Effect of Lacquer (Rhus verniciflua) Supplementation on Growth Performance, Nutrient Digestibility, Carcass Traits and Serum Profile of Broiler Chickens

  • This experiment was conducted to explore the efficacy of lacquer (Rhus verniciflua) supplementation on growth performance, nutrient digestibility, carcass traits and serum biochemical values in commercial broilers. Eight hundred and forty Hubbard broiler chicks (1d old) were randomly distributed and allotted to four dietary treatments for five weeks. Each treatment had 5 replicates with 42 birds each. The dietary treatments were (1) control (2) 1% lacquer, (3) 2% lacquer and 4) 4% lacquer supplied as meal in the diet. Supplementation with lacquer improved weight gain (p = 0.0960) showing a linear trend during the starter phase (0-3 wk), but weight gain, feed intake and feed conversion ratio remained unaffected at the finisher phase (4-5 wk) and overall (0-5 wk). The nutrient digestibility studies conducted after 18 and 35 days of experimental feeding showed a linear (p<0.05) increase in digestibility of CP and ether extract at both measurement timess. The carcass studies were conducted on two birds per replicate (10 per treatment) at the end of both 3 and 5 wk. The dressing percentage, liver weight, heart weight, abdominal fat and the breast meat expressed as a percentage of live weight, did not differ significantly due to treatments at both phases. The serum cholesterol and high-density lipoprotein (HDL) showed a linear decrease (p = 0.0683 and p = 0.0322, respectively) as the level of supplementation increased at 3 wk; at 5 wk serum cholesterol, HDL and triglyceride levels decreased significantly showing a positive linear effect of lacquer on fat metabolism. The meat color values such as lightness, redness and yellowness did not reveal any significant trend. Overall, it could be concluded that lacquer supplementation at higher levels did not affect growth performance, but had a positive impact on fat metabolism by influencing fat digestibility and reducing serum cholesterol and triglyceride.

Therapeutic Effect of the Impatiens balsamina Linne Extract on the Membranous Nephropathy (봉선화(鳳仙花) 추출물의 막성신증(膜性腎症)에 대한 치료효과(治療效果))

  • Objective : Membranous nephropathy(MN) is one of the most common causes of nephrotic syndrome in adults. MN has been defined as granular subepithelial deposition of IgG immune complexes along the glomerular basement membrane(GBM). However, there is not a satisfactory treatment for MN. We aimed to identify the effect of Impatiens balsamina Linne(IBL) treatment on cationic bovine serum albumin(cBSA)-induced MN in a mouse model. Methods : Mice were divided into 4 groups. The normal group was injected with saline. The Control group was treated with cBSA(50 mg/kg i.p) only. The third group IBL-100, was treated with cBSA(50 mg/kg, i.p) and IBL(100 mg/kg, p.o). The fourth group IBL-400, was treated with cBSA(50 mg/kg, i.p) and IBL (400 mg/kg, p.o). After cBSA and IBL treatment for 6 weeks, we measured change of body weight, proteinuria, serum albumin, total cholesterol, triglyceride, BUN, creatinine, IgA, IgM, IgG, TNF-${\alpha}$, IL-6 and IL-$1{\beta}$ levels. The morphologic changes of renal glomeruli were also observed with a light microscope. Results : The level of proteinuria significantly decreased and serum albumin increased in groups treated with cBSA and IBL extract compared with the control. The levels of serum triglyceride, BUN, IgG, TNF-${\alpha}$, IL-$1{\beta}$ significantly decreased in both IBL groups. In histological findings of kidney tissue, thickening of GBM decreased in both IBL groups. Conclusions : This study shows that IBL might be effective for treatment of acute stage MN. More clinical data and studies are to be done for efficient application.

Effect of Dietary Vitamin E on Lipid Metabolism in Caffeine-Fed Rats (카페인 첨가식이로 사육한 흰쥐의 지질대사에 미치는 식이성 비타민 E의 영향)

  • This study was designed to investigate the effect of dietary vitamin E levels on lipid metabolism in caffeine-fed rats. Male Sprague-Dawley rats were fed on diet containing one of three levels of vitamin E(37.5, 750, or 1,500mg/kg diet0 which was with or without 0.3% caffeine. The rats were sacrificed after 5 and 10 weeks of the feed periods. Results obtained from this study were as follows ; Net weight gain, feed intake and FER in the caffeine added groups were significantly decreased as compared to those of only vitamin E fed group. Liver and kidney weights tended to increase in the caffeine added groups, but spleen and heart weights were not affected. Total lipid contents in serum and liver tended to decrease as dietary vitamin E became increasing and caffeine diet adding. Serum total cholesterol content tended to increase in the caffeine added group. But, liver cholesterol content were decreased. And the higher the dietary vitamin E level became, the lower serum and liver cholesterol contents were. Serum and liver triglyceride contents were significantly lower in the caffeine added groups than in the only vitamin E fed groups. The degree of decrease was more evident as dietary vitamin E level became more increasing. Serum aminotransferase activity was not affected in all experimental groups.

Hypoglycemic Effect of Tabebuia avellandae on Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨 흰쥐에서 Tabebuia avellandae의 항당뇨 효과)

  • Hypoglycemic effect of Tabebuia avellandae was investigated in the streptozotocin(STZ)-induced diabetic rats. Diabetes was induced in male Sprague-Dawley rats by injections of STZ (45 mg/kg, i.v.). Rats weighing 200-250 g were divided into 6 groups: normal, STZ-control, hexane fr., CHCl$_3$fr., BuOH fr. and $H_2O$ fr. group. Normal and STZ-control rats received 3% Tween 80 only. Four groups of diabetic rats were administered orally at doses of 100, 400, 300 and 400 mg/kg/day of hexane, CHC1$_3$, BuOH and $H_2O$ fr. respectively. Fractions were administered orally to the rats for 7 days after STZ injection. All rats were anesthetized with ether, blood samples were taken by cardiac puncture for clinical chemistry and the rats were killed by exsanguination. Liver, kidney, heart and spleen were removed, weighed and analyzed. We measured glucose, protein, cholesterol and triglyceride levels in the plasma and glycogen, cholesterol and triglyceride levels in liver. The extent of blood glucose decrement in rats administered $H_2O$ fraction was greater than that in the STZ-control rats. The serum cholesterol and triglyceride levels were significantly lowered by administration of $H_2O$ fraction compared with those of STZ-control group. Treatment of rats with Tabebuia avellandae fractions caused decreases in STZ-induced elevation of cholesterol and triglyceride. Liver triglyceride level was significantly lowered hexane and BuOH fraction group compared with STZ-control group. These results suggest that $H_2O$ fraction of Tabebuia avellandae has the hypoglycemic action against STZ-induced diabetic rats.

Effects of Cardiotonic Pills (CP) on Serum Lipid Level in Hyperlipidemic Rats (심적환이 고지혈증 흰쥐의 혈중 지질 변화에 미치는 영향)

  • Objectives : SimJeok-Hwan (CP, Cardiotonic Pills) is the traditional remedy to treat patients with coronary arteriosclerosis, angina pectoris and hyperlipidemia. This study was designed to investigate the effects of CP on changes in serum cholesterol levels in hyperlipidemic rats. Methods : In this study, we investigated the effects of CP on changes in body weights, food and water uptake, fasting blood glucose, total cholesterols, HDL-cholesterol, and triglyceride in hyperlipidemic rats. Results : In our results, body weights of hyperlipidemic rats induced by high fat diet did not changed, and treatment with CP did not affect body weights in hyperlipidemic rats. For experimental period, food and water uptake in CP administered group were the same as those in hyperlipidemic control group. In this experiment, treatment with CP decreased total cholesterol and triglyceride in serum which elevated by high fat diet respectively. Conclusions : These results suggest that CP is useful to treat patients with disease related to hyperlipidemia because CP can decrease cholesterol in serum

Effects of Dietary Fiber Sourecs on Lipid Metabolism and Kidney Function in Rats Fed High Cholesterol Diet (식이섬유 종류가 고콜레스테롤 식이를 급여한 흰쥐의 체내 지질대사 및 신장기능에 미치는 영향)

  • This study was conducted to investigate the effects of dietary fibers (cellulose ; polydextrose ; pectin ; ricebran) supplementation on the kidney fucntion of hyperlipidemic-induced rats on a high cholesterol diet (5% cholesterol). Serum lipid concentrations were not influenced by dietary fiber sources, but the pectin group was effective in reducing serum lipid levels. Total serum protein and albumin concentrations tended to higher in the polydextrose group, and serum creatinine and urea-N concentrations were higher in the pectin group than those in the other groups. Even though total urinary protein was high in the pectin group, GFR was reduced (18~30% decrease) as compared to other groups. The urea-N level was elevated in the polydextrose group as compared to that of cellulose group(25% increase). Total lipid, triglyceride and cholesterol concentrations in the liver of pectin group were lower than those of other groups. Total lipid and cholesterol concentrations in the kidneys were reduced in pectin and ricebran groups, respectively. The total cholesterol concentration infecal was significantly high in the polydextrose and pectin groups(p<0.05), and the triglyceride was highest in the pectin group. These results indicate no significant effects of dietary fiber supplementation on the kidney function of hyperlipidemic-induced rats on a high cholesterol diet, but pectin was very effective to improve lipid metabolism and to reduce GFR.

Anti-atherogenic Effects of Curcumin in Hypercholesterolemic Rabbits (고콜레스테롤혈증 토끼에서 Curcumin의 항동맥경화 효과)

  • Curcumin, the yellow pigment in turmeric, curry and mustard, has anti-inflammatory and anti-carcinogenic activities. In this study; we investigated the hypocholesterolemic and anti-atherogenic effect of curcumin in rabbits. Hypercholesterolemia was induced by feeding high cholesterol diet to male rabbits for 30 days, and the animals were then fed high cholesterol diet containing 0.1% (w/w) or 0.5% (w/w) curcumin for additional 30 days. Supplementation of 0.l% curcumin tended to lower serum total cholesterol and low density lipoprotein (LDL)-cholesterol levels and inhibit serum lipid peroxidation. In the 0.5% curcumin-supplemented group, serum total cholesterol was significantly lowered by 11.7%, LDL-cholesterol by 12.8% and lipid peroxidation by 47.9% compared to the control group. Hepatic cholesterol and triglyceride contents were also significantly lowered by 50.6% and 37.4%, respectively compared to the control group. Lipid staining of the arteries isolated from the curcumin-treated rabbits showed that curcumin significantly decreased formation of fatty streaks and atheromatous plaques on the intima of the arteries. These results demonstrated that curcumin lowered serum cholesterol concentration, hepatic cholesterol and triglyceride contents, and accumulation of cholesterol in the artery These cholesterol lowering effects of curcumin, together with its anti-oxidative and anti-inflammatory activities, may play some important roles in preventing atherosclerosis.

Dietary Soy Protein and Calcium Reduce Serum Lipid and Cholesterol in Rats Fed Eat-Enriched Diets

  • This study investigated the effects of dietary soy protein and Ca on the lipid profiles of rats fed fat-enriched diets. Rats were divided into two groups and fed either a casein-low Ca (Exp I) or ISP-high Ca diet (Exp II) for a control period of 4 weeks. The two groups of rats were again subdivided into 4 groups and fed one of 4 experimental diets for another 4 weeks. The experimental diets consisted of 18% beef tallow and 1 % cholesterol, in which either 20 % casein or ISP with one of two levels of Ca, high (1 %) or low (0.1 %). The concentrations of total lipid, cholesterol and triglyceride in serum, liver and feces were determined. At the end of the control period, the serum total lipid and cholesterol concentrations were low in the rats fed ISP-high Ca diet (67~76% and 83~86%). During the next 4 week period, these concentrations remained significantly lower in rats fed the diets containing ISP and high Ca compared with those on casein and low Ca diets (p < 0.05). Total lipid and cholesterol concentrations in feces were significantly higher in the ISP-high Ca dietary group at 4 weeks, and high in both high Ca groups at 8 weeks. This study demonstrates that both soy protein and Ca reduce serum and liver cholesterol, triglyceride, and total lipid in rats fed fat-enriched diet, and that they have an additive effect when combined.

Effects of Sayeoksanhap-Pyeongweisan-Gamibang(SPG) on Hepatological Changes in Animals Damaged by $CCl_4$ (사역산합평위산가미방(四逆散合平胃散加味方)의 경구투여와 약침의 병용이 백서의 간손상에 미치는 영향)

  • Objectives: This study was designed to investigate protective effects of Sayeoksanhap-Pyeongweisan-Gamibang(SPG) on hepatological changes in rats damaged by $CCl_4$. Methods : We investigated the effects of SPG on hepatological changes in terms of AST/ALT levels, Total cholesterol, HDL-cholesterol and triglyceride in serum and lipid peroxidation in rat liver. Results : Combination treatment with acupuncture and oral administration of SPG decreased AST and ALT in serum. Combination treatment also decreased lipid peroxidation in liver and triglyceride in serum. In addition, HDL-cholesterol in serum was increased significantly by combination treatment. Contrary, level of total cholesterol was not changed by combination treatment. Conclusion : These results suggest that Combination treatment with acupuncture and oral administration of SPG was useful to protect liver against $CCl_4$ by its antioxidant action in liver tissues.

Effect of Saponin from the Shoot of Aralia elata in Normal Rats and Steptozotocin Induced Diabetic Rats (두릅추출물이 정상쥐 및 당뇨쥐에 미치는 영향)

  • The purpose of this study was to investigate the effect of saponin in the shoot of Aralia elata on serum lipid level and glucose in streptozotocin(STZ) induced diabetic rats. Sprague Dawley normal male rats weighing 70$\pm$5g were randomly assigned to normal(control group), crude saponin(S group), and shoot of Aralia elata(D group). Experimental diets have been fed for 6 weeks. STZ induced diabetic rats were classified to diabetic control(DC group) and crude saponin(DS group). Diabetic rats were experimentally induced by intravenous injection of STZ(65mg/kg of body weight) dissolved in citrate buffer(pH 4.5). DS group has been i.p. injected with crude saponin solved in phosphate buffer(pH 7.0, 10mg/100g body weight) and DC group fed for 10 days. Body weight decreased significantly in crude saponin group. Feed intakes and feed efficiency ratio were not significantly different among C, S, and D group. The crude saponin group has indicated the lowest values of serum total cholesterol, glucose, and triglyceride. However, the values of serum glucose and triglyceride were not significant. Insulin levels among the crude saponin group, the shoot powder group, and the control group were not significantly different. When STZ induced diabetic rats have i.p. injection of crude saponin, the crude saponin has reduced the serum glucose but it is not been significant.
