Excessive endogenous endotoxin, especially lipopolysaccharide (LPS) reflux from gastrointestinal (GI) tract to the liver tissue is one of the most serious reasons of severe and acute liver injury which is mainly mediated by Kupffer cell activations. However, there is no clear molecular clues to explain the exact pathophysiological mechanism and effective drugs available till nowadays. We aimed to comprehend the pathophysiological features of LPS-induced liver injury and evaluate the efficacies of potential therapeutic drug, Ga-mi-Yuk-Mi-Jihwang-Tang (GYM), which is composed of herbal plants. GYM remarkably caused to normalize hepatic inflammation and oxidations against LPS-induced liver injury by evidence of serum liver enzymes, histopathological analysis, both hepatic protein and gene expression levels of pro-inflammatory cytokines, nitric oxide levels, and hepatic tissue levels of reactive oxygen species (ROS) levels, malondialdehyde (MDA), and 4-hydroxyneoneal, respectively. To assess molecular events in the hepatic tissue, we further found hepatic Sirtuin6 (Sirt6) levels were considerably depleted by LPS injection with aberrant alterations of Nrf2/HO-1 signaling pathways, whereas administration with GYM notably exerted to normalize these abnormalities. Our results exhibited that GYM would be one of target drug to diminish hepatic inflammation as well as oxidative stress by regulation of hepatic Sirt6 levels.
Aim This study was done to investigate whether SPP has inhibitory effects on the activation of RAW 264.7 cells. Method In tumor necrosis factor-a (TNF-a)/ interleukin-1b (IL-1b) and IL-6, the mRNA expression of molecular indicators related to inflammatory changes of the Reumatoid Arthritis (RA) were examined using quantitative real-time PCR. Results The treatment of SPP significantly suppressed the expression of proinflammatory cytokines and chemokines such as TNF-a, IL-1b, IL-6 compared with the control. The expression of NOS-II was considerably reduced, which was accompanied by a reduction in the production of nitric oxide (NO). It also reduced the expression of $TNF-{\alpha}$ in serum of Balb/c mice compared with control group. Conclusion SPP is an effective herbal material for suppressing the inflammation related cytokines of RAW 264.7 cells.
Objective : Yegangji-tang (YJT) is noted in China as effective medicine for hyperlipidemia. This experiment was performed to determine the effects of Yegangji-tang (銳降脂湯: YJT) on antioxidant activity and hyperlipidemia induced by hypercholesterolemic diet in mice. Methods : We observed three experimental groups: normal, control, and treatment. Rats were provided a normal diet in the normal group, and the other groups were provided a hypercholesterolemic diet to induce hyperlipidemia. After 2 weeks, YJT was treated to the treatment group. We observed the change of body and liver weight, change of serum total cholesterol, LDL cholesterol, HDL cholesterol and triglycerides, lipid peroxidation and antioxidant activity of liver tissue, cholesterol gene revelation control efficiency, and histologic change of liver and abdominal aorta. Results : Total cholesterol, LDL cholesterol and triglycerides decreased significantly with YJT, whereas HDL cholesterol increased significantly. Lipid peroxidation decreased and antioxidant activity of the liver increased significantly. Gene revelation of ACAT and HMG-CoA reductase in hepatic tissue decreased significantly with YJT. We also found that tissue defects of the liver and abdominal aorta were controlled by YJT on histologic study. Conclusion : These results suggest that YJT might be effective in treatment and prevention of hyperlipidemia.
the pupose of this study was to investigate the effects of gonadotropin and nitric oxide (NO) on the expression of mouse follicular bad and bax genes that are known induce apoptosis. Large and midium size follicles of immature mice were obtained at 0, 24, and 48 hours time intervals after Pregnant Mare's Serum gonadotropins(PMSG, 5 I.U.) injection. Preovulatory follicles collected at 24 hrs after PMSG injection were cultured with or without various chemicals such as gonadotropin, gonadotropin Releasing hormone(GnRH), testosterone, Sodium nitroprusside (SNP) for 24 hrs at $37^{\circ}C$. After 24 hrs culture, the culture media was used for nitrite assay and total RNA was extracted, subjected to RT-PCT for the analyses of bad and bax expression. We found that expression of bad and bax genes in follicles was markedly reduced before and after in vivo priming with hCG. When the preovulatory follicles were cultured for 24 hrs in culture media with PMSG and hCG, the expression of bad and bax genes was decreased. Moreover, SNP (NO generating agent) can significantly suppress the expression of bad and bax genes in follicles when apoptosis was induced by GnRH agonist and testosterone. At the same time, nitrite production of culture media was increased in GnRH agonist + SNP, testosterone + SNP and SNP treated groups than control group. These data demonstrated for the first time that peptide hormones and NO may play important roles in the regulation of mouse follicular differentiation and may prevent apoptosis via supressing the expression of bad and bax genes.
Botulism in wild waterfowl has been reported in many areas of the world. In 18 October 2008 to 22, 101 dead wild ducks were found in Anyangcheon. 24 of dead birds were spotbill (Anas poeilorhyncha), 56 were Mallard (Anas platyrhynchos), 18 were green- winged teal (Anas carolinensis) and 3 were others. Clinical sign of the sick birds were flaccid paralysis of the neck. At autopsy, most birds had no specific lesions. Stomach is empty. In the mouse bioassay, Samples of serum and intestinal contents taken from 9 dead birds killed all the injected mice, and their toxicity was neutralized by the antitoxin against C. botulinum type C toxin.

The aim of the present investigation was to evaluate the effect of A.nilotica extract against Dalton's ascitic lymphoma (DAL) induced solid and ascitic tumors in BALB/c mice. Experimental animals received A.nilotica extract (10 mg/kg.bw) intraperitoneally for 10 and 14 consecutive days before induction of solid and ascitic tumors, respectively. Treatment with A.nilotica extract significantly decreased the development of tumor and percentage increase in body weight when compared to DAL induced solid tumor control group, also increasing the life span, restoring the total white blood cell count and hemoglobin content and significantly decreasing the levels of serum aspartate transaminase (SGPT), alanine transaminase (SGOT), alkaline phosphatase (ALP), gamma glutamyl transferase (GGT) and nitric oxide (NO) when compared to DAL induced ascitic tumor controls. The treatment also reduced significantly the cellular glutathione (GSH) and nitric oxide levels in treated animals. Histopathological studies also confirmed protective influence. The outcome of the present work indicates that A.nilotica extract could be used as natural anticancer agent for human health.
The purpose of this research was to investigate the anti-inflammatory effects of Gagamtunong-san(GTNS) which has been medicated the patient such as mastitis. The results were as follows. The cytotoxicity on mouse lung fibroblast Cells(mLFC) was not served at all concentration of GTNS. GTNS in RAW264.7 cell inhibited $IL-1{\beta}$, IL-6, $TNF-{\alpha}$, COX-2 and NOS-II mRNA genes expression in a concentration-dependent manner. Specially GTNS inhibited NOS-II production very significantly at 100 ${\mu}g/ml$. GTNS inhibited NO production significantly in a concentration-dependent manner. GTNS inhibited ROS production in a concentration-dependent manner. GTNS inhibited $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ production significantly in serum of acute anti-inflammation-induced mice. GTNS increased the survival rate from the 3rd day on LPS-induced lethal endotoxemia. These results suggest that Gagamtunong-san(GTNS) can be useful in treating a lot of women mammary diseases caused by inflammation such as acute and chronic mastitis.
Background & Objectives : Allergic rhinitis is an inflammation or irritation of the mucous membranes that line the nose. Common symptoms include sneezing, a stuffy or runny nose, itchy eyes, nose and throat, and watery eyes. We aimed to determine therapeutic effects of Tribulus terrestris by observing changes of total IgE level and the scratching or rubbing around the nose behavior. Materials and Methods : Fifteen BALB/c mice were divided into three groups : AR group(allergic rhinitis elicitated), TT group(which ate herbal medicine) and AH group(which ate antihistamine). Allergic rhinitis was induced in the three groups by intraperitoneal and intranasal sensitization with 0.1% ovalbumin solution. Then Tribulus terrestris was orally administered only to the TT group of 28days, while the AH group was given antihistamine. We observed changes of total IgE level and the scratching or rubbing around the nose behavior. We used Kruskal-Wallis test statistically(p<0.05). Results : 1. Tribulus terrestris showed significantly decreased IgE level on the serum of the rat model. 2. Tribulus terrestris showed significantly decreased scratching or rubbing around the nose behaviors of the rat model. Conclusion : According to the above results, it is considered that Tribulus terrestris has an inhibitory effect on the process of allergic rhinitis and it can be used in relieving symptoms of allergic rhinitis.
BACKGROUND/OBJECTIVES: Grifola frondosa, commonly referred to as the maitake mushroom, has been studied extensively to explore its potential health benefits. However, its anti-inflammatory effects in skin disorders have not been sufficiently elucidated. This study aimed to elucidate the anti-inflammatory role of the ethanol extract of G. frondosa in atopic dermatitis (AD) using in vivo and in vitro models. MATERIALS/METHODS: We investigated its impact on skin and spleen inflammatory responses in Dermatophagoides farinae extract (DFE)/1-chloro-2,4 dinitrochlorobenzene (DNCB)-induced AD-like skin lesions in a mouse model. Additionally, we determined the immunosuppressive response and mechanism of G. frondosa by inducing atopic-like immune reactions in keratinocytes through tumor necrosis factor (TNF)-α/interferon (IFN)-γ stimulation. RESULTS: Our study revealed that G. frondosa ameliorates clinical symptoms in an AD-like mouse model. These effects contributed to the suppression of Th1, Th2, Th17, and Th22 immune responses in the skin and spleen, leading to protection against cutaneous inflammation. Furthermore, G. frondosa inhibited the production of antibodies immunoglobulin (Ig)E and IgG2a in the serum of AD mice. Importantly, the inhibitory effect of G. frondosa on inflammatory cytokines in TNF-α/IFN-γ-stimulated AD-like keratinocytes was associated with the suppression of MAPK (Mitogen Activated Protein Kinase) pathway activation. CONCLUSIONS: Collectively, these findings highlight the potential of G. frondosa as a novel therapeutic agent for AD treatment and prevention.
Objectives : Atractylodes chinensis(AC) and Phellodendron amurense Ruprecht(PAR) have been used as anti-inflammatory medicines. However, the effects of AC, PAR and Yimyosan on AP were not examined. To prove this, We decocted the dried prescription of AC and PAR with boiling distilled water and freeze-dried to be powdered. AC, PAR and Yimyosan was administrated intraperitoneally. Methods : 1h after administration, cerulein was injected hourly six times. 6hrs after last cerulein injection, mice were sacrificed, then the pancreas and blood were harvested. Serum amylase and lipase, neutrophil infiltration, pancreatic cytokines were used as the parameter of severity of AP. Results : As a result of assessment the parameters of AP, AC alone treatment did not inhibit the severity of AP, however PAR treatment inhibited the severity of AP significantly. Yimyosan also showed the protective effects against AP at lower doses, however AC alone plus PAR alone extract did not show the protective effects significantly. Conclusions : In conclusion, PAR extract has a protective effects on AP, and the effects could be increased by co-treatment with AC.
