The Effect of Laserpuncture and Aquapuncture with Methionine on the Recovery in Artificially Induced Hepatic Damaged Rats (肝損傷 랫트에 있어서 Laser針療法 및 Methionine 水針療法이 肝損傷 回復에 미치는 影響)

  • The present experiment was performed in order to know the therapeutic effect of laserpuncture and aquapuncture with methionine on recovery in artificially induced hepatic damaged rats by carbon tetrachloride. The animals were divided into control, laserpuncture and aquapuncture groups. The changes of serum enzyme activities(ALT and AST), serum total protein contents, protein fractions(ALB and GLB) and A/G ratio were examined before and after application of laserpuncture and aquapuncture. In change of serum ALT activity, recovery with treatment in laser and aquapuncture groups was more rapid than that of control. The significant decreased value was shown on 3rd(P < 0.05) and 7th day(P < 0.01) after treatment in laserpuncture group and on 7th day(P < 0.05) after treatment in aquapuncture group. In addition, significance was detected on 2nd day(P < 0.05) between laserpuncture and aquapuncture groups. In change of serum AST activity, recovery with treatment in laserpuncture and aquapuncture groups was more rapid than that of control. The significant lower values on 7th day(P<0.01) in aquapuncture groups. In addition, significant low value was detected on 7th day(P < 0.05) in aquapuncture group comparing with that of laserpuncture group. The change of serum TP contents showed similar pattern in control, laser and aquapuncture groups. The significances were detected on 7th day(P < 0.05) in both experimental groups. In addition, significant high value was detected on 2nd day(P < 0.05) in laserpuncture group comparing with that of aquapuncture group. The change of serum ALB content with treatment in laser and aquapuncture groups was more rapid than that of control. The significant high value was shown on 2nd(P < 0.01), 5th(P < 0.05) and on 7th day(P < 0.01) in laserpuncture group. In addition, significant high value was detected on 3rd day(P<0.01) only in aquapuncture group comparing with laserpuncture group. The change of serum GLB content showed similar pattern among groups. The significant low values were detected on 2nd day(P < 0.05) in aquapuncture group. In addition significant high value was detected on 5th day(P < 0.01) only in laserpuncture group comparing with that of aquapuncture group. In change of A/G ratio laser and aquapuncture groups showed higher values than that of control. The significant high values were detected on 1st day(P < 0.Of) and 2nd day(P < 0.05) in laserpuncture group, and on 1st day(P < 0.05) and 2nd day(P < 0.01) in aquapuncture group. In addition, significant high value was detected on 5th day(P < 0.05) in aquapuncture group comparing with that of laserpuncture. Considering above findings collectively, it was considered that both laserpuncture and aquapuncture were effective; aquapuncture was more effective than laserpuncture for recovery of hepatic damage.

Changes of Chemical Composition in Blood Serum during the Antler Growth Period in Spotted Deer (Cervus nippon)

  • The aim of this study was to provide basic haematological information to allow improved nutritional management for velvet production in spotted deer (Cervus nippon) by investigating biochemical changes in blood values during the antler growth period. Blood samples, obtained from the jugular vein of twenty-five deer, were taken every 10 days from casting (day 0) to harvesting (day 50) of velvet antler. Negligible changes were found in the concentrations of total protein, albumin, and creatinine during the antler growth period, but there were significant changes in the concentrations of urea (p<0.05) and uric acid (p<0.01). The concentration of triglyceride was significantly higher (p<0.05) during the antler growth period compared to casting time, while serum high-density lipoprotein concentrations were low and remained unchanged during the antler growth period. Serum glucose concentration increased (p<0.05) significantly and was slightly changeable during antler growth. The serum concentrations of Ca and P did not fluctuate during antler growth, while those of Na, K and Cl showed slight differences between the time of casting and the rest of the antler growth period. No significant changes in concentrations of AST, ALT, amylase, CK, GGT and LDH were detected during the antler growth period. However, the concentration of ALK-P increased during antler growth reaching its peak on day 50 after casting. We found a significant difference in the concentration of ALK-P between the time of casting and the rest of the antler growth period (p<0.01). Consequently, antler growth was associated with mild changes in measured serum biochemical values with the exception of ALK-P activity in spotted deer.

Hepatoprotective Effect of Coccinia indica Against $CCl_4$ Induced Hepatotoxicity

  • The hepatoprotective effect of the ethanolic extract of Coccinia indica fruits in rats treated with carbon tetrachloride. In hepatotoxic rats, liver damage was studied by assessing parameters such as aspartate aminotransferase (AST), alanine aminotransferase (AlT), alkaline phosphatase (AIP) and gamma glutamyl transpeptidase (GGT) in serum, and concentrations of total proteins, total lipids, phospholipids, triglycerides and cholesterol in both serum and liver. The effect of co-administration of ethanolic extract on the above parameters was further investigated. Histopathological study of the liver in experimental animals was also undertaken. Hepatic damage as evidenced by a rise in the levels of AST, AIT, AIP and GGT in serum, and also changes observed in other biochemical parameters In serum and liver showed a tendency to attain near normalcy in animals co-administered with the extract. The normal values for AST (IU/L), AIP (IU/I), protein (g/100 ml) and total lipids (mg/100 ml) in serum (i.e.,20.24, 70.04, 5.72 and 135.54 respectively) were found to alter towards values 32.61, 127.11, 3.83 and 265.91 in hepatotoxic rats. These parameters Attained near normal values (I.e.,22.82, 79.30, 5.22 and 151.24 for AST, AIP protein and total lipids respectively) in ethanolic extract co-administered rats. Profound steatosis, ballooning degeneration and nodule formation observed in the hepatic architecture of $CCl_4$ treated rats were found to acquire near-normalcy in drug co-administered rats, thus corroborating the biochemical observations. Thus the study substantiates the hepatoprotective potential of ethanolic extract of Coccinia indica fruits.

Determination of Inorganic Elements in Women Blood Serum using Instrumental Neutron Activation Analysis (중성자방사화분석법을 이용한 성인여성 혈청중의 무기 원소 분석)

  • In this study, instrumental neutron activation analysis was used to assess the concentration level of inorganic trace elements in Korean women blood serum. It was found out that high concentration of Na and Cl incurs analytical interference, but 12 elements of Br, Ca, Cl, Co, Cr, Cs, Fe, K, Na, Rb, Se, Zn can be determined under the condition of interference minimization. Serum samples collected from 63 women were analyzed and the concentration level and range of the elements were evaluated. NIST SRMs were analyzed simultaneously for analytical quality control. The average values of Na and Cl determined in serum samples are around 3000 mg/L, Ca is 100 mg/L and K is 200 mg/L. Besides, Br, Se and Zn have concentration level of 6.0, 0.1 and 1.0 mg/L, respectively. It was found that there is no significant difference between the present values and reported values.

Changes of Blood Chemical Values and Vit. E Concentration in Calves Born from Hanwoo Dams Administrated Se and Vit. E at Late Pregnancying Periods (임신말기에 Se과 Vit. E 투여 후 태어난 한우 송아지의 혈액성분과 Vit. E 농도의 변화)

  • The objective of this study were to investigate the effects of Se and Vit. E administration on blood chemical values and Vit. E concentrations of serum in calves born from Hanwoo dams injected Se and Vit. E at late pregnancying periods. The effects of Se and Vit. E administration of blood chemical values in calves born from Hanwoo dams injected Se and Vit. E at the last month of pregnancying periods were examined. In albumin concentration of blood, the Vit. E 2000IU group were slightly higher than other groups and the cholesterol concentrations were also higher in Vit. E 15001u group than other groups. But, another components examined of blood chemical values were not significantly different in all experimental groups. The blood chemical values in calves born from Hanwoo dams administrated Se and Vit. E at two months before parturition were examined. However, there were not significantly different in all experimental groups in concentrations of albumin, cholesterol, BUN, creatinine, glucose, inorganic phosphorous, calcium, total protein and triglycerides. The concentrations of serum Vit. E in calves born from Hanwoo dams administrated Se and Vit. E at 30 and 15 days before parturition were examined. The concentrations of Vit. E in serum were higher in treatment groups than in control group(P>0.05). The concentrations of serum Vit. E in calves born from Hanwoo dams administrated Se and Vit. E at 60 days before parturition were examined, the concentrations of Vit. E in serum were slightly higher in groups administrated than in control group(P>0.05), but there were not significantly difference in all experimental groups.

Evaluation of Variation Method to Improve the Sensitivity of Immunoradiometric Assay (면역방사계수측정법의 민감도 향상을 위한 변법의 평가)

  • Purpose The concentration of PSA (Prostate Specific Antigen) after radical prostatectomy in prostate cancer patients is a predictor of biochemical recurrence, and the AUA (American Urological Association) is defined as biochemical recurrence when the concentration of PSA is measured at 0.2 ng/mL or more, and when the concentration is measured at 0.2 ng/mL or more at the retest. This standard is also applied our hospital. In this laboratory, the PSA reagent using IRMA (Immunoradiometric Assay) is used, and the sensitivity at a very low value was not as good as the reagent used in the department of laboratory medicine. This study aims to increase the reliability of the results by improving the precision and sensitivity of very low values. Materials and Methods As a reagent for the study, PSA reagent using IRMA was used. As a method to improve the precision and sensitivity of very low values, a variation method on the serum volume(25 uL, 50 uL, 100 uL, 200 uL) was studied, and variation usefulness evaluation was conducted. The evaluation items were compared the results of precision, analytical sensitivity, recovery rate, dilution test, high-dose hook effect test, parallel test and very low concentration values(n = 20). Results The validation results were displayed in the order of 25 uL, 50 uL, 100 uL, 200 uL. As the serum volume increased, it was confirmed that CV (Coefficient of Variation)(%) improved. Analytical sensitivity(ng/mL) was 0.038, 0.041, 0.017, 0.015 and recovery rate(%) was 101±3, 101±3, 99±2, 97±4. very low concentration values(ng/mL) between each volume(n=20) were 0.135±0.068, 0.076±0.050, 0.048±0.034, 0.046±0.034. and high dose hook effect appeared as the serum volume increased. Conclusion Through the variation usefulness evaluation, it was confirmed that as the serum volume increased, the precision and sensitivity improved at very low concentration values. However, it is necessary to pay special attention to the occurrence of high-dose hook effect as the serum volume increases. In the case of tests that requires very low concentration values, it is thought that the reliability of the result will be increased if the variation method is properly used after the variation usefulness evaluation.

  • Heavy metals like Mercury and Cadmium cause various kinds of toxicities in the organs of Liver and Kidney. To observe the results of toxicity in the liver, kidney, and serum when the rats were injected subcutaneously with HgCl$_2$ and CdC1$_2$ and sacrificed after 24 hours and 72 hours from the last injection, we measured variation of lipidperoxide values in rat liver homogenate, variation of aspartate aminotransferase and alanine aminotransferase in rat serum. Variation of lipidperoxide values in rat kidney homogenate and variation of BUN in rat serum. It was found that Mercury and Cadmium administered subcutaneously to the skin in the air could cause the damages of liver and kidney.

Effects of Intravenous Administration of Taurocholate on Hepatic Catechol-O-Methyltransferase Activity in Common Bile Duct Ligated Rats

  • Possible mechanism of decreased catechol-O-methyltransferase (COMT) activity in cholestatic rat liver was studied. Hepatic and serum COMT activities were determined from the experimental rats with common bile duct ligation (CBDL). The Michaelis-Menten constants in this hepatic enzyme were also measured. The activities of cytosolic, mitochondrial and mircosomal COMT as well as their Vmax values were found to be decreased significantly in CBDL plus taurocholic acid (TCA) injected group than in the control group, such as CBDL alone groups. However, their Km values in the experimental groups did not vary. Serum COMT activity increased slightly in the CBDL plus TCA injected group than in the control group. The above results suggest that TCA represses biosynthesis of the COMT in the liver. The elevated activity of the serum COMT is believed to be caused by the increment of membrane permeability of hepatocytes upon TCA mediated liver cell necrosis.

Thiol Methyltransferase Activity in Cholestatic Rat Liver Induced by Commom Bile Duct Ligation

  • Changes of thiol methyltransferase (TMT) activity in cholestatic rat liver were studied. Hepatic subcellular and serum TMT activities were determined in cholestatic rat induced by common bile duct (CBD) ligation over a period 28 days. The mitochondrial and microsomal TMT activities in cholestatic rat liver were found to be significantly increased between the 1st and the 28th day after CBD ligation. The TMT activity in serum was significantly increased throughout the experiments. The Vmax values of the above hepatic TMT in cholestatic rat were significantly increased at the 7th day after CBD ligation. However, the Km values of the above hepatic enzymes did not vary in all the experimental groups. Therefore, the results indicate that the biosynthesis of TMT was increased in cholestatic rat liver. The elevated serum TMT activity is most likely caused by increased hepatocytes membrane permeability due to cholestasis mediated liver cell necrosis.

Observations on the Cholinesterase Activity in Normal Blood of Korean Male Cattle (한국모우(韓國牡牛)의 Cholinesterase 활성도(活性度)에 대(對)하여)

  • The serum and erythrocyte cholinesterase activity and their seasonal variations were determined with 8 males of healthy Korean cattles during one year period from September, 1966 to august, 1977. The results obtained were as follows : 1. The serum cholinestrase activity showed a range of 0.64mg/hr/ml to 8.26mg/hr/ml with a mean value of $3.69{\pm}0.25mg/hr/ml$. There was highly significant differences in the seasonal variations. 2. The erythrocyte cholinesterase activity ranged from 29.78mg/hr/ml to 48.88mg/hr/ml with a mean value of $38.37{\pm}1.11mg/hr/ml$. Seasonal variations was not found to be significant. 3. The results showed that the serum cholinesterase activity of Korean cattle had great variations among individual values and significant seasonal variations. On the other hand, cholinesterase activity of erythrocyte showed a little variations among individual values and no significant seasonal variations.
