Effects of Handayulso-tang for Taeum-in on the Body Weight and Lipid Metabolism of induced Obesity Rats (태음인(太陰人) 한다열소탕(寒多熱少湯)이 비만유도(肥滿誘導) 백서(白鼠)의 체중(體重) 및 지질대사(脂質代謝)에 미치는 영향(影響))

  • Objects. There was an increase in obese population due to eating habits resulted in modernization of lifestyle and the change of the tendency to conduct the physical activity, in order words, the reduction in the range of kinetic energy. Obesity is viewed as a major risk factor for serious health problems, including heart disease, high blood pressure, stroke, diabetes, and certain forms of cancer as well as for psychological and social problems owing to our fat-phobic society. In the present study, we investigate the weight loss thanks to giving Handayulso-tang(HD) medication of Taeum-in to induced obesity rats and its effects on lipid metabolism, given that their behavior patterns are changed according to their constitution. Methods: The extracts of Handayulso-tang was orally administered to the rats of HD group 200mg/200g of rats weight every day for 6 weeks, and the control group were fed with equal amount of saline. And measured the body weight and total cholesterol, triglyceride, free fatty acid and total lipid contents in serum in obese rats. Results: 1. The body weight in HD group were decreased. 2. The contents of serum total cholesterol and triglyceride in HD group were decreased and statistically showed the significant difference compared with the control group. 3. The contents of serum free fatty acid and total lipid in HD group were decreased, but did not statistically show the significant difference compared with the control group. Conclusion: Based on the above results, we know that Handayulso-tang have effects on the decrease of the body weight and the contents of serum total cholesterol, triglyceride, free fatty acid and total lipid So it could be concluded that Handayulso-tang can help the treatment of obesity.

Influence of the Feeding Mixed Perilla Oil and Red Pepper Oil on Fatty Acid Compositions of Serum and Platelet in Rats (들깨유와 고추 종자유의 혼합 급이가 흰쥐의 혈청 및 혈소판 지방산 조성에 미치는 영향)

  • This study aims at revealing the effects of the feeding mixed $\omega$-3 perilla oil and $\omega$-6 red pepper oil on fatty acid compositions of serum and platelet in rats. The diet administered to the male rats of Sprague - Dawley was composed of the peril1a oil containing about 60% $\omega$-3 linolenic acid as well as the red pepper oil with roughly 64% of $\omega$-6 linoleic acid. In the total fatty acid composition of serum lipid, SFA had no great significance among the groups, while MUFA ranged from 14.4% to 17.0% and PUFA stood from 48.0% to 53.0%. PUFA compositions of phospholipid, triglyceride and cholesteryl ester fractions, $C_{18:2}$ ($\omega$-6) was the highest in cholesteryl ester than the other components. In platelet's fatty arid composition, SFA were low in groups 2 (7.5% perilla oil+2.5% red pepper oil) and 3 (5.0% perilla oil +5.0% red pepper oil), and the highest in group 5 (10% red pepper oil). MUFA stood the highest in group 3 and the lowest in group 5, while PUFA was the lowest in group 2. Ratio of EPA / AA was the highest in group 2, but group 5 was the lowest.t.

Effects of Stearic, Oleic and Elaidic Acid on Cellular Lipids and Their Fatty Acid Composition in Hep-$G_2$ Cells (단일지방산 첨가에 의한 간세포의 지질조성과 지방산조성에 미치는 영향)

  • The effects of stearic(18 : 0, SA), oleic(18 : 1 cis, OA) and elaidic acid(18 : 1 trans, EA) on the cell growth, contents of cellular lipids, and the fatty acid composition of cellular and medium lipids in Hep-G$_2$cells were evaluated. The cells were incubated in serum-free medium containing 25, 50, 100 and 200$\mu$M of a fatty acid combined with albumin for 2 days. The fatty acid concentration up to 100$\mu$M showed the normal growth, but the cell growth decreased in the presence of 200$\mu$M fatty acid. The treatment of cells with 100$\mu$M of a fatty acid for two days significantly(p<0.05) increased the cellular triglyceride(TG) content in all fatty acid groups compared to control, but TG contents was not significantly different among all treatment group, but total cholesterol(TC) was the highest level in EA group. The level of free cholesterol(FC) and cholesteryl ester(CE) was similar to those of TC in all fatty acid treated group. The cellular phospholipid(PL) contents were similar between the control and all fatty acid groups. The treatment of cells with SA has no notable effects on the fatty acid composition of TG, CE and PL. The OA treatment caused significant increases in CE(51.2%) and PL(29.8%), but not in TG. The EA treatment resulted in 10.1, 10.7 and 7.8% of $C_{18:1\;trans}$ content in cellular TG, CE, and PL. The TG, CE and PL of medium were relatively similar between SA and OA groups. In EA treated group, TG, CE and PL of medium contained 17.0%, 0.7% and 5.6% of $C_{18:1\;trans}$, respectively.

  • This experiment was conducted to compare the fatty acid composition in the backfat and the levels of testosterone and androstenone in the serum of boars, barrows and gilts. The results obrained are summarized as follows : 1. The contents of palmitic acid and stearic acid in the backfat of barrows increased as their body weight increased and the levels of them were higher in barrows than in boars. However, the contents of oleic acid and linoleic acid were lower in barrows than in boars. 2. The levels of testosterone in boars at 70, 90 and 110 kg of body weight were 3.25, 5.66 and 4.5ng/ml respectively, while those of barrows or gilts were 0.01-0.43ng/ml. 3. The level of androstenone in boars at 110kg of body weight was higher(1.04ng/ml) than that at 70kg of body wight(0.44ng/ml), but there was little difference in the levels of it by body weight or age in barrows and gilts.

  • This study was conducted to identify the effects of caffeine or combinations of caffeine and iron or vitamin E on the lipid and protein components or blood chemistry levels of the serum as well as the total homogenate, mitochondrial and microsomal fraction of the rat(Sprague-Dawley, female) liver. Chronic test were conducted to determine those effects. The chronic test was conducted by dividing rats into 5 groups according to the type of drugs and dosages administrated as follows; the control(group A), and group B was given 25mg/kg caffeine orally once daily for 30 days, group C was given 50mg/kg caffeine orally once daily for 30 days, group D was given 25mg/kg caffeine and orally ferric chloride once daily for 30 days and group E was given 25mg/kg caffeine and 25mg/kg vitamin E once daily for 30 days. The concentrations of glucose, urea nitrogen, uric acid, creatinine, total protein, albumin, A/G ratio, triglyceride, total cholesterol, HDL-cholesterol, free fatty acid, phospholipid as well as the activities of alanine aminotransferase(ALT), aspartate aminotransferase(AST) and alkaline phosphatase(ALP) were measured in the serum of each experimental groups. The concentrations of the carbonyl group and malondiaidehyde(MDA) and the patterns of the SDS-PAGE(Sodium Dodecyl Sulfate - Polyacrylamide Gel Electrophoresis) and fatty acid compositions in free fatty acids and phospholipids were analyzed to determine the oxidative damages and metabolic changes on the lipid and protein components in the serum, and total homogenate, mitochondrial and microsomal fractions of the rat liver. The results obtained from this study were summarized as follows; 1. Body weights of groups B, C, D and E were significantly decreased(p < 0.01) in comparison with that of the control in the chronic test. 2. The concentrations of serum glucose in groups B(124.5mg/dl), C(130.1mg/dl), D(122.1mg/dl), E(119.3mg/dl) were significantly higher(p < 0.01) in comparison to that of the control(101.5mg/dl). But, there were no significant differences in the concentrations of urea nitrogen, uric acid, creatinine, total protein, albumin and A/G ratio in comparison to that of the control. 3. The concentrations of total cholesterol and HDL-cholesterol in serum of groups B(69.6, 53.4mg/dl), C(73.0, 56.3mg/dl), D(68.9, 51.1mg/dl) and E(68.2, 51.3mg/dl) were significantly higher(p < 0.01) in comparison to that of the control(52.6, 38.8mg/dl). On the other hand, the concentrations of triglyceride in serum of groups B(45.0mg/dl), C(40.4mg/dl), D(33.8mg/dl) and E(47.2mg/dl) were significantly lower(p < 0.01) in comparison to that of the control(66.2mg/dl). There were no significant differences in the activities of ALT, AST and ALP in comparison to that of the control. 4. The concentrations of free fatty acid and phospholipid in serum of groups B(45.7, 154.4mg/dl), C(50.0, 167.2mg/dl), D(52.5, 148.4mg/dl) and E(41.1, 159.2mg/dl) were higher(p < 0.01) in comparison to that of the control(35.2, 125.3mg/dl). And the concentrations of the carbonyl group and malondialdehyde in serum of group D(1.82, 0.52nM/mg protein) were significantly higher(p < 0.01) in comparison to the control(1.53nM/mg protein). 5. The concentrations of carbonyl group in total homogenate, mitochondrial and microsomal fraction of group D(1.45, 0.94, 1.67nM/mg protein) were significantly higher (p < 0.01) in comparison to the control(1.16, 0.66, 1.27nM/mg protein). And the concentrations of malondialdehyde in the total homogenate, mitochondrial and microsomal fraction of group D(6.70, 6.10, 1.36nM/mg protein) were significantly higher(p < 0.01) in comparison to the control(5.17, 3.64, 0.68nM/mg protein). 6. As the analytical results of the fatty acid compositions of free fatty acid in serum, the proportions of stearic acid and arachidonic acid of groups B(16.52, 12.62%), C(17.52, 15.18%), D(19.73, 13.47%) and E(17.62, 13.28%) were significantly higher(p < 0.01) in comparison to the control(14.75, 7.88%), but the proportions of oleic acid and linoleic acid of groups B(12.97, 32.59%), C(10.88, 31.23%), D(12.37, 30.66%) and E(11.95, 32.41%) were significantly lower(p < 0.01) in comparison to the control(16.44, 35.12%). Otherwise, as the results of the fatty acid compositions of phospholipid in serum, the proportions of stearic acid and arachidonic acid of groups B(39.37, 16.39%), C(40.63, 17.83%), D(42.73, 15.39%) and E(39.16, 15.70%) were significantly higher(p < 0.01) in comparison to the control(37.74, 14.24%), but the proportions of oleic acid and linoleic acid of groups B(4.03, 14.38%), C(3.54, 12.38%), D(4.52, 11.68%) and E(4.29, 13.64%) were significantly lower(p < 0.01) in comparison to the control(5.53, 16.14%). 7. As the analytical results of the fatty acid compositions of free fatty acid in total homogenate, mitochondrial and microsomal fraction of liver, the proportions of oleic acid of groups B(7.8**, 8.73**, 6.88%) and C(6.89**, 7.75**, 6.58%) were lower(**:p < 0.01) in comparison to the control(8.67, 10.08, 7.81%), but the proportions of arachidonic acid of group C(22.62, 19.79, 23.71%) were significantly higher(p < 0.01) in comparison to the control(20.93, 18.47, 22.24%). And the proportions of palmitic acid of group D(25.95**, 26.16, 26.34**%) were significantly higher(**:p < 0.01) in comparison to the control(24.43, 25.42, 23.34%). In addition, the proportions of linoleic acid of group D(23.43, 25.02, 23.95%) were also significantly higher(p < 0.01) in comparison to the control(22.17, 23.75, 21.26%). The proportions of stearic acid of group D(19.87, 19.76**%) in mitochondrial and microsomal fraction were lower(**:p < 0.01) in comparison to the control(21.01, 24.18%), and the proportions of stearic acid of group E(16.71*, 19.65**%) in mitochondrial and microsomal fraction were significantly lower(**:p < 0.01, *:p < 0.05) in comparison to the control(21.01, 24.18%), and the proportions of linoleic acid of group E(25.04, 29.20, 26.48%) in total homogenate, mitochondria and microsome were significantly higher(p < 0.01) in comparison to the control(22.17, 23.75, 21.26%). 8. As the results of the fatty acid compositions of phospholipid in total homogenate, mitochondrial and microsomal fraction of liver, the proportions of palmitic acid of group D(17.58**, 18.78*, 18.23%**) were significantly higher(**:p < 0.01, *:p < 0.05) in comparison to the control(16.28, 17.22, 16.38%), and the proportions of stearic acid of group D(36.41, 37.23, 39.53%) were also significantly higher(p < 0.01) in comparison to the control(34.18, 34.16, 36.04%). But the proportions of oleic acid(3.41*, 3.11**, 3.12**%) and linoleic acid (18.03**, 15.79**, 14.74**%) of group D were significantly lower(**:p < 0.01, *:p < 0.05) in comparison to the control(oleic : 3.63, 3.72, 3.79%, linoleic : 20.03, 18.71, 18.48%). 9. In order to determine the oxidative damages to the protein in serum, mitochondrial and microsomal fraction of the rat liver, the patterns of the SDS-PAGE were identified, but the results of SDS-PAGE were not significantly different between the control and experimental groups.

Excessive Dietary Conjugated Linoleic Acid Affects Hepatic Lipid Content and Muscular Fatty Acid Composition in Young Chicks

  • The effects of dietary conjugated linoleic acid (CLA) on lipid concentrations and fatty acid composition of various tissues were studied in young chicks. From 7 days of age, a total of 160 chicks were divided into 4 groups, placed into 4 pens per group (10 birds per pen) and fed one of four experimental diets containing 6% tallow (TO 6%), 4% tallow plus 2% CLA (TO 4%-CLA 2%), 2% tallow plus 4% CLA (TO 2%-CLA 4%) or 6% CLA (CLA 6%) for 3 weeks. There were no significant differences in growth performances and the relative weights of various organs, but relative liver weight of chicks fed dietary CLA at 4 and 6% levels was significantly higher (p<0.05) than that of TO 6% group. The chemical compositions of leg muscle were not affected by CLA feeding. However, hepatic total lipid of chicks fed 6% CLA diet was significantly higher (p<0.05) than those of TO 6% and TO 4%-CLA 2% groups. The concentrations of various lipid fractions in serum were not affected by CLA feeding. With the increase in dietary CLA levels, cis 9-trans 11 CLA, trans 10-cis 12 CLA and total CLA of leg muscle increased linearly. The relative proportions of C18:1 $\omega$ -9 and C20:4 $\omega$-6 fatty acids in the leg muscles of chicks fed the CLA containing diets were significantly lower (p<0.05) than those of TO 6% group. These results indicate that the levels of CLA isomers were increased linearly in dose-dependent manner after feeding of synthetic CLA source. But it was also observed that excessive amount of dietary CLA resulted in the possible adversely effects, such as increase of liver weight, hepatic lipid accumulation and serum GOT level.

Lipid Contents of Serum and Fatty Acid Composition of Serum Cholesteryl Esters in Mothers and Newborns (산모와 신생아의 혈청 지질 분획과 콜레스테릴 에스텔의 지방산 조성)

  • Lipid consumption in early life has been implicated with arteriosclerosis in later life. It has been also claimed that lipid contents of serum in early life would reflect coronary atherosclerosis in later life. Present study has been undertaken to look for serum lipid contents of mother and newborn simultaneously to determine the difference of various lipid classes. Lipids were extracted by Folch method. Total lipid was determined gravimetrically; cholesterol by Zak's method, phospholipids by Fiske-Subba-Row's method, and triglycerides by Sugiura's method. Fatty acids were analysed by gas liguid chromatography. Values of the total lipids of mother was significantly higher than those of newborn. HDI-cholesterol/total cholesterol ratio of newborn's was higher than mother's. The amount of saturated fatty acids in the serum cholesteryl ester of newborn was significantly higher than mother's.

Effect of Prunus persica Batsch var. davidiana Max. Extract on the Free Fatty Acid, Creatine Phosphokinase and LCAT Activities in Hypercholesterolemic Rats (고콜레스테롤혈증 흰쥐의 유리지방산, Creatine Phosphokinase 및 LCAT 활성에 돌복숭아(Prunus persica Batsch var. davidiana Max.)추출액이 미치는 영향)

  • This study was performed to observe the effects of the feeding Prunus persica Batsch var davidiana Max. extract on the improvement of the free fatty acid, creatine phosphokinase and lecithin cholesterol acyltransferase(LCAT) activities in the serum of dietary hypercholesterolemic rats(S. D. strain, male) fed the experimental diets for. 5 weeks. Concentration of electrolyte(Na, K, Cl) in serum were fairly reduced in the group BCP(basal diet + cholesterol + Prunus persica, $5.0\;g\%$ extract) than in the group BCW(basal diet + cholesterol + water). However, no significance was found in the effect of an electrolyte concentration among the groups. Concentrations of free fatty acid, lipid peroxide and creatine phosphokinase activity in serum were significantly higher in the cholesterol administration groups(group BCW, BCP) than those in the control group(group BW, basal diet + water). However, concentrations of free fatty acid, lipid peroxide and creatine phosphokinase activity in serum were remarkably lower in the group BCP than those in the group BCW The LCAT activity in serum was increased in the Prunus persica $5.0g\%$ extract administration group(group BCP) than in the cholesterol of diet group(group BCW). From these results, the Prunus persica Batsch var davidiana Max. extracts were found to be effective on the improvement of the lipid compositions in serum of dietary hypercholesterolemic rats.

Relationship Among Apolopoprotein E Phenotypes, Dietary Fat, Serum Lipoprotein Concentrations and Erythrocyte Membrane Fatty Acid Composition in Young Healthy Women (건강한 젊은 여성의 Apolipoprotein E 의 다형성 , 식이 지방 섭취, 혈청 지단백 농도와 적혈구 막의 지방산 조성과의 관계)

  • As fat consumption has increased in Korea , serum cholesterol concentrations have risen and the prevalence of atherosclerosis increased. The aim of the present study was to investigate the association among apolipoprotein E (Apo E) phenotypes, dietary fat consumption, plasma lipoprotein levels and fatty acid compositions of red blood cells in young healthy women. Seventy-one percent of participants had Apo E3/3, 14.9 percent had ApoE3/2 , 11.8% had Apo E4/3 and 1.2% had Apo E4.2. The subjects daily consumed approximately 1760kcal containing 29 energy % of fat. The ratio of polyunsaturated fat to saturated fat in a diet was 0. 73 . There were no significant differences of nutrient intakes according to Apo E phenotypes. Total cholesterol concentrations of subjects averaged approximately 160mg/dl , but 12 percent of them had over 200mg/dl, which is higher than the range recommended by the National Cholesterol Deucation Program. Notably, most subjects with 210mg/dl had Apo E4 isoforms. Subjects with Apo E4 isoforms had significantly higher total and LDL-cholesterol concentrations than those with Apo E3/2. Also subjects carrying Apo E4 isoforms tended to accumulated more fat in the body. Their BMI , WHR and arm fat area appeared to be how can arm fat area be greater no you mean grater, please check work vsage than subjects with Apo E3 isoforms, but not significantly. Fat intakes slightly influenced serum cholesterol levels. Myristic aicd intakes were positively correlated to serum total and LDL-choelsterol levels. Polyunsaturated fatty acid intakes were negatively correlated to serum total choelsterollevels. The results of 소냐 study suggest that , people with Apo E4 isoforms need to prevent the raising of serum total and LDL -cholesterol concentrations by reducing calorie and fat intakes and by maintaining a normal weight.

  • In order to determine the effect of Sopungchukdamtang experimental studies were performed in Hyperlipidemia Rats. The contents of serum Total cholesterol, Triglyceride, Free fatty acid, Phospholipid, HDL-cholesterol and LDL-cholesterol were measured The results were summarized as follows; 1. The content of Total cholesterol in the serum compared with control group tended to be decreased in Sopungchukdamtang(34mg/l00g) group, but did not show a significance. Sopungchukdamtang (17mg/l00g) group showed a significant value. 2. The content of Triglyceride in the serum compared with control group tended to be decreased in Sopungchukdamtang(17mg/l00g) group and Sopungchukdamtang(34mg/l00g) group, but did not show a significance. 3. The content of Free fatty acid in the serum was significantly decreased in Sopungchukdamtang(17mg/l00g) group and Sopungchukdamtang(34mg/l00g) group. 4. The content of Phospholipid in the serum compared with control group tended. to be decreased in Sopungchukdamtang(34mg/l00g) group, but did not show a significance. Sopungchukdamtang(17mg/l00g) group showed a significant value. 5. The content of HDL-cholesterol in the serum compared with control group tended to be increased in Sopungchukdamtang(17mg/l00g) group, but did not show a significance. Sopungchukdamtang(34mg/l00g) group showed a significant value. 6. The content of LDL-cholesterol in the serum compared with control group tended to be decreased in Sopungchukdamtang(34mg/l00g) group, but did not show a significance. Sopungchukdamtang(17mg/100g) group showed a significant value. According to the above results, it is assumed that Sopungchukdam-tang has a valid effect on Hyperlipidemia.
