars was investigated. Thirty-one primer pairs showed high polymorphism within cucumber cultivars with different fruit types. These markers were applied for the constructing DNA profile data base of 110 commercial cucumber cultivars through multiplex PCR and fluorescence based automatic detection system. A total of 139 polymorphic amplified fragments were obtained by using 31 microsatellite markers. The average of PIC value was 0.610 ranging from 0.253 to 0.873. One hundred and thirty nine microsatellite loci were used to calculate Jaccard's distance coefficients for UPGMA cluster analysis. A clustering group of varieties, based on the results of microsatellite analysis, were categorized into plant shape and fruit type. Almost the cultivars were discriminated by marker genotypes. This information may be useful to compare through genetic relationship analysis between existing variety and candidate varieties in distinctive tests and protection of plant breeders' intellectual property rights through variety identification.

Effect of Temperature and Nutrition Affecting Oospore Formation of Phytophthora capsici Causing Red Pepper Fruit Rot (고추 역병균(疫病菌) (Phytophthora capsici)의 난포자(卵胞子) 형성(形成)에 미치는 온도(溫度) 및 영양(營養)의 효과(效果))

  • Sexual reproductive structure of Phytophthora capsici in vitro was round shape with thick wall and $24.4\;{\mu}m$ of diameter ranging $20-32.5\;{\mu}m$. Oogonium was $26.7\;{\mu}m$ $(21-37.5\;{\mu}m$) and $6.5-{\times}5.4m$ $(6-8{\times}5-6{\mu}m)$ for antheridium as doughnut shaped. Since mycelial contact of the paired cultures initiated right after inoculation, mycelial expansion phase was followed. Oospore morphogensis could be divided into the four phases for reproducing adult oospores. The optimum temperature for oospore reproduction was $20-24^{\circ}C$, whereas a retard trend for oospore formation was at the temperature above $25^{\circ}C$. Korean squash agar medium showed a higher oospore formation than the existing V-8 agar medium. Red pepper fruit agar medium was next. No oospore was reproduced on the red pepper leaf medium. Diurnal light with $1800{\pm}300$ and $800{\pm}300$ Lux showed rather retardation for oospore formation than dark conditions.

  • For agriculture is very highly dependent on climate and weather condistions, global warming seems to have a great impact on it, including its productivity, cultivation condition, product quality, and optimum cultivation location. In this study, we adopted geographical information system (GIS) in order to investigate the changes of Korea's cultivation area which are caused by global warming, especially with the examples of such tropical and sub-tropical fruits as lemon, fig, kiwi, orange, pomegranate, and mandarin. In terms of GIS techniques, we utilized the interpolate function for temperature changes, surface analysis function for slope, and raster calculator. Currently, these fruits's cultivation areas are in Jeju island and southern part of Korea. But these areas will be expanded according as our GIS model assumes $3^{\circ}C$ and $4.5^{\circ}$ increases of average and lowest temperature by the global warming in Korea. Optimum cultivation areas of these six fruits have two patterns; one is expansion and the other is belt shape shift. From the results of the study, we call for an urgent need of Korea government's policy and farmers' reasonable responses about global warming, which will be able to give more opportunities and better foods to Korea society in general.
