EXPERIMENTAL STUDIES OF EUCOMMIAE CORTEX ACCORDING TO PROCESSING (두중(杜仲)의 수치(修治)에 따른 실험적(實驗的) 연구(硏究))

  • In order to estimate the effects of Eucommiae Cortex, which is one of the most important herb medicines used for invigorating the liver and kidney and strengthening ones and muscles, fetus-soothing etc., this experiment was conducted the quantitative analysis of geniposidic acid and geniposide by HPLC, and the analgesic effects of mice, the lipid metabolism of rats, the catecholamine concentration in the brain and the plsma of rats stressed by immobilization. The results obtained are as follows. 1. The contents of geniposidic acid and geniposide were 22.3mg/g, 5.4mg/g in Eucommiae Cortex without processing, and 119.8mg, 10.4mg/g in Eucommiae Cortex fried with salts. 2. In analgesic effects of mice, there were all significant in Eucommiae Cortex without processing and Eucommiae Cortex fried with salts. 3. In the effects of lipid metabolism of rats, there were not significant on the change of organ weight and the level of serum total lipid and total cholesterol, triglyceride, phospholipid, HDL-cholesterol. But, there were significant on the level of free fatty acid. 4. In tile effects of the catecholamine concentration in the brain and the plsma of rats stressed by immobilization, there were all significant in Eucommiae Cortex without processing and Eucommiae Cortex fried with salts, especially in case of norepinephrine. From the results mentioned above, there were significant on the analgesic effects of mice, and the catecholamine concentration in the brain and the plsma of rats stressed by immobilization. And there were different from Eucommiae Cortex without processing and Eucommiae Cortex fried with salts in the contents of geniposidic acid and geniposide. These results are relative to invigorating the liver and kidney and strengthening ones and muscles, and fetus-soothing, therefore when we use Eucommiae Cortex, it is more desirable to use Eucommiae Cortex with processing.

Anti-Lipogenic Effect of Functional Cereal Samples on High Sucrose Diet-Induced Non-Alcoholic Fatty Liver Disease in Mice (고당식이로 유도된 비알코올성 지방간 마우스에서 기능성 잡곡의 지질 대사 개선 효과)

  • The anti-lipogenic effect of cereal samples on high sucrose diet (HSD)-induced non-alcoholic fatty liver disease (NAFLD) in mice was studied. We divided C57BL/6 mice into various groups based on 8 weeks of treatment with three types of cereal samples (HSD+WR, HSD diet containing 40% white rice; HSD+MCG, HSD diet containing 40% mixed cereal grain; HSD+AO-MCG, HSD diet containing 40% mixed antiobesity-cereal grain). After the experimental period, body weight changes, liver weights, serum lipid profiles, and hepatic fatty acid metabolism-related gene expression levels were determined. We found that HSD+WR, HSD+MCG, and HSD+AO-MCG treatments reduced body weight and liver weight, especially HSD+MCG and HSD+AO-MCG effectively reduced levels of serum triglycerides, total cholesterol, and low-density lipoprotein cholesterol. However, high density lipoprotein cholesterol levels increased compared to the control group. Furthermore, expression of hepatic lipogenic genes such as sterol regulatory element-binding protein-1c, acetyl-coenzyme A carboxylase, fatty acid synthase, stearoyl-coenzyme A desaturase-1, cluster of differentiation, and $PPAR-{\gamma}$ (peroxisome proliferator activated receptor ${\gamma}$) decreased, whereas expression of ${\beta}-oxidation$ genes such as $PPAR-{\alpha}$ and carnitine palmitoyl transferase-1 increased following HSD+MCG and HSD+AO-MCG treatment compared with levels in HSD+WR and control groups. These results suggest that the functional cereal samples, especially HSD+AO-MCG treatment, improved hepatic steatosis triggered by an HSD-induced imbalance in hepatic lipid metabolism.

Dietary Intakes and Serum Lipids and Iron Indices in Obese Children (비만아동의 영양섭취와 혈청 지질농도 및 철분영양지표)

  • This study was performed to compare nutrient intakes, diet quality and serum indices (TG, Total cholesterol, LDL-cholesterol, HDL-cholesterol, RBC, Hb, Hct, Serum iron, ferritin, MCV) of obese and normal weight children. The subjects were 149 children living in the Seoul and Gyeonggi areas. Each subject was assigned to one of such as normal weight group ($15th{\le}BMI$ percentile < 85th, n = 82) or obese children ($85th{\le}BMI$ percentile, n = 67) according to their percentile of BMI by The Korean Pediatric Society. Data on dietary intakes, body composition and serum indices were obtained. Differences of all the above variables were assessed. Energy, carbohydrate, fat, cholesterol, total fatty acid, SFA, MUFA, PUFA, phosphate, potassium, zinc, vitamin $B_1$ and vitamin $B_6$ intakes of girls in the obese group were higher than those of the normal weight group (p<0.05). The intake of fiber was as insufficient as below 50% of KDRIs in both groups. Nutrient adequacy ratio of calcium, iron, phosphate, zinc and folate in obese boys were lower than those of normal weight boys (p<0.05). Energy intakes of grain and milk and dairy food in the obese girl group ($905.9{\pm}344.5kcal,\;210.9{\pm}166.4kcal$) were higher than those of normal weight girls ($671.2{\pm}360.7kcal,\;184.0{\pm}103.5kcal$) (p<0.05). HDL-cholesterol level of obese boys ($52.7{\pm}6.3mg/dL$) was significantly lower than that of normal weight boys ($65.3{\pm}15.6mg/dL$). Serum iron level of obese boys and girls (boys: $79.4{\pm}32.6mg/dL$, girls: $98.3{\pm}16.2mg/dL$) was significantly lower than those of normal weight children (boys: $104.8{\pm}38.6mg/dL$, girls: $106.7{\pm}28.9mg/dL$) (p<0.05). These results suggest that there should be sensible food selection and more intakes of fruit and vegetable are needed to improve the nutritional status in obese children.

  • Objective : This research was performed to investigate the effect of herbal acupuncture(Atratylodes japonica, Coix lachrymajobi, Ephedra sinica, Atratylodes japonica mixed with Coix lachrymajobi and Ephedra sinica mixed with Green tea) at Pungnyung(ST40) and Umnungchon(SP9) on weight gain, food intake, food efficiency, serum of lipid concentrations, liver function and HDL to total cholesterol ratio of rats fed high fat diet for 5weeks. Method : Experimental groups were divided into normal group(Normal), high fat diet group(Control), high fat diet and Atractylodes japonica-herbal acupuncture group(AJ), high fat diet and Coix lachrymajobi-herbal acupuncture group(CL), high fat diet and Ephedra sinica-herbaI acupuncture group(ES), high fat diet and Atractylodes japonica+Coix lachrymajobi-herbal acupuncture group(AJ+CL), Ephedra sinica+Green tea-herbal acupuncture group(ES+GT). Herbal acupuncture was bilaterally treated at the level of 132.5mg/kg body weight per 2day. Results : Body weight and food efficiency were decreased in AJ, ES, AJ+CL, ES+GT. The level of serum total cholesterol, triglyceride and free fatty acid were increased in AJ, ES, ES+GT. That of serum HDL-cholesterol was increased in AJ. The change of food intake, the level of serum phospholipid and ALP were not significant. The HDL to Total cholesterol ratio was increased in AJ and ES. Conclusion : Atractylodes japonica-herbal acupuncture in ST40 SP9 is effective on Body weight, food efficiency ratio, the level of serum lipid, protection of liver function and prevention cardiovascular risk by obesity induced by high fat diet. Herbal acupuncture mixed Ephedra sinica with Green tea can control the body weight, food efficiency ratio and the level of serum lipid.

Effects of lycopene on abdominal fat deposition, serum lipids levels and hepatic lipid metabolism-related enzymes in broiler chickens

  • Objective: The present study was conducted to investigate the effects of lycopene on growth performance, abdominal fat deposition, serum lipids levels, activities of hepatic lipid metabolism related enzymes and genes expression in broiler chickens. Methods: A total of 256 healthy one-day-old male Arbor Acres broiler chicks were randomly divided into four groups with eight replicates of eight birds each. Birds were fed basal diet supplemented with 0 (control), 100, 200, and 400 mg/kg lycopene, respectively. Results: Dietary 100 mg/kg lycopene increased the body weight at 21 day of age compared to the control group (p<0.05). Compared to the basal diet, broilers fed diet with 100 mg/kg lycopene had decreased abdominal fat weight, and broilers fed diet with 100 and 200 mg/kg lycopene had decreased abdominal fat percentage (p<0.05). Compared to control, diets with 100, 200, and 400 mg/kg lycopene reduced the levels of total triglyceride and total cholesterol in serum, and diets with 100 and 200 mg/kg lycopene reduced the level of serum low density lipoprotein cholesterol (p<0.05). The activity of fatty acid synthase (FAS) in 400 mg/kg lycopene treated broilers and the activity of acetyl-CoA carboxylase (ACC) in 100, 200, and 400 mg/kg lycopene treated broilers were lower than those fed basal diet (p<0.05). Lycopene increased the mRNA abundance of adenosine monophosphate activated protein kinase α (AMPK-α), whereas decreased the mRNA abundance of sterol regulatory element-binding protein 1, FAS, and ACC compared to the control group (p<0.05). Conclusion: Dietary lycopene supplementation can alleviate abdominal fat deposition and decrease serum lipids levels, possibly through activating the AMPK signaling pathway, thereby regulating lipid metabolism such as lipogenesis. Therefore, lycopene or lycopene-rich plant materials might be added to poultry feed to regulate lipid metabolism.

Growth Performance of Lambs Fed Diet Supplemented with Rice Bran Oil as Such or as Calcium Soap

  • Forty two Malpura lambs (21 d old) were divided into three groups of 14 each consisting of 8 females and 6 males. Lambs were allowed to suckle their respective dams twice daily up to weaning (13 wks) and offered free choice concentrate and roughage in a cafeteria system. The lambs in control group were fed conventional concentrate mixture, in RBO group concentrate mixture fortified with 4% industrial grade rice bran oil and in Ca-soap rice bran oil (as in RBO group) was supplemented in the form of calcium soap. The concentrate intake decreased($p{\leq}0.05$) in RBO group as a result total dry matter, crude protein and metabolizable energy intake decreased compared to control whereas Ca-soap prepared from the same rice bran oil stimulated the concentrate intake leading to higher total dry matter, crude protein and energy intakes. The digestibility of dry matter ($p{\leq}0.05$), organic matter ($p{\leq}0.05$) and crude protein ($p{\leq}0.05$) was higher in RBO group followed by Ca-soap and control whereas no effect was observed for ether extract digestibility. Higher cholesterol ($p{\leq}0.05$) content was recorded in serum of oil supplemented groups (RBO and Ca-soap) while no effect was recorded for other blood parameters. Rice bran oil as such adversely affected and reduced the body weight gain ($p{\leq}0.001$) of lambs in comparison to control whereas the Ca-soap of rice bran oil improved body weight gain and feed conversion efficiency in lambs. Fat supplementation decreased total volatile fatty acids ($p{\leq}0.05$) and individual volatile fatty acid concentration which increased at 4 h post feeding. Fat supplementation also reduced ($p{\leq}0.05$) total protozoa count. Ca-soap of rice bran oil improved pre slaughter weight ($p{\leq}0.05$) and hot carcass weight ($p{\leq}0.05$). It is concluded from the study that rice bran oil in the form of calcium soap at 40 g/kg of concentrate improved growth, feed conversion efficiency and carcass quality as compared to rice bran oil as such and control groups.

Study on the Effect of Concentrated Extracts of Pteridium sp. Tube Feeding on the Serum Lipid Peroxides and Hepatic Homogenate Lipid Peroxides value (Pteridium sp.의 농축추출액(濃縮抽出液)이 동물혈청(動物血淸) 및 간조직(肝組織)의 지질과산화(脂質過酸化)에 미치는 영향(影響)에 관(關)한 연구(硏究))

  • This experiment was carried out to investigate variation of plasma lipid level and liver peroxide level(Thiobarbituric acid, T.B.A) caused by tube feeding of Pteridium sp. extracts for seven weeks in rats. A seven week rat's plasma triacylglyceride (T.G.) level showed remarkably higher than that of the control group and that of the three week rats. On the other hand, no remarkable differences of plasma phospholipid, free fatty acid and thiobarbituric acid level have been recognized. Liver homogenate lipid peroxides value of seven week feeding rats showed remarkably higher level compared with that of the control group and the three weeks rats. So it could not be agreed that short time feeding of Pteridium sp. plant or even it's concentrated extracts influenced to the levels of liver homogenate lipid peroxides, plasma lipids and its peroxides.

Effects of Yukmijihwang-tang on Maximal Exercise Performance (육미지황탕 투여가 운동 지구력 향상에 미치는 영향)

  • Yukmijihwang-tang (YM) is a well-known prescription to treat kidney inflammation on Korea. However, the effects of YM on maximal exercise performance (MEP) is not-well investigated. In this study, we aimed to evaluate the effects of YM on MEP using forced swimming test (FST) model. Mice were largely divided into three group; a non-swimming group, saline treated-swimming load test group, and YM treated-swimming load group. Swimming load test groups were re-divided into swimming control group (Control), YM fed group (YM). YM was administrated orally for 2 weeks before FST. After FST, immobility time, oxygen consumption was measured by physiological test, and serum was collected for biochemical analysis. FST induced MEP identified by increased immobility, oxygen consumption, and biochemical factors. Immobility time was significantly reduced by YM treatment compared with control group. Oxygen consumption was also significantly reduced by YM treatment. The increase on lactic acid and lactate dehydrogenase after FST was inhibited by YM treatment. In addition, consumption of energy source (free fatty acid, and triglyceride) and recovery of energy was enhanced by YM treatment after FST. In conclusion, YM inhibited the elevation of immobility time, oxygen consumption, and biochemical factors after FST. YM could be used as a beneficial agents to enhance maximal endurance exercise performance and an effective ergogenic aid in exercise training.

A Novel Homozygous LIPA Mutation in a Korean Child with Lysosomal Acid Lipase Deficiency

  • Patients with lysosomal acid lipase (LAL) deficiency and glycogen storage disease (GSD) demonstrated hepatomegaly and dyslipidemia. In our case, a 6-year-old boy presented with hepatosplenomegaly. At 3 years of age, GSD had been diagnosed by liver biopsy at another hospital. He showed elevated serum liver enzymes and dyslipidemia. Liver biopsy revealed diffuse microvesicular fatty changes in hepatocytes, septal fibrosis and foamy macrophages. Ultrastructural examination demonstrated numerous lysosomes that contained lipid material and intracytoplasmic cholesterol clefts. A dried blood spot test revealed markedly decreased activity of LAL. LIPA gene sequencing identified the presence of a novel homozygous mutation (p.Thr177Ile). The patient's elevated liver enzymes and dyslipidemia improved with enzyme replacement therapy. This is the first report of a Korean child with LAL deficiency, and our findings suggest that this condition should be considered in the differential diagnosis of children with hepatosplenomegaly and dyslipidemia.

The Effects of Total Mixed Ration Feeding with High Roughage Content on Growth Performance, Carcass Characteristics, and Meat Quality of Hanwoo Steers

  • This study investigated the dietary effect of total mixed ration (TMR) based on high roughage content on the growth performance, carcass characteristics, and meat quality of Hanwoo steers. Twenty-four Hanwoo steers (average body weight, 195.3±4.7 kg; age, 8.5 mon) were randomly allocated to three experimental groups according to forage and concentrate ratio (DM basis): 25:75 (control), 50:50 (T50), and 70:30 (T70). Productivity in the fattening period and final body weight were significantly higher in the control. Average daily gain and feed conversion ratio were the same among treatments. Serum parameters, cholesterol, blood urea nitrogen, and total protein were higher in the control. Carcass weight was comparable in the control and T50 but feeding more roughage was significantly correlated with a higher intramuscular fat. Shear strength and drip loss were higher while n-6/n-3 was lower in T70 compared to the other groups. However, meat color was not significantly different among treatments. In terms of free amino acid contents, glutamic acid and glycine were higher in the control than T50 and T70. Overall, feeding Hanwoo steers with high forage content TMR had the lowest n-6/n-3 ratio of fatty acid content but highest intramuscular fat, shear strength, and drip loss. High forage content TMR is the best feed for Hanwoo steers that gives more benefits for human health and consumption but also provides the best meat grade and quality, which is important in the beef market in Korea.
